Study protocol for One Health data collections, analyses and intervention of the Sino-Swedish integrated multisectoral partnership for antibiotic resistance containment (IMPACT)

Qiang Sun,Yang Wang,Anette Hulth,Yonghong Xiao,Lennart E Nilsson,Xuewen Li,Zhenwang Bi,Yuqing Liu,Hong Yin,Yanbo Luo,Maud Nilsson,Chengtao Sun,Yiqing Zhu,Beiwen Zheng,Baoli Chen,Pan Sun,Lilu Ding,Xi Xia,Jakob Ottoson,Sonja Löfmark,Oliver James Dyar,IMPACT Consortium,Stefan Börjesson,Cecilia Stalsby Lundborg

Introduction To effectively minimise the emergence and dissemination of antibiotic resistant bacteria, a holistic One Health approach is called for. The Sino-Swedish Integrated Multisectoral Partnership for Antibiotic Resistance Containment is a cross-sectoral and integrated project on antibiotic resistance, conducted in Shandong Province in China. This paper outlines the overall study protocol for the project. To our knowledge, this is the first research programme aiming to take a true holistic approach across multiple sectors simultaneously in China, and the first to incorporate both antibiotic use and infection prevention and control in addition to antibiotic resistance patterns. The project aims to address gaps in current knowledge and seeks to improve the situation through a system-wide intervention. By using a One Health approach we can address important research questions that individual discipline investigations are unable to. The results obtained should thus more closely reflect the world in which human health, animal health and the environment are inextricably and intimately interlinked. Methods and analysis Both quantitative and qualitative studies are included for households from 12 villages, their surrounding environment and a tertiary care hospital in a nearby town. The studies include analyses of antibiotic consumption for humans and pigs; qualitative and quantitative data on perceptions, knowledge and attitudes; faecal carriage of extended spectrum β-lactamase and carbapenemase-producing Enterobacteriaceae from pigs and humans, and occurrence in household drinking water, surface water, waste water and clinical bacterial isolates from the hospital. Carriage of methicillin-resistant Staphylococcus aureus in humans, household pigs and clinical bacterial isolates is also investigated. Furthermore, potential inter-relationships between these sources are analysed. A multifaceted One Health intervention is designed and implemented in 6 of the 12 villages. Repeated and continuous data collections take place over 2 years, where the repeated data collection is performed after 1 year of intervention. Comparisons are made between intervention and control villages, before and after the intervention. Ethics Ethics approval was obtained from the first Affiliated Hospital, College of Medicine, Zhejiang University, China, reference number 2015#185 and 2015#283.

Arsenic exposure analysis for children living in central China: From ingestion exposure to biomarkers

Beibei Wang,Hongguang Cheng,Chunye Lin,Xuan Zhang,Xiaoli Duan,Qin Wang,Dongqun Xu

Emerging evidence indicates that chronic low-dose arsenic (As) exposure can pose adverse health effects to children. This study aimed to systematically study the exposure risk induced by As ingestion in children living in Hubei Province, central China. The feasibility of first morning spot urine instead of 24-h urine as an environmental exposure biomarker was also explored. A total of 120 children aged 2–17 years were recruited from an urban area for the collection of biomarker samples (first morning and 24-h urine samples), environmental exposure samples (duplicate diets, drinking water , and soil), and related child-specific exposure factors. The external exposure risk, internal exposure level, and source of exposure to As in children were analyzed. The results indicated that As concentration in duplicated diets, water, and soil were 29.2 μg kg −1 , 1.3 μg L −1 , and 9.3 mg kg −1 , respectively; these were all below the corresponding maximum allowable levels in China (the threshold value of As in most food, drinking water and soil are 0.5 mg⸱kg −1 , 0.01 mg L −1 , and 20 mg⸱kg −1 , respectively). Dietary intake was the predominant exposure route, accounting for 90% of the total daily dose. The combined oral non-carcinogenic and carcinogenic risks all exceeded the corresponding maximum acceptable risk level . Therefore, As bioavailability should be investigated and used in health risk assessment. Multiple linear regression analysis indicated that urinary As was positively associated with dietary As, with a one-unit increase in daily As intake from the diet associating with 4.82 and 5.21 μg g −1 increases in 24-h urine and first morning urine, respectively. Furthermore, significant correlations with 24-h urine and external exposure metrics suggested that creatine-adjusted As concentrations in first morning urine could be an appropriate substitute of 24-h urine as exposure biomarkers.

