The ideal treatment for myocardial infarction should address the critical issues of oxygen supply improvement in the infarcted area and prevention of myocardial cell death. Myocardial necrosis is a reversible process, but the challenge lies in mitigating the irreversible pyroptosis of cardiomyocytes, which significantly impedes the recovery of cardiac function. To tackle these complexities, here we have developed MnO2@INN@Alb (MIA) nanoparticles. MIA nanoparticles possess the unique ability to generate oxygen and release glibenclamide (INN) in response to the acidic and H2O2-rich environments present in the infarcted region. MIA plays a dual role by inhibiting the pyroptosis process of cardiomyocytes through the suppression of NLRP3 and by providing vital oxygen to alleviate hypoxia. The results shows that MIA can effectively reverse cardiomyocyte hypoxia, reduce infarct fibrosis, and aid in the reconstruction of myocardial vessels. Moreover, MIA demonstrates high biocompatibility, making it a promising avenue for the future of myocardial infarction improvement. We anticipate that MIA can pave the way for novel solutions and research paradigms in the field.

Oral biofilm is the leading cause of dental caries, which is difficult to completely eradicate because of the complicated biofilm structure. What's more, the hypoxia environment of biofilm and low water-solubility of conventional photosensitizers severely restrict the therapeutic effect of photodynamic therapy (PDT) for biofilm. Although conventional photosensitizers could be loaded in nanocarriers, it has reduced PDT effect because of aggregation-caused quenching (ACQ) phenomenon. In this study, we fabricated an oxygen self-sufficient nanodroplet (PFC/TPA@FNDs), which was composed of fluorinated-polymer (FP), perfluorocarbons (PFC) and an aggregation-induced emission (AIE) photosensitizer (Triphenylamine, TPA), to eradicate oral bacterial biofilm and whiten tooth. Fluorinated-polymer was synthesized by polymerizing (Dimethylamino)ethyl methacrylate, fluorinated monomer and 1-nonanol monomer. The nanodroplets could be protonated and behave strong positive charge under bacterial biofilm acid environment promoting nanodroplets deeply penetrating biofilm. More importantly, the nanodroplets had extremely high PFC and oxygen loading efficacy because of the hydrophobic affinity between fluorinated-polymer and PFC to relieve the hypoxia environment and enhance PDT effect. Additionally, compared with conventional ACQ photosensitizers loaded system, PFC/TPA@FNDs could behave superior PDT effect to ablate oral bacterial biofilm under light irradiation due to the unique AIE effect. In vivo caries animal model proved the nanodroplets could reduce dental caries area without damaging tooth structure. Ex vivo tooth whitening assay also confirmed the nanodroplets had similar tooth whitening ability compared with commercial tooth whitener H 2 O 2 , while did not disrupt the surface microstructure of tooth. This oxygen self-sufficient nanodroplet provides an alternative visual angle for oral biofilm eradication in biomedicine.

Anticancer treatment is largely affected by the hypoxic tumor microenvironment (TME), which causes the resistance of the tumor to radiotherapy. Combining radiosensitizer compounds and O2self-enriched moieties is an emerging strategy in hypoxic-tumor treatments. Herein, we engineered GdW10@PDA-CAT (K3Na4H2GdW10O36·2H2O, GdW10, polydopamine, PDA, catalase, CAT) composites as a radiosensitizer for the TME-manipulated enhancement of radiotherapy. In the composites, Gd (Z = 64) and W (Z = 74), as the high Z elements, make X-ray gather in tumor cells, thereby enhancing DNA damage induced by radiation. CAT can convert H2O2to O2and H2O to enhance the X-ray effect under hypoxic TME. CAT and PDA modification enhances the biocompatibility of the composites. Our results showed that GdW10@PDA-CAT composites increased the efficiency of radiotherapy in HT29 cells in culture. This polyoxometalates and O2self-supplement composites provide a promising radiosensitizer for the radiotherapy field.Keywords:radiosensitizer;polyoxometalates;catalase;O2self-supplement;radiotherapy

