Breast cancer remains a significant global health concern owing to the limitations of conventional therapies, such as side effects, drug resistance, and high costs. Nanocatalytic therapy has emerged as a promising alternative due to its tumor specificity, spatiotemporal controllability, and noninvasiveness. However, its effectiveness is limited by endogenous antioxidants like glutathione (GSH) and the low catalytic activity of nanocatalysts. Herein, an Cu/Au-doped polypyrrole nanocatalyst, Cu-AuPP, with multiple catalytic activities is developed by sequentially polymerizing pyrrole monomers using CuCl2 and HAuCl4, followed by PEGylation. The obtained Cu-AuPP catalyzes the production of hydroxyl radicals (·OH) and facilitates the oxidation of GSH to GSSG via redox reactions mediated by multivalent Cu ions, leading to oxidative damage, mitochondrial dysfunction, and tumor cell apoptosis. Upon 1064 nm laser irradiation, these catalytic activities were enhanced by elevated temperature and electron–hole separation mediated by Au nanoclusters, resulting in more intense oxidative damage to tumor cells. Collectively, the developed Cu-AuPP nanocatalytic medicine capable of simultaneously catalyzing GSH depletion and ·OH production via several improved catalytic mechanisms, has significant promise for the treatment of malignant tumors.

Multimodal tumor therapy based on “all-in-one” nanoplatforms enhances therapeutic efficacy, simplifies the construction process, and improves material utilization. Elemental selenium was successfully supported on upconversion nanoparticles (UCNPs) to constitute the near-infrared (NIR) light-responsive UCNP@Se heterostructures for the first time. Under 980 nm irradiation, the UCNP@Se heterostructures could not only produce holes and superoxide radicals •O2– but also catalyze the generation of hydroxyl radicals •OH by highly elevated levels of H2O2 in tumor cells to kill the tumor cells. In addition to the superior photocatalytic performance, elemental selenium itself also exhibited inherent inhibition activity against tumor cells. On the basis of the binding of anthracycline anticancer drugs such as doxorubicin (DOX) to the supported elemental selenium through Cu2+ bridging coordination, the UCNP@Se–Cu-DOX drug delivery system was constructed. The introduction of Cu2+ not only improved the efficient loading of DOX but also achieved the “AND” logic-controlled release of DOX under the combined stimuli of low pH and overexpressed glutathione (GSH) in tumor cells. Moreover, the loaded Cu2+ reacted with the overexpressed GSH to generate the active species CuII-GSSG upon NIR light irradiation, which further promoted tumor cell apoptosis. The NIR light-responsive UCNP@Se–Cu-DOX drug delivery system achieved the combination tumor therapy of selenium therapy, photocatalytic therapy, and logic-gated chemotherapy and has great potential applications in tumor therapy.

Reactive oxygen species (ROS) play a major role in driving acute kidney injury (AKI) by causing oxidative stress and triggering inflammatory responses. However, treatment of AKI with traditional nanomedicines is still challenging because of low ROS scavenging efficacy and poor inflammatory chemotactic. Herein, we have constructed a novel cerium single-atom catalyst (A-CeSACs) for AKI catalytic therapy which targets inflammation and mimics several enzymatic redox activities. After injection of A-CeSACs into AKI mice via tail vein, targeting damaged kidney sites is realized by hitchhiking neutrophils that naturally target sites of inflammation via chemotaxis. After entering the AKI inflammatory environment, A-CeSACs rapidly scavenge multiple ROS via the Ce 3+ /Ce 4+ redox reaction, thus reducing the release of inflammatory factors. The designed A-CeSACs displayed remarkably catalytic therapy efficacy in glycerol-induced AKI mice models. Overall, the present study describes a novel therapeutic strategy for targeted AKI catalytic therapy that is also potentially applicable to other inflammation-related diseases.

