Application of Additives in Platycladus orientalis (L.) Franco Tending Shreds Compost in Forest

Xuan Li,Huijuan Bo,Jialei Zhu,Jiwei Zhang,Jun Hu,Fuyong Mu,Lishui Nie

This study aimed to explore the effects of different additives on tending shreds ofPlatycladus orientalis(L.) Franco. Two different additives (priming 0.2% and common compost 0.2%) combined with C, N, and P adjustment of raw material treatments were tested on the temperature, moisture, EC, pH, lignocellulose degradation rate, nutrient content, and toxicity of compost. Priming made the compost temperature rise rapidly, and the peak temperature of the composting group with priming reached 51 °C. At the end of composting, the moisture in each treatment from high to low was in the order: common compost > priming > C/N, C/P adjustment only > control group. The increase of EC in the treatments with additives was great, and the peak value of EC in the treatment of priming was 1.30 ms·cm−1, which was 3.9 times higher than that of the control group. At the end of composting, the decomposition rate of cellulose in priming compost was 1.7 times higher than that in the control group, and the hemicellulose decomposition rate in the common compost group was 3.2 times higher than that in the control group. By the end of composting, the pH value of the composts in additive treatments was above 7.0, and the pH value of the priming treatment was the highest (7.5). The highest content of organic matter was found in the priming treatment, which was 52%, 1.7 times higher than that in the control group. The total nutrient content (TN + K2O + P2O5) of additive treatments was higher than 5.0%, and the priming treatment was 2.7% higher than that of the control group. By the end of composting, the germination rate and germination index ranged from 88% to 91% and 60% to 81%. Except for the control group, the C/N ratio of other treatments decreased to below 25. Additives can accelerate the decomposition of raw materials, shorten the composting cycle, and improve the quality of composts, and the effect of adding priming is the most significant. Keywords:Platycladus orientalis(L.) Deep eutectic solvents as green media for extraction of flavonoid glycosides and aglycones from Platycladi Cacumen

Bo Zhuang,Li-Li Dou,Ping Li,E-Hu Liu

Deep eutectic solvents (DESs) are emerging as alternatives to conventional ionic liquids and organic solvents due to their unique advantages. In the present study, the tuneability of DESs as tailor-made solvents to efficiently extract polar and non-polar bioactive compounds from Platycladi Cacumen was detailedly investigated. Totally 12 types of choline chloride-, betaine-, and l-proline-based DESs were synthesized for initial screening, and extraction conditions was optimized by single-factor experiment. Experiments with different DESs and principal components analysis demonstrated that the extractability of both flavonoid glycosides and aglycones was greater with certain designed DESs than conventional solvents. In addition, the water content in DESs led to significantly different extraction yields of flavonoid compounds. The target compounds were recovered from DESs by macroporous resin LX-38 with a satisfactory yield between 77.44% and 98.92%. The knowledge acquired in this study could contribute to further DES application in extraction of bioactive compounds from natural sources.

Discrimination of wood borers infested Platycladus orientalis trunks using quartz crystal microbalance gas sensor array

Zhenhe Wang,Wei Chen,Shuang Gu,Jun Wang,Yongwei Wang

Trunk borers infestation is difficult to detect because larvae always mine inside trees. However, plants volatile organic compounds (PVOCs) usually change markedly after pests damage which provide us an opportunity to identify trunk borers infestation. Herein, a four-element quartz crystal microbalance (QCM) gas sensor array based on molecularly imprinted polymers (MIPs) was reported for the discrimination of Semanotus bifasciatus (Motschulsky) and Phloeosinus aubei Perris (Coleoptera: Scolytidae) infested Platycladus orientalis (L.) Franco trunks. MIPs were prepared using the methacrylic acid (MAA) polymer matrix and four characteristic PVOCs (α-pinene, β-phellandrene, 3-carene and cis-thujopsene) as templates molecules, repectively. The amounts of MIPs dropped on the QCM chips were investigated and optimized according to the sensitivities and response times. Finally, the sensor array was applied to detect PVOCs of P. orientalis trunks with S. bifasciatus and P. aubei infestation, and the response matrix were processed with principal component analysis (PCA) and linear discrimination analysis (LDA) for visualization, and further discrimination was performed by k-nearest neighbor (KNN), probabilistic neural network (PNN) and support vector machine (SVM). SVM exhibited satisfying identification accuracy rates in both calibration set (97.62 %) and validation set (93.75 %). This study may contribute to gas sensor arrays development in areas of pest control and forest protection.

