Integrated Wound Recognition in Bandages for Intelligent Treatment

Xuecheng He,Shijie Yang,Conghui Liu,Tailin Xu,Xueji Zhang

The mismatch between irregular wounds and fixed dressings is an important but long-neglected problem in wound management, which may cause imprecise or incomplete care outcomes. Continuous advances in the internet, Internet of Things, and materials science put forward higher requirements in precise wound treatment. Herein, an integrated wound recognition strategy is demonstrated by extracting digital geometric information of specific irregular wounds. Such online wound image scanning and recognition integrated with offline intelligent material fabrication can greatly reduce incomplete wound closure or potential irritation toward normal tissue around the wound site. As a proof-of-concept, an active matrix of Ag-modified gelatin on bandages demonstrates promising antibacterial properties for practical application. In vivo mouse experiment reveals that wound-customized bands enable precisely fitting in the irregular wound bed, resulting in accelerating wound healing efficiency. Such wound recognitions in a bandage involve multidisciplinary technology including image identification, computer modeling nanomaterial fabrication and modification, presenting a broad potential of personalized wound healthcare for a wide variety of clinical applications.

Microneedle Patch for Transdermal Sequential Delivery of KGF-2 and aFGF to Enhance Burn Wound Therapy

Huacheng He,Wen Huang,Shihui Zhang,Jie Li,Jian Zhang,Bingxin Li,Jie Xu,Yuting Luo,Huiling Shi,Yue Li,Jian Xiao,Odinaka Cassandra Ezekiel,Xiaokun Li,Jiang Wu

Severe burn wounds usually destroy key cells’ functions of the skin resulting in delayed re-epithelization and wound regeneration. Promoting key cells’ activities is crucial for burn wound repair. It is well known that keratinocyte growth factor-2 (KGF-2) participates in the proliferation and morphogenesis of epithelial cells while acidic fibroblast growth factor (aFGF) is a key mediator for fibroblast and endothelial cell growth and differentiation. However, thick eschar and the harsh environment of a burn wound often decrease the delivery efficiency of fibroblast growth factor (FGF) to the wound site. Therefore, herein a novel microneedle patch for sequential transdermal delivery of KGF-2 and aFGF is fabricated to enhance burn wound therapy. aFGF is first loaded in the nanoparticle (NP aFGF ) and then encapsulated NP aFGF with KGF-2 in the microneedle patch (KGF-2/NP aFGF @MN). The result shows that KGF-2/NP aFGF @MN can successfully get across the eschar and sequentially release KGF-2 and aFGF. Additional data demonstrated that KGF-2/NP aFGF @MN achieved a quicker wound closure rate with reduced necrotic tissues, faster re-epithelialization, enhanced collagen deposition, and increased neo-vascularization. Further evidence suggests that improved wound healing is regulated by significantly elevated expressions of hypoxia-inducible factor-1 alpha (HIF-1ɑ) and heat shock protein 90 (Hsp90) in burn wounds. All these data proved that KGF-2/NP aFGF @MN is an effective treatment for wound healing of burns.

MXene-based polysaccharide aerogel with multifunctional enduring antimicrobial effects for infected wound healing

Hui Li,Yuanli Yang,Min Mu,Chenqian Feng,Di Chuan,Yangmei Ren,Xiaoxiao Wang,Rangrang Fan,Jiazhen Yan,Gang Guo

Wound infection is a predominant etiological factor contributing to delayed wound healing in open wounds. Hence, it holds paramount clinical significance to devise wound dressings endowed with superior antibacterial properties . In this study, a Schiff base-crosslinked aerogel comprising sodium alginate oxide (OSA), carboxymethyl chitosan (CMCS), and Nb 2 C@Ag/PDA (NAP) was developed. The resultant OSA/CMCS-Nb 2 C@Ag/PDA (OC/NAP) composite aerogel exhibited commendable attributes including exceptional swelling characteristics, porosity, biocompatibility, and sustained antimicrobial efficacy. In vitro antimicrobial assays unequivocally demonstrated that the OC/NAP composite aerogel maintained nearly 100 % inhibition of Staphylococcus aureus and Escherichia coli under an 808 nm laser even after 25 h. Crucially, the outcomes of in vivo infected wound healing experiments demonstrated that the wound healing rate of the OC/NAP composite aerogel group reached approximately 100 % within a span of 14 days, which was significantly greater than that of the blank control group. In vitro and in vivo hemostatic experiments also revealed that the composite aerogel had excellent hemostatic properties. The results of this study demonstrate the remarkable potential of OC/NAP aerogel as a multifunctional clinical wound dressing, especially for infected wounds.

