Effects of glyphosate on survival, developmental rate, larval weight, and midgut bacterial diversity of Apis mellifera were tested in the laboratory. Larvae were reared in vitro and fed diet containing glyphosate 0.8, 4, and 20 mg/L. The dependent variables were compared with negative control and positive control (dimethoate 45 mg/L). Brood survival decreased in 4 or 20 mg/L glyphosate treatments but not in 0.8 mg/L, and larval weight decreased in 0.8 or 4 mg/L glyphosate treatments. Exposure to three concentrations did not affect the developmental rate. Furthermore, the intestinal bacterial communities were determined using high-throughput sequencing targeting the V3–V4 regions of the 16S rDNA. All core honey bee intestinal bacterial phyla such as Proteobacteria (30.86%), Firmicutes (13.82%), and Actinobacteria (11.88%) were detected, and significant changes were found in the species diversity and richness in 20 mg/L glyphosate group. Our results suggest that high concentrations of glyphosate are deleterious to immature bees.

BACKGROUND Obesity, a global chronic disease, has been recognized as a severe risk to health. In our study, a novel polysaccharide named ARS was isolated and purified from aerial part of salviae miltiorrhizae radix . Our aim is to investigate the weight-reducing effect of a polysaccharide from salviae miltiorrhizae radix on mice fed a high-fat diet. RESULTS The novel polysaccharide ARS mainly consisted of glucose and galactose with a molar ratio of 0.59:1.00. We found that treatment with ARS could inhibit weight gain of mice fed a high-fat diet via modulating the intestinal bacteria. Moreover, we surveyed its mechanism in mice, and the gut microbiota sequencing results demonstrated that ARS can reverse or resist high-fat-diet-induced significant weight gain or obesity by increasing the diversity of gut microbiota and optimizing the ratio of Firmicutes to Bacteroidetes. Phylum and species analysis of gut microbiota demonstrated that obesity caused by a high-fat diet was accompanied by significant changes in the microbial communities, but ARS could reverse the disturbance of gut microbiota induced by the high-fat diet to maintain homeostasis. CONCLUSION Overall, our findings suggested a new function of ARS in regulating gut microbiota, which provides a theoretical basis for the development of high-quality ARS functional foods and the application of dietary supplements. © 2023 Society of Chemical Industry.

Antibody-drug conjugate (ADC) plays a crucial role in the treatment of various diseases and intravenous injection is the primary administration route for ADCs due to its high bioavailability. However, this method requires drug preparation and dilution, which can compromise the stability of the solution and increase the risk of aggregation as excipient concentrations are reduced during dilution. Antibody drug dosing is often based on patient body weight and typically diluted in either 0.9% (w/v) sodium chloride or 5% (w/v) dextrose injections. Variations in patient body weight lead to differences in the final concentrations of both the drug and excipients, affecting stability. In this study, we examined the stability of intravenous dilutions for patients with varying body weights using trastuzumab and ado-trastuzumab emtansine ADCs. Analytical techniques including size-exclusion chromatography (SEC), fluid imaging microscopy (FIM), bicinchoninic acid (BCA) assay, and turbidity testing were employed to assess monomer, oligomer, and aggregate levels before and after dilution. Machine learning was applied to distinguish the morphology of sub-visible particles. Our findings reveal that weight-based dosing leads to significant stability differences in intravenous preparations. This study offers important insights into the formulation stability of biopharmaceuticals and their clinical use.

