Preparation and characteristics of sucrose-resistant emulsions and their application in soft candies with low sugar and high lutein contents and strong antioxidant activity

Li-Hua Pan,Cui-Ling Wu,Shui-Zhong Luo,Jian-Ping Luo,Zhi Zheng,Shao-Tong Jiang,Yan-Yan Zhao,Xi-Yang Zhong

Soft candies are popular confections but contain high-sugar and low-functional ingredients. The inclusion of xanthan gum (XG, 0.2–0.5%) in a water-in-oil emulsion stabilized by tea polyphenol palmitate (TPP) particles made structured lutein-loaded emulsions suitable for high-sugar foods . With the increase in XG concentration, the droplet size of the lutein-loaded emulsion decreased, but the lutein loading capacity, G′, G″, and apparent viscosity increased. The G′, G″, G* and η* values of the most desirable emulsion stabilized by 2.5% TPP particles and 0.5% XG were reduced, but tanα increased when 0.25–1.00 g/mL sucrose was added; however, the emulsion remained stable based on the droplet size and zeta potential , suggesting that the lutein-loaded emulsion was sucrose resistant. The emulsion was partially added into a confection formula for soft candy preparation. As the replacement ratio of emulsions increased from 20% to 100%, the moisture, lutein content, and DPPH• inhibition of soft candies increased by 61.98%, 325.55% and 135.66%, respectively, but hardness, chewiness , and resilience decreased by 61.68%, 67.81% and 47.76%, respectively, while color and sensory stabilities increased first, reaching the peaks at the level of 60%, and then decreased; however, the T g value first decreased and then increased. The emulsions that were filled evenly and stably in the jelly matrix enhanced the heat stability of the candies. This research provides an ingredient system for preparing sugar-resistant emulsions, which will be filling an unprecedented array of needs in confectionary development, such as the replacement of sugar and the delivery of fat-soluble bioactive compounds .

A novel strategy for producing low-sugar pomegranate jam with better anthocyanin stability: Combination of high-pressure processing and low methoxyl & amidated pectin

Yurou Yun,Jian Li,Fei Pan,Yijun Zhou,Xiangqiao Feng,Jun Tian,Shengbao Cai,Junjie Yi,Linyan Zhou

Low-sugar jams (LSJ) are promoted as a healthier food option due to the healthy trend, and significant color changes negatively affects the evaluation of these products. To reveal protective effect of low methoxyl & amidated pectin (LMA) on anthocyanin in jam treated by high-pressure processing (HPP), this study compared the changes in microbes and quality attributes of LSJ prepared with LMA and high-sugar pomegranate jam (HSJ) prepared with high methoxyl pectin (HMP) after HPP at 600 MPa for 5 min during accelerated storage at 35 °C for 30 days, and molecular dynamics simulation was conducted to reveal the interaction between LMA and anthocyanin. LSJ-HPP had better color quality with higher values of L* , a* , and total anthocyanin content (TAC), the corresponding percent polymeric color and browning index were 41% and 16% lower than those of HSJ-HPP, respectively. TAC in LSJ-HPP, HSJ, and HSJ-HPP decreased to 9.5, 8.6, and 6.0 mg/kg (pomegranate puree) after storage, respectively. Cyanidin-3- O -glucoside formed more H-bonds with LMA than HMP, while their total interaction energy and the binding frequency and number were also higher. This study provided a foundation for the processing and storage of high quality LSJ, and also broadened the application field of HPP technology.

Dual specificity phosphatase 22 suppresses mesangial cell hyperproliferation, fibrosis, inflammation and the MAPK signaling pathway in diabetic nephropathy

Na Ren,Shanshan Shi,Na Zhao,Lingyan Zhang

Dual specificity phosphatase 22 (DUSP22) regulates fibrosis and inflammation, which may be implicated in the development of diabetic nephropathy (DN). Hence, the current study aimed to assess the effect of DUSP22 on cell proliferation, apoptosis, fibrosis and inflammation in mouse mesangial cell line (SV40‑MES13) under both high glucose (HG) and low glucose (LG) conditions. SV40‑MES13 cells were treated with HG and LG, then HG‑group cells were transfected with DUSP22 overexpression and control plasmids, meanwhile LG‑group cells were transfected with DUSP22 and control siRNAs. Then, cell proliferation using Cell Counting Kit‑8, cell apoptosis by TUNEL assay, protein expression using western blotting, inflammatory cytokines using ELISA and RNA using reverse transcription‑quantitative PCR were determined. DUSP22 mRNA and protein were decreased in SV40‑MES13 cells under the HG condition compared with those under the LG condition. Under the HG condition, DUSP22 overexpression suppressed SV40‑MES13 cell proliferation at 48 and 72 h as well as Bcl2, but it facilitated TUNEL‑reflected apoptotic rate and cleaved‑caspase‑3; besides, DUSP22 overexpression restrained proteins of fibronectin 1, collagen I, transforming growth factor beta 1, and their corresponding mRNAs. As to the inflammation, DUSP22 overexpression downregulated TNF‑α, IL‑1β, IL‑6 and IL‑12 under the HG condition. By contrast, DUSP22 siRNA promoted SV40‑MES13 cell proliferation, fibrosis and inflammation, but attenuated apoptosis in SV40‑MES13 cells under the LG condition. Additionally, DUSP22 overexpression inactivated phosphorylated (p)‑ERK, p‑JNK, and p‑P38 in HG‑treated SV40‑MES13 cells; differently, DUSP22 small interfering RNA facilitated them under the LG condition. In conclusion, DUSP22 suppresses HG‑induced mesangial cell hyperproliferation, fibrosis, inflammation and the MAPK pathway, implying its potency in DN treatment.
