Graphene Quantum Dots-Band-Aids Used for Wound Disinfection

Hanjun Sun,Nan Gao,Kai Dong,Jinsong Ren,Xiaogang Qu

Herein, an antibacterial system combining the “safe” carbon nanomaterials, graphene quantum dots (GQDs), with a low level of H2O2 has been put forward. It has been found that the peroxidase-like activity of GQDs originates from their ability to catalyze the decomposition of H2O2, generating ·OH. Since the ·OH has a higher antibacterial activity, the conversion of H2O2 into ·OH improves the antibacterial performance of H2O2, which makes it possible to avoid the toxicity of H2O2 at high levels in wound disinfection. All the experiments in vitro display that this intrinsic activity exerts a high enhancement of antibacterial activity of H2O2, and the designed system possessed broad spectrum of antibacterial activity against both Gram-negative (Escherichia coli) and Gram-positive (Staphylococcus aureus) bacteria. More importantly, to assess the antibacterial efficacy of the designed system in actual wound disinfection, the GQD-Band-Aids are prepared and show excellent antibacterial property with the assistance of H2O2 at low dose in vivo.

Ag functionalized SnS2 with enhanced photothermal activity for safe and efficient wound disinfection

Jun Yang,Lin Sun,Shuhan Hui,Peng Zhang,Jian Li,Dong Wang,Xuelin Wang,Shan Jiang

Severe bacterial infections have brought an urgent threat to our daily life, and photothermal therapy (PTT) has acted as an effective method to kill bacteria. Herein we decorated Ag on the surface of SnS2 (Ag@SnS2), which has outstanding photothermal conversion capability and good biocompatibility. The decoration of Ag on SnS2 improved the absorption of near-infrared (NIR) light in comparison to SnS2, resulting in a temperature increase of 50 °C after 5 min of NIR light irradiation (1.9 W cm−2) and a photothermal conversion efficiency of 31.3%. Ag@SnS2 exhibits almost 100% growth inhibition of E. coli and S. aureus bacteria due to hyperthermia, with a concentration larger than 0.5 mg mL−1 and 5 min of NIR irradiation. Meanwhile, SEM images of treated bacterial cells showed the attachment of Ag@SnS2 on the cell surface and obvious cellular membrane destruction. Ag@SnS2 can also accelerate in vivo wound healing through PTT-induced bacterial disinfection. Therefore, Ag@SnS2 exhibits great potential for photothermal antibacterial application and wound disinfection.

Salen-manganese complex-based nanozyme with enhanced superoxide- and catalase-like activity for wound disinfection and anti-inflammation

Xiaomei Dai,Qingqing Xu,Yu Li,Lele Yang,Yongjie Zhang,Xiaojun Liu,Feng Gao

Skin wound healing remains a key challenge owing to its vulnerability to pathogen infection, oxidative stress and persistent inflammatory response. Herein, a salen-manganese complex (EUK)-based nanozyme was designed to simultaneously circumvent these barriers. The nanozyme was prepared by the amidation reaction between degradable chito-oligosaccharide (COS) and EUK. Positive charged COS and metallic compounds (EUK) could allow the nanozyme (COS-EUK) synergistic antibacterial activity. Due to the protection and synergistic catalytic enhancement effect provided by COS, superoxide (SOD)- and catalase (CAT)-like capability of COS-EUK was significantly improved compared with EUK. The nanozyme could eradicate not only O 2 − but also the generated H 2 O 2 , further supplying O 2 to promote fiber cell proliferation. Moreover, COS-EUK could prevent macrophages from transforming into classically activated macrophages (M1 phenotype), and endow them anti-inflammatory effects. COS-EUK can accelerate bacteria-infected skin wound healing. Thus, COS-EUK provides a promising strategy for bacteria-infected wound healing with great potential in clinical infection.

