A one-stop integrated natural antimicrobial microneedles with anti-inflammatory, pro-angiogenic and long-term moisturizing properties to accelerate diabetic wound healing

Aili Wang,Xi Ruan,Xuejiao Wang,Yuyu Ren,Chunjiao Shen,Kaiyi Zhang,Zhenjie Song,Bai Xiang,Yinling Ma,Feng Zhao

Diabetic ulcers present a formidable obstacle in diabetes management, typically leading to high mortality and amputation rates. To overcome traditional monotherapy drawbacks, We developed a novel microneedle strategy for combined antimicrobial action: ingeniously integrating quercetin with Platelet-derived Growth Factor-BB(PDGF-BB) and Sucrose Octasulfate(SOS) into the microneedle system(QPS MN). This method allows to penetrate through biofilms, administering quercetin nanocrystals and PDGF-BB deep into the tissue to combat microbial infection, mitigate inflammation, and promote angiogenesis. The accompanying backing material contains SOS, which absorbs wound exudate and forms a dressing that provides a moist environment for wound healing In an in vitro wound-scratch assay demonstrated that co-cultivating Human Umbilical Vein Endothelial Cells(HUVEC) with QPS MN for 48 h (90.3 ± 2.51 %) significantly enhanced cell migration compared to the control group (20.2 ± 1.41 %). Moreover, treatment of streptozotocin-induced diabetic wounds in rats with QPS MN for 14 days resulted in a wound healing rate of 96.56 ± 3.44 %, far surpassing the healing rate of only 40.34 ± 7.26 % observed in the untreated control group. Furthermore, the QPS MN treated wounds exhibited a notable increase in skin appendages and neovascularisation, indicating promising potential for achieving complete wound healing. These results suggest that QPS MN may offer substantial therapeutic benefits for addressing diabetic wounds.

Metal-ion Co-doped nanofilm with anti-bacterial and pro-angiogenic activities to accelerate infected wound healing

Kun Wang,Hui Wang,Yanqun Liu,Zhiqin Li

The ability of nanofilms to mimic the natural fiber architecture and form a physical structure that is best suited for skin tissue regeneration makes it an appealing vehicle for skin tissue engineering. However, developing a facile nanofilm with both anti-bacterial and pro-angiogenic activities is yet to be addressed. Herein, two metal ions, including copper (Cu) and silicon (Si), were loaded into Wharton's jelly/polycaprolactone (WJ/PCL) system to prepare a Cu/Si@WJ/PCL nanofilm. These metal ions could impart both anti-bacterial and pro-angiogenic properties to the nanofilms, which could improve their ability in promoting wound healing. The Cu and Si ions were released continuously from the Cu/Si@WJ/PCL nanofilm, which had a nanofibrous structure and random organization. Results derived from the in vitro experiment revealed that the incubated fibroblasts could successfully survive and proliferate under lab conditions, suggesting that the Cu/Si@WJ/PCL nanofilm was cytocompatible. Additionally, the Cu ions released from the nanofilm exhibited effective antibacterial activities against both the gram-positive and gram-negative bacteria, and they acted in conjunction with the released Si ions for improving the tube formation in the human umbilical vein endothelial cells. Additionally, compared to pure WJ/PCL nanofilm and Cu or Si ion-loaded nanofilm, the Cu/Si@WJ/PCL nanofilm significantly accelerated the infected wound healing process in the rat model. The Cu/Si@WJ/PCL nanofilm showed improved epidermal development, vascularization, and collagen deposition compared to the other nanofilms. In this study, a Cu/Si@WJ/PCL nanofilm with pro-angiogenic and anti-bacterial activities was successfully developed, and it could be used as a metal-ion-based therapeutic medium for the treatment of infected wound healing.

