Different thermal processed qingke: characteristics of in vitro simulated digestion and fermentation

Lu Wang,Qingyue Hong,Huijing Chen,Xiaowei Peng,Zhirong Wang,Jianquan Kan

Qingke, a special crop, is primarily cultivated in Chinese plateau. Qingke is rich in bioactive compounds, but different thermal processes alter the structure and contents of these bioactive compounds. And the characteristics of in vitro simulated digestion and fermentation also changed under the consumption of processed qingke. This study investigated the effects of traditional roasting, steam explosion, steaming, and microwave baking on qingke's bioactive compounds contents during in vitro digestion, and their effects on intestinal environment and gut microbiota for human during in vitro fermentation. Results indicated that traditional roasted and steam-exploded qingke exhibited higher bioactive compounds contents and antioxidant activity during in vitro digestion. Additionally, decreased pH, reduced ammonia nitrogen production, and increased short-chain fatty acids (SCFAs) contents in steam-exploded qingke during in vitro fermentation. And traditional roasted and steam-exploded qingke can better regulated the gut microbiota. These findings highlight traditional roasting and steam explosion as the optimal processing methods for qingke.

In vivo and in vitro anti-inflammatory effects of Sophora flavescens residues

Hongyan Ma,Qi Huang,Wenshan Qu,Linyuan Li,Min Wang,Shao Li,Fujiang Chu

Ethnopharmacological relevance The dried roots of Sophora flavescens Ait. (Leguminosae) is traditionally used as antipyretic medicine to reduce inflammation. It is well known that alkaloids and flavonoids are the main constituents of S. flavescens . However, the clinical researches and applications of S. flavescens is mainly based on its water-extracted alkaloids, its flavonoids may still remain in residues and have been underused. With development and manufacturing of S. flavescens in recent years, more herb residues are being produced. Since they are typically treated as waste and dumped openly in landfill sites, which can cause pollution, there is a great need to explore these wastes as recyclable resources and increase their added value. To date, whether other bioactive components would be found in the residues of S. flavescens is still unknown. If the extraction method of these active ingredients was established, the residues of S. flavescens could be turned from the harm to a benefit and make great sense of the comprehensive utilization of S. flavescens resources. This study aimed to establish an extraction method of the residues of S. flavescens and investigate the anti-inflammatory effect of it both in vivo and in vitro . Materials and methods Dried S. flavescens were decocted with distilled water firstly, then the residues were powdered and extracted with ethyl acetate by using ultrasonic wave. HPLC was utilized to analyze the chemical constituents of the water extracts of S. flavescens (WSF) and the ethyl acetate extracts of residues of S. flavescens (RSF). In vivo , the anti-inflammatory effect of WSF and RSF were evaluated using the xylene-induced auricle edema, acetic acid-induced peritoneal permeability and carrageenan-induced hind paw edema methods. In vitro, the inhibitory activities of WSF and RSF on NO, TNF-α, IL-6 and MCP-1 production of LPS-treated RAW264.7 cells were measured. Results The major ingredients of RSF were flavonoids, while WSF almost had no flavonoids. In vivo , WSF and RSF (200 mg/kg) could significantly inhibit the edema in the xylene-induced mice auricle edema and carrageenan-induced hind paw edema as well as the peritoneal permeability increased by acetic acid. They can also lower production levels of PGE 2 in inflamed paw tissues. In vitro experimental results showed that RSF (25-100 μg/mL) could significantly inhibit the release of pro-inflammatory cytokines NO, TNF-α, IL-6 and MCP-1 on LPS-induced RAW264.7 cells. The in vitro suppress effect of WSF had no dose-response relationship. Conclusions The residues of S. flavescens had obvious flavonoids with anti-inflammatory activity. This study provided evidence for the reuse of residues from S. flavescens in the food additive , medicine and cosmetic industries.