The impact of Bifidobacterium breve BBr60 (BBr60) on metabolic and gastrointestinal health in healthy adults: A combined in vitro metabolomic and randomized, double-blind, placebo-controlled study

Yao Dong,Yihui Cai,Hairui Tian,Jian Wen,Mei Han,Yan Tan,Kang Zou

Summary Purpose This study aims to evaluate the physiological and psychological health impacts of Bifidobacterium breve BBr60 (BBr60) on healthy adults. Methods The study was divided into preclinical research and a clinical trial phase. In the preclinical study, the bioactive components of BBr60 were analyzed through LC-MS targeted metabolomics. In the clinical trial, 109 healthy adults aged 19–45 were recruited to participate in an 8-week double-blind, randomized, placebo-controlled study. Participants were randomly assigned to receive either a BBr60 supplement (10 10 CFU/day) or a placebo (maltodextrin). Throughout the study, liver function, metabolic markers, cardiovascular and body composition metrics were assessed. Volunteer scores on the Alcohol Dependence Scale (ADS), Nepean Dyspepsia Index (NDI), Gastrointestinal Symptom Scale (GIS), and Positive and Negative Affect Schedule (PANAS) were recorded, and the impact on the intestinal microbiota structure and function of healthy participants was analyzed through 16S rRNA sequencing. Results The preclinical study indicated the role of BBr60 in modulating key metabolic pathways, including those involved in ABC transporters, arginine, proline, and tryptophan metabolism. Clinical trial results demonstrated significant improvements in high-density lipoprotein (HDL) levels and reductions in total cholesterol with BBr60 supplementation. Gastrointestinal symptoms and emotional states also improved, with significant reductions in ADS scores ( p = 0.000), particularly in symptoms related to alcohol consumption such as post-drinking memory loss and tremulous delirium. Additionally, significant enhancements in gastrointestinal health were observed, as indicated by a notable decrease in NDI scores ( p = 0.002), suggesting reduced impact of stomach issues on daily life. GIS scores further confirmed improvements in gastrointestinal function, particularly in early satiety and appetite. While overall emotional changes were not significant, positive emotional scores increased post-intervention, and negative emotional scores decreased. The probiotic demonstrated good safety and tolerability throughout the 8-week trial period. Conclusion Supplementation with BBr60 not only significantly alleviated neuropsychiatric symptoms associated with alcohol consumption but also showed potential benefits in enhancing gastrointestinal and metabolic health. These findings support further research into BBr60 as a potential non-pharmacological intervention to improve both psychological and physiological health. Trial Registration Number : NCT06196892 ( ClinicalTrials.gov ).

Integrative analysis of microbiome and metabolome reveals the effect of deoxynivalenol on growth performance, liver and intestinal health of largemouth bass (Micropterus salmoides)

Yi lian Yin,You xing Xu,Meng ting Wang,Dao zhi Yang,Xiao yan Wang,Xian ke Zhou,Meng meng Huang,Shun Yang,Hui Fei

This study explored the toxic effects of deoxynivalenol (DON) intake at low (100 µg·Kg −1 , LD group) and high doses (300 µg·Kg −1 , HD group) on largemouth bass ( Micropterus salmoides ). After a 56-day feeding trial, the HD group exhibited significantly reduced growth performance and weakened antioxidant capacity, along with elevated activities of enzymes related to metabolic dysregulation. Histopathological analysis showed an increase in hepatic cell vacuoles, as well as a shortened intestinal villi in both LD and HD groups. Notably, high-dose DON intake markedly down-regulated the expression of intestinal epithelial tight junction-related genes and proteins. Microbiome analysis indicated a significant increase in the Delftia and Acinetobacteria relative abundance, accompanied by diminished in Mycoplasma abundance in the HD group. Metabolomic profiling demonstrated that the dysregulated metabolites were mainly associated with the lysine biosynthesis pathway. Integrative multi-omics analysis revealed that pyridoxamine and diferuloylputrescine served as key biomarkers associated with Acinetobacteria . Collectively, we confirmed that high-dose DON intake induces hepatointestinal damage, thereby impairing the growth performance of largemouth bass. Our findings also further highlighted the link between DON-induced gut microbiota alterations and metabolic disorders, offering new intervention targets for alleviating DON toxicity.

