Developing optical oxygen-sensitive nanoparticles with good biocompatibility and low cost to rapidly detect molecular oxygen in a signal turn-on mode is highly desirable. Herein, we present a phenylenediamine-derived carbon dot (CD), which has been found to display a fluorescence-switchable property, with fluorescence quenching by Na2S2O4 and fluorescence recovery triggered by O2. Because of this unique property, we directly employed the quenched CDs as a reversible probe for the fluorescence turn-on detection of molecular oxygen and found that this detection mode possessed a short response time of 12 s and high sensitivity. Moreover, this CD solution could be used as a writeable ink for the patterning of paper strips and security service without any loss in the fluorescence-switchable property. We thus achieved CD-based test strips for oxygen detection in vacuum packaging. This study not only provides a convenient, low-cost and practical way for oxygen detection but also reveals a fluorescence-switchable CD that will find its applications in numerous O2-involved fields.

Dopamine (DA) is a very imperative neurotransmitter in our body, since it contributes to several physiological processes in our body, for example, memory, feeling, cognition, cardiovascular diseases, and hormone secretion. Meanwhile, tyrosinase is a critical biomarker for several dangerous skin diseases, including vitiligo and melanoma cancer. Most of the reported chemiluminescent (CL) methods for monitoring DA and tyrosinase are signal-off biosensors. Herein, we introduce a new chemiluminescent “signal-on” system, lucigenin-tris(hydroxypropyl)phosphine (THPP), for the selective determination of DA and tyrosinase. THPP is well known as a versatile and highly water–soluble sulfhydryl-reducing compound that is more highly stable against air oxidation than common disulfide reductants. By employing THPP for the first time as an efficient lucigenin coreactant, the lucigenin-THPP system has shown a high CL response (approximately 16-fold) compared to the lucigenin-H2O2 classical CL system. Surprisingly, DA can remarkably boost the CL intensity of the lucigenin-THPP CL system. Additionally, tyrosinase can efficiently catalyze the conversion of tyramine to DA. Therefore, lucigenin-THPP was employed as an ultrasensitive and selective signal-on CL system for the quantification of DA, tyrosinase, and THPP. The linear ranges for the quantification of DA, tyrosinase, and THPP were 50–1000 nM, 0.2–50 μg/mL, and 0.1–800 μM, respectively. LODs for DA and tyrosinase were estimated to be 24 nM and 0.18 μg/mL, respectively. Additionally, the CL system has been successfully employed for the detection of tyrosinase in human serum samples and the assay of DA in human serum samples as well as in dopamine injection ampules with excellent obtained recoveries.

As a representative of the third generation of bisphosphonate drugs, zoledronic acid (ZA) is clinically used to treat osteolytic bone metastases from malignant tumors and osteoporosis. Herein, a novel signal-on strategy for highly selective detection of ZA is proposed, employing Fe 3+ -mediated fluorescent sulfur quantum dots (SQDs) as a probe. Specifically, Fe 3+ can induce the aggregation of SQDs in aqueous solution and strongly quench their fluorescence, however, which recovers again in the presence of ZA owing to its emulative complexation with Fe 3+ . The complex ratio and binding mode between ZA and Fe 3+ are also theoretically simulated. This method exhibits the merits of rapidness, high selectivity, reliability, and with no need of derivatization for ZA detection, with a detectable range of 5–200 μM and a detection limit of 1.7 μM. It has also been successfully applied to determine ZA in real samples including human serum, urine, and drugs.

As a first-line tuberculostatic drug, isoniazid (INH) plays effective and irreplaceable role in prevention and treatment of tuberculosis. In this work, a rapid and simple signal-on fluorescence approach is established for INH assay by employing a platform composed of silver nanoclusters (AgNCs) and MnO 2 nanosheets. In the proposed sensing system, strong red fluorescence of poly (methacrylic acid)-stabilized AgNCs can be greatly quenched after they attach to the surfaces of MnO 2 nanosheets. With the addition of INH, MnO 2 nanosheets are reduced to Mn 2+ and subsequently release the AgNCs, which leads to obvious fluorescence recovery again. Based on this mechanism, highly sensitive detection of INH in the range of 0.8–200 μM is realized (detection limit: 476 nM). The present strategy shows remarkable advantages including simplicity, rapidness, high sensitivity and wide detectable range. This method is also practical and comparable to high-performance liquid chromatography, which can be applied to detect INH in human urine and serum samples as well as pharmaceutical products.