Iodine-doped carbon dots with inherent peroxidase catalytic activity for photocatalytic antibacterial and wound disinfection

Wang XiuLi,Lu Yinguang,Hua Kunwei,Yang Dezhi,Yang Yaling

A new type of nitrogen-iodine co-doped carbon dot (N/I-CD) was synthesized by a one-step hydrothermal method with a fluorescence quantum yield of 37%. The prepared N/I-CDs exhibit peroxidase-like activity, can catalyze bio-safety levels of H 2 O 2 to generate hydroxyl radicals (•OH) under visible light and enhance the level of reactive oxygen species (ROS) in bacteria cells. All in vitro experiments showed that the designed system has strong photocatalytic antibacterial activity against both E. coli and S. aureus bacteria. The light-induced antibacterial function of N/I-CDs was evaluated under the conditions of changing other experimental parameters. When the visible light irradiation time was extended to 60 min, the antibacterial efficiency of N/I-CDs (0.21 mg·mL −1 ) against S. aureus and E. coli reached 100% in the presence of exogenous H 2 O 2 (0.07 mM). More importantly, wound disinfection in vivo demonstrates the high antibacterial efficiency, low toxicity and application potential of good biocompatibility due to the nanozyme activity of N/I-CDs.

ICG@ZIF-8/PDA/Ag composites as chemo-photothermal antibacterial agents for efficient sterilization and enhanced wound disinfection

Shuhan Hui,Qiqi Liu,Yandong Han,Lijuan Zhang,Jun Yang,Shan Jiang,Haisheng Qian,Wensheng Yang

Bacterial infection has increasingly affected people's lives, therefore it is significant to explore novel antibacterial agents and strategies for efficient disinfection. Herein, we designed ZIF-8 based composites ICG@ZIF-8/PDA/Ag, which encapsulate photothermal agent indocyanine green (ICG) and grow polydopamine (PDA) on their surface for in situ reduction to generate Ag nanoparticles. With 20 min of 808 nm laser irradiation at 1.5 W cm−2, 100 μg mL−1 ICG@ZIF-8/PDA/Ag exhibited 100% bactericidal effects toward E. coli and S. aureus bacteria resulting from both hyperthermia of ICG and PDA and chemical toxicity of the released Ag and Zn ions. When the bacterial incubation period was extended to 12 h, the minimum bactericidal concentration (MBC) of ICG@ZIF-8/PDA/Ag was reduced to 6.25 μg mL−1, and this extremely low MBC was due to the long-term chemo-photothermal combinational effect induced by NIR irradiation. Additionally, the composites successfully promote the healing of S. aureus infected wounds on mice. This work constructed photo-responsive antibacterial composites that realize chemo-photothermal synergistic therapy.

Activation of biologically relevant levels of reactive oxygen species by Au/g-C3N4 hybrid nanozyme for bacteria killing and wound disinfection

Zhenzhen Wang,Kai Dong,Zhen Liu,Yan Zhang,Zhaowei Chen,Hanjun Sun,Jinsong Ren,Xiaogang Qu

As common reactive oxygen species, H 2 O 2 is widely used for bacterial inactivation and wound disinfection . However, the concentrations used are always higher than physiological levels , which frequently result in potential toxicity to healthy tissue and even delay wound healing. Here we report highly efficient nanozyme hybrids that are capable of activating biologically relevant concentrations of H 2 O 2 for defending bacterial infections. The integration of AuNPs with ultrathin graphitic carbon nitride (g-C 3 N 4 ) provides excellent peroxidase-activity, which can catalyze the decomposition of H 2 O 2 to OH radicals much more efficiently, allowing the use of bio-safety levels of H 2 O 2 for the first time. Furthermore, our system not only exhibits striking bactericidal performance against both DR Gram-negative and DR Gram-positive bacteria, but also shows high efficiency in breaking down the existing DR-biofilms and prevented formation of new biofilms in vitro. More importantly, in vivo experiments indicate that our system could significantly prevent bacterial infections and accelerate the healing rate of wounds.

Graphdiyne–hemin-mediated catalytic system for wound disinfection and accelerated wound healing

Arbab Ali,Jiaming Liu,Huige Zhou,Tao Liu,Muhammad Ovais,Huibiao Liu,Yukui Rui,Chunying Chen

Surgical site infections are the most common and serious concern in health care settings. Currently, systemic administration of prophylactic antibiotics or local disinfection treatment is prescribed after surgical procedures. However, prolonged use of antibiotics may pose adverse side effects, bacterial resistance and hospitalization costs. Recently, the use of nanomaterials with peroxidase enzyme-mimic activity has drawn higher consideration due to their high antibacterial potential and minimum biotoxicity. Hemin is a highly active molecule and also the active source of many biological enzymes, such as peroxidase, which could efficiently catalyze various biochemical reactions. However, the insolubility of hemin makes it easy for it to aggregate and lose its catalytic ability. Here, we designed a well-dispersed hemin/graphdiyne (GDY) nanocomposite (GDY–hemin) with high peroxidase activity. GDY–hemin can decompose hydrogen peroxide (H2O2) to generate highly-toxic hydroxyl radicals that impair bacterial cell membranes, ultimately leading to bacterial death in both in vitro and in vivo experiments. The results reveal that GDY–hemin has good biocompatibility and is suitable for wound infection treatment in vivo.

