Rationale Tongfu Xiexia Decoction (TFXXD), a traditional Chinese medicine formula, comprises six herbs: semen raphani, Rehmannia glutinosa, rhubarb, Magnolia officinalis , trifoliate orange, and mirabilite. The aim of this study was to investigate the effects of TFXXD on its biotransformation and microbial abundance in the gut of constipated rats. Methods Ultrahigh-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UHPLC-Q-TOF-MS/MS) was used to analyze the biotransformation of TFXXD in the colonic contents, and 16S rRNA sequencing was used to assess the structure and diversity of the gut microbiota across various rat groups. Results We identified and analyzed 25 biotransformations of TFXXD in the colonic contents, which undergo various reactions such as deglycosylation, ring opening, and hydration in intestinal bacteria, with 14 originating from trifoliate orange, six from rhubarb, three from Rehmannia glutinosa, one from Semen raphani, and one from M. officinalis . 16S rRNA analysis revealed that TFXXD significantly enhanced the relative abundance of beneficial bacteria, such as Lactobacillus and Bacteroides while significantly reducing Oscillospira abundance. Moreover, TFXXD considerably affected the carbohydrate and amino acid metabolism. Correlation analyses revealed a significant negative correlation between Bacteroides and ACH (Acetylcholine), NO (nitric oxide), D-Lac (D-Lactate), IL-6 (Interleukin-6), TNF-α (tumor necrosis factor-α), and IL-1β (Interleukin-1β) and a significant positive correlation between Bacteroides , hesperetin, and rhein. Conclusions In conclusion, our findings indicate that TFXXD can modulate the structure and diversity of the gut microbiota and enhance the metabolic balance in constipated rats.

Brown seaweed oligosaccharides (BSO) have demonstrated potential as nutraceuticals with cholesterol-lowering, anti-obesity, and anti-constipation properties. In this study, we initially examined how BSO impact body weight, blood lipid levels, and adipose tissue in a rat model of obesity induced by a high-fat diet. Our findings revealed that BSO administration significantly attenuated body weight gain, ameliorated dyslipidemia, and reduced visceral adiposity. Additionally, we evaluated the prophylactic efficacy of BSO in a loperamide hydrochloride-induced constipation mouse model by assessing defecation parameters and gastrointestinal motility. Our results indicated that BSO intervention significantly increased fecal pallet output and fecal water content, as well as improved intestinal transit. In conclusion, this study lays the foundation for a deeper understanding of how BSO may influence obesity and constipation, offering novel insights into its potential safe and efficacious clinical applications.

Constipation is a common gastrointestinal condition. Lactiplantibacillus plantarum T34 has been demonstrated efficacy in relieving loperamide-induced constipation in mice model. Oral intervention of L. plantarum T34 (1 × 10 8 CFU/mouse/day) resulted in improved defecation and enhanced intestinal morphology. Mechanistic studies revealed that L. plantarum T34 strengthend the intestinal barrier through the upregulation of tight junction protein genes, specifically Claudin-1 , as well as mucosal barrier proteins such as Muc2 and Muc3 . Additionally, L. plantarum T34 modulated gut homeostasis by influencing the expression of aquaporin genes, levels of the gut neurotransmitter serotonin (5-HT), and oxidative stress markers. Furthermore, L. plantarum T34 effectively restored the composition of the gastrointestinal microbiota, addressing dysbiosis linked to constipation. These findings suggested that L. plantarum T34 may serve as a promising probiotic intervention for managing constipation, offering a safe and effective alternative to traditional treatments.

