Study on the Anticoagulant or Procoagulant Activities of Type II Phenolic Acid Derivatives

Xuan Luo,Chuanrong Du,Hui Cheng,Jian-hua Chen,Cuiwu Lin

In this study, three type II phenolic acids (caffeic acid,p-hydroxycinnamic acid, and ferulic acid) were used to synthesize a total of 18 phenolic acid derivatives. With molecular docking for molecule design and target protein (factors) screening, in combination with the confirmation of target proteins (factors) by surface plasmon resonance, and the evaluation of haemostatic and anticoagulant activities with five blood assays (plasma recalcification time, prothrombin time, activated partial thromboplastin time, fibrinogen, and thrombin time), the data indicated that caffeic acid derivatives showed certain anticoagulant or procoagulant activities and that two other series contained compounds with the best anticoagulant activities. Using Materials Studio analysis, particular functional groups that affect anticoagulant or procoagulant activities were revealed, and these conclusions can guide the discovery of compounds with better activities. Keywords:anticoagulant;procoagulant;haemostatic;phenolic acid;surface plasmon resonance

Superabsorbent Chitosan-Based Nanocomposite Antibacterial Hydrogel for Biofilm Disruption and Accelerated Healing of Infected Wounds

Shengnan Zhang,Lidan He,Shuting Fu,Yingying Zhou,Li Wang,Le Deng

Chitosan-based hydrogels, owing to their unique barrier protection properties, have garnered significant attention as wound dressings. Improving the liquid-absorption and antimicrobial capabilities of chitosan hydrogels will further broaden their applications in this field. In this study, chitosan was modified with quaternary ammonium salt and glycidyl methacrylate and then combined with negatively charged gold–platinum nanodots (AuPt NDs) to construct a type of nanocomposite antibacterial hydrogel. Structurally, the hydrogel possesses a loose and porous structure. It can absorb about 800% of its own mass in pure water and has excellent pro-coagulant activity. These properties are beneficial for wound healing. Mechanistically, the positively charged hydrogel is capable of adsorbing negatively charged bacteria, disturbing the potential balance of the bacterial cell membranes, which can reduce the activity of the bacteria and lead to dissipation of the bacterial biofilm. Furthermore, the hydrogel antimicrobial effect can be greatly enhanced by the incorporation of AuPt NDs, enabling it to catalyze the production of ·OH from a low concentration of H2O2 and generate heat under 808 nm laser irradiation, thereby further killing the bacteria and promoting the dissipation of the biofilm effectively and rapidly. More importantly, the hydrogel has excellent biocompatibility and facilitates the healing of wounds infected with bacterial biofilms, holding great potential for application as a medical wound dressing.

HNTs assembled sponge with enhanced mechanical properties and bioactivity for uncontrollable hemorrhage and wound healing

Xiao Yang,Miao Chen,Yuzhen Wang,Xiaoqin Si,Jiahui Hao,Changcan Shi

Self-expanding sponges with excellent water absorption capacity hold unique prospects for treating uncontrollable and noncompressible hemorrhage. However, fabrication of sponges with robust mechanical properties, effective pro-coagulation potential and desired infection prevention enabling for irregular or deep wound management including hemostasis and wound repair are the urgent challenge. Here, the halloysite nanotubes (HNTs) were assembled with modified starch (MSt) by hydrogen bond formation, and the composite sponge (HNTs/MSt) was finally developed after foaming technique, crosslinking reaction and lyophilization. The prepared sponge not only retained excellent water absorption and self-expanding ability, but also exhibited a two-fold increase in the maximum compressive stress and a 53 % reduction in clotting time in vitro due to assembled silicon-based nanotubes. As a result, the composite sponge had better hemostatic performance in SD rat/rabbit liver volume defect and artery transection hemorrhage models as compared with control and commercial hemostatic sponges. Furthermore, due to the drug-loading capacity of HNTs, composite sponge loaded with gentamycin could inhibit the growth of microorganism and promote rat full-thickness skin wound healing. All the results indicated that HNTs assembled sponge would be promising biomaterial for treating uncontrollable hemorrhage and wound repair.

An anti-infective and pro-coagulant chitosan-based composite sponge with shape memory for effective hemostasis in multi-tissue defects

Xinchen Du,Ruxiang Li,Yadong Liu,Binghao Hou,Taosheng Sun,Lianyong Wang

The development of an anti-infective and pro-coagulant sponge with shape memory for hemostasis in multi-tissue defects remains a challenge. Herein, a novel composite sponge was developed using quaternized and phosphated chitosan, disuccinimide carbonate modified polyethylene glycol, and attapulgite. The sponge featured a macroporous structure, sufficient mechanical strength, high affinity for water and blood, and shape recovery when exposed to water and blood. The sponge promoted blood cell adhesion, aggregation, and activation, as well as the activation of intrinsic and extrinsic coagulation pathways. The sponge exhibited potent antibacterial efficacy against Staphylococcus aureus and Escherichia coli in a bacteria-infected rat skin defect model. More importantly, the biocompatible sponge demonstrated superior hemostatic capacity than commercial hemostats in rat femoral artery injury, rat liver and rabbit femora defect models. The developed sponge shows great potential for hemostasis and anti-infection in multi-tissue defects. This study provides a new strategy for designing and preparing novel chitosan-based hemostatic sponges.

