Here, hydro/solvent thermal method was applied to synthesize lanthanide doped NaYF 4 upconversion nanoparticles (UCNPs). In order to overcome the quenching effect of (CH 2 ) n due to their high energy stretching vibration , a series of new chelating ligands such as, butanoic acid , hexanoic acid , decanoic acid , and tetradecanoic acid were selected for hydrothermal synthesis of NaYF 4 :Yb, Er nanocrystals . For comparison, commonly used oleic acid was also used as ligand for hydrothermal synthesis of UCNPs. The effect of the carbon chain length of chelating ligands on the NaYF 4 nanocrystals’ emission intensity was systematically investigated. It was found that relative short length of the carbon chain in chelating ligands resulted in enhancement of luminescent intensity and modulation of the ratios of green to red (RGR) of NaYF 4 :Yb, Er nanocrystals. In contrast to the product prepared with oleic acid as ligand, the enhancement factors of 540 nm emissions were 25, 21, 6.5, and 3 for samples prepared with butanoic acid, hexanoic acid, decanoic acid, and tetradecanoic acid as ligands, respectively. The FT-IR spectra of NaYF 4 :Yb, Er UCNPs prepared with different ligands indicated that ligands with relatively short chain length indeed have weak IR absorption in 2800–2950 cm −1 , which resulted in weak quenching effect of Er 3+ . We also synthesized NaYF 4 :Yb, Tm UCNPs with these ligands. The results showed that chain length of ligands has similar effect on emission intensity of NaYF 4 :Yb, Tm. It is expected that this method can also be applied to enhance the luminescence intensity of other luminescent materials.

Simple SummaryMyostatin is a negative regulator of muscle growth and development, and mutational inactivation of this gene increases muscle mass. Knockout of themstnbgene inPelteobagrus fulvidracodid not increase muscle mass, although our previous study of themstnagene did. Instead, it reduced the body size and growth performance ofPelteobagrus fulvidraco. Through the present study, we found that the mutantPelteobagrus fulvidracohad a reduction in the number of vertebrae, the length, and the intervertebral distance inPelteobagrus fulvidraco, and these changes may be the underlying cause of the shorter body length in mutantPelteobagrus fulvidraco. Histological comparison of the same sites in themstnmutant and wild groups ofPelteobagrus fulvidracoalso revealed that the number and density of osteocytes were greater inmstnbknockoutPelteobagrus fulvidracothan in wild-typePelteobagrus fulvidraco.AbstractBased on obtainingmstnbgene knockout inPelteobagrus fulvidraco, a study on the effect of themstngene on skeletal morphology and growth was performed by comparing the number and length of the vertebrae of mutant and wild-type fish in a sibling group ofP. fulvidraco, combined with the differences in cells at the level of vertebral skeletal tissue. It was found thatmstnbgene knockdown resulted in a reduction in the number of vertebrae, the length, and the intervertebral distance inP. fulvidraco, and these changes may be the underlying cause of the shorter body length in mutantP. fulvidraco. Further, histological comparison of the same sites in themstnmutant and wild groups ofP. fulvidracoalso revealed that the number and density of osteocytes were greater inmstnbknockoutP. fulvidracothan in wild-typeP. fulvidraco. Our results demonstrated that when using genome editing technology to breed new lines, the effects of knockout need to be analyzed comprehensively and may have some unexpected effects due to insufficient study of the function of certain genes. Keywords:myostatin b;genome editing;Pelteobagrus fulvidraco;body length;skeleton;vertebral centrum

The selective deposition of noble metals on catalysts is an attractive modification method for improving catalytic efficiency. However, it is sometimes hard to achieve when facet energies of nano-catalysts show negligible differences. Here, we reported a piezoelectrochemical method which can realize the selective deposition of Ag nanoparticles on the positively polar end of {001}-enclosed BaTiO 3 (BTO) nanocubes/cuboids. Furthermore, BTO nanocubes/cuboids with selectively-deposited Ag nanoparticles show approximately 2 times higher piezocatalytic activity than those with randomly-loaded Ag nanoparticles, and much higher than pure BTO nanocubes/cuboids. The piezocatalytic mechanism revealed that the Ag nanoparticles deposited on positively polar end act as “fast lanes” for electrons to transfer to catalysts/solution interfaces, while those loaded on negatively polar end serve as holes trappers hindering ∙OH formation and pollutant degradation. This work confirms an efficient way to improve piezocatalytic performance and provides an insightful discussion of piezocatalytic mechanism.