Biofilms are responsible for most bacterial infections and significantly threaten human health. Designing effective and less harmful materials to inhibit and remove biofilms remains a significant challenge. Complex biofilm structures make conventional antibiotic treatments ineffective, and in recent years, photothermal therapy (PTT) and photodynamic therapy (PDT) have attracted attention for their ability to treat drug-resistant bacterial infections effectively. Therefore, a synergistic nanoantimicrobial system (PFH/TCPP@SL@PDA) enhances the effect through PTT and PDT. It is synthesized by loading meso-tetra(4-carboxyphenyl) porphine (TCPP) on soybean lecithin (SL), further coating perfluorohexane (PFH) and finally coating polydopamine (PDA) on the outer layer. The designed nanoantimicrobial system alleviates the hypoxic properties of the biofilm microenvironment by loading oxygen and enhances the effect of PDT to generate more reactive oxygen species. It significantly enhanced biofilm permeability and improved antimicrobial properties, with over 95% inhibition of multidrug-resistant (MDR) Escherichia coli and MDR Staphylococcus aureus while inhibiting and removing over 90% of biofilms. PDA not only acts as a good nanoshell but also induces relatively mild PTT and can facilitate the reaction of PDT. PFH/TCPP@SL@PDA has good biocompatibility, and the hemolysis test showed less than 5% hemolysis of erythrocytes, effectively relieving inflammatory symptoms and accelerating wound healing in mice model. In conclusion, this study provides an efficient and safe combined therapeutic strategy for the bacterial inhibition, suppression, and removal of biofilms.

Limited cells and growth factors, insufficient mechanical properties, and the hypoxic status in defect areas are the severe challenges faced by bone tissue engineering (BTE) scaffolds, currently. In this work, a 3D printable composite scaffold (Gel-MCM-41-CaO 2 ) without cells or growth factors is designed to promote vascularized bone regeneration through sustained release of oxygen and bioactive ions, utilizing a 1D tube encapsulation release technology to achieve slow release of oxygen from both ends of MCM-41-CaO 2 . Calcium silicate is used as a cross-linking agent and a bioactive agent to cross-link with sodium alginate, combined with polyacrylamide to form a hybrid double network, which endows the hydrogel with excellent compressive strength and 3D printable properties. Based on amounts of data in vitro and in vivo, the oxygen and silicon ions released from this hydrogel scaffold can promote cell proliferation and differentiation, upregulate the expression of osteogenic and angiogenic-related genes, and promote angiogenesis and osteogenesis. This work presents a strategy for fabricating BTE scaffolds without the need for cells or growth factors, with the potential to be applied in clinical bone regeneration and repair.

Periodontitis, the chronic inflammation of the periodontal tissues caused by bacteria in plaque, is the leading cause of tooth loss in adults in the world. Currently, periodontitis is effectively treated with mechanical cleaning and the use of antibiotics. However, these treatments only temporarily remove plaque, which can rapidly proliferate and multiply in periodontal pockets over time. Although antibiotics have positive antimicrobial effects, their long-term use increases the risk of the emergence of drug-resistant strains. The emergence of resistant strains reduces the effectiveness of periodontitis treatment and makes the disease more difficult to control. Herein, this paper reports the development of an injectable self-oxygenating composite hydrogel for periodontal therapy, which was produced by loading CaO2 nanoparticles and ascorbic acid into an injectable alginate hydrogel. CaO2 can improve the periodontal pocket microenvironment by reacting with water to generate oxygen, calcium ions can be used as a bone regeneration material, and ascorbic acid protects cells. The authors further showed that the composite hydrogel inhibited growth and colonization of anaerobic bacteria, reduced the degree of inflammation, and promoted alveolar bone regeneration. In conclusion, these findings suggest that the composite hydrogel can be used as a biocompatible, convenient, and effective method for periodontitis treatment.

