Main Chemical Components, Activity and Mechanism of Repellence of Cyperus esculentus Essential Oil Against Tribolium confusum

Xu Feng,Cheng-Bin Shan,Jian-Nan Ma,Yue Ma,Na Li,De-Jian Zhang,Zhan-Yuan Lu,Chao-Mei Ma

Tribolium confusum is a major stored-product pest that exhibits resistance to chemically synthesized pest repellents. This study investigated the potential of essential oil (EO) extracted from the roots of Cyperus esculentus as a natural alternative for pest management. The EO was obtained through steam distillation, and its chemical composition was elucidated using gas chromatography–mass spectrometry. The primary compounds, cyperotundone and cyperene, were further isolated from the EO through silica gel column chromatography. The efficacy of the EO and its isolated compounds as pest repellents was evaluated against a flaxseed pest, which was identified as T. confusum through DNA sequence analysis. The results demonstrated that at 86.12 μg/cm 2 , the EO and its two main components maintained significant repellent activity for up to 24 h. In contrast, the effectiveness of the positive control, N, N-diethyl-3-methylbenzamide (DEET) declined rapidly after 8 h. At 16 h, the repellent activity of the EO and one of its main components, cyperotundone, was significantly greater than that of DEET. Furthermore, at a lower concentration of 43.06 μg/cm 2 , cyperotundone’s repellent activity was significantly stronger than DEET’s at 16 h. Additionally, cyperotundone outperformed DEET significantly from 4 to 16 h at 21.53 μg/cm 2 and at 16 h at 10.76 μg/cm 2 . Among the two compounds, cyperotundone exhibited a longer-lasting repellent effect compared to cyperene, which is consistent with the lower evaporation rate of cyperotundone. Biochemical assays revealed that exposure to the EO of C. esculentus and its major compounds significantly reduced ( p < 0.05) the activities of acetylcholinesterase and glutathione-S-transferase in T. confusum . Molecular docking experiments indicated that the compounds could bind to olfactory receptors with low binding energies. qRT-PCR analysis revealed that the EO and its two compounds significantly altered ( p < 0.05) the expression levels of odorant receptor genes in the pest. These findings suggest that the repellent action of C. esculentus EO and its major compounds on T. confusum may be mediated through the modulation of the pest’s olfactory system, as well as by inhibiting essential enzymatic activities in the pests. This research contributes valuable insights into the development of sustainable, long-lasting, and eco-friendly pest repellents, harnessing the potential of the rich botanical resource C. esculentus .

Volatiles of the Predator Xylocoris flavipes Recognized by Its Prey Tribolium castaneum (Herbst) and Oryzaephilus surinamensis (Linne) as Escape Signals

Shaohua Lu,Li Yang,Zonglin Wu,Mingshun Chen,Yujie Lu

Simple SummaryThe sense of smell helps insects perform essential behaviors like finding mates and food and avoiding dangers. Predators use unique smells to locate prey, but it is less understood if prey can recognize these predator odors and use them to escape. In this study, we examined howTribolium castaneum(Coleoptera: Tenebrionidae) andOryzaephilus surinamensis(Coleoptera: Silvanidae), two common pests, react to the predatorXylocoris flavipes, widely used in pest control. We found that these pests avoidX. flavipesodors, particularly the volatiles, linalool and geraniol, which decrease their attraction to food sources. These compounds show strong potential as natural repellents for pest management. The olfactory sensory system plays vital roles in daily activities, such as locating mate partners, foraging, and risk avoidance. Natural enemies can locate their prey through characteristic volatiles. However, little is known about whether prey can recognize the volatiles of their predators and if this recognition can increase the efficiency of prey escaping from predators.Xylocoris flavipesis a predator ofTribolium castaneum(Herbst) andOryzaephilus surinamensis(Linne) that has been widely used in stored pest control. Herein, we analyze the volatile components ofXylocoris flavipesand their impacts on the olfactory behavior ofT. castaneumandO. surinamensis. We found thatT. castaneumandO. surinamensispreferred blank air rather than odors ofX. flavipesandX. flavipesemissions, which significantly decreased the orientation preference ofT. castaneumandO. surinamensisto wheat.X. flavipesemits three major volatiles, including linalool, α-terpineol, and geraniol. Y-tube bioassays showed thatT. castaneumandO. surinamensiscan recognize linalool and geraniol at certain concentrations, especially at 200 μg/mL. EAG recordings verified that linalool and geraniol elicit higher olfactory responses in the two pests, but very small EAG responses were observed in the insects to α-terpineol. A further repellency evaluation also proved that linalool and geraniol are repellent to the two pests, and this repellency can be slightly enhanced by mixing them together.T. castaneumandO. surinamensiscan recognize the predatorX. flavipesby perceiving its volatiles and using them as signals for escaping. The two most potent volatiles, linalool and geraniol, may have potential values as repellents in controlling pests in these two stored products.