The utilization of chitosan/Bletilla striata hydrogels to elevate anti-adhesion, anti-inflammatory and pro-angiogenesis properties of polypropylene mesh in abdominal wall repair

Di Yuntao,Wang Lu,He Wei,Liu Shuyan,He Yuqi,Liao Jie,Zhang Ruihong,Yin Lan,Xu Zhiwei,Li Xiaoming

Polypropylene (PP) mesh is commonly used in abdominal wall repair due to its ability to reduce the risk of organ damage, infections and other complications. However, the PP mesh often leads to adhesion formation and does not promote functional tissue repair. In this study, we synthesized one kind of aldehyde Bletilla striata polysaccharide (BSPA) modified chitosan (CS) hydrogel based on Schiff base reaction. The hydrogel exhibited a porous network structure, a highly hydrophilic surface and good biocompatibility. We wrapped the PP mesh inside the hydrogel and evaluated the performance of the resulting composites in a bilateral 1 × 1.5 cm abdominal wall defect model in rats. The results of gross observation, histological staining and immunohistochemical staining demonstrated the positive impact of the CS hydrogel on anti-adhesion and wound healing effects. Notably, the addition of BSPA to the CS hydrogel further improved the performance of the composites in vivo, promoting wound healing by enhancing collagen deposition and capillary rearrangement. This study suggested that the BSPA-modified CS hydrogel significantly promoted the anti-adhesion, anti-inflammatory and pro-angiogenesis properties of PP meshes during the healing process. Overall, this work offers a novel approach to the design of abdominal wall repair patches.

Dual-functional injectable hydrogel as antimicrobial and angiogenic therapeutics for dental pulp regeneration

Nhu-Quynh Thi Nguyen,Cuong Hung Luu,Ngoc-Dan Ho Nguyen,Duy Quoc Vo,Ngoc Hong T. Luu,Nhu-Y Ngoc Ha,Thanh-Truc Ngoc Vo,Que-Phuong Le Huynh,Thavasyappan Thambi,Chau T.T. Ngo,V. H. Giang Phan

Pulpitis is a painful inflammatory condition of the dental pulp, commonly triggered by bacterial infection, trauma, or repeated dental procedures. Patients often experience heightened sensitivity to temperature changes, spontaneous pain, and, in severe cases, necrosis of the pulp tissue, leading to tooth loss if left untreated. Traditional treatment primarily involves root canal therapy, which removes infected pulp and seals the canal with gutta-percha; however, this approach lacks antimicrobial properties and does not support tissue regeneration. To address these limitations, this study investigates an injectable hydrogel system composed of hyaluronic acid (HA) and carboxymethyl chitosan (CMCs), chemically crosslinked with 1,4-butanediol diglycidyl ether (BDDE). The hydrogel demonstrated excellent injectability, self-healing ability, and mechanical stability. In vitro and in vivo assessments confirmed its biocompatibility, biodegradability, and capability for controlled release of therapeutic agents. Notably, amoxicillin-loaded hydrogel (HACM/AX) showed significant antibacterial properties; while erythropoietin-loaded hydrogel (HACM/EPO) significantly enhanced endothelial cell proliferation and migration, facilitating angiogenesis for tissue regeneration. These findings highlight the potential of HACM hydrogel as a dual-function biomaterial with antimicrobial and pro-angiogenic properties, offering a promising alternative for pulpitis treatment and dental tissue engineering.

Biomimetic dually cross-linked injectable poly(l-glutamic acid) based nanofiber composite hydrogels with self-healing, osteogenic and angiogenic properties for bone regeneration

Bo Wang,Jia Liu,Chao Guo,Xiaogang Bao,Liangfa Qi,Jingbo Yin,Guohua Xu,Shifeng Yan