Novel lignin–chitosan–PVA composite hydrogel for wound dressing

Yiwen Zhang,Miaomiao Jiang,Yuqing Zhang,Qiping Cao,Xing Wang,Ying Han,Guangwei Sun,Yao Li,Jinghui Zhou

Although PVA–chitosan composite hydrogel has excellent biocompatibility and antibacterial ability, its poor mechanical strength limits its application for wound dressings. Furthermore, PVA–chitosan composite hydrogel cannot satisfy the requirements of wound dressing as an environmental conditioner to accelerate wound healing. In this work, a novel lignin–chitosan–PVA composite hydrogel was prepared as wound dressing. The introduction of lignin effectively improved the mechanical strength (tensile stress is up to 46.87 MPa), protein adsorption capacity, and wound environmental regulation ability of the hydrogel. In a murine wound model, the lignin–chitosan–PVA composite hydrogel significantly accelerated wound healing. The developed hydrogel provides new opportunities for highly efficient skin wound care and management.

An Electrostatic-Induction-Enabled Anti-Touching Hydrogel Dressing for Chronic Wound Care

Ren Yan,Zhaoqing Sun,Wanyu Du,Nian Liu,Qizeng Sun,Peng Li,Fei Liang,Yangzhi Zhu,Wei Huang,Hai-Dong Yu

Chronic wounds are inherently vulnerable to external mechanical contact owing to their impaired physiological states. Accidental touching may lead to secondary damage to delicate wound tissues, including deterioration of tissue and inflammation, thereby prolonging the healing process. In addition, patients encounter nociceptive sensations, indicative of physical discomfort and potential distress. However, the traditional dressings for chronic wounds fail to convey adequate perceptual information to prevent mechanical trauma. An electrostatic-induction-enabled anti-touching hydrogel dressing (EAHD) is proposed, which promotes wound healing and reduces touching-induced injuries. The EAHD combines a proximity sensor and a pro-healing hydrogel. The proximity sensor can detect approaching objects, enabling remote perception. More importantly, EAHD and a wireless alarm system are integrated to warn against the detected potential hazards such as unintentional touching. Meanwhile, the EAHD significantly accelerates wound healing in preclinical diabetic mice, ≈38% faster than that of the control group. This study proposes a novel strategy for multifunctional wound dressing that promotes wound healing and prevents accidental touching-induced secondary damage to the wound.

Color-changing smart fibrous materials for naked eye real-time monitoring of wound pH

Nan Pan,Juanrong Qin,Pingping Feng,Zeke Li,Botao Song

Real-time monitoring of wound pH may provide information about the wound healing status and potential bacterial infection. Herein, we integrated the biocompatible color changing substance curcumin into a fibrous material, capable of in situ real-time visually monitoring the wound pH. The results indicate that the curcumin-loaded fibrous mat exhibits an obvious pH-dependent color change from yellow to red brown with a change in pH from 6.0 to 9.0, which can be easily detected by the human naked eye. Moreover, the wound pH conditions can be determined with the aid of a smart phone App after image analysis. Due to their flexibility, the fibrous materials have been further processed into various shapes from 1D to 3D for fitting the irregular wounds. It is believed that smart fibrous materials that can simultaneously real-time monitor the wound pH and repair the wound may change wound management to a convenient and comfortable way.

Injectable ADM temperature-sensitive hydrogel loaded with bFGF in diabetic rat wound healing study

Haojiao Chen,Jianping Sun,Wenyang Liu

Background: Diabetic wound is one of the most common diabetic chronic complications. Effective treatments of diabetic wound remain limited. Here, we explored the effects of basic fibroblast growth factor (bFGF)-acellular dermal matrix (ADM) hydrogel on the diabetic wound. Methods: The bFGF-ADM hydrogel was manufactured by mixing 180 µL ADM hydrogel and 20 µL bFGF aqueous solution (10 mg/mL). The morphology of ADM hydrogel and bFGF-ADM hydrogel was observed under scanning electron microscope. The release property of bFGF-ADM hydrogel was determined by ELISA. CCK-8 assay was utilized to estimate the cell viability of mouse skin fibroblasts. The diabetes mellitus (DM) model was established in rats. The four wounds on the back of each DM rat were treated with the ADM hydrogel, bFGF-ADM hydrogel, bFGF aqueous solution and no solution (control), respectively. The wound healing rate of each rat was estimated. The traumatized skin tissue of each rat was observed by H&E staining and Sirius red staining. Results: The bFGF-ADM hydrogel displayed an interconnected pore structure and bFGF was gradually released from the bFGF-ADM hydrogel over time. The bFGF-ADM hydrogel could enhance the cell viability of skin fibroblasts and promote the wound healing rate, the re-epithelialization of wound and increase the collagen fiber content of dermis. And the bFGF-ADM hydrogel exhibited better therapeutic effects of diabetic wound than either bFGF or ADM alone. Conclusions: Our study revealed that the bFGF-ADM hydrogel could promote diabetic wound healing.

