Biodegradable electronics dissolve in vivo , eliminating retention risks, but they are often not self-contained due to limited energy storage, relying on tethered or wireless power. Here, we report a biodegradable flexible supercapacitor (BFSC) that can be completely dissolved to achieve symbiosis with the body and modulate neural excitation and inhibition without external energy. Its low stiffness and favorable biocompatibility allow it to minimize interface mismatch and rejection to be integrated with the body like a biological tissue. The BFSC has a farad surface capacitance (0.36 F/cm 2 ), high energy density (73.1 μWh/cm 2 ), and 1-month operating life in vivo due to the MXene nanolayered electrode and solid-state hydrogel electrolyte. The BFSC can drive bioelectric stimulation circuits to enable neuromodulation and cardiac pacing in rats and pigs. The scaffold-like BFSC almost completely inhibits neural activity. These advances are expected to facilitate the development of self-contained biodegradable and symbiotic bioelectronics.

Encouraging progress in multifunctional nanotheranostic agents that combine photothermal therapy (PTT) and different imaging modalities has been made. However, rational designed and biocompatible multifunctional agents that suitfable for in vivo application is highly desired but still challenging. In this work, we rationally designed novel ultrasmall multifunctional nanodots (FS-GdNDs) by combining the bovine serum albumin (BSA)-based gadolinium oxide nanodots (GdNDs) obtained through a biomineralization process with a small-molecule NIR-II fluorophore (FS). The as-prepared FS-GdNDs with an ultrasmall hydrodynamic diameter of 9.3 nm exhibited prominent NIR-II fluorescence properties, high longitudinal relaxivity (10.11 mM −1 s −1 ), and outstanding photothermal conversion efficiency (43.99%) and photothermal stability. In vivo studies showed that the FS-GdNDs with enhanced multifunctional characteristics diaplayed satisfactory dual-modal MR/NIR-II imaging performance with a quite low dose. The imaging-guided PTT achieved successful ablation of tumors and effectively extended the survival of mice. Cytotoxicity studies and histological assay demonstrated excellent biocompatibility of the nanodots. Importantly, this novel FS-GdNDs can undergo efficient body clearance through both hepatobiliary and renal excretion pathways. The novel ultrasmall multifunctional FS-GdNDs with excellent features hold tremendous potential in biomedical and clinical applications.

Mechanical cues are critical regulators of cell fate and behavior through the orchestrated and continual conversion of physical forces into biochemical responses. However, due to the poor compatibility between mechanical and biochemical techniques, existing methods are often limited in characterizing the occurring biochemical signals during mechanical stimulation. Herein, this work presents a magneto-responsive nanomesh (MRnM) biosensor capable of mechanically stimulating cells in vitro and tissues in vivo and simultaneously detecting the triggered biomolecules. Under external magnetic fields, the sensor exhibits excellent magnetic responsiveness with remote, controllable and tailored deformation, while maintaining prominent and stable electrochemical sensing performance. As a proof of concept, this MRnM sensor achieves the magnetically-actuated deformation of osteoblasts and real-time monitoring of the ensuing nitric oxide release, revealing the role of Piezo1 channels in nitric oxide synthase signaling pathways. Furthermore, we demonstrate the capability of MRnM sensor for in vivo applications. Ultimately, the developed MRnM biosensor holds great potential for mechanical stimulation and real-time monitoring of various biological systems, ranging from living cells to soft tissues and in vivo organs.

Although existing nanomedicines have focused on the tumor microenvironment with the goal of improving the effectiveness of conventional chemotherapy, the penetration of a tumor’s core still represents a formidable barrier for existing drug delivery systems. Therefore, a novel multifunctional hypoxia-induced size-shrinkable nanoparticle has been designed to increase the penetration of drugs, nucleic acids , or probes into tumors. This cooperative strategy relies on three aspects: (i) the responsiveness of nanoparticles to hypoxia, which shrink when triggered by low oxygen concentrations; (ii) the core of a nanoparticle involves an internal cavity and strong positive charges on the surface to deliver both doxorubicin and siRNA; and (iii) a reactive oxygen species (ROS) probe is incorporated in the nanoparticle to monitor its preliminary therapeutic response in real time, which is expected to realize the enhanced efficacy together with the ability to self-monitor the anticancer activity . A more effective inhibition of tumor growth was observed in tumor-bearing zebrafish, demonstrating the feasibility of this cooperative strategy for in vivo applications. This research highlights a promising value in delivering drugs, nucleic acids, or probes to a tumor’s core for cancer imaging and treatment. Statement of Significance Hypoxia-induced chemoresistance of tumor cells still represents a formidable barrier, as it is difficult for existing drug delivery systems to penetrate the tumor hypoxia core. This study involves the hypoxia-responsive size-shrinkable nanoparticle co-delivery of DOX and siRNA to enhance the penetration of DOX deep within tumors and subsequently disturb crucial pathways of cancer development induced by hypoxia and to improve sensitization to DOX chemotherapy. Furthermore, the nanopreparation can combine the ROS probe as a self-reporting nanopreparation to realize the function of real-time feedback efficacy, which has a good application prospect in the diagnosis and treatment of cancer.

Genetic engineering faces persistent challenges in achieving precise Deoxyribonucleic acid (DNA) cleavage, especially with the limitations associated with current enzyme-based methods, exemplified by issues in CRISPR technologies. This study introduces a groundbreaking approach: utilizing reactive oxygen species (ROS) generated by Multiphoton Absorption (MPA)-excited Black Phosphorus Quantum Dots (BPQDs) under femto-second laser irradiation. This innovative method not only allows excitation with lower energy but also enhances overall efficiency. The integration of complementary RNA sequences facilitates high-efficiency, site-selective DNA cleavage in the system, named “TADPOLE” (Targeted DNA Precision Oriented Laser Excision). Beyond its precision in targeting arbitrary DNA sequences using quantum dots, TADPOLE harnesses the unique multiphoton absorption property of BPQDs, enabling excitation with lower-energy light sources suitable for in vivo applications in the future. Moreover, the approach integrates guiding RNA and ultrafast laser technology to provide precise control over local ROS generation and minimal heat production. This guarantees site-specific DNA cleavage while mitigating the risk of damage to non-targeted sequences. In summary, this study catalyzes advancements in enzyme-free DNA cleavage technologies, with transformative implications for genetic engineering, biotechnology, and medicine. The holistic precision, versatility, and endurance presented by TADPOLE open new avenues for targeted gene therapies and transformative applications in related fields.