Color, Starch Digestibility, and In Vitro Fermentation of Roasted Highland Barley Flour with Different Fractions

Zixuan Zhao,Jian Ming,Guohua Zhao,Lin Lei

Highland barley (HB) is commonly milled into flour for direct consumption or further processed with other food formulations. Nevertheless, the association between milling and HB flour properties remains lacking. This work studied the effect of particle sizes (coarse, 250–500 μm; medium, 150–250 μm; fine, <150 μm) on physicochemical and nutritional properties of raw and sand-roasted HB flour. Gelatinization enthalpy decreased with increasing particle sizes of raw HB flour, while no endothermic transitions were observed in sand-roasted flour. Sand roasting destroyed starch granules and decreased short-range molecular order. Starch digestibility increased while total short-chain fatty acids (SCFAs) production decreased with decreasing particle sizes in all samples. The relative crystallinity of sand-roasted HB flour decreased by 80–88% compared with raw samples. Sand roasting raised in vitro starch digestibility, while total SCFAs during in vitro fecal fermentation decreased. Sand-roasted HB flour with particle sizes <150 μm had the highest starch digestibility (94.0%) but the lowest production of total SCFAs (1.89–2.24 mM). Pearson’s correlation analysis confirmed the relationship between the nutritional qualities of HB flour and milling. Keywords:highland barley flour;particle sizes;in vitro starch digestibility;in vitro fermentation

In vitro digestion and fermentation characteristics of soluble dietary fiber from adlay (Coix lacryma-jobi L. var. ma-yuen Staft) bran modified by steam explosion

Shuxin Zhou,Xinjing Tang,Ferenc Hegyi,András Nagy,Krisztina Takács,Zsolt Zalán,Guangjing Chen,Muying Du

Adlay bran is known for its nutrient-rich profile and multifunctional properties, and steam explosion (SE) is an emerging physical modification technique. However, the specific effects of SE on the activity composition and antioxidant capacity of adlay bran soluble dietary fiber (SDF) during in vitro digestion, as well as its influence on gut microbiota during in vitro fermentation, remain inadequately understood. This paper reports the in vitro digestion and fermentation characteristics of soluble dietary fiber from adlay bran modified by SE (SE-SDF). Compared with the untreated samples (0-SDF), most of the phenolic compounds and antioxidant capacity were significantly increased in the SE-SDF digests. Additionally, SE was beneficial for adlay bran SDF to increase the content of acetic acid, propionic acid and total short-chain fatty acids (SCFAs) in fermentation broth during in vitro fermentation. SE-SDF could promote the growth of beneficial bacteria while inhibiting the proliferation of pathogenic microbes. Our research indicates that SE-SDF shows strong antioxidant properties after in vitro digestion and plays a pivotal role in regulating gut microbiota during in vitro fermentation, ultimately enhancing human intestinal health.

Danilo de Souza Costa,Rayssa A. Cajas,Carla Monteiro Leal,Lara S. Aleixo de Carvalho,Lara Cruz de Souza,Lucas Fukui-Silva,Josué de Moraes,Ademar A. Da Silva Filho

Ethnopharmacological relevance Acmella oleracea (L.) R. K. Jansen, commonly referred to as “Jambu”, is a valuable medicinal plant native to the Amazon regions. Inflorescences of A. oleracea is utilized as local anesthetic properties and for its insecticidal, antiparasitic, and anthelmintic attributes. Aims of this study This study aimed to evaluate the in vitro and in vivo antiparasitic properties of A. oleracea extract and spilanthol (SPL) against Schistosoma mansoni . Materials and methods The ethanolic extract of A. oleracea inflorescences (AoE) was prepared, analyzed by HPLC-DAD, and characterized by UHPLC-HRMS/MS. SPL was isolated from AoE by chromatographic fractionation. The antischistosomal properties of AoE and SPL were evaluated in vitro against adult schistosomes and in preclinical assays using murine models of patent S. mansoni infection. Results Through UHPLC-HRMS/MS analysis, 14 alkamides were annotated in AoE. HPLC-DAD analysis of AoE revealed a peak with a substantial relative area of ∼85%, which was isolated and identified as SPL. AoE and SPL caused mortality of adult schistosomes ex vivo , showing EC 50 values of 32.6 μg/mL and 27.8 μM, respectively, without affected Vero cells or Caenorhabditis elegans . In preclinical studies, the oral administration (400 mg/kg) of AoE and SPL resulted in significant reductions in worm burden of 28% and ∼42%, respectively. Additionally, SPL exhibits remarkable effects on oogram, decreasing egg burden and the number of immature eggs by over 80%, while also promoting significant reductions in hepato- and splenomegaly. Conclusions The research underscores the antischistosomal activity of A. oleracea highlighting the potential of SPL as a lead for the development of new schistosomicidal drugs and encouraging further studies to validate the traditional anthelmintic use of A. oleracea inflorescences.

