Body-temperature responsive hydrogel adhesive for active wound closure

Xiaowei Liu,Haojie Yu,Li Wang,Ruixue Liang,Yanxin Xiang,Xiaohang Song,Ying Yang,Zhikun Huang,Jian Yang,Di Shen,Yan Sheng

Hydrogels find extensive application in sutureless wound closure and healing. For relying solely on the natural recovery rate of skin or tissue often results in prolonged healing periods due to the gradual process of wound contraction facilitated by cell migration, our approach involves the integration of two distinct layers to create a cohesive and innovative body-temperature responsive hydrogel tape (TRAT). The shape memory layer of P (PTHF-co-PEG-co-PETMP) owns excellent mechanical properties and adjustable temperature-responsive characteristics. Upon integration with the adhesive layer of consisting of a silk fibroin semi-interpenetrating network and poly (acrylic acid) covalent network, the resulting TRAT composite maintains remarkable body temperature responsive shape memory attributes while retaining robust wet surface adhesion capabilities (~150 kPa). In virtue of the exceptional temperature-responsive and wet-adhesion features, TRAT is capable of achieving varying degrees of wound contraction, and spontaneously unfolds from an initial folded state and securely attaches to the inner esophageal surface upon temperature stimulation. This holds promising implications for the development of anti-migration trachea stents, bordering TRAT's potential in this application.

Photothermal-manipulatable shape memory polyacrylamide/gelatin Janus hydrogel with drug carrier array for invasive wound closure and responsive drug release

Yujie Wang,Yuxue Feng,Hongyu Tu,Huifang Zheng,Yunyun Xiang,Tongyao Zhang,Xinxin Huang,Fei Lu,Kun Yu,Enling Hu,Guangqian Lan,Liang-Ju Ning,Ruiqi Xie

Traditional wound closure methods often present several issues, including additional puncture wounds, adverse effects from anesthesia, and noticeable scarring. Inspired by embryonic wound healing, a Janus hydrogel (PG/Au-Asp@PCM) is designed to manipulate non-invasive wound closure by photothermal-responsive self-contraction of PG/Au-Asp@PCM, which is attributed to the shape memory behavior of PG/Au-Asp@PCM under near-infrared (NIR). Wherein, gelatin acts as a thermally reversible “switch” and polyacrylamide creates stable and cross-linked “net-points”. The elongated PG/Au-Asp@PCM can be temporarily fixed at 4 °C, and subsequently self-contracts upon NIR irradiation, generating a recovery force of 10 kPa, adequate for the closure of wound spontaneously. The Janus hydrogel also incorporates a drug carrier loaded with phase change material (PCM) and aspirin. The PCM absorbs heat during its phase transition above 42 °C, offering the photothermal-responsive release of aspirin on-demand; additionally, it also reduces the risk of skin burn during NIR exposure. Animal studies confirm the effectiveness of PG/Au-Asp@PCM in wound closure. Moreover, wounds treated with PG/Au-Asp@PCM exhibit reduced inflammation, increased thickness of epidermal and dermal layers, and a smoother appearance without scarring. These findings reinforce the feasibility of the photothermal strategy utilizing PG/Au-Asp@PCM for non-invasive wound closure, resulting in enhanced cosmetic appearance and improved wound healing outcomes.

Water-driven noninvasively detachable wet tissue adhesives for wound closure

Hongjian Huang,Renfeng Xu,Peng Ni,Zhenghong Zhang,Caixia Sun,Huaying He,Xinyue Wang,Lidan Zhang,Ziyi Liang,Haiqing Liu

Tissue adhesive with on-demand detachment feature is critically important since it can minimize hurt to patient when it is stripped away. Herein, a water-driven noninvasively detachable wet tissue adhesive hydrogel (w-TAgel) was produced by UV-initiated radical copolymerization of N-isopropylacrylamide (NIPAM), acrylamide (AAm), gelatin methacrylate (GelMA), and urushiol. As a w-TAgel, its robust and tough mechanical property makes it suitable for dynamic wound tissue. The polyurushiol segments of it are crucial to the formation of tough adhesion interface with various wet tissues, while polyNIPAM units play an indispensable role in on-demand detachment via thermo-responsive swelling behavior because the hydrophobic aggregation among isopropyl groups is destroyed upon water treatment with temperature of 25 °C or less. Additionally, it exhibits multiple merits including good hemocompatibility, cytocompatibility as well as pro-coagulant activity and hemostasis. Therefore, our w-TAgel with strong adhesion and facile detachment is an advanced prospective dressing for wound closure and rapid hemostasis. The wet tissue adhesion and water-driven detachable mechanism may shed new light on the development of on-demand noninvasively detachable wet tissue adhesives.