Polydopamine-Doped Porous Nano-CaCO3-Supported Green Tablets for the Humidity-Responsive Intelligent Release of Active Ingredients and Control of Grain Storage Pests

Wen-kui Li,Xiang-yun Fu,Yu-zhen Wu,Yong-hong Wang,Jing Zhang,Zhi-qing Ma

Green tablets for grain storage are urgently required, and intelligently controlled release of active ingredients based on functional nanomaterials is a promising strategy for the preparation of green and high-efficiency tablets. In this work, a polydopamine-doped nano-CaCO3 (nanoPCaCO3) with a diameter of 60–88 nm was prepared by a gas diffusion pore-opening strategy. The porous nanoPCaCO3 exhibited superior encapsulation capacity toward phenolic and ketone compounds based on pore filling, hydrogen bonding, and π–π interactions. After functional disintegrants and excipient addition, two kinds of nanoPCaCO3 tablets containing carvacrol (18.0%) and carvone (16.7%) were prepared, which had a novel humidity-controlled release mode based on the humidity-responsive reaction between nanoPCaCO3 and hygroscopic solid acids. At low relative humidity (RH), grain storage pests were hard to survive, and the release rates of active ingredients in the tablets were slow; at high RH, grain storage pests grew well, and the release rates of active ingredients were accelerated correspondingly. As a result, the 7 day cumulative release ratios of carvacrol and carvone from the tablets at 75% RH were approximately 3 times that at 40% RH. Besides, the tablets exhibited desirable fumigation activity; at 75% RH, the 19 day inhibition ratio against Aspergillus flavus attained 73.7%, and the 7 day fumigation mortality against Sitophilus zeamais reached 92.7%. The tablets can be efficiently used for grain storage pest control, and the porous nanoPCaCO3-supported humidity-controlled release mode can be extended for more high efficiency and functional medicament development.

Bioactivities of 3-Butylidenephthalide and n-Butylbenzene from the Essential Oil ofLigusticum jeholense against Stored-product Insects

Cheng Luo,Dong-Liang Li,Yang Wang,Shan-Shan Guo,Shu-Shan Du

The essential oil extracted from roots and rhizomes of Ligusticum jeholense Nakai et Kitagawa was investigated for its chemical composition by GC-MS analysis, and evaluated for its contact toxicity and repellency against Tribolium castaneum and Lasioderma serricorne, along with some of its individual components. The essential oil was rich in aromatics (65.34%) with low molecular weight. Major components included sedanolide (33.95%), 3-butylidenephthalide (18.76%), spathulenol (8.90%) and myristicin (6.76%). The results of bioassays indicated that the essential oil of L. jeholense and 3-butylidenephthalide possessed significant repellent activities against T. castaneum at 2 and 4 h post-exposure. Meanwhile, 3-butylidenephthalide had potent contact toxicity against L. serricorne (LD50 = 13.64 µg/adult). The minor component n-butylbenzene in the oil was highly toxic to T. castaneum (LD50 = 23.99 µg/adult) and L. serricorne (LD50 = 7.86 µg/adult) in contact assays, but failed to repel these beetles at all testing concentrations. Spathulenol and myristicin exerted good insecticidal and repellent effects on the two target insects. This work suggests that the essential oil of L. jeholense has promising potential for development as natural insecticide or repellent to control pest damage in warehouses.