Antibiotics are emerging hazardous small molecules, requiring urgent need for fast signal-on analytical methods to control antibiotic abuse. Signal-on fluorescence sensing strategies utilizing nanoquenchers and aptamers are fascinating but rarely accomplished on paper-based platforms. Here, a novel multilayer MXene sensing platform was established by leveraging Nb 2 C-MXene as a multilayer nanoquencher and zero-dimensional carbon dots-labeled aptamer (B-CDs@Apt) as a stable and bright recognition probe. The Nb 2 C-MXene has a multilayer nanosheet stack-like structure and efficient mass transfer channels. It can efficiently adsorb abundant B-CDs@Apt probes and quench their fluorescence. Importantly, the Nb 2 C-MXene/B-CDs@Apt system can release the B-CDs@Apt in response to the analyte with high sensitivity, thereby restoring the fluorescent signal. The developed aptasensor achieved sensitive and selective detection of chloramphenicol (CAP) and showed satisfactory anti-interference ability, stability, and practicability. Notably, the multilayer Nb 2 C-MXene/B-CDs@Apt system was successfully transferred to a paper-based sensing platform, with a low-density distribution of multilayer nanoquenchers carrying sufficient aptamer probes for analyte access. In comparison, the monolayer Nb 2 C nanosheets were unable to adsorb enough probes to output analyte-induced signals. The established paper-based analytical device (PAD) showed a LOD of 0.360 ng mL −1 for CAP, which is the first paper-based MXene aptasensor reported for fluorescence detection. By replacing the aptamer and carbon dot, the strategy was further extended to detect another analyte oxytetracycline (OTC), with LODs of 0.399 in tube and 0.867 ng mL −1 on PAD, respectively. Furthermore, concurrent detection of CAP and OTC was achieved using a dual-color PAD, demonstrating the potential to meet multi-target analytical requirements.

A signal-on photoelectrochemical (PEC) biosensing platform was developed here for the highly sensitive detection of a cancer marker type IV collagenase, which relied on the exciton energy transfer (EET) between CdTe quantum dots (QDs) and Ag nanoparticles (NPs) combined with the catalytic hydrolysis of a protease. A special sequence peptide chain with positively charged arginines at one end and a cysteine-labeled terminal at the other end, which could be modified with Ag NPs, was designed and synthesized. CdTe QDs and synthetic peptide were assembled onto the surface of indium–tin oxide (ITO) electrodes by means of a layer-by-layer self-assembly technology. The as-obtained modified electrodes generated a lower photocurrent under irradiation due to the effect of EET. The photocurrent intensity increased due to the addition of type IV collagenase, which could specifically hydrolytically cleave Gly–Pro–Ala, leading to Ag NPs being away from the electrode surface. Thus, a signal-on PEC biosensor could be obtained. Under optimum conditions, the PEC biosensor for type IV collagenase exhibited a linear range from 0.5 to 50 μg mL−1 and sensitivity of 96 ng mL−1. The peptide-QD-based PEC detection against type IV collagenase showed a promising potential for other protease or kinase detection when we changed the substrate of the peptide.

α-synuclein oligomer is a marker of Parkinson's disease. The traditional enzyme-linked immunosorbent assay for α-synuclein oligomer detection is not conducive to large-scale application due to its time-consuming, high cost and poor stability. Recently, DNA-based biosensors have been increasingly used in the detection of disease markers due to their high sensitivity, simplicity and low cost. In this study, based on the DNAzyme-driven DNA bipedal walking method, we developed a signal-on electrochemical sensor for the detection of α-syn oligomers. Bipedal DNA walkers have a larger walking area and faster walking kinetics, providing higher amplification efficiency compared to conventional DNA walkers. The DNA walker is driven via an Mg 2+ -dependent DNAzyme, and the binding-induced DNA walker will continuously clamp the MB, resulting in the proliferation of Fc confined near the GE surface. The linear range and limit of detection were 1 fg/mL to 10 pg/mL and 0.57 fg/mL, respectively. The proposed signal-on electrochemical sensing strategy is more selective. It will play a significant role in the sensitive and precise electrochemical analysis of other proteins.

Hydrogen sulfide (H2S) is a dramatically toxic gas molecule; long term exposure to H2S environment poses a serious threat to human health. It is highly imperative to develop sensitive and selective methods for H2S detection. In this study, a signal-on fluorescence biosensor for the H2S (used to represent the total amount of H2S, HS− and S2−) detection was developed based on silver-mediated base pairs (C–Ag+–C). Two separate C-rich single-stranded DNAs (ssDNAs) were synthesized, namely, one was labeled with fluorescein (6-carboxyfluorescein, FAM) and the other was labeled with quencher (black hole quencher, BHQ1). The two ssDNAs were in a flexible single strand owing to the mismatch of the ssDNA, and hence, significant fluorescence intensity was observed. In the presence of Ag+, DNA probes could selectively capture Ag+ to form double stranded DNA (dsDNA) via the C–Ag+–C base pairs, which made the quench of FAM by BHQ1 and low fluorescence signal was detected. With the addition of H2S to the system, the dsDNA was decomposed into ssDNA since Ag+ could be released from C–Ag+–C by the formation of a more stable complex Ag2S, which resulted in the recovery of the fluorescence intensity. The enhanced fluorescence intensity had a linear relationship with H2S concentration in the range from 0.5 to 20 μM with a detection limit (LOD) of 0.16 μM (S/N = 3). The proposed system was applied to detect H2S (used to represent the total amount of H2S, HS− and S2−) in air near a garbage station with satisfying results.