Preparation and characterization of BSA-loaded microspheres based on polyanhydrides

Yang Yu,Tingli Lu,Wen Zhao,Weiguang Sun,Tao Chen

A series of biodegradable PEG-containing polyanhydrides composed of sebacic acid, 1, 6-bis( p -carboxyphenoxy) hexane, and poly (ethylene glycol) (PEG) were used as matrix material for BSA-loaded microspheres. The effects of polymer composition on the microsphere size, entrapment efficiency, in vitro degradation, and in vitro protein release were studied. Microspheres in the size range of 0.8–10 μm were fabricated via a modified double emulsion method and were characterized using scanning electron microscopy. As the content or the molecular weight of PEG was increased in the copolymer, a proportional increase was found in the particle size and the efficiency of BSA entrapment. The in vitro degradation rate of particles could be controlled by varying the polymer composition, increasing as the PEG proportion increased. In vitro release studies of BSA from polyanhydride microspheres revealed that the increased amounts of PEG within microspheres could accelerate the release rate of protein. These studies indicate that the PEG-containing polyanhydrides hold potential for protein delivery applications. © 2011 Wiley Periodicals, Inc. J Appl Polym Sci, 2011

Combinational Overexpression of Foxa3 and Hnf4a Enhance the Proliferation and Prolong the Functional Maintenance of Primary Hepatocytes

Fan J. Y.,Dama G.,Liu Y. L.,Guo W. Y.,Lin J. T.

Abstract In an in vitro culture system, primary hepatocytes usually display a low proliferation capacity, accompanied with a decrease of viability and a loss of hepatocyte-specific functions. Previous studies have demonstrated that the combination introductions of certain hepatocyte-specific transcription factors are able to convert fibroblasts into functional hepatocyte-like cells. However, such combinational usage of transcription factors in primary hepatocytes culture has not yet sufficiently studied. The forkhead box protein A3 (FoxA3) and hepatocyte nuclear factor 4α (Hnf4α) are liver-enriched transcription factors that play vital roles in the differentiation, and maintenance of hepatocytes. Thus, we simultaneously overexpressed the two genes, Foxa3 and Hnf4a , in rat hepatocytes and observed that the combinational augmentation of these two transcription factors have enhanced the proliferation and stabilized the hepatocyte-specific functions of primary hepatocytes over a long-term culture period.

Predicting the Glycemic Index of Biscuits Using Static In Vitro Digestion Protocols

Xingguang Peng,Hongsheng Liu,Xuying Li,Huaibin Wang,Kejia Zhang,Shuangqi Li,Xianyang Bao,Wei Zou,Wenwen Yu

In vitro digestion methods that can accurately predict the estimated GI (eGI) values of complex carbohydrate foods, including biscuits, are worth exploring. In the current study, standard commercial biscuits with varied clinical GI values between 9~30 were digested using both the INFOGEST and single-enzyme digestion protocols. The digestion kinetic parameters were acquired through mathematical fitting by mathematical kinetics models. The results showed that compared with the INFOGEST protocol, theAUR180deduced from digesting using either porcine pancreatin or α-amylase showed the best potential in predicting the eGI values. Accordingly, mathematical equations were established based on the relations between theAUR180and the GI values. When digesting using porcine pancreatin,GI=1.834 + 0.009×AUCR180(R2=0.952), and when digesting using only α-amylase,GI=6.101 + 0.009×AUCR180(R2=0.902). TheAUR180represents the area under the curve of the reducing-sugar content normalized to the total carbohydrates versus the digestion time in 180 min. The in vitro method presented enabled the rapid and accurate prediction of the eGI values of biscuits, and the validity of the formula was verified by another batch of biscuits with a known GI, and the error rate of most samples was less than 30%.Keywords:biscuits;in vitro digestion;INFOGEST;single enzyme;glycemic index

In Vitro Evaluation Reveals Effect and Mechanism of Artemether against Toxoplasma gondii