Oysters ( Crassostrea gigas ) are rich in proteins and a source of valuable peptides. Previous studies have revealed that oral administration of oyster peptides (OP) obtained by simulating gastrointestinal digestion reduced the weight of mice . However, the underlying regulatory mechanisms are unclear. We speculate that OP may be able to reduce fatty acid absorption by inhibiting pancreatic lipase activity. Therefore, we investigated the regulatory effects of OP on lipid metabolism in mice with high-fat diet-induced obesity. The body weight (BW), fat weight, lipid parameters, lipid metabolism-related gene expression, and serum lipid profiles were determined by MS-based lipidomics . Daily supplementation with 500 mg of OP/kg BW in obese mice reduced their BW gain and organ weights , improved dyslipidemia , and downregulated the genes expression of encoding proteins involved in fatty acid uptake , including acetyl coenzyme A carboxylase 1 ( Acc1 ), fatty acid synthase ( Fas ), and sterol regulatory element binding protein 1c ( Srebp-1c ). Moreover, administration of OP resulted in significant changes in the lipid profile of mice. We screened 129 differential metabolites to identify biomarkers for OP intervention and analyzed the effects of OP on the metabolism of sphingolipids , glycerophospholipids , linoleic acid , and α-linolenic acid. In summary, OP can improve obesity-induced lipid metabolism dysfunctions by inhibiting enzyme activity and regulating fatty acid uptake.

Unisexual animals are commonly found in some polyploid species complexes, and most of these species have had a long evolutionary history. However, their method for avoiding genomic decay remains unclear. The polyploid Carassius complex naturally comprises the sexual amphidiploid C. auratus (crucian carp or goldfish) (AABB) and the gynogenetic amphitriploid C. gibelio (gibel carp) (AAABBB). Recently, we developed a fertile synthetic amphitetraploid (AAAABBBB) male from C. gibelio by incorporating a C. auratus genome. In this study, we generated novel amphitriploids (AAABBB) by backcrossing the amphitetraploid male with the amphidiploid C. auratus. Whole-genome resequencing revealed the genomic changes, including recombination and independent assortment between homologs of C. gibelio and C. auratus. The fertility, sex determination system, oocyte development, and fertilization behaviors of the novel amphitriploids were investigated. Approximately 80% of the novel amphitriploid females recovered the unisexual gynogenesis ability. Intriguingly, two types of primary oocyte (with and without homolog synapsis) were discovered, and their distinct development fates were observed. Type I oocytes entered apoptosis due to improper synaptonemal complex assembly and incomplete double-strand break repair, whereas subsequent type II oocytes bypassed meiosis through an alternative ameiotic pathway to develop into mature eggs. Moreover, gynogenesis was stabilized in their offspring, and a new array of diverse gynogenetic amphitriploid clones was produced. These revealed genomic changes and detailed cytological data provide comprehensive evidence that changes in ploidy drive unisexual and sexual reproduction transition, thereby resulting in genomic diversity and allowing C. gibelio avoid genomic decay.

Background: Obesity and high fasting blood glucose (FBG) resulting from high-fat diets (HFDs) have emerged as significant public health concerns, garnering increasing attention. Recently, gut microbiota has been linked with metabolic diseases such as type 2 diabetes (T2DM), and its mediating role in dietary supplements has been confirmed. Seeking various dietary supplements to lose body weight (BW) and decrease FBG and explaining the underlying mechanism have become the research hotspots in T2DM studies. Methods: In this study, rutin and proanthocyanidins (PA) were selected as dietary supplements (200 mg/kg × day, oral gavage, 6 weeks) in T2DM mice induced with HFD to assess their efficacy in weight loss, FBG reduction, gut microbiota alterations, and the associated underlying mechanisms. Results: Our findings indicate that rutin was more effective than PA in relieving inflammation and fat hypertrophy, although both significantly reduced BW and FBG within 2 weeks after the intervention. Analysis of 16S rRNA amplicons revealed substantial alterations in the gut microbial community composition of mice administered with PA and rutin compared to HFD-fed mice. Importantly, several core microbes, particularly a series of probiotics, such as Akkermansia, Lactococcus, Odoribacter, Faecalibaculum, and Roseburia were identified, which were significantly correlated with the changes in BW and FBG.Conclusion: Overall, our study highlights that rutin and PA can reduce BW, FBG, and inflammation by modulating the gut microbiota composition, providing novel perspectives for managing and treating weight and FBG concerns in obesity and T2DM patients through dietary supplements in clinical treatment.