In this work, a “signal on” electrochemiluminescence (ECL) strategy was proposed to detect Apolipoprotein-A1 (Apo-A1), a common tumor biomarker for bladder cancer. The tris(2,2′-bipyridyl)-ruthenium(II) [Ru(bpy) 3 2+ ] was used as luminophor captured by three-dimensional (3D) zinc oxalate metal–organic frameworks (MOFs), which constructed as the ECL nanoprobe (Ru-MOF) for detecting Apo-A1. Meanwhile, L-cysteine (L-Cys) as the co-reactant was connected to the [email protected] core–shell nanostructure through Au-S bond, which was directly modified on the glassy carbon electrode (GCE) to significantly reduce the electron transfer distance between the luminophor and co-reactant. The ECL intensity was “signal on” when the ECL probe of Ru-MOF was linked to the electrode by the immunoreaction of antigen–antibody. Based on these principles, the quantitative relationship between the concentration of Apo-A1 and the ECL intensity was obtained in the range of 1.00 fg·mL −1 to 1.00 ng·mL −1 with a linear coefficient of 0.9968. The proposed ECL immunosensor showed some great advantages, such as simplification, rapidity, stability and selectivity. It was also successfully applied for the determination of Apo-A1 in real serum and urine samples with the recovery of 98.0% ∼ 115.0% (RSD < 5.2%). To sum up, a “signal on” ECL immunosensor was successfully constructed for Apo-A1 detection, and it may facilitate the diagnosis of bladder cancer in clinic.

In this work, we tactfully constructed an electrochemiluminescence (ECL) aptasensing platform with ferrocene-graphene nanosheets (Fc-GNs) using the ferrocene (Fc) as quench unit to tris(2.2′-bipyridyl) ruthenium (II) [Ru(bpy) 3 2+ ] and the Ru(bpy) 3 2+ tagged thrombin binding aptamer (Ru-TBA) to recognize the thrombin molecules. Duing to the unique π-π interaction between nucleotides and graphene, the Ru-TBA could be preferentially adsorbed on the surface of ferrocene-graphene nanosheets with the signal generator into off-state. The conformational transformation of Ru-TBA leads to the desorption of Ru-TBA from Fc-GNs after the biosensing electrode incubating with the thrombin solution and the ECL “signal-on” was triggered. With the transformation of luminescence signal from “off” to “on”, the biosensor exhibited high sensitivity for the determination of thrombin with a detection limit of 0.21 nM. Particularly, the proposed method could be widely applied to the aptamer-based determination of other target analytes.

In this work, an in situ growth protocol is introduced to fabricate three-dimensional graphene films (3D GFs) on gold substrates, which are successfully utilized as working electrode for electrochemical detection of nucleic acid (microRNA) and protein (lysozyme) based on a signal-on sensing mechanism. To realize the bridge between the gold substrate and graphene film, a monolayer of 4-aminothiophenol is self-assembled on the substrate, which is then served as connectors for the growth of 3D GFs on the gold substrate by the hydrothermal reduction (HR) technique. Moreover, given the excellent properties, such as enlarged surface area, strong binding strength between 3D GFs and gold substrate, and improved conductivity, the proposed 3D GF-fabricated gold substrate is readily employed to the construction of electrochemical biosensing platforms through introduction of magnetic nanoparticles (MNPs) as probe carriers. On the basis of the strand displacement reaction and specific binding between aptamer and its target, the developed biosensors achieve signal-on detection of microRNA-155 (miR-155) and lysozyme (Lyz) with high sensitivity and selectivity and further successfully applied to human serum assay. Overall, the proposed strategy for in situ growth of 3D GFs provides a powerful tool for a wide range of applications, which is not limited to electrochemical biosensors and can be extended to other areas, such as electrocatalysis and electronic energy-related systems.

Amino-modified carbon dots (C-dots) with positively charged surface were prepared. They display strong blue fluorescence and are shown to act as quenchers of the green fluorescence of FAM-labeled ssDNA such as the F-probe used in this work that was immobilized on the C-dots. On the addition of highly negatively charged heparin (Hep), it will interact with the C-dots and displace the F-probe from C-dots. Once the F-probe is displaced by Hep, its green fluorescence is restored. The intrinsic blue fluorescence of the C-dots remains stable after addition of Hep. Thus, a signal-on ratiometric fluorometric assay was developed for the ultra-sensitive detection of Hep. The underlying mechanisms of quenching and recovery are discussed. Under optimized conditions, the recovery of the ratiometric fluorescence of the system composed of C-dots and quenched F-probe is proportional to the Hep concentration in the range of 0.01–2.0 μg·mL −1 (= 0.00125–0.25 U·mL −1 ). The method was successfully applied to the determination of Hep in spiked serum samples.
Schematic of a signal-on ratiometric fluorometric method for the ultra-sensitive detection of heparin on the basis of the displacement and fluorescence enhancement of adsorbed FAM-labeled ssDNA from amino-modified carbon dots (C-dots) by heparin.
