Streptococcosis disease caused by Streptococcus agalactiae (Group B Streptococcus, GBS) results in a huge economic loss of tilapia culture. It is urgent to find new antimicrobial agents against streptococcosis. In this study, 20 medicinal plants were evaluated in vitro and in vivo to obtain medicinal plants and potential bioactive compounds against GBS infection. The results showed that the ethanol extracts of 20 medicinal plants had low or no antibacterial properties in vitro, with a minimal inhibitory concentration ≥256 mg/L. Interestingly, in vivo tests showed that 7 medicinal plants could significantly inhibit GBS infection in tilapia, and Sophora flavescens (SF) had the strongest anti-GBS activity in tilapia, reaching 92.68%. SF could significantly reduce the bacterial loads of GBS in different tissues (liver, spleen and brain) of tilapia after treated with different tested concentrations (12.5, 25.0, 50.0 and 100.0 mg/kg) for 24 h. Moreover, 50 mg/kg SF could significantly improve the survival rate of GBS-infected tilapia by inhibiting GBS replication. Furthermore, the expression of antioxidant gene cat , immune-related gene c-type lysozyme and anti-inflammatory cytokine il-10 in liver tissue of GBS-infected tilapia significantly increased after treated with SF for 24 h. Meanwhile, SF significantly reduced the expression of immune-related gene myd88 and pro-inflammatory cytokines il-8 and il-1β in liver tissue of GBS-infected tilapia. The negative and positive models of UPLC-QE-MS, respectively, identified 27 and 57 components of SF. The major components of SF extract in the negative model were α, α-trehalose, DL-malic acid, D- (−)-fructose and xanthohumol, while in the positive model were oxymatrine, formononetin, (−)-maackiain and xanthohumol. Interestingly, oxymatrine and xanthohumol could significantly inhibit GBS infection in tilapia. Taken together, these results suggest that SF can inhibit GBS infection in tilapia, and it has potential for the development of anti-GBS agents.

The nematode Caenorhabditis elegans is a useful model host for pathogenesis research that can be infected by a large number of human pathogens. Conventionally, nematode–pathogen infection assays are mainly performed on agar medium which are labor-intensive and time-consuming. To overcome these challenges, we develop for the first time an integrated microfluidic device for evaluating in vivo antimicrobial activity of natural compounds, which allows infection and anti-infection assays to be sequentially and automatically carried out in liquid medium. The device consists of a worm dispenser with 32 trap-construction chambers and concentration gradient generators, in which the processes of introduction, dispensation, confinement of worms in the chamber and drug delivery to the chamber can be integrated into a single device. In addition, the operation of the device is simple and does not require any expensive robotic fluid handling systems to dispense samples. To demonstrate the ability of this device, we devise an on-line screening experiment using a Caenorhabditis elegans–Staphylococcus aureus infection model and characterize the survival rate of the infected worms treated with antibiotics. Then, we applied the system to evaluate the antibacterial activity of several components of rhubarb: aloe-emodin, rhein and emodin at various concentrations. The device is able to load uniform worms into each chamber within 10 min and then generate various chemical concentrations automatically and simultaneously. Furthermore, the on-chip method only requires 6 h to establish the infection model and 48 h to perform the subsequent treatments. Based on the excellent advantages and scalable properties of microfluidics, the microfluidic platform holds a great potential in high-throughput screening for antimicrobials.

Antimicrobial resistance has emerged as a significant threat to global public health. To develop novel, high efficiency antibacterial alternatives to combat multidrug-resistant bacteria, A total of thirty-two novel amphiphilic benzopyran derivatives by mimicking the structure and function of antimicrobial peptides were designed and synthesized. Among them, the most promising compounds 4h and 17e displayed excellent antibacterial activity against Gram-positive bacteria (MICs = 1–4 μg/mL) with weak hemolytic activity and good membrane selectivity. Additionally, compounds 4h and 17e had rapid bactericidal properties, low resistance frequency, good plasma stability, and strong capabilities of inhibiting and eliminating bacterial biofilms. Mechanistic studies revealed that compounds 4h and 17e could effectively disrupt the integrity of bacterial cell membranes, and accompanied by an increase in intracellular reactive oxygen species and the leakage of proteins and DNA, ultimately leading to bacterial death. Notably, compound 4h exhibited comparable in vivo antibacterial potency in a mouse septicemia model infected by Staphylococcus aureus ATCC43300, as compared to vancomycin. These findings indicated that 4h might be a promising antibacterial candidate to combat antimicrobial resistance.