Targeting excess H 2 O 2 in the tumor microenvironment, nanotheranostic agents for catalytic therapy are designed based on Fenton reaction, catalyzing H 2 O 2 into oxygen and hydroxyl radical ( OH). But the catalytic efficiency in tumor microenvironment is not satisfactory. In order to solve the problem, a series of bimetallic-dual ligands metal-organic frameworks Fe/Tm-MOFs were designed, that Fe 3+ and Tm 3+ as metalions, 2-methylimidazole and trimesic acid as ligands. Due to the doped Tm 3+ in Fe/Tm-MOFs and the conjugated structures formed by two ligands, the rate of electron transfer was improved, thus promoting the generation of OH at some extent. In addition, the photothermal effect of Fe/Tm-MOFs further promotes the generation of OH, which was evidenced by the 3,3′,5,5′-tetramethylbenzidine(TMB). Combining the drug loading and release capabilities of Fe/Tm-MOFs, synergetic therapy of photothermal/chemo-/catalytic therapy can be achieved. In vitro results reveal that DOX release behaviors are both pH- and thermal-responsive. In vivo anti-cancer results show that the tumors of mice almost disappeared within 10 days, which were injected with Fe/Tm-MOFs/DOX and irradiated with 808 nm for 10 min. Thus, an excellent therapeutic performance has been achieved. Besides, Fe/Tm-MOFs can serve as a multimodality bioimaging contrast agent, covering fluorescence imaging, photothermal imaging and magnetic resonance imaging. Thus, an all-in-one nanotheranostic agent is constructed, improving the catalytic efficiency and providing a novel method to design an efficient nanotheranostic agent.

Foreseen as foundational in forthcoming oncology interventions are multimodal therapeutic systems. Nevertheless, the tumor microenvironment (TME), marked by heightened glucose levels, hypoxia, and scant concentrations of endogenous hydrogen peroxide could potentially impair their effectiveness. In this research, two-dimensional (2D) Ti3C2 MXene nanosheets are engineered with CeO2 nanozymes and glucose oxidase (GOD), optimizing them for TME, specifically targeting cancer therapy. Following our therapeutic design, CeO2 nanozymes, embodying both peroxidase-like and catalase-like characteristics, enable transformation of H2O2 into hydroxyl radicals for catalytic therapy while also producing oxygen to mitigate hypoxia. Concurrently, GOD metabolizes glucose, thereby augmenting H2O2 levels and disrupting the intracellular energy supply. When subjected to a near-infrared laser, 2D Ti3C2 MXene accomplishes photothermal therapy (PTT) and photodynamic therapy (PDT), additionally amplifying cascade catalytic treatment via thermal enhancement. Empirical evidence demonstrates robust tumor suppression both in vitro and in vivo by the CeO2/Ti3C2-PEG-GOD nanocomposite. Consequently, this integrated approach, which combines PTT/PDT and enzymatic catalysis, could offer a valuable blueprint for the development of advanced oncology therapies.

Recently, extending single-atom catalysts from mono- to binary sites has been proved to be a promising way to realize more efficient chemical catalytic processes. In this work, atomically dispersed Fe, Pt dinuclear catalysts ((Fe, Pt)SA-N-C) with an ca. 2.38 Å distance for Fe1 (Fe-N3) and Pt1 (Pt-N4) could be precisely controlled via a novel secondary-doping strategy. In response to tumor microenvironments, the Fe-N3/Pt-N4 moieties exhibited synergistic catalytic performance for tumor catalytic therapy. Due to its beneficial microstructure and abundant active sites, the Fe-N3 moiety effectively initiated the intratumoral Fenton-like reaction to release a large amount of toxic hydroxyl radicals (•OH), which further induced tumor cell apoptosis. Meanwhile, the bonded Pt-N4 moiety could also enhance the Fenton-like activity of the Fe-N3 moiety up to 128.8% by modulating the 3d electronic orbitals of isolated Fe-N3 sites. In addition, the existence of amorphous carbon revealed high photothermal conversion efficiency when exposed to an 808 nm laser, which synergistically achieved an effective oncotherapy outcome. Therefore, the as-obtained (Fe, Pt)SA-N-C-FA-PEG has promising potential in the bio-nanomedicine field for inhibiting tumor cell growth in vitro and in vivo.