Improving arsenic and cadmium contaminated paddy soil health and rice quality with plant-animal-based modified biochar: A mechanistic study

Md Shafiqul Islam,Hui Deng,Youming Dong,Junhua Zhu,Minling Gao,Zhengguo Song

Arsenic and cadmium (As/Cd) are toxic elements which negatively impact the health and productivity of rice plants by subjecting them to stress. To address these issues, we developed three biochar amendment materials derived from plant-based corn-stover with iron-magnesium modification (FM@CB), char sourced from animal-based eggshells with iron-magnesium modification (FM@EB), and a blend (1:1, w/w) of both (FM@CEB). Batch adsorption experiment revealed that FM@CB exhibited a Langmuir sorption capacity of 77 mg g −1 for As(III) and 107 mg g −1 for Cd(II). In contrast, FM@EB showed values of 63 mg g −1 for As(III) and 115 mg g −1 for Cd(II), both observed in mixed systems. The exothermic dual adsorption mechanisms could include ion exchange, cation-anion bridging, complexation with surface functional groups, and precipitation processes. Pot trials were conducted, and the findings unveiled a substantial enhancement in rice grain yield with a 2% application of FM@CB, FM@EB, and FM@CEB. Specifically, there was a notable increase of 21.9%, 34.9%, and 37.6%, respectively. Simultaneously, the application led to a reduction in grain As levels by 34.4%, 53.2%, and 42.6%, and grain Cd levels by 48.7%, 71.2%, and 59.9%, respectively, in comparison to the no-amendment control condition. Our study observed a transformation in the soil's microbial community, as beneficial bacterial genera such as Anaeromyxobacter , Citrifermentans , and Desulfovibrio emerged to assist in the detoxification of these metal contaminants. Concurrently, the application of these materials led to a reduction in harmful bacterial genera like Pseudomonas , Flavobacterium , and Massilia . In addition, these amendments decreased the bioavailable As/Cd fractions in the soil and promoted the root-iron-plaque, which effectively sequesters these contaminants, rendering rice quality improvement. Ultimately, the use of these amendments, particularly FM@CEB, holds the promise of protecting rice from harmful metal(loid)s contaminants, increasing yields, and revitalizing the soil. The presence of these unique bacterial genera, especially those capable of detoxifying As/Cd, plays a crucial role in ensuring a healthier and more secure environment for all living organisms.

Transport, pollution, and health risk of heavy metals in “soil-medicinal and edible plant-human” system: A case study of farmland around the Beiya mining area in Yunnan, China

Jiayi Hu,Xiaofang Yang,Huajian Chi,Xin Liu,Ning Lu,Ya Liu,Shengchun Yang,Xiaodong Wen

Heavy metals (HMs) pose a significant threat to soil quality, plant growth, and human health. However, the impacts (transport, pollution, and health risk) of HMs in medicinal and edible plants with rich value growing in high background soil for a long time remain unclear. So the effects of HMs on the “soil-medicinal and edible plant-human” system were comprehensively studied. Houttuynia cordata Thunb. (HCT) and growing soil were collected from the farmland around Beiya mining area located in Three Parallel Rivers of Yunnan Protected Areas, China. The transport, pollution, and health risks of HMs in the system were investigated by determining the content of active ingredients, HMs, and physicochemical properties in HCT and soil. The results of comprehensive analysis using various measurement and analysis methods showed that Zn, Cu, Mn, and Co in soil could affect the contents of available nutrients (available P (AP), available K (AK), and organic matter (OM)), while AP, OM, pH, Co, and Fe in soil were closely related to the formation of HCT quality. Cr and Fe had the strongest transport capacity in stems and leaves of HCT, respectively. Furthermore, it is worth noting that Cr, Pb, and Cu had caused serious pollution to HCT in the area. Pb and Cu are tolerable levels for human health and may pose a carcinogenic risk. Our study demonstrates that HCT is expected to be a marker of heavy metal pollution in the area and provides a scientific basis for making effective regulations on HMs control and scientific planting.