An anti-infective hydrogel adhesive with non-swelling and robust mechanical properties for sutureless wound closure

Xinchen Du,Yujie Hou,Le Wu,Shilin Li,Ao Yu,Deling Kong,Lianyong Wang,Guiling Niu

A non-swelling hydrogel adhesive is urgently needed in clinical application for wound closure; however, preparing a non-swelling hydrogel adhesive with superior mechanical and tissue adhesion properties remains a challenge. In this study, we developed a new family of non-swelling hydrogel adhesives composed of Pluronic F127 diacrylate, poly(ethylene glycol) diacrylate, modified sodium alginate, and tannic acid. Physical and biological properties of the hydrogels were systematically evaluated in vitro/vivo. The results indicated that the hydrogels exhibited non-swelling features, robust mechanical properties and good adhesion abilities toward various tissues. The hydrogels also exhibited good cytocompatibility and strong antibacterial activities against S. aureus and E. coli. Additionally, the hydrogel could be used for sutureless wound closure and displayed better advantages compared to sutures and commercial adhesive pads. The above results demonstrated that our non-swelling hydrogel adhesive with robust mechanical properties holds great promise for applications in clinical surgery.

Hydrogel–mesh composite for wound closure

Yang Gao,Xiuyuan Han,Jiaojiao Chen,Yudong Pan,Meng Yang,Linhe Lu,Jian Yang,Zhigang Suo,Tongqing Lu

During operations, surgical mesh is commonly fixed on tissues through fasteners such as sutures and staples. Attributes of surgical mesh include biocompatibility, flexibility, strength, and permeability, but sutures and staples may cause stress concentration and tissue damage. Here, we show that the functions of surgical mesh can be significantly broadened by developing a family of materials called hydrogel–mesh composites (HMCs). The HMCs retain all the attributes of surgical mesh and add one more: adhesion to tissues. We fabricate an HMC by soaking a surgical mesh with a precursor, and upon cure, the precursor forms a polymer network of a hydrogel, in macrotopological entanglement with the fibers of the surgical mesh. In a surgery, the HMC is pressed onto a tissue, and the polymers in the hydrogel form covalent bonds with the tissue. To demonstrate the concept, we use a poly(N-isopropylacrylamide) (PNIPAAm)/chitosan hydrogel and a polyethylene terephthalate (PET) surgical mesh. In the presence a bioconjugation agent, the chitosan and the tissue form covalent bonds, and the adhesion energy reaches above 100 J⋅m−2. At body temperature, PNIPAAm becomes hydrophobic, so that the hydrogel does not swell and the adhesion is stable. Compared with sutured surgical mesh, the HMC distributes force over a large area. In vitro experiments are conducted to study the application of HMCs to wound closure, especially on tissues under high mechanical stress. The performance of HMCs on dynamic living tissues is further investigated in the surgery of a sheep.

Single-component hyaluronic acid hydrogel adhesive based on phenylboronic ester bonds for hemostasis and wound closure

Ming Li,Xinglei Shi,Bo Yang,Jiajun Qin,Xiaobao Han,Wanjia Peng,Yiyan He,Hongli Mao,Deling Kong,Zhongwei Gu

Hydrogel tissue adhesives that currently available are often fabricated by mixing two or more polymeric components. Single-component hydrogels afford injectability, strong and reversible adhesion remain a formidable challenge. This research describes the creation of the first single-component hyaluronic acid hydrogel adhesive-based on phenylboronic acid-diol ester linkages. Phenylboronic acid can not only serve as a cross-linker to form hydrogel, but also act as an adhesion site for glycosyl compounds found in biological cell membranes. The rheological and compressive tests for the hydrogel show that it has excellent self-healing properties, good injectability and strong compressive strength . Adhesion tests demonstrated that the hydrogel has significantly greater adhesion strength than commercial fibrin glue. These findings suggest that the rational design of hydrogel precursors facilitates the formation of single-component networks and multiple functionalities. In vivo studies further proved the hydrogel was an ideal bio-adhesive with biocompatibility, absorbed wound exudate and hemostasis , and accelerated wound closure.