Background The critical issue in the competitiveness between bioengineering and chemical engineering is the products titer and the volume productivity. The most direct and effective approach usually employs high-density biocatalyst, while the weakened mass transfer and evoked foam problem accompany ultrahigh-density biocatalyst loading and substrate/product titer. In high-density obligate aerobic bioconversion, oxygen as electron acceptor is a speed-limiting step in bioprocesses, but sufficient oxygen supply will lead to the foaming which results in a significant reduction in oxygen utilization and the use of additional defoamers. In this study, we designed a novel sealed-oxygen supply (SOS) biotechnology to resolve the formidable barrier of oxygen transferring rate (OTR), for bio-based fuels and chemical production process. Results Based on systemic analysis of whole-cell catalysis in Gluconobacter oxydans, a novel sealed-oxygen supply technology was smartly designed and experimentally performed for biocatalytic oxidation of alcohols, sugars and so on. By a simple operation skill of automatic online supply of oxygen in a sealed stirring tank bioreactor of SOS, OTR barrier and foaming problem was resolved with great ease. We finally obtained ultrahigh-titer products of xylonic acid (XA), 3-hydroxypropionic acid (3-HPA), and erythrulose at 588.4 g/L, 69.4 g/L, and 364.7 g/L, respectively. Moreover, the volume productivity of three chemical products was improved by 150–250% compared with normal biotechnology. This SOS technology provides a promising approach to promote bioengineering competitiveness and advantages over chemical engineering. Conclusion SOS technology was demonstrated as an economic and universally applicable approach to bio-based fuels and chemicals production by whole-cell catalysis. The novel technology greatly promotes the competitiveness of bioengineering for chemical engineering, and provides a promising platform for the green and environmental use of biofuels.

Transplantation-related bacterial infections often slow new tissue regeneration and even cause transplant failure. Sonodynamic therapy is a promising antibacterial method, which kills bacteria by triggering sonosensitizers to generate reactive oxygen species (ROS). Nevertheless, the generation efficiency of ROS is hindered by the fast electron-hole pair recombination in sonosensitizers and the hypoxic of bacteria microenvironment. Herein, ruthenium nanoparticles (Ru NPs) are grown in-situ on the oxygen vacancies-rich barium titanate nanoparticles (BTOv NPs) to form a Schottky heterojunction. The resulting BTOv-Ru NPs as sonosensitizers is uniformly distributed with poly-L-lactic acid (PLLA) powders to fabricate ultrasound-responsive scaffolds via selective laser sintering. On the one hand, the Schottky heterojunction facilitates electron injection from BTOv to Ru, while the resultant charge separation significantly suppresses electron-hole recombination. On the other hand, the catalase-like activity of Ru catalyzes the overexpressed hydrogen peroxide to generate O 2 to achieve O 2 self-supply. This not only alleviates hypoxia but also facilitates the oxygen vacancies of BTOv adsorb more O 2 to generate ROS. The results confirmed PLLA/BTOv-Ru scaffold generated numerous ROS and exhibited excellent antibacterial effects under ultrasonic irradiation, with inhibiting E. coli (89.3 %) and S. aureus (88.1 %). This O 2 self-supplying sonodynamic therapy shows great promise in antibacterial treatment.

Diabetic foot ulcers (DFUs), a serious and increasingly common chronic issue among diabetics, often do not respond well to generalized treatment strategies. Hypoxia and the overexpression of reactive oxygen species (ROS), resulting in inflammatory dysregulation and subsequent imbalance in macrophage phenotypes, are critical factors contributing to the prolonged non-healing of DFU wounds. These two issues often interact in a continuous, problematic cycle. Presently, there is a lack of comprehensive strategies aimed at addressing both of these factors simultaneously to interrupt this detrimental cycle. Herein, an immunomodulatory hydrogel (PHG2) is developed for reshaping the hostile DFU microenvironment. The engineered PHG2 not only removes excess internally-produced ROS but also generates O 2 , with its efficiency further boosted by local hyperthermia (42.5 °C) activated by near-infrared light. Through both in vitro and in vivo studies, along with transcriptomic assessment, it is confirmed that PHG2 disrupts the detrimental feedback loop between ROS and inflammation while also lowering the M1/M2 macrophage ratio. Such discoveries contribute to a significant enhancement in the healing process of injuries that undergo a gradual increase in movement, covering wounds from the back, mouth, to the foot. Ultimately, this method provides an easy, safe, and highly effective solution for treating DFUs.