Comprehensive evaluation of the risk system for heavy metals in the rehabilitated saline-alkali land

Xin Xu,Lin Guo,Shaobo Wang,Meng Ren,Pengjie Zhao,Ziyi Huang,Hongjun Jia,Jinhang Wang,Aijun Lin

A comprehensive assessment of the heavy metal system in the rehabilitated saline-alkali land holds significant importance, as the in-situ remediation process utilizing amendments substantially alters the initial physicochemical properties of the soil, which could lead to the migration or reactivation of previously stabilized heavy metals. In this context, the present study aims to evaluate the heavy metal content and health risk within the improved saline-alkali soil-plant system. Moreover, a comprehensive evaluation based on the TOPSIS-RSR method is carried out to accurately gauge the soil health status. The findings indicate that the modification process has an impact on the concentrations of heavy metals in the soil and crops, causing either an increase or decrease. However, the level of heavy metal pollution in the improved saline-alkali soil and rape remains within safe limits. The results of the migration of heavy metals after amendment application indicated that the migration of heavy metals in the soil was influenced by the properties of the heavy metals, the composition of the amendment, and leaching. Furthermore, the total non-carcinogenic hazard quotients in the soil and rape were within the safe threshold for all populations. The findings provided novel insights into the status and risk assessment of the pollution of improved saline-alkali soil.

Accumulation behavior and risk assessment of heavy metals and arsenic in tissues of white bream (Parabramis pekinensis) from the Xiang River, southern China

Jia Yuyu,Kong Qian,Yang Zhaoguang,Wang Lin

Aquatic environment suffered from serious heavy metal and As pollutions due to rapid industrialization and urbanization in the last decades. In order to evaluate the heavy metal and As contamination in aquatic organism in southern China, the levels of As, Cd, Co, Cr, Cu, Fe, Mn, Pb, and Zn in muscle, gill, and liver of white bream ( Parabramis pekinensis ) collected from Xiang River were analyzed. The statistically significant differences of the analyzed element concentration (wet weight) between tissues were determined through one-way analysis of variance (ANOVA). The liver presented high affinity for accumulation of Cd, Cu, and Zn due to the binding activity of metallothionein proteins. Mn and Pb mainly accumulated in the gill indicated the major uptake route through the gill. Lack of correlation between analyzed element and fish size might imply no or weak biomagnification of the analyzed elements in fish tissues. The potential health risk associated with exposure to heavy metals and As was evaluated. The uncertainty of target hazard quotient was calculated by Monte Carlo simulation. The results indicated that the analyzed element contents in the white bream muscle posed relatively low-chronic toxic risk to consumers.

Arsenic speciation and heavy metal distribution in polished rice grown in Guangdong Province, Southern China

Li Ma,Lin Wang,Jie Tang,Zhaoguang Yang

Arsenic speciation and heavy metal distributions have been investigated in locally grown rice grains from Guangdong Province, Southern China. A total of 41 polished rice grain samples were collected throughout Guangdong Province. Arsenite (As(III)), as the predominant form found in the rice, was positively correlated ( p   <   0.01) with total As (tAs) concentration. However, the percentage of As(III) reduced while tAs concentration increased (r   =   −0.361, p   <   0.05), due to restricted accumulation and translocation of As(III) in rice grains at high level of tAs. Statistical and geostatistical analyses were applied to investigate potential origins of heavy metals in rice. Only Cd, Cu and Ni were identified as influenced by anthropogenic sources such as industrial and commercial activities. As and Pb were primarily controlled by natural occurrence. The results of health risk assessment implied that continuous intake of rice grown in Guangdong Province could cause considerably non-carcinogenic and carcinogenic risk to local inhabitants.

A comparison of accumulation and depuration effect of dissolved hexavalent chromium (Cr6+) in head and muscle of bighead carp (Aristichthys nobilis) and assessment of the potential health risk for consumers

Jiaojiao Yin,Li Wang,Yuanyuan Chen,Dianfu Zhang,Abeer M. Hegazy,Xuezhen Zhang

In this study, bighead carp ( Aristichthys nobilis ) were exposed to waterborne Cr 6+ of 0.01, 0.1, 1 and 5 mg/L for 14 days and subsequently transferred to clean water for another 14 days. The Cr 6+ contents in some edible parts, such as dorsal muscle, ventral muscle and head were detected. The Cr 6+ concentrations in the three parts were in the order of: head > ventral muscle > dorsal muscle with significant increase during exposure period and remarkable decrease when kept at clean water during depuration stage. The head contained higher fat than that of muscle and the Cr 6+ levels of these parts showed significantly positive correlation with fat content, however, the Cr 6+ contents in the separated fat were extremely low. The Cr 6+ levels determined in tissues can tell a real story rather than the correlation coefficient. Fish head poses a higher potential health risk than muscle due to heavy metals pollution.