To investigate the role of gastrointestinal (GI) polysaccharide fermentation in alleviating constipation, two polysaccharide fractions were isolated from a soluble fiber extract with determined anti-constipation activity: a 2.04 kDa neutral fraction (SSP-1) contained 99.29% glucose, and a 41.66 kDa acidic fraction (SSP-2) contained 63.85% uronic acid. After mice were given loperamide for 14 d to induce constipation, the GI transit rate increased significantly in the SSP-1 group (p< 0.05) but not in the SSP-2 group. The stool weight in the SSP-2 group was significantly higher than that in SSP-1 (383.60 mg vs. 226.23 mg) (p< 0.05). Both SSP-1 and SSP-2 groups had significantly increased serum gastrin and motilin levels (p< 0.05) and changes in their fecal short-chain fatty acid (SCFA) profiles, while SSP-1 showed better fermentation properties than SSP-2 in terms of statistically higher fecal contents of acetic acid and total SCFAs (p< 0.05). Bioinformatic analysis indicated that SSP-1 upregulated bacteria such asOscillibacterto improve SCFA metabolism and stimulate GI hormone secretion, while SSP-2 had less influence on the gut microbiota. These results suggest that the neutral polysaccharide with superior GI fermentation properties exerted beneficial effects on constipation, while the less fermentable pectic fraction might act as a stool-bulking agent. Keywords:sweet potato;polysaccharide;constipation;short-chain fatty acids (SCFAs);gut microbiota;fermentation

Constipation is a prevalent intestinal disorder that significantly diminishes patients' quality of life and predisposes them to various serious health conditions. Current therapeutic approaches for constipation frequently result in drug dependence and adverse effects. In this study, we present evidence that licorice water extract (LWE), a naturally derived compound from Glycyrrhiza uralensis Fisch, effectively ameliorates constipation in mice induced by diphenoxylate (DIP). Our findings indicate that oral administration of LWE (82 mg/kg/d) substantially reduces defecation time, enhances intestinal motility, and increases fecal water content in constipated mice. Furthermore, LWE elevates the levels of excitatory neurotransmitters such as serotonin (5-HT) and motilin (MTL), while concurrently decreasing the levels of inhibitory neurotransmitters including vasoactive intestinal peptide (VIP), nitric oxide (NO) as well as Lipopolysaccharide (LPS). Mechanistically, compared to the model control group, the LWE treatment upregulates the expression of ZO-1, Occludin , and MUC2 genes; repairs DIP-induced damage to the intestinal barrier; increases aquaporin 3 ( AQP3 ) protein expression; downregulates Toll-like receptor 4 ( TLR4 ); mitigates low-grade intestinal inflammation; and restores colonic goblet cell populations. Additionally, LWE is metabolized by gut microbiota, thereby restoring dysbiosis associated with DIP-induced constipation. In conclusion, our results suggest that LWE alleviates constipation through mechanisms involving intestinal barrier repair and gut microbiota modulation, highlighting its potential as an alternative therapeutic option for this condition. In response to this, we have developed a convenient gel-type candy (LWEI) with licorice extract and inulin as the main ingredients, which has shown good efficacy in relieving constipation.

Constipation is a gastrointestinal disorder related to intestinal flora imbalance and inflammation. Numerous clinical studies have affirmed the efficacy of psyllium husk powder (PHP) in alleviating constipation; however, the efficacy and specific mechanism of PHP in alleviating constipation still lack comprehensive and in-depth discussions. In this study, we comprehensively investigated the mechanism by which round-bracted psyllium husk powder improves loperamide hydrochloride-induced constipation in mice, focusing on gut microbiota and fecal metabolic profiles while comparing the effects of PHP interventions at varying doses to polyethylene glycol 4000 bulk treatment for a comprehensive evaluation of PHP's efficacy in alleviating constipation. Our findings revealed that PHP significantly reduced defecation time and increased fecal output, weight, water content, intestinal motility, and short-chain fatty acid (SCFA) concentrations while positively influencing intestinal inflammation and promoting repair of the intestinal barrier, and higher doses of PHP demonstrate optimal efficacy. Furthermore, PHP markedly modified the microbial composition in constipated mice by enhancing the abundance of beneficial bacteria, particularly Akkermansia muciniphila . UPLC-QTOF-MS/MS analyses indicated that PHP contributed to elevated levels of Cucurbitacin B, BIX 01294 Trihydrochloride along with peptides like Asp-Gly-Lys-Ile-Leu and Val-Met-Met-Val-Arg among other potentially beneficial metabolites. KEGG pathway analysis revealed that PHP alleviates constipation by modulating metabolic pathways associated with SCFA production, beneficial microbiota colonization, and the reduction of intestinal inflammation, encompassing ABC transporters, biosynthesis of unsaturated fatty acids, and galactose metabolism etc. In conclusion, PHP can effectively relieve constipation by remodeling the structure of the gut microbiota while enhancing its associated metabolic pathways and products.

