Affecting analysis of the rheological characteristic and reservoir damage of CO2 fracturing fluid in low permeability shale reservoir

Li Qiang,Wang Fuling,Forson Kobina,Zhang Jinyan,Zhang Chenglin,Chen Juan,Xu Ning,Wang Yanling

The fracturing property of liquid CO 2 fracturing fluid varies greatly due to the rheology of fracturing fluid during fracturing process. The main objective of this investigation is to study the rheology property of thickened liquid CO 2 by measuring the viscosity of thickened liquid CO 2 in different physical parameters of this prepared thickener and explain the causes of rheological changes. The results show that thickener content, branching content, and molecular weight of a thickener for all could significantly improve the rheology of liquid CO 2 ; the consistency coefficient K increased as they rose, but the rheological index n presented a decreased trend. Meanwhile, the mesh structure is proposed as a model to explain the rheological changes, and the large wetting angle means an excellent backflow, low reservoir damage, and low adsorption property. These results herein provide a basic reference to improve the CO 2 fracturing technology and molecular design of CO 2 thickener.

Evaluation of antinociceptive and anti-inflammatory effects of aqueous extract of Armadillidium vulgare Latreille

Guo Shen-shen,Ren Meng-yue,Song Shuai,Wei Ping,Luo Jia-bo

Objective To assess the antinociceptive and anti-inflammatory properties of the aqueous extract of Armadillidium vulgare (AV). Methods The antinociceptive effect of AV (400, 600 and 800 mg/kg) was investigated in mice using the acetic acid-induced writhing, formalin-induced nociceptive, and hot plate tests. Phlogogen-induced paw edema using carrageenan, dextran, or compound 48/80 as phlogogen was used as inflammatory models to evaluate AV's anti-inflammatory effect. Additionally, the bioactive substances glucosamine (GLcN) and taurine in AV were determined using high-performance liquid chromatography. Results Oral treatment of the mice with AV (600 and 800 mg/kg) significantly reduced the number of writhes in the acetic acid-induced writhing test ( P <0.01) but not the hot plate test ( P >0.05). All doses tested significantly inhibited paw-withdrawal during the second phase of the formalin-induced nociceptive model ( P <0.01). AV demonstrated a strong anti-inflammatory effect in all those inflammatory models ( P <0.05). Conclusions AV has antinociceptive and anti-inflammatory effects, providing scientific evidence of the efficacy of its traditional use in pain treatment. Furthermore, GLcN and taurine contribute, at least in part, to the anti-inflammatory activity of AV.

Antinociceptive effects of sinomenine in a rat model of postoperative pain

Qing Zhu,Yuehua Sun,Lina Mao,Chengpeng Liu,Bo Jiang,Wei Zhang,Jun-Xu Li

Background and Purpose This study examined the antinociceptive effects of sinomenine in a rat model of postoperative pain. Experimental Approach Male and female rats were subjected to a surgical incision in the right hind paw, and the von Frey filament test was used to measure mechanical hypersensitivity after drug or vehicle treatment (p.o. or i.p.). Rats were treated daily with sinomenine before or after the surgery and the AUCs of the antinociceptive effects measured during a 4 h period were calculated to determine the ED 50 values of sinomenine. The anti-hyperalgesic effects of different doses of a combination of sinomenine and acetaminophen (paracetamol) were assessed in another group of rats. Dose combinations were determined by using a fixed ratio dose-addition analysis method. Key Results Sinomenine (5–80 mg·kg −1 ) produced dose-dependent antinociceptive effects in rats that had been subjected to surgery and this effect lasted for 4 h. The potency of sinomenine, given i.p. or p.o., did not differ between male and female rats. However, sinomenine was fourfold more potent when given i.p. than p.o. The GABA A receptor antagonist bicuculline blocked the antinociceptive effects of sinomenine. The antinociceptive effect of a daily treatment with sinomenine remained stable throughout the course of postoperative pain. Pretreatment with sinomenine did not alter the mechanical hypersensitivity post-surgery. The combination of sinomenine with acetaminophen produced an infra-additive interaction. Conclusions and Implications Sinomenine demonstrated significant antinociceptive activity against postoperative pain and may be a useful novel pharmacotherapy for the management of postoperative pain.