Bio-interface engineering of MXene nanosheets with immobilized lysozyme for light-enhanced enzymatic inactivation of methicillin-resistant Staphylococcus aureus

Daorui Zhang,Lunjie Huang,Da-Wen Sun,Hongbin Pu,Qingyi Wei

Being a unique biocatalyst to hydrolyse structural polysaccharides on the cell wall of bacteria, lysozyme can slow down bacterial resistance caused by antibiotic overdose treatments, however, efficient activation of lysozyme activities under pathogenic microenvironments remains challenging. Herein, a bio-interface engineering strategy was proposed for the remote modulation of thermoresistant lysozyme upon rationally designed photothermal nanoplatforms. For this, Ti 3 C 2 T X MXene nanosheets were functionalized by polydopamine (PDA) surface chemistry to enhance photothermal effects and performance durability, during which lysozyme biomacromolecules were immobilized at such a two-dimensional hybrid interface via intermolecular electrostatic affinity. The integrated nanoplatform (denoted as [email protected] @Lyso), with an optimal high light-to-heat conversion efficiency of 46.88%, realized not only precision control of local heat but also photo-responsive up-regulation for bio-catalysis of laden lysozyme. As a result, in vitro and in vivo antibacterial experiments revealed that [email protected] @Lyso could effectively inhibit the proliferation of methicillin-resistant Staphylococcus aureus and accelerated wound disinfection of mice with negligible biological toxicities. The outstanding antibacterial activities of [email protected] @Lyso were attributed to the photo-enhanced lysozyme activity, assisted by bacterial death caused by the mild local hyperthermia and the physical destruction derived from the [email protected] This work exemplified a solution to the bacterial resistance threats via stimuli-responsive enzymatic nanoplatforms.

Cu2O Nanocubes Embedded in Polycaprolactone Nanofibers for Photo–chemotherapeutic Wound Disinfection and Regeneration

Hui Zhao,Yang Yang,Jing An,Huilong Xin,Yinan Xiao,Zhaojun Jia,Yuandong Wu,Liyuan Sheng,Min Wen

Wound-associated infections represent an arduous global healthcare challenge, especially in the “post-antibiotic” era, necessitating the development of advanced antimicrobial dressing materials that simultaneously dictate the use of rapid yet effective disinfection, low drug resistance and side effects, and the potency to foster tissue healing. Nanoantimicrobial-armored nanostructured functional dressing systems have piqued extensive interest to satisfy these criteria. Herein, fusing electrospinning nanotechnology and an emerging photo–chemo synergistic sterilization paradigm, a smart “all-in-one” multifunctional wound-managing dressing platform was constructed, through incorporating photochemically active nanocrystalline cuprous oxide (Cu2O) into modified polycaprolactone (mPCL) electrospun fibrous mats. The resulting materials were systematically characterized pertaining to physiochemical analysis, photoabsorption and photocurrent measurements, and photocatalytic reactive oxygen species (ROS) generation. The capability of this nanoplatform to elicit rapid, photoactivated, catalytically augmented bacterial disinfection was corroborated, displaying ∼99–100% broad-spectrum antibacterial efficacies against Staphylococcus aureus and Escherichia coli in 25 min. Benefiting from the release of Cu2+, mPCL-Cu2O impairs bacterial contamination even under dark conditions. Furthermore, these materials also exhibited favorable in vitro/-ovo biocompatibility toward NIH3T3 fibroblasts, red blood cells, and chick chorioallantoic membrane, alongside appreciable pro-healing capacity in bridging scratch-based wounds, which could be attributed to nanofiber-afforded biomimetic topography and controllable liberation of micronutrient Cu ions. This study provides insights into the design and fabrication of simple yet smart antibacterial nanoplatforms for biomedical applications.