Biomimetic hydrogels with satisfactory mechanical properties and osteogenic/angiogenic abilities have attracted extensive attention in recent years. Injectable hydrogels with the advantages of minimally invasive implantation and adaptability to irregular defects exhibit superior application potential in the field of bone regeneration. However, poor mechanical properties and limited self-healing ability hinder their potential as supporting materials. Furthermore, the absence of osteogenic and angiogenic properties significantly affect their application in bone regeneration. For injectable hydrogels, simulating natural bone tissue ECM presents an opportunity for developing innovative materials for bone repair. In this work, a strategy to prepare biomimetic nanofiber-reinforced dually cross-linked (DC) injectable hydrogels with self-healing, osteogenic and angiogenic properties was proposed. The PLGA/MDex/CaP@PBLG nanofiber hydrogels were crosslinked through two reversible dynamic interactions including Schiff base reaction and physical chelation, thereby demonstrating good self-healing property. Meanwhile, the incorporation of CaP@PBLG mineralized nanofibers and alendronate sodium (BP) grafted on PLGA could endow the hydrogels with favorable osteogenic function. Moreover, the introduction of antioxidant ferulic acid (FA) could significantly stimulate angiogenic activity of the hydrogels. The successful regeneration of cranial defects in rats demonstrated a potential application of PLGA/MDex/CaP@PBLG nanofiber hydrogels in the field of bone regeneration.

Application of Mg-MOF-loaded gelatin microspheres with osteogenic, angiogenic, and ROS scavenging capabilities in bone defect repair

Zhiheng Luo,Jianxiong Ma,Yan Wang,Yuhan Du,Yujie Liu,Wei Zhang,Yuxiao Lai,Xinlong Ma

The management of bone defects, particularly those with irregular geometries resulting from osteoporotic fractures, remains fraught with challenges. Microspheres have emerged as a promising vehicle for tissue engineering, distinguished by their controlled release, safety, and ease of application. Various bioactive components are integrated into microspheres to improve their performance. Metal-organic frameworks, formed from metal ions interconnected by organic ligands, are increasingly utilized in tissue engineering. Specifically, magnesium-based MOFs are notable for their broad applicability; Mg 2+ ions are instrumental in bone reconstruction and repair, facilitating osteogenesis, angiogenesis, antibacterial effects, and anti-inflammatory properties. Mg-MOF was synthesized using magnesium chloride and gallic acid, and it was incorporated into gelatin microspheres to create Gel@Mg-MOF composite microspheres. Leveraging gelatin's biocompatibility, controlled release, and biodegradability, the composites' biocompatibility was evaluated through toxicity and adhesion assays. Moreover, the osteogenic and angiogenic potentials of the Gel@Mg-MOF microspheres were assessed, alongside their capacity for ROS scavenging. Results suggest that controlled Mg 2+ release from Gel@Mg-MOF microspheres promotes osteogenic activity in RBMSCs and enhances angiogenic potential in HUVECs. Additionally, the gallic acid-containing composite microspheres exhibited antioxidative properties. Collectively, the findings suggest that Gel@Mg-MOF microspheres could provide effective support for bone defect repair, with potential for clinical deployment.

Microenvironment-Regulated Hydrogels Prepared with a Brand-New Small Molecule Cross-Linker for Stepwise Treatment of Myocardial Infarction

Yu Wu,Wen Zhang,Linlin Huang,Xinping Xu,Shuyi He,Zhicun Wang,Wenqi Liu,Ya Yao,Li Yang,Cheng Hu,Yunbing Wang

Despite the development of many injectable hydrogels intended for the repair of myocardial infarction (MI), their effectiveness is often compromised because they target merely one or two phases of MI's pathological progression. Here, a multifunctional hydrogel delivery platform is prepared with a brand-new small molecule cross-linker for stepwise treatment of MI. The synthesis and reporting of a novel small-molecule phenylboronic acid cross-linker ((N(BA) 3 )) with a precise molecular structure is conducted for the first time, and it is successfully utilized with polyvinyl alcohol (PVA) dopamine to prepare a hydrogel with infarct microenvironmental responsiveness and anti-oxidant. Further, considering the multistage of MI repair, hydrogel contains both hyperoside bioactive nanoparticles (EGCG@Hyp&Arg NPs) and PLGA microspheres loaded with galunisertib (PLGA@Gal Ms). The EGCG@Hyp&Arg NPs are rapidly released and demonstrate anti-inflammatory and pro-angiogenesis effects, while in the long term, Gal is released from the PLGA@Gal Ms to inhibit myocardial fibrosis and improve cardiac function. Results from both in vitro and in vivo studies reveal that the hydrogel is engineered with programmed capabilities for anti-oxidation, reducing inflammation, promoting new blood vessel formation, and inhibiting fibrosis, thereby significantly enhancing heart function post-MI. Overall, this multifunctional hydrogel delivery platform has great potential for application as a therapeutic strategy for MI.