Chronic wound management: a liquid diode-based smart bandage with ultrasensitive pH sensing ability

Wang Xueqi,Cheng Jing,Wang Han

Chronic wounds, which require prolonged healing periods, pose significant impacts on individuals with diabetes, vascular diseases, and high blood pressure. Simultaneous drainage and monitoring of wound exudate are vital for advanced wound management. However, recently reported smart dressings either lack integration of wound cleaning and monitoring functions or fail to achieve dynamic in situ monitoring of wound status, which hinders their ability to meet the demands of wound care. In this study, a smart bandage is introduced, which integrates a biocompatible liquid diode membrane with an ultrasensitive 3D polyaniline mesh (M-PANI)-based pH biosensor. The smart bandage allows for unidirectional drainage of wound exudate while dynamically sensing the wound pH environment. Specifically, the proposed smart bandage effectively cleans excessive wound exudate while providing real-time information on the wound status during the drainage process. The M-PANI-based pH biosensor demonstrates a high sensitivity of 61.5 mV/pH and a wide pH detection range from 4.0 to 10.0, encompassing the pH range of normal and infected wounds. Moreover, the sensing module exhibits excellent stability after 48 hours of dynamic testing and 28 days of storage, with only a 4.8% decline in the detected signal, and high repeatability with a device-to-device relative standard deviation (RSD) of 3.1%. To evaluate the practicality of this smart bandage, simulated skin and rats have been employed, and the results indicate the immense potential of this smart bandage for clinical applications. In conclusion, the present smart bandage demonstrates considerable promise for wound exudate cleaning and monitoring in advanced wound care and offers a promising method for home-based wound management.

A Novel Multilayer Composite Membrane for Wound Healing in Mice Skin Defect Model

Yuyu Qiu,Qingqing Wang,Yajun Chen,Shufang Xia,Wei Huang,Qufu Wei

To develop a wound dressing material that conforms to the healing process, we prepared a multilayer composite (MC) membrane consisting of an antibacterial layer (ABL), a reinforcement layer (RFL), and a healing promotion layer (HPL). Biocompatible zein/ethyl cellulose (zein/EC) electrospun nanofibrous membranes with in situ loaded antibacterial photosensitizer protoporphyrin (PPIX) and healing promotion material vaccarin (Vac) were, respectively, chosen as the ABL on the surface and the HPL on the bottom, between which nonwoven incorporated bacterial cellulose (BC/PETN) as the HPL was intercalated to enhance the mechanical property. Photodynamic antibacterial activity againstStaphylococcus aureusandPseudomonas aeruginosawas confirmed by the enlarged inhibition zones; meanwhile, satisfactory biocompatibility of the HPL was verified by scanning electronic microscopy (SEM) of L929 cells cultured on its surface. The potential effects on wound healing in a mice skin defect model of the MC membranes were also evaluated. The animal experiments demonstrated that the wound healing rate in the MC group was significantly increased compared with that in the control group (p< 0.05). Histopathological observation revealed an alleviated inflammatory response, accompanied with vascular proliferation in the MC group. The MC membranes significantly promoted wound healing by creating an antibacterial environment and promoting angiogenesis. Taken together, this MC membrane may act as a promising wound dressing for skin wound healing. Keywords:multilayer composite membrane;wound dressing;wound healing;antibacterial activity;biocompatibility

A sprayable alginate hydrogel based on copper-tannic acid metal-polyphenol for promoting acute and infected wound healing

Ruigang Zhou,Mengyan Zhou,Weimei Wang,Junjie Huang,Weilong Peng,Lianggang Wang,Jun Chen,Ruonan Bo,Mingjiang Liu,Jingui Li