Organometal halide perovskite single crystals (PSCs) have drawn intensive attention in X-ray detection due to their outstanding optoelectronic properties. However, they currently suffer from a short lifespan caused by a phase transition or water and oxygen erosion. Sustainably recycling end-of-life PSCs is essential for reducing environmental impact and saving costs on raw materials and time. Herein, we propose a “trash to treasure” economically recycling strategy, which utilizes ammonium salt-assisted octahedral [PbI6]4– reconstruction and rearrangement to in situ treat various degraded PSCs. X-ray detectors based on the recycled PSCs demonstrate significantly enhanced performances comparable to those of previously reported fresh PSCs. For example, the recycled FAPbI3 detectors show a high sensitivity of 4.32 × 104 μC Gyair–1 cm–2 and a low limit of detection of 8.3 nGyair s–1, along with a high spatial resolution of 7.0 lp mm–1. Furthermore, this method has also been successfully employed to construct 2D/3D heterojunctions. This method offers universality, effectiveness, and simplicity, thereby inspiring the recycling and reuse of degraded perovskites.

Pancreatic malignancies rank among the most aggressive and fatal carcinomas in modern oncology, presenting significant therapeutic challenges due to their inherent chemoresistance. In the context of our experimental investigations, we have developed a mitochondria-targeted hydrogel formulation, termed SWMU690@PUE, which demonstrates the ability to modulate mitochondrial homeostasis effectively. The hydrogel construct was precisely designed via the spontaneous supramolecular assembly of puerarin (PUE) and SWMU690 components, facilitated under carefully controlled thermodynamic conditions. Comprehensive in vitro analyses of the SWMU690@PUE complex were conducted to rigorously evaluate its efficacy in regulating mitochondrial homeostasis. The experimental findings revealed that SWMU690@PUE exhibited pronounced selectivity for mitochondrial targeting, subsequently diminishing mitochondrial membrane potential—a pivotal parameter in cellular bioenergetics. Our study further demonstrated that SWMU690@PUE effectively mitigates the chemoresistance phenotype associated with mitochondrial dysfunction. This therapeutic potential was exemplified by its capacity to induce mitochondria-mediated apoptotic and ferroptotic cell death, critical oncocidal pathways, in pancreatic carcinoma cells across both in vitro and in vivo models. Additionally, SWMU690@PUE displayed significant antineoplastic activity, as evidenced by its robust inhibition of pancreatic cancer cell proliferation in both experimental frameworks. In conclusion, this study presents an innovative therapeutic approach specifically designed to overcome the persistent challenge of chemoresistance in the treatment of pancreatic neoplasms.

Mn4+-activated fluoride red phosphors exhibit excellent luminescence properties. However, a persistent technical challenge lies in their poor moisture resistance. Current strategies primarily focus on surface modifications to effectively shield the [MnF6]2– species from water molecules while neglecting the underlying structure of the fluoride matrix. In this study, we introduce Si4+ and Ge4+ ions into the K2TiF6:Mn4+ crystal to create covalent fluoride solid solutions, namely, K2Ti1–xSixF6:Mn4+ and K2Ti1–yGeyF6:Mn4+, through crystal reconstruction. The findings reveal that the incorporation of Si4+ leads to increased particle size, enhanced luminescence intensity (by 40%), and improved moisture resistance. Furthermore, after undergoing 1000 h of aging at high temperature and high humidity conditions, the white LED featuring the K2Ti0.97Si0.03F6:Mn4+ phosphor demonstrates remarkable durability by retaining 90% of its initial luminous efficacy. This performance surpasses that of the device utilizing the K2TiF6:Mn4+ phosphor, which only retains 74% of its original efficacy. The crystal reconstruction method and covalent enhancement strategy proposed in this work contribute to enhancing the luminescence efficiency and moisture resistance of fluoride phosphors, thereby offering new insights for advancing the development of high-efficiency and highly stable white light LED devices.