Early identification of fungal leaf blight disease (Alternaria alternate) on Platycladus orientalis plants by using gas chromatography-ion mobility spectrometry

Chengyu Zheng,Qin'an Zhou,Wenqing Shao,Jing Zhang,Jun Wang

Platycladus orientalis (L.) is a typical urban greening tree in Asia, but the young leaves of P. orientalis are   vulnerable to pathogenic fungi with hidden early   signs. In this study, we investigated the volatile organic compounds (VOCs) emissions of the Alternaria alternate infected P. orientalis plants, for the first time, using the gas chromatography-ion mobility spectrometry (GC-IMS) combined with chemometric approaches. Results showed that the A. alternate infection could reduce the VOCs release of P. orientalis leaves. The α-pinene and β-pinene emissions were down to 70% and 55% of controls on day 10, respectively. The destructive inoculated plants responded more to the A. alternate , both   in the VOCs emissions and the fungal growth. The (Z)-3-hexenol, n-hexanol, α-pinene, and β-pinene can serve as potential markers for infection discrimination, and their IMS signal intensities were calibrated with corresponding volume concentrations ranging from 0 to 250 μg/L. The PLS-DA model based on GC-IMS fingerprints can correctly classify at least 83% of samples in infection durations after fungal inoculation for the first 10 days. The PLSR model can predict the fungal colony counts in the P. orientalis needles (R c 2 >0.97 and R p 2 >0.92) with good robustness, whose RMSEP was lower than 0.68 (×10 6 CFU g −1 ) in predict set. In a word, the GC-IMS-based approach had the potential to evaluate A. alternate infection in P. orientalis , and the variation of potential markers can provide reference to its fungal infection monitor with   millisecond response in the forest.

Chemical composition, odor-active compounds and in vitro antioxidant activity of essential oils from leaves, fresh fruits and seed shells of Platycladus orientalis (L.) Franco

Runhu Xin,Miao Liang,Rui Wang,Yajian Wu,Wei Guan,Yuping Liu

To investigate the volatiles and odor-active compounds (OACs) in essential oils (EOs) from leaves, fresh fruits and seed shells of Platycladus orientalis (L.) Franco, three EOs were extracted and analyzed by hydrodistillation combined with gas chromatography-mass spectrometry (GC-MS), gas chromatography-mass spectrometry-olfactometry (GC-MS-O) and GC. The yields of three EOs were 0.22%, 0.48% and 1.94%, respectively; a total of 77 compounds were identified by comparing their mass spectra and retention index with those of the authentic standards or the related data reported in the references, and 29 odorants were determined as OACs. Among them, 58 volatile components were identified in the leaf EO, accounting for 98.19% of the total volatiles, of which 18 compounds were screened as OACs; 47 volatile components were determined in the fruit EO, representing 98.98% of the entire volatile oil, of which 12 compounds were sniffed as OACs; 49 volatiles were found in seed shell EO, covering 97.26% of the whole volatiles, of which 19 compounds were detected as OACs. OACs in three EOs were significantly different, and the odorants with higher concentrations included α-pinene, sabinene, δ-3-carene, myrcene, β-caryophyllene, α-terpinyl acetate, cedrol, etc., which could be used as indexes for optimizing extraction process parameters of POF EOs and evaluating EO quality. The in vitro antioxidant activity of three EOs was tested by DPPH assay, ABTS assay and FRAP assay.

Essential oil from Sabina chinensis leaves: A promising green control agent against Fusarium sp.