Bacterial-infected wounds may lead to delayed repair due to increased inflammation, especially wounds infected by drug-resistant bacteria, which remains a major clinical challenge. Sprayable hydrogels can treat large and irregular dynamic wounds and have attracted significant attention as wound dressings. Herein, sprayable hydrogels were formed by rapid crosslinking of atomized sodium alginate (SA) and high-concentration calcium ion solution, and copper-tannic acid metal-polyphenol network (CuTA) was loaded as the active ingredient. The hydrogel system achieved rapid crosslinking (10 s) through interaction with 10 % CaCl₂ solution, while deep eutectic solvent (DES) enhanced moisturizing capacity. In vitro studies demonstrated the broad-spectrum antibacterial effect of DES + Gel/CuTA sprayable hydrogel, with an inhibition rate of over 99 %. The excellent hemostatic performance of DES + Gel/CuTA sprayable hydrogel was also demonstrated, with the bleeding volume being less than 0.2 g, which was lower than 0.46 g in the control group. In three animal models of acute, infected and burn-infected skin wounds, DES + Gel/CuTA sprayable hydrogel showed significant effects in promoting wound healing. In conclusion, the developed sprayable hydrogels offered several advantages, including antibacterial and anti-inflammatory properties and promoting key stages of wound healing, such as cell migration, M2 polarization, collagen deposition, and angiogenesis. This sprayable hydrogel can resist infection and promote healing, showing its great potential in treating infected wounds.

Sprayable oxidized cellulose nanofiber hydrogel with rapid hemostatic ability for skin wound healing

Le Wang,Ya Liu,Wentao Duan,Feng Cao,Qianxin Lv,Sijia Zhang,Jianzhen She,Luying Yang,Boling He,Yan Hou,Liang Kong,Taiqiang Dai,Ruizhi Ning,Bolei Cai

Wound healing, especially hemostasis, requires rapid bleeding control for tissue regeneration. Although TEMPO-mediated oxidized cellulose nanofibers (TOCN) are commonly used in hemostatic materials clinically, Scarce research has explored TOCN as the sole component in sprayable hydrogels for hemostasis and wound repair. In this study, we successfully formulated a sprayable hydrogel composed solely of TOCN and water. We evaluated its effectiveness in achieving rapid hemostasis and promoting skin wound healing via comprehensive in vitro and in vivo experiments. Our findings indicate that the uniformly sized nanoscale oxidized cellulose, obtained via ultrasonic fragmentation, can form a non-crosslinked three-dimensional network in water, resembling a hydrogel. The TOCN hydrogel exhibits excellent biocompatibility and coagulation properties. When compared to sodium alginate, TOCN hydrogel demonstrates superior hemostatic results, effectively stopping bleeding in a rat liver hemostasis model. Additionally, the sprayable TOCN hydrogel showed outstanding efficacy in a rat skin-defect model, achieving complete wound closure and regeneration of dermal and epidermal tissues, including sebaceous glands and hair follicles, by day 15. Its significant hemostatic effect and promotion of wound healing were further validated in a porcine wound model. In conclusion, the study highlights the great clinical potential of TOCN-based sprayable hydrogel for hemostasis and skin healing.

Inhibition and Rescue of Hyperglycemia-Induced Cellular Senescence by Mitochondrial Transfer from Enucleated Mesenchymal Stem Cell-Derived Microvesicles for Chronic Wound Healing

Zixuan Dong,Xiaobing Liu,Shichun Li,Xiaoling Fu

The aberrant cellular senescence in chronic wounds presents a significant barrier to healing. Mitochondrial dysfunction is critical in initiating and maintaining cellular senescence, underscoring therapeutic potential in restoring mitochondrial function by delivering healthy mitochondria to wound cells. However, approaches for delivering mitochondria to achieve optimized wound repair remain lacking. Herein, enucleated MSCs-derived microvesicles containing functional mitochondria (Mito@euMVs) via simple extrusion are developed. By controlling the size of microvesicles within a small micron-scale range, the mitochondrial encapsulation efficiency is optimized. Mito@euMVs effectively delivered mitochondria into fibroblasts and HUVECs, inhibiting and rejuvenating hyperglycemia-induced cellular senescence. To enhance the clinical applicability, soluble PVA microneedle patches for the transdermal Mito@euMVs delivery are utilized. In diabetic rats with pressure sores, the senescence-inhibiting and -rescuing properties of Mito@euMVs are further validated, along with their therapeutic efficacy, demonstrating their potential for chronic wound repair. Moreover, as a versatile delivery vehicle for mitochondria, Mito@euMVs hold promising for treating mitochondrial dysfunction and aging-related conditions.