Polysaccharide-derived functional food ingredients are increasingly prevalent in the food industry for their health benefits and functional versatility. Among them, sodium alginate (SA), a natural polysaccharide, is widely valued for its sustainability, renewability, non-toxic nature, and broad applicability. However, its poor water solubility has limited its use as biomedical materials and food additives. Here, we enhanced SA through carboxymethylation to produce carboxymethylated SA (CMSA), aiming to develop functional food ingredients with improved physicochemical properties for health management. The synthesized CMSA was characterized for its physical properties and ability to form a hydrogel through cross-linking with Ca 2+ . We assessed its encapsulation efficiency for food particles in simulated gastric and intestinal fluids and evaluated its physiological effects in a rat model. Our findings demonstrated that CMSA-based hydrogels effectively encapsulate ingredients in the stomach, reducing nutrient diffusion in the intestine, and helping to manage postprandial hypoglycemia. Additionally, the hydrogel expands in the stomach and small intestine, contributing to modest weight loss. These findings suggest that CMSA-based hydrogels offer significant potential as functional food ingredients for managing postprandial hypoglycemia and supporting weight management. The development of such gelling systems presents promising applications in both medical and nutritional sciences, offering innovative strategies for addressing diet-related health concerns.

The modification of inert boron nitride by carbon doping to make it an efficient photocatalyst has been considered as a promising strategy. Herein, a highly efficient porous BCN (p-BCN) photocatalyst was synthesized via precursor reconfiguration based on the recrystallization of a new homogeneous solution containing melamine diborate and glucose. Two crystal types of the p-BCN were obtained by regulating the recrystallization conditions of the homogeneous solution, which showed high photocatalytic activities and a completely different CO2reduction selectivity. The CO generation rate and selectivity of the p-BCN-1 were 63.1 μmol·g−1·h−1and 54.33%; the corresponding values of the p-BCN-2 were 42.6 μmol·g−1·h−1and 80.86%. The photocatalytic activity of the p-BCN was significantly higher than those of equivalent materials or other noble metals-loaded nanohybrids reported in the literature. It was found that the differences in the interaction sites between the hydroxyl groups in the boric acid and the homolateral hydroxyl groups in the glucose were directly correlated with the structures and properties of the p-BCN photocatalyst. We expect that the developed approach is general and could be extended to incorporate various other raw materials containing hydroxyl groups into the melamine diborate solution and could modulate precursors to obtain porous BN-based materials with excellent performance.Keywords:carbon-doped BN;homogeneous;precursor reconfiguration;photocatalysis;selectivity

Decabromodiphenyl ethane (DBDPE) as a novel brominated flame retardant is frequently detected in environmental media due to its widespread use. Studies have shown that exposure to environmental pollutants in utero could lead to weight loss in newborns and obesity in adulthood. However, the mechanisms of how the cubs grow rapidly from low birth weight to obesity remain unclear. Although it has been reported that perinatal DBDPE exposure caused obesity in the offspring of mice in adulthood, its metabolic changes in offspring juvenile are unknown. Here, we monitored changes of body weight in cubs following exposure to DBDPE in utero. Furthermore, 1 H NMR-based metabolomics was used to investigate the effects of in utero DBDPE exposure on the metabolic profile of neonates included days 1 and 25. Additionally, the expression levels of key genes in the relevant pathways were quantified. The results showed that exposure to high levels of DBDPE in utero caused cubs to be smaller at birth and grow rapidly during lactation. Metabolomics analysis showed that cubs in the exposed group consistently accelerated glycogenolysis and lipid synthesis to promote appetite and energy absorption, and achieved rapid growth in the subsequent lactation, which was further evidenced by the expression levels of genes related to glycolipid metabolism. These results reveal the mechanism for the first time how the weight loss of newborns in utero environmental pollutant exposure transformed into obesity in adulthood, although the exposures here were much higher than would be anticipated from the environment.