G(1-5)-EM2, a multi-targeted agonist to opioid and growth hormone secretagogue receptors exhibited nontolerance forming antinociceptive effects in a mouse model of burn pain

Songxia Cheng,Jiali Ding,Biao Xu,Yan Wang,Xiaoyu Shen,Yanhua Xia,Lei Wu,Jie Wei

Burn induced-pain (BIP) is one of the most common pain symptoms, which seriously affects the quality of sufferer life. Researches show that multi-targeted drug therapies offer superior efficacy and fewer side effects compared to single-target drug therapies. Consequently, in this study, we developed G(1-5)-EM2, a multi-targeted peptide designed to target μ-opioid receptor and the growth hormone secretagogue receptor 1α (GHS-R1α), and explored its antinociceptive effects on burn injury pain. Calcium mobilization experiments revealed that G(1-5)-EM2 demonstrated weak multi-agonist activities to μ-opioid receptor and κ-opioid receptor as well as GHS-R1α in vitro. Near-infrared fluorescence imaging experiments demonstrated that G(1-5)-EM2 could penetrate the blood-brain barrier (BBB) and access the brain following intravenous injection. The enzymatic stability of G(1-5)-EM2 was significantly enhanced compared to EM2. Our results indicated that intrathecal administration of G(1-5)-EM2 mitigated mechanical allodynia and thermal hyperalgesia in BIP. These antinociceptive effects of G(1-5)-EM2 were partially mediated through μ-opioid receptor and GHS-R1α. Moreover, intrathecal administration of G(1-5)-EM2 significantly decreased burn-induced up-regulation of phosphorylated p38 MAPK, phosphorylated NF-κBp65 and TRPV1 in the ipsilateral spinal cord, reduced the levels of IL-1β, IL-6 and TNF-α in serum, and enhanced wound healing in burned skin. Repeated intrathecal administration of G(1-5)-EM2 produced a non-tolerance-forming antinociception in BIP. These results suggest that the multi-targeted peptide G(1-5)-EM2 exhibits a novel role in alleviating BIP with fewer side effects and may represent a promising strategy for developing new analgesic drugs.

Live imaging and quantitation of insect feeding-induced Ca2+ signal using GCaMP3-based system in Nicotiana benthamiana

Yunjing Wang,Qian Gong,Fan Huang,Linfang He,Yule Liu

Summary Wounding evokes transient increases in cytosolic calcium (Ca 2+ ) concentration. Visualizing real-time Ca 2+ flux provides new insights into Ca 2+ -signaling pathways. Here, we outline a protocol to detect insect feeding-induced Ca 2+ flux elevation in Nicotiana benthamiana leaves based on the GCaMP3 reporter system by Leica fluorescence stereo microscopes (LFSM). LFSM combines super-fast manual screening with high-end imaging capabilities. Through this protocol, we can clearly observe the calcium flow after aphid’s piercing-sucking. Additionally, we describe a protocol to quantify Ca 2+ level using LFSM. For complete details on the use and execution of this protocol, please refer to Wang et al. (2021) .

Performance evaluation of oil-displacement viscoelastic zwitterionic surfactant fracturing fluid

Hang Xu,Fujian Zhou,Yuan Li,Che Liu,Liyan Zhu,Erdong Yao

Viscoelastic surfactant (VES) fracturing fluid has advantages in sand-carrying and micelle-breaking without residues, while its practical applications are always limited due to the poor temperature and shearing resistance. Accordingly, a kind of stearicamide propyl hydroxylsulfobetaine (SPHB) was self-synthesized by using stearic acid, N, N-dimethyl-1,3-propane diamine, and sodium 3-shloro-2-hydroxypropane sulfonate as raw materials, and a novel VES fracturing fluid (HCF) was prepared by optimizing the ratio of SPHB and counterion. The composition of the novel fracturing fluid system was determined according to the viscosity varies with temperature. Moreover, the temperature and shearing resistance, viscoelastic, sand-carrying, drag reduction, micelle-breaking, and oil displacement properties of the system were systematically studied. The results showed that the optimum molar ratio of SPHB to salicylate counterion in the fracturing fluid system was 1:1. Surface tension test showed the critical micelle concentration (CMC) of SPHB surfactant solution is 6.27 × 10 -4 mol /L and the surface tension γ cmc is 40 mN/m. The apparent viscosity of the novel fluid was kept above 50 mPa·s after continuous sheared for 60 min under the condition of 170 s −1 at 130 ℃. Besides, it has good viscoelastic property and sand-carrying performance, and the experimental results showed that the drag reduction rate of the novel fluid reaches more than 60% at different temperatures. The micelle-breaking performance of the novel system was perfect. There is basically no solid residue in the micelle-breaking solution, and the permeability damage rate of the micelle-breaking liquid to the natural core was only 9.41%, which showed the novel fluid has the characteristics of low damage. Finally, low-permeability natural cores were used to carry out micelle-breaking fluid oil displacement experiments, including single core flooding and double-tube parallel core flooding, and the mechanism of micelle-breaking fluid improving oil recovery was revealed.