Intestinal inflammation frequently occurs alongside dysmotility, which is characterized by altered myosin light chain phosphorylation levels. Curcumin, an active component from the ginger family, is reported to confer anti‑inflammatory effects. However, the effects of curcumin on both diarrhea and constipation associated inflammation remains to be elucidated. The present study was designed to investigate the effects of curcumin on diarrhea and constipation and to determine the related mechanisms. Sprague‑Dawley rats were used to establish diarrhea and constipation models via intracolonic acetic acid (4%) instillation or cold water gavage for 2 weeks, respectively. Blood samples were collected to measure the serum levels of the cytokines TNF‑α and IL‑1β using ELISA kits. Western blotting was performed to measure NF‑κB, RhoA, Rho‑related kinase 2, phosphorylated MLC20, phosphorylated myosin phosphorylated target subunit 1, 130k Da‑MLC kinase (MLCK), c‑kit tyrosine kinase protein expression, and reverse transcription‑quantitative PCR was conducted to measure MLCK expression levels. The results indicated that curcumin reversed the elevations in the pro‑inflammatory cytokines IL‑1β and TNF‑α by inhibiting the NF‑κB pathway in rats with diarrhea and constipation. The results also indicated that myosin light chain (MLC) phosphorylation in intestinal smooth muscle was positively and negatively associated with the motility of inflammation‑related diarrhea and constipation in rats, respectively. Curcumin significantly reversed the increased MLC phosphorylation in the jejunum of the rats with diarrhea, significantly enhanced the reductions in inflammatory mediators, including TNF‑α and IL‑1β, of rats with constipation and significantly ameliorated the related hyper‑motility and hypo‑motility in rats with both diarrhea and constipation. In conclusion, the potential roles of the MLC kinase, c‑kit tyrosine and Rho A/Rho‑associated kinase 2 pathways, which are involved in curcumin‑induced amelioration of inflammation‑related diarrhea and constipation, were explored in the present study. Results from the present study suggested that curcumin has potential therapeutic value for treating intestinal inflammation and inflammation‑related motility disorders.

Different dietary fibers have distinct structures, leading to significant variations in their laxative effects. To explore how these structural differences impact constipation intervention, a 14-day study was conducted on loperamide-induced constipated mice using five dietary fibers: soluble dietary fiber from steamed sweet potato (SDF-S), oat β-glucan (OB), polydextrose (PD), arabinogalactan (AG), and inulin (IN). The results showed that four fibers, excluding PD, significantly improved gastrointestinal (GI) transit rate (p< 0.05), although PD had the highest fecal moisture, it was significantly different from the lowest IN (p< 0.05). AG and IN resulted in higher 6 h fecal weights compared to other fibers. SDF-S and OB were more effective in modulating serum levels of gastrointestinal hormones. The different monosaccharide compositions and glycosidic bonds of these fibers led to distinct changes in gut microbiota composition and SCFA profiles. Galactose and arabinose in AG were linked to increased abundance ofLachnospiraceae_UCG-006,Bacteroides, andOdoribacter, promoting butyrate fermentation, which is positively correlated with GI transit rate. Glucose in SDF-S, OB, and PD favored acetate fermentation positively correlated with fecal moisture. Fructose in IN encouraged the proliferation ofMuribaculaceae_unclassifiedandRuminococcus, associated with butyrate fermentation and increased 6 h stool weight, respectively. The β-glycosidic bonds in OB may lead to high butyrate production through the selective proliferation ofLachnospiraceae_unclassified. Minor components like fucose, rhamnose, and ribose were positively correlated with the abundance ofOscillospiraceae_unclassified,Anaerotignum, andLachnospiraceae_unclassified. In conclusion, the unique monosaccharide compositions and glycosidic bond differences in dietary fibers selectively promote the proliferation of fiber-degrading and butyrate-producing bacteria, resulting in varied effects on constipation relief.