In this work, tandem oxygen delivery systems were built by coating cube-like Co 3 O 4 with Ag/CeO 2 , which exhibited high soot oxidation activity that overwhelmed the Fe 2 O 3 -based catalysts. The results of TPR and activity tests indicated that a large amount of Co 3 O 4 lattice oxygen could be transferred into O x − with the assistance of Ag/CeO 2 . These active oxygen species accelerated the oxidation of soot and NO at low temperature. Furthermore, the Co 3 O 4 -CeO 2 synergy facilitated Co 3+ → Co 2+ transformation during reactions, resulting in boosted NO conversion and thereby NO 2 -assisted soot combustion at mild and high temperatures. Given the Ag/Co 3 O 4 @CeO 2 -type catalysts were low-cost, hydrothermally stable and could be fabricated via co-precipitation methods easily, they were highly promising for application in both gasoline and diesel particulate filters.

Large bone defects resulting from fractures and disease are a major clinical challenge, being often unable to heal spontaneously by the body's repair mechanisms. Lines of evidence have shown that hypoxia-induced overproduction of ROS in bone defect region has a major impact on delaying bone regeneration. However, replenishing excess oxygen in a short time cause high oxygen tension that affect the activity of osteoblast precursor cells. Therefore, reasonably restoring the hypoxic condition of bone microenvironment is essential for facilitating bone repair. Herein, we designed ROS scavenging and responsive prolonged oxygen-generating hydrogels (CPP-L/GelMA) as a “bone microenvironment regulative hydrogel” to reverse the hypoxic microenvironment in bone defects region. CPP-L/GelMA hydrogels comprises an antioxidant enzyme catalase (CAT) and ROS-responsive oxygen-releasing nanoparticles ( [email protected] /PPS) co-loaded liposome (CCP-L) and GelMA hydrogels. Under hypoxic condition, CPP-L/GelMA can release CAT for degrading hydrogen peroxide to generate oxygen and be triggered by superfluous ROS to continuously release the oxygen for more than 2 weeks. The prolonged oxygen enriched microenvironment generated by CPP-L/GelMA hydrogel significantly enhanced angiogenesis and osteogenesis while inhibited osteoclastogenesis. Finally, CPP-L/GelMA showed excellent bone regeneration effect in a mice skull defect model through the Nrf2-BMAL1-autophagy pathway. Hence, CPP-L/GelMA, as a bone microenvironment regulative hydrogel for bone tissue respiration, can effectively scavenge ROS and provide prolonged oxygen supply according to the demand in bone defect region, possessing of great clinical therapeutic potential.

Background Colorectal cancer (CRC) is the second most deadly cancer worldwide, with chemo-resistance remaining a major obstacle in CRC treatment. Notably, the imbalance of redox homeostasis-mediated ferroptosis and the modulation of hypoxic tumor microenvironment are regarded as new entry points for overcoming the chemo-resistance of CRC.
Methods Inspired by this, we rationally designed a light-activatable oxygen self-supplying chemo-photothermal nanoplatform by co-assembling cisplatin (CDDP) and linoleic acid (LA)-tailored IR820 via enhanced ferroptosis against colorectal cancer chemo-resistance. In this nanoplatform, CDDP can produce hydrogen peroxide in CRC cells through a series of enzymatic reactions and subsequently release oxygen under laser-triggered photothermal to alleviate hypoxia. Additionally, the introduced LA can add exogenous unsaturated fatty acids into CRC cells, triggering ferroptosis via oxidative stress-related peroxidized lipid accumulation. Meanwhile, photothermal can efficiently boost the rate of enzymatic response and local blood flow, hence increasing the oxygen supply and oxidizing LA for enhanced ferroptosis.
Results This nanoplatform exhibited excellent anti-tumor efficacy in chemo-resistant cell lines and showed potent inhibitory capability in nude mice xenograft models. Conclusion sTaken together, this nanoplatform provides a promising paradigm via enhanced ferroptosis and alleviated hypoxia tumor microenvironment against CRC chemo-resistance.Graphical Abstract