A shape memory cationized chitosan sponge loaded with thrombin for high-effective hemostasis and antibacterial activity

Ming Ma,Yi Guo,Ruotong Ji,Luwei Wang,Yan Zhang,Xinchen Du,Sujuan Zeng

This study presents a thrombin-loaded cationized chitosan (TCCS) sponge with highly effective hemostatic and antibacterial activity. The TCCS sponge, prepared using a multistep method, features a porous structure, favorable mechanical properties, excellent water absorption ability, and shape recovery triggered by water or blood. The TCCS sponge exhibited strong antibacterial activity against Methicillin-resistant Staphylococcus aureus and Escherichia coli . Additionally, it demonstrated enhanced procoagulant and hemostatic efficacy in rat tail amputation and rat liver perforation wound models compared to commercial hemostats. Furthermore, the sponge exhibited favorable biocompatibility and biosafety. These findings suggest that the TCCS sponge has substantial potential for practical applications in managing severe hemorrhages and bacterial infections.

Procoagulant CaCO3-embedded embolic microspheres can potentiate transcatheter arterial embolization of hepatocellular carcinoma

Ziliang Dong,Lei Zhang,Dongxu Zhao,Chunjie Wang,Yu Hao,Quguang Li,Yunyun Zhang,Zhijuan Yang,Caifang Ni,Zhuang Liu,Liangzhu Feng

Transcatheter arterial embolization (TAE) represents a preferred therapeutic modality with minimal invasiveness for unresectable intermediate and advanced hepatocellular carcinoma (HCC) patients. However, its therapeutic outcome is restricted by incomplete occlusion of the whole tumor vasculature and exacerbated tumor immunosuppression due to the lack of ideal embolic agents. Herein, procoagulant embolic microspheres with tunable sizes are prepared by loading thrombin into calcium carbonate (CaCO 3 ) nanoparticle-embedded gelatin microspheres (Th-CaCO 3 @gelatin MSs) via a microfluidic process. In this system, the yielded Th-CaCO 3 @gelatin MSs are capable of occluding main tumor-feeding blood vessels, while the released thrombin and Ca 2+ triggered by the acidic tumor microenvironment can synergistically promote the formation of intravascular blood clots to occlude tumor capillaries. Meanwhile, the embedded CaCO 3 nanoparticles can work as proton sponges to neutralize tumor acidity and thus effectively reverse tumor immunosuppression. Resultantly, TAE treatment with Th-CaCO 3 @gelatin MSs exhibits the most effective tumor suppression efficacy on orthotopic N1S1 HCC rat xenografts without causing obvious toxic effects. Therefore, this study highlights that our biocompatible embolic microspheres are capable of potentiating conventional TAE treatment by promoting pH-responsive tumor vascular infarction and neutralizing the acidic tumor microenvironment, with great potential in clinical translation.

Injectable hemostatic hydrogel adhesive with antioxidant, antibacterial and procoagulant properties for hemorrhage wound management

Jiahao Yang,Tianjiao Wang,Liang Zhang,Peng Fan,Jiulong Zhao,Xiaoyi Zheng,Yongkang Lai,Hongrui Liu,Shige Wang

In emergencies, uncontrolled severe bleeding can result in undesired complications and even death of the injured. Designing advanced hemostatic agents is a potential solution for emergency hemostasis, yet it remains challenging to realize the persistent adhesion in a wet wound environment. In this study, based on dynamic reversible Schiff base bond and photo-initiated double-bond polymerization, a novel injectable hemostatic hydrogel ( L -COC) consisting of methacrylated carboxymethyl chitosan (CMCSMA), oxidized konjac glucomannan (OKGM) and (+)-catechin hydrate (CH) was synthesized for emergency hemostasis. To our delight, the incorporated CH imparted enhanced blood procoagulantion to the L -COC hydrogel by intensifying the hydrogel-red blood cell interactions. As a result, the hemostatic effect of the engineered L -COC hydrogel was significantly superior to that of fluid gelatin Surgiflo TM for liver bleeding wounds in rats (Blood loss: 0.62 ± 0.11 g ( L -COC), 0.90 ± 0.08 g (Surgiflo TM ); hemostasis time: 69.0 ± 2.9 s ( L -COC), 84.0 ± 2.2 s (Surgiflo TM )). With the favorable antioxidant and antibacterial activities, as well as multifunctional properties, the bio-adhesive L -COC hydrogel and the underlying design principles may facilitate further development of practical hemostatic hydrogels.