Vidarabine (VID) is an antiviral medication that is commonly utilized to treat conditions such as hand, foot, and mouth disease and herpes. Constipation is a prevalent complication of these diseases. Could VID treat these diseases by influencing defecation behavior? To date, no studies have been conducted on the potential of VID to relieve constipation. Therefore, a systematic investigation was conducted into the laxative effects and mechanisms of VID using loperamide-induced functional constipated mice. The findings indicate that the oral administration of VID promoted gastrointestinal peristalsis, improved fecal properties, facilitated defecation, and demonstrated a significant laxative effect on functional constipated mice. It has been demonstrated that VID may increase the water content of feces by regulating the expression of aquaporins (AQP3, AQP4, andAQP8) in the colon and promote intestinal motility by regulating the expression of neurotransmitters (AChE and VIP) and the adenosine A2A receptor–myosin light chain (A2AR-MLC) signaling pathway in constipated mice. Concurrently, VID may also reduce colonic inflammation in constipated mice, reinforce the gut barrier function, and alter the composition and structure of the gut microbial community. Some microbial taxa, including Firmicutes andLactobacillus, were found to be associated with the alleviation of constipation, while other taxa, including Bacteroidetes, Proteobacteria, Muribaculaceae,Muribaculum,norank__f__Desulfovibrionaceae, andParasutterella, were found to be associated with constipation. These results indicate that the gut microbiota may play a significant role in the alleviation of constipation by VID. These findings confirm the efficacy of VID in a constipated animal model, which justifies further investigation into its potential clinical applications.

This study investigated the effects of Lactobacillus paracasei ( L. paracasei )-fermented Goji juice supernatant (FGS) on constipation. GC–MS data showed that the critical metabolites of FGS were nicotinamide, squalene, L-proline, melatonin, and sarcosine. FGS intervention significantly promoted intestinal peristalsis. Meanwhile, FGS ameliorated intestinal inflammation and mucosal barrier damage by inhibiting the expressions of pro-inflammatory factors (TNF-α, IL-1β, COX-2), promoting the expressions of Mucin 1, Mucin 3, ZO-1, AQP4, and ENac-γ. In addition, FGS treatment modulated gut microbiota composition, reduced the abundance of harmful bacteria ( Faecalibaculum , Bacteroides , Alloprevotella and Alistipes ), induced a bloom of probiotics, including Marvinbryantia , Eubacterium _ xylanophilum _group, Colidextribacter , and Candidatus _ Soleaferrea . Conclusively, this study demonstrated that FGS significantly reduced inflammatory response, prevented intestinal barrier damage, and modulated microbiota dysbiosis to improve constipation. Compared to unfermented Goji juice (UFGJ), FGS has shown a more effective anti-constipation effect, indicating that FGS could relieve constipation as a dietary therapeutic.

Lactiplantibacillus plantarum M660 (M660) was isolated from the human milk of a healthy mother. M660 could survive the simulated gastrointestinal conditions without haemolytic and biogenic amine-producing ability. The faecal water content was reduced, and the recoveries of the cecum and intestinal barrier were accelerated by M660 in the antibiotic-associated diarrhoea model. In the constipation model, M660 enhanced intestinal motility, improved serum neurotransmitter levels, and reduced colonic injury. Analysis of gut microbiota showed that M660 increased the abundance of potentially beneficial bacteria and decreased the number of potentially pathogenic bacteria. Overall, M660 could be a probiotic candidate to promote infant health.

Chitosan (CTS) and its oligosaccharide (COS) can protect against excessive protein consumption (EPC)-induced constipation by regulating gut microbiota and metabolites. The imbalanced gut microbiota was observed in whey protein isolate (WPI) group by 16S rRNA gene sequencing. However, CTS and COS treatments reversed the abundance changes of several gut microbiota, which led to the balance in the production of metabolites. Interestingly, some metabolites associated with constipation were also regulated by CTS and COS in in vitro colonic fermentation model and EPC-induced constipated rats, including nicotinamide , glutamine, proline , cysteine, choline , tryptophan , and pipecolic acid , which were mainly associated with aminoacyl-tRNA biosynthesis , arginine biosynthesis, and d -glutamine and d -glutamate metabolism pathways. In conclusion, the present study shed light on the effects of CTS and COS on EPC by regulating gut microbiota and metabolites and explored the correlation between gut microbiota and metabolites, reinforcing its potential application as a prebiotic in maintaining intestinal health.
