Due to the heterogeneity of the tumor, it is difficult to achieve complete ablation of the tumor by single photothermal treatment. Moreover, their broader use is constrained by the intrinsic high uptake in the reticuloendothelial system (RES) organs, resulting in slow elimination from the body and potential toxicity. In this study, we report a single-step synthetic approach to fabricate versatile FeCP-NPs, leveraging the coordination bonds formed between bioactive polyphenolic compounds (curcumin), Fe III ions, and polyvinyl pyrrolidone (PVP). The FeCP-NPs possess the following favorable characteristics: (1) The components of FeCP-NPs originate from natural products, an endogenous element, and an FDA-approved polymer, guaranteeing good biosafety. (2) FeCP-NPs exhibit excellent photothermal properties. (3) After the photothermal treatment is completed, the dynamic disassembly strategy triggered by deferoxamine mesylate (DFO) can induce the dissociation of nanoparticles and achieve rapid clearance in vivo. (4) Curcumin released during the disassembly of FeCP-NPs can be combined with photothermal therapy to achieve complete tumor ablation. A comprehensive analysis of the safety, biodistribution, and clearance patterns of FeCP-NPs is conducted in vitro or in vivo. This work provides a nanoparticle with great clinical translational potential and provides a new solution for inhibiting tumor growth.

Aim: To study the difference in biodistribution of gold nanoprisms (NPr) and nanorods (NR), PEGylated to ensure colloidal stability. Materials & methods: Surface changes were studied for nanoparticles in different media, while the biodistribution was quantified and imaged in vivo. Results: Upon interaction with the mouse serum, NR showed more abrupt changes in surface properties than NPr. In the in vivo tests, while NPr accumulated similarly in the spleen and liver, NR showed much higher gold presence in the spleen than in liver; together with some accumulation in kidneys, which was nonexistent in NPr. NPr were cleared from the tissues 2 months after administration, while NR were more persistent. Conclusion: The results suggest that the differential biodistribution is caused by size-/shape-dependent interactions with the serum.

To retain the agents in tumors for cancer diagnosis and therapy, and then to remove them from the body, are key for the clinical applications of ideal inorganic theranostic agents. To meet these needs, we have developed a transformed theranostic platform, employing PVP coated Fe3S4 tetragonal nanosheets (TNSs), which could effectively accumulate in the tumor under magnetic targeting, whilst gradually transforming to small particles (∼5 nm) over three weeks. These were then effectively excreted from the body in normal physiological conditions after exerting their therapeutic effect. The aqueous dispersion of PVP coated Fe3S4 TNSs had an intense near-infrared absorption, excellent photothermal conversion efficiency (64.3%) and great T2 weighted magnetic resonance imaging properties (71.3 mM−1 S−1). In addition, Fe3S4 TNSs could realize a synergistic photothermal therapy (PTT)/chemodynamic therapy (CDT), because the localized heat produced by PTT from the defect-rich structure could enhance the Fenton process by utilizing the overproduced H2O2 in the tumor microenvironment, and in return, the produced ˙OH could inhibit tumor growth and recurrence after PPT. We thus developed a high-efficiency inorganic theranostic platform which was effectively cleared from the body. This will open up a new avenue for the design of inorganic agents for clinical applications in the future.

Encouraging progress in multifunctional nanotheranostic agents that combine photothermal therapy (PTT) and different imaging modalities has been made. However, rational designed and biocompatible multifunctional agents that suitfable for in vivo application is highly desired but still challenging. In this work, we rationally designed novel ultrasmall multifunctional nanodots (FS-GdNDs) by combining the bovine serum albumin (BSA)-based gadolinium oxide nanodots (GdNDs) obtained through a biomineralization process with a small-molecule NIR-II fluorophore (FS). The as-prepared FS-GdNDs with an ultrasmall hydrodynamic diameter of 9.3 nm exhibited prominent NIR-II fluorescence properties, high longitudinal relaxivity (10.11 mM −1 s −1 ), and outstanding photothermal conversion efficiency (43.99%) and photothermal stability. In vivo studies showed that the FS-GdNDs with enhanced multifunctional characteristics diaplayed satisfactory dual-modal MR/NIR-II imaging performance with a quite low dose. The imaging-guided PTT achieved successful ablation of tumors and effectively extended the survival of mice. Cytotoxicity studies and histological assay demonstrated excellent biocompatibility of the nanodots. Importantly, this novel FS-GdNDs can undergo efficient body clearance through both hepatobiliary and renal excretion pathways. The novel ultrasmall multifunctional FS-GdNDs with excellent features hold tremendous potential in biomedical and clinical applications.

Currently used in vitro models for estimating liver metabolism do not take into account the physiologic structure and blood circulation process of liver tissue. The Bio-PK metabolic system was established as an alternative approach to determine the in vitro intrinsic clearance of the model drug tolbutamide. The system contained a peristaltic pump, recirculating pipeline, reaction chamber, and rat liver microsomes (RLMs) encapsulated in pluronic F127-acrylamide-bisacrylamide (FAB) hydrogel. The metabolism of tolbutamide at initial concentrations of 100, 150, and 200 μM was measured in both the FAB hydrogel and the circular medium. The data from the FAB hydrogel and the circular medium were fitted to a mathematical model to obtain the predicted intrinsic clearance of tolbutamide after different periods of preincubation. The in vitro clearance value for tolbutamide was incorporated into Simcyp software and used to predict both the in vivo clearance value and the dynamic process of elimination. The predicted in vivo clearance of tolbutamide was 0.107, 0.087, and 0.095 L/h/kg for i.v. injection and 0.113, 0.095, and 0.107 L/h/kg for oral administration. Compared with the reported in vivo clearance of 0.09 L/h/kg (i.v.) and 0.10 L/h/kg (oral), all the predicted values differed by less than twofold. Thus, the Bio-PK metabolic system is a reliable and general in vitro model, characterized by three-dimensional structured RLM and circulation and perfusion processes for predicting the in vivo intrinsic clearance of low-extraction compounds, making the system more analogous with the rat in terms of both morphology and physiology