Nanozyme-based synergistic catalytic therapies for tumors have attracted extensive research attention. However, the unsatisfactory efficiency and negative impact of the tumor microenvironment (TME) hinder its clinical applications. In this study, we provide an easy method to prepare transition metals loaded onto pyrrolic nitrogen-rich g-C 3 N 4 (PN-g-C 3 N 4 ) for forming metal-N4 sites. This N-rich material effectively transfers electrons from g-C 3 N 4 to metal-N4 sites, promotes the oxidation–reduction reaction of metals with different valence states, and improves material reusability. Under TME conditions, copper ions loaded onto PN-g-C 3 N 4 (Cu-PN-g-C 3 N 4 , CPC) can produce ·OH through a Fenton-like reaction for tumor inhibition. This Fenton-like reaction and tumor cell inhibition can be improved further by a photodynamic effect caused by light irradiation. We introduced upconversion nanoparticles (UCNPs) into CPC to obtain nano-enzymes (UCNPs@Cu-PN-g-C 3 N 4 , UCPC) for effectively penetrating the tissue, which emits light corresponding to the UV absorption region of CPC when excited with 980 nm near-infrared (NIR) light. The nanoplatform can reduce H 2 O 2 concentration upon exposure to NIR light; this induces an increase in dissolved oxygen content and produces a higher supply of reactive oxygen species (ROS) for destroying tumor cells. Owing to the narrow bandgap (1.92 eV) of UCPC under 980 light irradiation, even under the condition of hypoxia, the excited electrons in the conduction band can reduce insoluble O 2 through a single electron transfer process, thus effectively generating O 2 •– . Nanoenzyme materials with catalase properties produce three types of ROS (·OH, O 2 •– and 1 O 2 ) when realizing chemodynamic and photodynamic therapies. An excellent therapeutic effect was established by killing cells in vitro and the tumor-inhibiting effect in vivo , proving that the prepared nanoenzymes have an effective therapeutic effect and that the endogenous synergistic treatment of multiple treatment technologies can be realized.

Catalytic therapy based on nanozymes is promising for the treatment of bacterial infections. However, its therapeutic efficacy is usually restricted by the limited amount of hydrogen peroxide and the weak acidic environment in infected tissues. To solve these issues, we prepared polyvinyl alcohol (PVA)–polyacrylic acid (PAA)–iron oxide (Fe3O4)/polyvinyl alcohol (PVA)–zinc peroxide (ZnO2) double-layer electrospun nanofibers (PPF/PZ NFs). In this design, PVA serves as the carrier for ZnO2 nanoparticles (NPs), Fe3O4 NPs, and PAA. The double-layer structure of nanofibers can spatially separate the PAA and ZnO2 to avoid their reaction with each other during preparation and storage, while in the wet wound bed, PVA can dissolve and PAA can provide H+ ions to promote the generation of hydrogen peroxide and subsequent conversion to hydroxyl radicals for bacteria killing. In vitro experimental results demonstrated that PPF/PZ NFs can reduce the methicillin-resistant Staphylococcus aureus by 3.1 log (99.92%). Moreover, PPF/PZ NFs can efficiently treat the bacterial infection in a mouse wound model and promote wound healing with negligible toxicity to animals, indicating their potential use as “plug-and-play” antibacterial wound dressings. This work provides a novel strategy for the construction of double-layer electrospun nanofibers as catalytic wound dressings with hydrogen peroxide/acid self-supplying properties for the efficient treatment of bacterial infections.

Reactive oxygen species (ROS) production efficiencies of the nanocatalysts are highly desired for cancer therapy, but currently the ROS generation efficiency is still far from defecting the tumors. Therefore, improving their ROS generation ability is highly desirable for cancer therapy. Herein, inspired by the electrostatic preorganization effect during the catalysis of natural protein enzymes, a human self-driven catalysis-promoting system, TENG-CatSystem is developed, to improve catalytic cancer therapy. The TENG-CatSystem is mainly composed of three elements: a human self-driven triboelectric nanogenerator (TENG) as the electric field stimulator to provide electric pulses with high biosafety, a nanozyme comprising a 1D ferriporphyrin covalent organic framework coated on a carbon nanotube (COF-CNT) to generate ROS, and a COF-CNT-embedded conductive hydrogel that can be injected into the tumor tissues to increase local accumulation of COF-CNT and decrease the electrical impedances of tissues. Under the human self-generated electric field provided by the wearable TENG, the peroxidase-like activity of the COF-CNT is fourfold higher than that without an electric field. Highly malignant 4T1 breast carcinoma in mice is significantly suppressed using the TENG-CatSystem. The human self-driven TENG-CatSystem not only demonstrates high catalytic ROS generation efficiency for improved cancer therapy, but also offers a new therapeutic mode for self-driven at-home therapy.