Camphor is a terpene ketone with aromatic and volatile properties in nature derived from the bark of Cinnamomum camphora or synthesized from turpentine. Camphor exhibits various biological properties such as anti-microbial, anti-viral, anti-coccidial, and anti-cancer. It is also used as a form of topical medication for skin irritation, joint pain, and as a relief for itching from insect bites. However, even though the high dose of camphor has been documented to be toxic/lethal in humans in different studies, camphor’s developmental toxicity has not yet been explored, and its extensive mechanism of action is still unclear. In the present study, we aimed to assess the toxic effects of camphor in zebrafish embryos in the initial developmental stages. The obtained results demonstrated that a sub-lethal dose of camphor caused a decrease in hatching rate, body length, and substantial elevation in malformation rate on zebrafish embryos. On further observation, in the following time frame, curved body and pericardial edema of zebrafish were also observed. Furthermore, exposure to a sub-lethal dose of camphor was also able to trigger cardiotoxicity in zebrafish larvae. Later, on subsequent biochemical analysis, it was found that the antioxidant capacity inhibition and oxidative stress elevation that occurred after camphor exposure might be associated with the inhibition of total superoxide dismutase (SOD) activity and an increase in reactive oxygen species (ROS) and malondialdehyde (MDA) concentration. In addition, compared to the control group, several apoptotic cells in treated zebrafish were also found to be elevated. Finally, after further investigation on marker gene expressions, we conclude that the developmental toxicity of camphor exposure might be associated with apoptosis elevation and oxidative stress. Taken together, the current study provides a better understanding of the developmental toxicity of camphor on zebrafish, a promising alternative animal model to assess the developmental toxicity of chemical compounds.

In the tumor microenvironment (TME), tumor associated macrophages (TAMs) often exhibit a preference for M2-like phenotype, which supports tumor progression under hypoxia. The underlying mechanisms driving the behavior of hypoxic TAMs are highly complex. Recently, exosomal long non-coding RNAs (lncRNAs) derived from non-cancerous cells within TME have attracted more attention from researchers. In the present study, we investigated the function of exosomal lncRNA MIR210HG derived from hypoxic TAMs in triple negative breast cancer (TNBC). Results showed that MIR210HG was significantly upregulated in hypoxic TAMs, their exosomes, and exosomes-treated TNBC cells. When MIR210HG was suppressed, the expression of M2-polarization markers and proteins associated with the PI3K/Akt/mTOR pathway in hypoxic TAMs was notably inhibited. Moreover, the suppression of TAM-derived exosomal MIR210HG effectively inhibited the migration, invasion and vasculogenic mimicry (VM) formation of TNBC cells. Notably, dual inhibition of both exosomal and endogenous MIR210HG in tumor cells synergistically restricted TNBC metastasis of in vivo . Mechanistically, we elucidated that MIR210HG promoted the malignant phenotypes of TNBC by upregulating the expression of HIF-1α protein, which in turn initiated RASSF7 transcription, resulting in the promotion of epithelial-mesenchymal transition (EMT) process. Collectively, our results identified MIR210HG as a key molecule mediating exosome-based communication from hypoxic TAMs to TNBC cells and revealed its pro-metastatic role, Consequently, MIR210HG emerges as a highly promising therapeutic target of TNBC.