Biomimetic Dual-Layer Architectural Hydrogel Bandage with Smart Thermally Self-Contraction for Enhanced Wound Closure and Burn Wound Healing

Qiaobo Wang,Wenqian Zheng,Jie Wang,Caicai Jiao,Dian Gao,Ji Liu,Baoyang Lu

Severe burn injuries disrupt cutaneous barrier integrity, leading to elevated infection susceptibility, substantial fluid loss, and delayed tissue regeneration, which collectively deteriorate patient outcomes. While early stage wound closure is critical for mitigating these complications, current hydrogel dressings often lack dynamic contractile properties to synergistically facilitate both physical closure and biological healing. Inspired by the mechanobiology of embryonic wound contraction, we develop a nested poly(N-isopropylacrylamide)-sodium alginate hydrogel bandage (PNS-HB) featuring a biomimetic dual-layer architecture. The outer layer comprises a polydopamine-poly(acrylic acid) (PDA–PAA) adhesive framework, enabling tissue-conformal fixation (interfacial toughness of 162 J/m2; Young’s modulus of 45 kPa; fracture strain of 85%). The inner layer is a thermoresponsive PNS-HB exhibiting excellent mechanical flexibility (Young’s modulus of 9.9–16.7 kPa) and programmable shape-morphing capabilities, including temperature-dependent bending (118° angle) and contraction (contraction rate of 56%). In a deep second-degree burn model, the PNS-HB demonstrated a three-stage therapeutic mechanism: thermally triggered wound edge closure through contractile strain generation, inflammatory modulation through downregulation of IL-6 and TNF-α (>60% reduction), and pro-regenerative microenvironment establishment evidenced by accelerated angiogenesis and re-epithelialization. This biomimetic dual-layer architecture offers a paradigm-shifting approach for bridging the gap between physical wound closure and biological tissue restoration in burn care.

A shear-thinning adhesive hydrogel reinforced by photo-initiated crosslinking as a fit-to-shape tissue sealant

Shaoquan Bian,Zhiqiang Zheng,Yuan Liu,Changshun Ruan,Haobo Pan,Xiaoli Zhao

Surgical sealants suitable for wounds with non-flat complex geometries are still a challenge to fulfill clinical requirements. Herein, a novel fit-to-shape sealant enhanced by photo-initiated crosslinking was developed utilizing maleic anhydride modified chitosan (MCS), benzaldehyde-terminated PEG (PEGDF) and polyethylene glycol diacrylate (PEGDA). Initially, the shear-thinning hydrogel prepared through the Schiff-base linkage between MCS and PEGDF could be injected into target sites, remolded to conform to a wound with non-flat complex geometry, and remain on the wound, avoiding adverse liquid leakage. Under illumination with ultra-violet (UV) light, the hydrogel was solidified in situ rapidly to adopt the wound contour and enhanced in adhesive strength to seal defects of the tissue. In addition, the hydrogel exhibits stability in extreme pH environments (pH = 1) and has potential to treat wounds inside the stomach with the existence of gastric acid. Moreover, the hydrogel can be applied as adhesive wound dressings through in situ 3D printing. Taken together, the fit-to-shape sealant enhanced by photo-initiated crosslinking can be considered as promising tissue adhesives for wound closure and other biomedical applications.

Robust but On-Demand Detachable Wet Tissue Adhesive Hydrogel Enhanced with Modified Tannic Acid

Xinyue Wang,Hongjian Huang,Renfeng Xu,Yan Fang,Yunxiang Weng,Zhengchao Wang,Xiaopeng Xiong,Haiqing Liu

Adhesives with robust but readily detachable wet tissue adhesion are of great significance for wound closure. Polyelectrolyte complex adhesive (PECA) is an important wet tissue adhesive. However, its relatively weak cohesive and adhesive strength cannot satisfy clinical applications. Herein, modified tannic acid (mTA) with a catechol group, a long alkyl hydrophobic chain, and a phenyl group was prepared first, and then, it was mixed with acrylic acid (AA) and polyethylenimine (PEI), followed by UV photopolymerization to make a wet tissue adhesive hydrogel with tough cohesion and adhesion strength. The hydrogel has a strong wet tissue interfacial toughness of ∼1552 J/m2, good mechanical properties (∼7220 kPa cohesive strength, ∼873% strain, and ∼33,370 kJ/m3 toughness), and a bursting pressure of ∼1575 mmHg on wet porcine skin. The hydrogel can realize quick and effective adhesion to various wet biological tissues including porcine skin, liver, kidney, and heart and can be changed easily with triggering urea solution to avoid tissue damage or uncomfortable pain to the patient. This biosafe adhesive hydrogel is very promising for wound closure and may provide new ideas for the design of robust wet tissue adhesives.