Artificially synthesized nanozymes exhibit enzymatic activity similar to that of natural enzymes. However, in the complex tumor microenvironment, their diversity and catalytic activity show significant variations, limiting their effectiveness in catalytic therapy. Developing artificial enzymes with multiple enzymatic activities and spatiotemporal controllable catalytic abilities is of great clinical significance. Herein, we propose a novel strategy for synergistic enzyme catalysis and sonocatalytic therapy of tumors using polyoxometalates-based nanozymes. Copper-doped molybdenum-based polyoxometalates (denoted as CP) were rapidly synthesized at room temperature through a one-step method. CP contains mixed-valence states of Cu + /Cu 2+ and Mo 5+ /Mo 6+ ions, endowing it with enzyme-like activities of peroxidase, catalase, and glutathione peroxidase. Additionally, the incorporation of copper ions introduces oxygen vacancies into the nano-polyoxometalate, which not only reduces the bandgap but also enhances carrier separation efficiency, thereby improving the sonocatalytic performance of CP as a semiconductor. The combined effects of enzyme-like catalysis and sonocatalysis generate multiple reactive oxygen species (ROS), synergistically depleting glutathione (GSH) and disrupting the redox homeostasis of the tumor, inducing ferroptosis in tumor cells and thereby inhibiting tumor proliferation. This study provides new insights into the design of artificial nanozymes with multiple enzymatic activities and ultrasound activation functions for combined tumor therapy.

Extracellular vesicles (EVs) hosting enzymatic activities that function as independent metabolic units are attractive natural biocatalytic platforms. However, directly using these metabolically active nanoreactors for effective biocatalytic applications remains challenging, mainly due to their constrained catalytic capabilities. Here, we construct an EV-templated nanobiohybrid system by engineering an EV surface with a photoresponsive zeolitic imidazolate framework (ZIF). The deposition of ZIF nanostructures on EVs not only contributes to improved biocatalytic stability but also enables interfacial coupling between photoexcited electrons from the ZIF and the enzymatic reaction of metabolically active EVs. Nearly 300% of biomass conversion efficiency increment could be achieved by EVs derived from macrophages. This enhanced biocatalysis, high catalytic stability, and low cytotoxicity endowed the EV@ZIF nanosystem with robust biosynthesis and antimicrobial activity. When evaluated in a mouse periodontitis model, we show that the autologous biocatalytic EV@ZIF demonstrated efficient therapeutic capability by killing bacteria and inhibiting inflammation. This nanoengineering strategy will benefit the future optimization of metabolically active EV nanoreactors as biocatalysts for a broad range of therapeutics.

The extremely low delivery efficiency of nanomedicine is an important obstacle in the process of tumor therapy, which is mainly caused by the abnormally high tumor interstitial fluid pressure (TIFP) in the tumor microenvironment. This is due to the tumor being short of the lymphatic system, resulting in the lymphatic drainage being blocked. The retained interstitial fluid raises TIFP, leading to the formation of a reverse pressure difference between the tumor tissue and blood vessels, which blocks the delivery of blood-borne drugs to the center of the tumor. Herein, we designed a ZnO–Au/CLM nanomedicine delivery system focusing on reducing TIFP, which is composed of ZnO–Au, bis(2,4,5-trichloro-6-carboxyphenyl) oxalate, liposome, and tumor cell membrane. The drugs were delivered to the tumor through cell membrane homologous targeting and then bis(2,4,5-trichloro-6-carboxyphenyl) oxalate reacted with hydrogen peroxide (H2O2) to generate high-energy intermediates 1,2-dioxetanedione, which activated the splitting water of ZnO to produce hydrogen and hydroxyl radical (•OH) for chemocatalytic treatment. Therefore, the volume of tumor interstitial fluid was reduced, thereby decreasing TIFP and enhancing the delivery of drugs to the depth tumor. As nanozyme Au NPs could effectively catalyze glucose to generate H2O2, which not only starved the tumor cells but also supplied H2O2 for chemocatalytic therapy. After 120 min of ZnO–Au/CLM treatment, the relative TIFP decreased to 59.16% in tumor-bearing mice, meanwhile, the drug accumulation in the tumor increased by 10.4 times compared with the control group. This work leads to a new direction of catalytic medicine by combining nanomedicine with catalysis.