Zhang Jianyun,Zhao Ziyi,Liang Wenyu,Bi Jingyi,Zheng Yuguang,Gu Xian,Fang Huiyong

Sabina chinensis is a woody plant with important ecological functions in different regions of China, but its essential oils (EO) against plant pathogenic fungi remain largely undetermined. The purpose of our study was to assess the chemical composition and antifungal activity of S. chinensis EO based on optimization of the extraction process. In this study, an actionable and effective model with the experimental results and identified optimum conditions (crushing degree of 20 mesh, liquid–solid ratio of 10.1:1, immersion time of 9.1 h) was established successfully to achieve an extraction yield of 0.54%, which was basically consistent with the theoretical value. A total of 26 compounds were identified using headspace gas chromatography–mass spectrometry (GC–MS) and showed that the major constituent was β-phellandrene (26.64-39.26%), followed by terpinen-4-ol (6.53-11.89%), bornyl acetate (6.13-10.53%), etc. For Petri plate assays, our experiments found for the first time that S. chinensis EO revealed high and long-term antifungal activity against the tested strains, including Fusarium oxysporum and Fusarium incarnatum, at EC50 values of 1.42 and 1.15 µL/mL, which especially reached approximately 76% and 90% growth inhibition at a dose of 0.2 µL/mL, respectively. Furthermore, the antifungal activity of EO from different harvest periods showed remarkable variation. The orthogonal partial least-squares discriminant analysis (OPLS-DA) method revealed 11 metabolites with chemical marker components, and 5 of its potential antifungal activities, terpinen-4-ol, α-terpineol, α-elemol, γ-eudesmol, and bornyl acetate, were strongly correlated with the mycelial inhibition rate. In total, this study explored the antifungal activity of EO against root rot fungus as a potential fungicide and provided valuable information into developing potential products from natural agents.

Photoinduced Associative Ionization Time-of-Flight Mass Spectrometry for the Sensitive Determination of Monoterpenes

Zuojian Zhang,Haijie Wang,Bo Yang,Jinian Shu,Zhangqi Yu,Zhiyang Wei,Jingyun Huang,Kui Jiang,Yedong Guo,Zhen Li

Recently, photoinduced associative ionization (PAI) has been demonstrated to be extremely efficient for the determination of oxygenated volatile organic compounds. However, its application for the determination hydrocarbons has not yet been reported. In this study, associative ionization of nine typical monoterpenes induced by dichloromethane (CH2Cl2) under vacuum ultraviolet (VUV) light was investigated using a laboratory-constructed PAI time-of-flight mass spectrometer (PAI-TOFMS). Without doping CH2Cl2, VUV light generated ordinary photoionization mass spectra of monoterpenes that were dominated by fragment ions, while the molecular ions were inconspicuous. In contrast, doping with CH2Cl2 resulted in the appearance of mass peaks of the characteristic protonated molecular ions of the monoterpenes. The PAI method significantly enhanced the signals for the monoterpenes. The intensities of protonated molecular ions derived from PAI were amplified by 52 to 108 fold compared with those of molecular ions produced by VUV photoionization (VUV PI). With the high ionization efficiency of PAI toward monoterpenes, the sensitivities of PAI-TOFMS toward limonene, β-ocimene, β-myrcene, β-pinene, and α-pinene were from 291 ± 6 to 1338 ± 20 counts pptv−1 using an acquisition time of 30 s. The limits of detection were from 0.32 to 1.45 pptv, equivalent to between 1.92 and 8.83 ng m−3. The applicability of this ionization mode was further characterized by the determination of monoterpenes from potted Sabina chinensis. This study provides a new and highly efficient ionization method for the determination of plant monoterpenes using mass spectrometry.

Screening of hypoglycemic components in Platycladi Cacumen by phytochemical investigation, spectrum-effect relationship, and chemometric methods

Peng-Bo Duan,Ping-Ting Xiao,Xing Yang,Jin-Hua Hao,Kai Li,E-Hu Liu

In this work, the hypoglycemic components in Platycladi Cacumen, an essential traditional Chinese medicine, were evaluated by combining phytochemical investigation, spectrum-effect relationship analysis, and chemometric methods. The phytochemical studies on Platycladi Cacumen extract lead to the isolation of 21 potential bioactive compounds. The chromatographic fingerprints of Platycladi Cacumen samples were established by high-performance liquid chromatography. The hypoglycemic effects of Platycladi Cacumen samples were further evaluated by inhibition of α-glucosidase and detected by the high-performance liquid chromatography method. The spectrum-effect relationship study by bivariate correlations analysis and orthogonal partial least squares regression revealed that myricitrin (P9), quercitrin (P13), afzelin (P18), and amentoflavone (P24) were more relevant to the α-glucosidase inhibitory activity. The results of α-glucosidase inhibitory activity of 21 isolated compounds and molecular docking studies also indicated these flavonoids had potent α-glucosidase inhibitory activity. Collectively, the present study established the spectrum-effect relationship mode of Platycladi Cacumen and discovered the major hypoglycemic components, which provides a feasible method for screening bioactive components.