Bioactive Metal Ion-Coordinated Dynamic Hydrogel with Antibacterial, Immunomodulatory, and Angiogenic Activities for Infected Wound Repair

Xinrui Li,Xu Chen,Lian Guan,Wenbo He,Weiling Yin,Dongcheng Ye,Jia Gao,Miao Wang,Guoqing Pan

The repair of infected wounds is a complex physiopathologic process. Current studies on infected wound treatment have predominantly focused on infection treatment, while the factors related to delayed healing caused by vascular damage and immune imbalance are commonly overlooked. In this study, an extracellular matrix (ECM)-like dynamic and multifunctional hyaluronic acid (HA) hydrogel with antimicrobial, immunomodulatory, and angiogenic capabilities was designed as wound dressing for the treatment of infected skin wounds. The dynamic network in the hydrogel dressing was based on reversible metal–ligand coordination formed between sulfhydryl groups and bioactive metal ions. In our design, antibacterial silver and immunomodulatory zinc ions were employed to coordinate with sulfhydrylated HA and a vasculogenic peptide. In addition to the desired bioactivities for infected wounds, the hydrogel could also exhibit self-healing and injectable abilities. Animal experiments with infected skin wound models indicated that the hydrogel dressings enabled minimally invasive injection and seamless skin wound covering and then facilitated wound healing by efficient bacterial killing, continuous inflammation inhibition, and improved blood vessel formation. In conclusion, the metal ion-coordinated hydrogels with wound-infection-desired bioactivities and ECM-like dynamic structures represent a class of tissue bionic wound dressings for the treatment of infected and chronic inflammation wounds.

Injectable glycyrrhizic acid-based hydrogel with Immunoregulatory and angiogenic properties for diabetic wound healing

Haisheng Wu,Zichao Xue,Zhigang Wang,Jiacai Mei,Mingzhe Shao,Jian Zhang,Haiyang Hu,Ye Pan

Diabetic wound healing (DWH) demands multifunctional dressings capable of delivering therapeutic agents to counteract the impaired repair microenvironment. Elevated oxidative stress and chronic inflammation not only suppress regenerative cell viability but also exacerbate apoptosis and vascular dysfunction, collectively hindering wound healing. To address these challenges, we developed an injectable dual-network hydrogel dressing based on immunomodulatory and pro-angiogenic glycyrrhizic acid (GA). This hydrogel combines   alkylated glycyrrhizinic acid (AGA)   and   methacrylated sericin (SFMA)   to form a natural extracellular matrix (ECM)-mimetic dual network, achieving both excellent injectability and enhanced mechanical properties. Furthermore, the hydrogel incorporates the pro-angiogenic agent   desferrioxamine (DFO)   through dynamic covalent bonding, enabling sustained drug release. The resulting   AGA/SFMA@DFO hydrogel   effectively modulates macrophage polarization and enhances vascular regeneration under inflammatory conditions. Additionally, the residual aldehyde groups in the   AGA/SFMA@DFO   gel form Schiff base linkages with amino groups on the skin surface, ensuring strong tissue adhesion. Consequently, this multifunctional hydrogel dressing demonstrates significant potential in accelerating diabetic wound repair.