Exposure of street sweepers to cadmium, lead, and arsenic in dust based on variable exposure duration in zinc smelting district, Northeast China

Xiaoqian Li,Yan Yu,Na Zheng,Sujing Wang,Siyu Sun,Qirui An,Pengyang Li,Yunyang Li,Shengnan Hou,Xue Song

Street dust is an important contributor to heavy metal exposure in street sweepers. In this work, the levels of cadmium (Cd), lead (Pb), and arsenic (As) in street dust were evaluated, and the corresponding health implications for street sweepers in the smelting district of Huludao city northeast China were assessed. The contributions of dietary sources and dust to total metal exposure in street sweepers were compared. Because street sweepers are exposed to street dust both during work and nonwork hours, the health risks faced by street sweepers are uncertain. Therefore, variable exposure duration was considered using a deterministic model. A probabilistic risk assessment model was developed to explore the health effects of street dust on street sweepers via Monte Carlo simulation. The various exposure parameters that affect risk were analyzed using sensitivity analysis. The average Cd, Pb, and As levels in the hair of street sweepers were 2.04, 20.12, and 0.52 mg·kg -1 , respectively. These values were higher than those for residents (i.e., not street sweepers) of Huludao. Strong correlations were found between the logarithms (base 10) of the Cd, Pb, and As contents in dust and hair ( r Cd  = 0.581, p  < 0.01; r Pb  = 0.428, p  < 0.01; r As  = 0.378, p  < 0.01; n  = 62). Based on analysis using deterministic models, the maximum exposures to Cd and Pb via dust through the alimentary canal were nearly three and six times higher than the dietary exposures, respectively. Sensitivity analysis indicated that exposure duration is an important parameter.

Benzophenone-type ultraviolet filters inhibit human and rat placental 3β-hydroxysteroid dehydrogenases: Structure-activity relationship and in silico docking analysis

Hao Lin,Shaowei Wang,Yunbing Tang,Zhiyan Hu,Xiaofang Chen,Huitao Li,Yang Zhu,Yiyan Wang,Yi Liu,Ren-shan Ge

Benzophenones (BPs) are a class of chemicals found in various personal care and cosmetic products, such as sunscreens and lotions. Their usage is known to cause reproductive and hormonal health risks, but the exact mechanism of action remains unknown. In this study, we investigated the effects of BPs on human and rat placental 3β-hydroxysteroid dehydrogenases (3β-HSDs), which play a crucial role in the biosynthesis of steroid hormones , particularly progesterone . We tested inhibitory effects of 12 BPs, and performed structure-activity relationship (SAR) and in silico docking analysis. The potency of BPs to inhibit human 3β-HSD1 (h3β-HSD1) is BP-1 (IC 50 , 8.37 μM)>BP-2 (9.06 μM)>BP-12 (94.24 μM)>BP-7 (1160 μM) >BP-8 (1257 μM) >BP-6 (1410 μM) > other BPs (ineffective at 100 μM). The potency of BPs on rat r3β-HSD4 is BP-1 (IC 50 , 4.31 μM)>BP-2 (117.3 μM)>BP-6 (669 μM) >BP-3 (820 μM)>other BPs (ineffective at 100 μM). BP-1, BP-2, and BP-12 are mixed h3β-HSD1 inhibitors and BP-1 is a mixed r3β-HSD4 inhibitor. LogP, lowest binding energy, and molecular weight were positively associated with IC 50 for h3β-HSD1, while LogS was negatively associated with IC 50 . The 4-OH substitution in the benzene ring plays a key role in enhancing the effectiveness of inhibiting h3β-HSD1 and r3β-HSD4, possibly through increasing water solubility and decreasing lipophilicity by forming hydrogen bonds . BP-1 and BP-2 inhibited progesterone production in human JAr cells. Docking analysis shows that 2-OH of BP-1 forms hydrogen bonds with catalytic residue Ser125 of h3β-HSD1 and Thr125 of r3β-HSD4. In conclusion, this study demonstrates that BP-1 and BP-2 are moderate inhibitors of h3β-HSD1 and BP-1 is a moderate inhibitor of r3β-HSD4. There is a significant SAR differences for 3β-HSD homologues between BPs and distinct species-dependent inhibition of placental 3β-HSDs.