Pregabalin can interact synergistically with Kv7 channel openers to exert antinociception in mice

Chao-Nan Huang,Yan-Ming Chen,Xin-Yi Xiao,Hui-Ling Zhou,Jin Zhu,Hui-Min Qin,Xue Jiang,Zongzheng Li,Tao Zhuang,Gui-Sen Zhang

Chronic pain is a common public health problem and remains an unmet medical need. Currently available analgesics usually have limited efficacy for the treatment of chronic pain, including neuropathic pain and persistent inflammatory pain , or they are accompanied by many adverse side effects . The voltage-gated calcium channel blocker (pregabalin) and potassium channel openers (flupirtine and retigabine) have been widely used for the management of chronic pain, but their effectiveness in combination is unclear. In this research, we evaluated the antinociceptive effects of pregabalin in combination with flupirtine or retigabine in carrageenan-induced inflammatory pain and paclitaxel-induced peripheral neuropathy in mice using the von Frey test. Isobolographic analysis indicated that pregabalin exerted synergistic antinociceptive effects when combined with flupirtine or retigabine in neuropathic and inflammatory pain models. Furthermore, the antinociceptive effects of pregabalin, flupirtine/retigabine, and their combinations were significantly attenuated by the Kv7 channel blocker XE991. The favored dose ratio between pregabalin and flupirtine/retigabine in combinations was also investigated. Finally, we evaluated the motor coordination of their combinations using the rotarod test, and the outcomes underpinned their safety. Collectively, our results support the potential use of pregabalin in combination with flupirtine or retigabine to alleviate chronic pain.

Novel chimeric peptides based on endomorphins and ghrelin receptor antagonist produced supraspinal antinociceptive effects with reduced acute tolerance in mice

Bing Wu,Songxia Cheng,Fuyan Liu,Jia Wei,Yongling Liu,Teng Qian,Jiali Ding,Biao Xu,Jie Wei

It is widely recognized that developing bi- or multifunctional opioid compounds could offer a valuable approach to pain management with fewer side effects compared to single-target compounds. In this study, we designed and characterized two novel chimeric peptides, EM-1-DLS and EM-2-DLS, incorporating endomorphins (EMs) and the ghrelin receptor antagonist [D-Lys3]-GHRP-6 (DLS). Functional assays demonstrated that EM-1-DLS and EM-2-DLS acted as κ-opioid receptor (κ-OR)-preferring agonists, weak μ-opioid receptors (μ-OR) and ghrelin receptor (GHSR) agonists. Upon intracerebroventricular (i.c.v.) administration in mice, both EM-1-DLS and EM-2-DLS exhibited dose- and time-dependent antinociceptive effects in the tail withdrawal test. EM-1-DLS demonstrated the highest antinociceptive potency among the peptides, with an ED 50 approximately 8-fold greater than EM-1, while EM-2-DLS showed comparable effects to EM-2. The antinociceptive actions of EM-1-DLS involved activation of GHS-R1α, μ-OR, and κ-OR, whereas EM-2-DLS acted via GHS-R1α, δ-OR, and κ-OR pathways. Additionally, acute antinociceptive tolerance was investigated, revealing that EM-1-DLS induced a tolerance ratio of 2.33-fold, significantly lower than the 5.19-fold ratio induced by EM-1. Cross-tolerance ratios between the chimeric peptides and EMs ranged from 0.92 to 1.76, indicating reduced tolerance compared to EMs alone. These findings highlight the potential of these chimeric peptides to mitigate pain with diminished tolerance development, suggesting a promising strategy for the development of new analgesic therapies with improved safety profiles.

A Cross-Scale Memristor-Based Neuromorphic Platform Bridging Physical Encoding and Medical Inference

Xiaobin Ren,Zhirong Liu,Song Wang,Zelin Cao,Kaikai Gao,Guangdong Zhou,Lu Ye,Bai Sun

Despite the progress in machine vision, existing image recognition systems still operate in isolation from the physical world, lacking dynamic temporal encoding, in-memory processing, and intrinsic interpretability. Here, a memristive vision architecture that integrates multimodal sensing, physical memory, and medical inference within a unified device-level framework is developed. Specifically, spatial gradients from retinal fundus images are encoded into Ag/Indigo/FTO memristor array via pulse modulation, generating conductance maps (“G maps”) that serve both as nonvolatile memory and as neuromorphic inputs for lesion segmentation via lightweight U-Net models. Beyond static vision, the same memristor exhibits strong temporal sensitivity, enabling biologically realistic nociceptive encoding and Morse code recognition. These results demonstrate a cross-task, cross-scale system that merges physical encoding with machine intelligence, bridging the gap between material dynamics and high-level perception. Therefore, this paradigm offers a scalable, interpretable hardware foundation for next-generation bio-perceptive computing and intelligent diagnostics.