Dual peptide-coordinated dynamic hydrogel with antibacterial and proangiogenic activities for infected skin wounds

Li Ni,Xiaoyu Zhang,Jia Gao,Junjiang Yue,Linlin Zhang,Guoqing Pan

Infected skin wounds represent a class of significant clinical challenges due to their complexity and the risk of delayed healing. Developing an effective wound dressing capable of promoting tissue regeneration while addressing infections is crucial. In this study, we designed a dual peptide-coordinated dynamic hydrogel with both antibacterial and proangiogenic properties, specifically tailored for treating infected skin wounds. The dynamic hydrogel was constructed using gelatin, a natural biomacromolecule with high biocompatibility, as the base material. Two functional peptides, the antibacterial melittin and angiogenic peptide, were incorporated to enhance both infection control and tissue repair. Hydrogelation was achieved through coordination interactions between gold (Au) ions and sulfhydryl groups, endowing the hydrogel with dynamic properties. The excellent biocompatibility, antibacterial efficacy and angiogenic capability of hydrogel allow it to provide structural support for wound healing while promoting cell proliferation and migration. By eradicating bacteria and stimulating new blood vessel formation, it accelerates the healing of infected wounds. Additionally, its injectability and self-healing properties make it suitable for treating irregularly shaped and deep wounds. This dual-functional hydrogel may offer a promising and minimally invasive treatment option for infected skin wounds, combining high biological safety with simplified surgical procedures.

H2S/Curcumin Coreleasing Biomineralized Zinc Sulfide Nanostructures for Diabetic Wound Healing

Shiqi Wu,Yuxuan Ge,Yuan Peng,Yunsheng Chen,Zheng Zhang,Yixin Zhang

Diabetic wound healing is a great challenge among diabetic patients, and it is a complex dynamic process involving inflammatory and angiogenesis responses. Anti-inflammation and pro-angiogenesis are key issues in diabetic wound healing. Herein, an H2S/curcumin coreleasing biomineralized zinc sulfide (named HZSC) is developed to promote diabetic wounds healing. The prepared HZSC features a nanoscale size of 166 ± 6.4 nm as well as a unique structure of HA loading Cur and coating ZnS biomineralized nanoclusters. After being applied in the mildly acidic diabetic wound tissue, the prepared HZSC controllably coreleases H2S and curcumin to promote macrophage polarization to the M2 phenotype, reduce reactive oxygen species levels, as well as enhance angiogenic factors. According to the results of both in vitro/vivo studies, HZSC can affect macrophage phenotype polarization, stimulate vascular endothelial cell migration and tube formation, accelerate the wound transition into the repair and proliferation stages, and then promote the diabetic wound healing. RNA sequencing analysis supports that HZSC downregulates TNF, NF-κB, and JAK-STAT pathways while upregulating genes associated with anti-inflammation and pro-angiogenesis. Our findings introduce a therapeutic approach to promote diabetic wound healing through H2S/curcumin coreleasing, improving anti-inflammation and pro-angiogenesis.

Near-Infrared Responsive Nanocomposite Hydrogel Dressing with Anti-Inflammation and Pro-Angiogenesis for Wound Healing

Wen Luo,Zhenzhen Li,Junjie Che,Xinyao Li,Huali Zhang,Jinxiu Tian,Chunyang Wang,GuiYing Li,Lin Jin

Anti-inflammatory and angiogenesis are two important factors in wound healing. Wound dressings with anti-inflammation and vascularization are essential to address complex interventions, expensive treatments, and uncontrolled release mechanisms. Based on the above considerations, we designed a near-infrared (NIR)-responsive hydrogel dressing, which is composed of mPDA-DFO@LA nanoparticles (mPDA: dopamine hydrochloride nanoparticles, DFO: deferoxamine, LA: lauric acid), valsartan (abbreviated as Va), and dopamine–hyaluronic acid hydrogel. The hydrogel dressing demonstrated injectability, bioadhesive, and photothermal properties. The results indicated the obtained dressing by releasing Va can appropriately regulate macrophage phenotype transformation from M1 to M2, resulting in an anti-inflammatory environment. In addition, DFO encapsulated by LA can be sustainably released into the wound site by NIR irradiation, which further prevents excessive neovascularization. Notably, the results in vivo indicated the mPDA-DFO@LA/Va hydrogel dressing significantly enhanced wound recovery, achieving a healing rate of up to 96% after 11 days of treatment. Therefore, this NIR-responsive hydrogel dressing with anti-inflammation, vascularization, and on-demand programmed drug release will be a promising wound dressing for wound infection.