Alginate-modulated continuous assembly of iron/tannic acid composites as photothermally responsive wound dressings for hemostasis and drug resistant bacteria eradication

Juanjuan Li,Jiani Han,Wenqin Yu,Kaiyuan Wang,Zhu Liu,Yong Liu

Multifunctional dressing materials are highly required to combat multidrug resistant bacteria in wound infections. Here an alginate-based aerogel dressing is reported that combines photothermal bactericidal activity, hemostatic property, and free radical scavenging for skin wound disinfection and accelerated wound healing. The aerogel dressing is facilely constructed by immersing a clean nail (Fe) in a mixed solution of sodium alginate (Alg) and tannic acid (TA), followed by freezing, solvent replacement, and air drying. The Alg matrix plays an essential role in modulating the continuous assembly process between TA and Fe to allow the homogenous distribution of TA-Fe metal-phenolic networks (MPN) in the resulting composite, without forming aggregates. The photothermally responsive Nail-TA/Alg aerogel dressing is successfully applied in a murine skin wound model infected with Methicillin-resistant Staphylococcus aureus (MRSA). This work provides a facile strategy to integrate MPN with the hydrogel/aerogel matrix through in situ chemistry, which is promising for developing multifunctional biomaterials and biomedicine.

An Ultrasmall Fe3O4-Decorated Polydopamine Hybrid Nanozyme Enables Continuous Conversion of Oxygen into Toxic Hydroxyl Radical via GSH-Depleted Cascade Redox Reactions for Intensive Wound Disinfection

Jiayu Xiao,Luo Hai,Yaoyao Li,Huan Li,Minhui Gong,Zefeng Wang,Zifeng Tang,Le Deng,Dinggeng He

Nanozyme-based chemodynamic therapy (CDT) for fighting bacterial infections faces several major obstacles including low hydrogen peroxide (H 2 O 2 ) level, over-expressed glutathione (GSH) in infected sites, and inevitable damage to healthy tissue with abundant nonlocalized nanozymes. Herein, a smart ultrasmall Fe 3 O 4 -decorated polydopamine (PDA/Fe 3 O 4 ) hybrid nanozyme is demonstrated that continuously converts oxygen into highly toxic hydroxyl radical (•OH) via GSH-depleted cascade redox reactions for CDT-mediated bacterial elimination and intensive wound disinfection. In this system, photonic hyperthermia of PDA/Fe 3 O 4 nanozymes can not only directly damage bacteria, but also improve the horseradish peroxidase-like activity of Fe 3 O 4 decorated for CDT. Surprisingly, through photothermal-enhanced cascade catalytic reactions, PDA/Fe 3 O 4 nanozymes can consume endogenous GSH for disrupting cellular redox homeostasis and simultaneously provide abundant H 2 O 2 for improving •OH generation, ultimately enhancing the antibacterial performance of CDT. Such PDA/Fe 3 O 4 can bind with bacterial cells, and reveals excellent antibacterial property against both Staphylococcus aureus and Escherichia coli . Most interestingly, PDA/Fe 3 O 4 nanozymes can be strongly retained in infected sites by an external magnet for localized long-term in vivo CDT and show minimal toxicity to healthy tissues and organs. This work presents an effective strategy to magnetically retain the therapeutic nanozymes in infected sites for highly efficient CDT with good biosafety.

Low-intensity near-infrared light-triggered spatiotemporal antibiotics release and hyperthermia by natural polysaccharide-based hybrid hydrogel for synergistic wound disinfection

Hongling Deng,Jie Sun,Zhongpeng Yu,Zhihua Guo,Chen Xu

Photothermal sterilization is a promising and effective treatment method in treating bacterial infection. Generally, a widely employed light source in photothermal sterilization inevitably damages the skin tissue due to the high-intensity irradiation dose. How to provide useful antibacterial outcomes without light-triggered skin damage is a challenge for photothermal sterilization. In this work, a novel antibacterial hydrogel (VAT hydrogel, the abbreviation for vancomycin-agarose-ferric tannate hydrogel) has been successfully constructed by the natural polysaccharide hydrogel (AG) encapsulating ferric tannate (TA-Fe) nanoparticles and vancomycin. The VAT hydrogel exhibited the outstanding photothermal properties and controllable antibiotics release. With the results of antibacterial assays, the VAT hydrogel revealed the superior effectiveness of synergistic wound disinfection by the low-intensity near-infrared light-triggered spatiotemporal antibiotics release and hyperthermia. More importantly, the VAT hydrogels possessed the good biocompatibility. With the outstanding synergistic sterilizing effect and excellent biocompatibility, the VAT hydrogel would be a promising candidate for bacteria-associated wound infections.