Strong and Degradable Adhesion of Hydrogels

Hang Yang,Chenghai Li,Jingda Tang,Zhigang Suo

Adhesives potentially offer convenient means to close wounds, but existing adhesives do not fulfill many common requirements. Here we demonstrate an approach to develop hydrogel adhesives that are strong initially, remain soft when adhered to soft tissues, and degrade over time. We demonstrate the approach by using a hydrogel that dissipates a large amount of energy during separation, forms strong and interlinks with the soft tissues, and degrades by breaking cross-links. The hydrogel achieves initial adhesion energies of 300–700 J/m2 when adhered to different tissues, can bear huge pressure, and completely degrades in 5 weeks under simulated physiological conditions.

Non-swelling polyelectrolyte complex hydrogels with tissue-matchable mechanical properties for versatile wet wound closure

Xinyi Li,Jiarong Wang,Yirong Guo,Honglin Qian,Yiduo Chen,Yanchen Chen,Jing Wang,Youxiang Wang,M. Cristina L. Martins,Xinyang Hu,Jian'an Wang,Jian Ji

The stable adhesion of hydrogel-based bioadhesives with tissue-matchable mechanical properties in biological environments remains a significant challenge. In this work, we propose a polyethyleneimine-polyacrylic acid (PEI-PAA, PEA) double-network polyelectrolyte hydrogel with swelling resistant capacity and tunable mechanical properties via one-step UV-initiated polymerization. Driven by electrostatic interactions and polymer-chain entanglement, this PEA hydrogel displays a distinctive microphase separation behavior, which facilitates a wide tunability in mechanical properties. Specifically, the modulus varies from 0.4 MPa to 106 MPa, and the toughness ranges from 1479 kJ/m 3 to 7641 kJ/m 3 , respectively. Besides, the microphase separation endows PEA hydrogel with notable anti-swelling properties in saline, TBS buffer, and blood, leading to consistent adhesion to diverse moist tissues. We further demonstrate that our PEA hydrogels provide matchable mechanical properties and long-lasting adhesion to rat skin and arteries, which promote skin injury healing and effectively halt artery rupture bleeding in vivo. This work presents a straightforward method to generate non-swelling hydrogels and offers novel insight into the development of bioadhesives to meet diverse mechanical requirements.

A silk fibroin based bioadhesive with synergistic photothermal-reinforced antibacterial activity

Xiang Ke,Shuxian Tang,Zhiyun Dong,Hao Wang,Xinyuan Xu,Rongmin Qiu,Jiaojiao Yang,Jun Luo,Jianshu Li

Bioadhesives have gained considerable popularity for application in wound closure. However, applying bioadhesives incurs risks associated with bacterial infection during wound healing. Hence, in this study, a silk fibroin based bioadhesive was constructed via employing natural macromolecule, silk fibroin (SF), to spontaneously coassemble with natural plant polyphenol, tannic acid (TA), and iron oxide nanoparticles (Fe 3 O 4 NPs). In the system, the natural macromolecule SF plays a key role in fabricating the macromolecular network matrix due to the change of the secondary structure of SF (from random coil to β-sheet) under the trigger of TA. Importantly, the strong hydrogen bonding interactions between SF and TA, and the coordination bonds between TA and Fe 3 O 4 NPs endow the bioadhesive with high extensibility, self-healing properties, and considerable wet adhesion. Meanwhile, the synergy between the inherent photothermal properties of Fe 3 O 4 NPs and TA/Fe 3+ complexes under near-infrared (NIR) radiation enables the bioadhesive superior photothermal-reinforced antibacterial activity. The multifunctional natural macromolecule bioadhesive is a potential candidate in clinical wound management for improved outcomes, especially in infected wounds.