Gabapentin (GAB) is an emerging contaminant that is frequently detected in water bodies across the globe. The present study used zebrafish as a model organism to investigate the effects of GAB on the early development of zebrafish and on its antioxidant system. Acute toxicity tests indicated that the 96 h LC50 value of GAB for zebrafish embryos was 59.9 g L−1. Further, it was observed that GAB causes malformation of embryos such as hemagglutination and pericardial edema. Compared to the control group, a significant enhancement (p < 0.05) of heartbeat rates was found at GAB concentrations exceeding 50 mg L−1, while the swimming frequency was clearly increased upon exposure to GAB at a concentration of 100 mg L−1 (p < 0.05). Additionally, the development of the zebrafish embryo was also negatively impacted after exposure to GAB as demonstrated by significantly decreased body lengths. Exposure to GAB at concentrations exceeding 50 mg L−1 significantly influenced the development of zebrafish, leading to malformation of organs and abnormal movements. Although no significant developmental effects of GAB were observed at environmentally relevant concentrations (0.1 and 10 μg L−1), further research about the antioxidant system confirmed that severe oxidant injury happened inside the organisms. catalase (CAT), lactate dehydrogenase (LDH), glutathione S-transferase (GST), glutathione (GSH) and the ability of inhibition of hydroxyl radicals (IHR) were used as biomarkers in the present study to illustrate GAB toxicity at environmentally relevant concentrations. The results showed that activities of CAT, LDH and GST as well as IHR were all elevated after GAB exposure, which proved that ROS were formed in the body as derived from GAB exposure. Among all of these biomarkers, CAT was the most sensitive one to evaluate the influence of GAB, and showed a significant increase even at a very low exposure concentration (0.1 μg L−1).

Platinum nanoparticles were prepared by sodium borohydride reduction of Pt(II) precursors and stabilized by the self-assembly of acetylene derivatives of different lengths (phenylacetylene, 4-ethylphenylacetylene, and 4-butylphenylacetylene). Transmission electron microscopic measurement showed that these nanoparticles exhibited similar average core sizes of 2.22 ± 0.83 nm, 2.09 ± 0.91 nm, and 2.18 ± 0.77 nm, respectively. FTIR study confirmed that the Pt cores were capped by these acetylene ligands with the breaking of the terminal C H vibrational stretch and a substantial red-shift of the C C vibration. X-ray photoelectron measurements showed that platinum is in the metallic form. Pt nanoparticles all exhibited typical exponential decay profiles with some difference in small absorption peaks before 360 nm in UV–vis measurements. In ( αhv ) 1/2 versus hv plot, band gap decrease is related with the increase of ligand chain length. The Pt nanoparticles all showed apparent photoluminescence with a clear red-shift of the excitation and emission peak positions as compared to those of the free monomers , due to intraparticle charge delocalization between the particle-bound acetylene moieties. Electrochemical measurements showed that the nanoparticles exhibited electrocatalytic activity towards the oxidation of methanol and hydroquinone oxidation, and the activity was found to increase with increasing ligand chain length.

Corn oil, sodium carboxymethyl cellulose (CMC-Na), and dimethyl sulfoxide (DMSO) are widely used as solvents or suspensions in animal experiments, but the effects of prenatal exposure to them on fetal development have not been reported. In this study, Kunming mice were given a conventional dose of corn oil (9.2 g/kg·d), CMC-Na (0.05 g/kg·d) or DMSO (0.088 g/kg·d) during gestation days 10–18, and the pregnancy outcome, fetal physical development, serum phenotype, and multi-organ function changes were observed. The results showed that corn oil decreased serum triglyceride level in males but increased their serum testosterone and CORT levels, and affected female placenta and female/male multi-organ functions (mainly bone, liver, kidney). CMC-Na increased female/male body lengths and tail lengths, decreased serum glucose and total cholesterol levels in males as well as increased their serum LDL-C/HDL-C ratio and testosterone level, decreased female serum bile acid level, and affected male/female placenta and multi-organ functions (mainly bone, liver, hippocampus). DMSO decreased male body weight and serum glucose level, decreased male/female serum bile acid levels, and affected male/female placenta and multi-organs functions (mainly bone, hippocampus, adrenal gland). In conclusion, prenatal exposure to a conventional dose of corn oil, CMC-Na or DMSO could affect fetal physical development and multi-organ functions, and has the characteristics of "multi-pathway, multi-organ and multi-target". This study provides the experimental basis for the rational selection of solvents or suspensions in pharmacology and toxicology studies.