Qiong Xu,Yin-Yan Duan,Ming Pan,Qi-Wang Jin,Jian-Ping Tao,Si-Yang Huang

Due to the limited effectiveness of existing drugs for the treatment of toxoplasmosis, there is a dire need for the discovery of new therapeutic options. Artemether is an important drug for malaria and several studies have indicated that it also exhibits anti-T. gondiiactivity. However, its specific effect and mechanisms are still not clear. To elucidate its specific role and potential mechanism, we first evaluated its cytotoxicity and anti-Toxoplasmaeffect on human foreskin fibroblast cells, and then analyzed its inhibitory activity duringT. gondiiinvasion and intracellular proliferation. Finally, we examined its effect on mitochondrial membrane potential and reactive oxygen species (ROS) inT. gondii. The CC50value of artemether was found to be 866.4 μM, and IC50was 9.035 μM. It exhibited anti-T. gondiiactivity and inhibited the growth ofT. gondiiin a dose-dependent manner. We also found that the inhibition occurred primarily in intracellular proliferation, achieved by reducing the mitochondrial membrane integrity ofT. gondiiand stimulating ROS production. These findings suggest that the mechanism of artemether againstT. gondiiis related to a change in the mitochondrial membrane and the increase in ROS production, which may provide a theoretical basis for optimizing artemether derivatives and further improving their anti-Toxoplasmaefficacy.Keywords:Toxoplasma gondii;artemether;mitochondria;mechanism;in vitro

Anthelmintic efficacy of febantel against a monogenean parasite, Gyrodactylus kobayashii

Shun Zhou,Liwei Xia,Jing Dong,Yongtao Liu,Qiuhong Yang,Ning Xu,Yibin Yang,Xiaohui Ai

Monogenean parasites pose a significant challenge to aquaculture, leading to adverse effects on fish health and yields. Current anthelmintic treatments for monogeneans have demonstrated limited efficacy and are further complicated by potential issues, which emphasize the necessity for effective and safe therapeutic strategies to manage monogeneans in aquaculture. In this study, in vivo and in vitro anthelmintic activity of febantel against Gyrodactylus kobayashii on goldfish ( Carassius auratus ), as well as its toxicity to goldfish were evaluated. In vivo assays indicated that febantel exhibited potent anthelmintic activity against G. kobayashii with an EC 50 value of 0.03 mg/L and 100 % anthelmintic efficacy at 0.1 mg/L after 48 h of exposure. Moreover, in vivo trials also revealed a notable post-treatment effect of febantel, where infected goldfish transferred to drug-free water after short 6-h exposure could still result in full eradication of the worms, indicating febantel might induce persistent perturbations in parasite physiology. In vitro assays showed a negative correlation between febantel concentrations and the survival of G. kobayashii . However, increasing the febantel concentration to 2.0 mg/L did not result in the complete death of all worms. Oral administration of febantel demonstrated limited anthelmintic activity, with only 49 % efficacy at a dosage of 200 mg/kg body weight daily over five days. Acute toxicity assays revealed the 48-h LC 50 value of febantel was 5.47 mg/L, which was 182.23 times higher than the 48-h EC 50 value, indicating that febantel has a favorable safety profile. However, febantel exposure potentially interfered with hepatic metabolism and oxidative status, as indicated by variations in SOD , GST , and P450 gene expression. In conclusion, treatment with 0.1 mg/L febantel for 24 h completely eradicated G. kobayashii infection on goldfish, demonstrating febantel’s potent anthelmintic activity. Coupled with its safety profile and extended post-treatment effectiveness, febantel is a promising candidate for controlling Gyrodactylus infections in aquaculture.

Prebiotic properties of extruded maize starch-caffeic acid complexes: A study from the small intestine to colon in vitro

Yuxue Zheng,Xingqian Ye,Yanyu Hu,Shaoyun Wang,Jinhu Tian

Recent studies show that starch-polyphenols complexes exert positive effects on gut health, but the probiotic effects of maize starch-caffeic complexes remain underexplored. Therefore, this study aimed to investigate the probiotic effect of maize starch–caffeic complexes from the small intestine to the colon. First, maize starch was extruded with caffeic acid and subjected to in vitro digestion, and the undigested parts were fermented in vitro , and the structural characteristics, short chain fatty acids (SCFAs) and microbiota communities were investigated. Results showed that caffeic acid reduced the long/short-range order of maize starch after extrusion, significantly increasing resistant starch to 30.35 ± 2.36 %. In vitro fermentation indicated that microbiota utilized the amorphous area of the residues first, promoting SCFAs production and the growth of Bifidobacterium and Lactococcus genus. Overall, the probiotic properties of extruded maize starch–caffeic acid complexes suggest they could serve as a functional food for health benefits.
