Enzymatic Cleanup of Formaldehyde in Aqueous Solutions

Zhenyu Zhai,Yunjie Li,Zhilei Li,Jingting Wang,Yuan Li,Yi-Heng P. Job Zhang

Graphical Formaldehyde poses a significant threat to human health. We developed an enzymatic technique for the cleanup of formaldehyde in aqueous solutions. This technique utilized an enzyme cocktail, which catalyzed the reaction of formaldehyde and ambient dioxygen into carbon dioxide and water, with degradation ratios of up to 100 %. Numerous methods have been developed to address gaseous formaldehyde pollution, but most of them cannot be applied directly to eliminate the pollution of formaldehyde in aqueous solutions. Formaldehyde in aqueous solutions can be leached from formaldehyde-containing solid materials (e. g., food, wood, clothes, resins) and absorbed from gaseous formaldehyde by water. Here we developed an enzymatic cleanup technique — the reconstitution of an enzyme cocktail consisting of three coenzyme-free oxidoreductases (i. e., formaldehyde dismutase, methanol oxidase, and formate oxidase) and catalase for the complete oxidation of formaldehyde. This enzyme cocktail catalyzed the reaction of formaldehyde and ambient dioxygen into carbon dioxide (CO 2 ) and water, which was demonstrated by the stable isotope tracer technique. Significant levels of formaldehyde were detected from aqueous solutions leached from the squid, pomfret, fabric, and curtain in the market. When this enzyme cocktail was applied to treat the leachates of contaminated samples above, formaldehyde was eliminated with degradation ratios of up to 100 %. This enzymatic cleanup technique, featuring excellent biosafety (for example, degradable catalysts and non-immunogenicity), independence from light, high degradation ratios, and no special equipment required, could be widely used to treat contaminated food, drinking water, and formaldehyde-containing leachate.

Shielding effects of Fe3+-tannic acid nanocoatings for immobilized enzyme on magnetic Fe3O4@silica core shell nanosphere

Jiandong Cui,Sizhu Ren,Tao Lin,Yuxiao Feng,Shiru Jia

Enzyme immobilization on solid supports is a valuable approach to address enzyme stability and reuse for continuous operations. However, the enzymes immobilized on the external surface of solid supports may not be protected by carriers and suffer inactivation caused by denaturing stresses and hazard external environment. Herein, we describe for the first time a enzyme-shielding strategy to prepare hybrid organic/inorganic nanobiocatalysts; it exploits the self-assembly of supramolecular metal-organic coordination complex (tannic acid (TA) and Fe 3+ ) at the surface of immobilized catalase on Fe 3 O 4 /silica core-shell nanospheres to grow a protective nanocoating (Fe 3+ -TA nanocoating). The nanocoatings around the immobilized catalase (Fe 3+ -TA@Fe 3 O 4 /SiO 2 -catalase) provide a “shield effect” to protect from biological, thermal and chemical degradation for enzyme. As a result, the stability of immobilized catalase against proteolytic agent, denaturants and heat were improved remarkably compared to the immobilized catalase without a protective nanocoating and free catalase. More importantly, the recycling of the immobilized catalase was improved remarkably. The Fe 3+ -TA@Fe 3 O 4 /SiO 2 -catalase still retained 55% of their original activity after 9 cycles, whereas the immobilized catalase without a protective nanocoating only retained 20% of original activity. These results demonstrated that the novel enzyme-shielding strategy is an efficient method to obtain stable and recycled biocatalyst with yolk-shell structure.

Highly sensitive visual detection of catalase based on the accelerating decomposition of H2O2 using Au nanorods as a sensor

Simin Lu,Ling Chen,Ping Yang,Katarzyna Matras-Postolek

The detection of catalase associated with various diseases is very important due to the fact that it is frequently downregulated in human cancer tissues. Noble metal nanomaterial-assisted sensors have achieved rapid development for wide applications but few for detecting catalase. Thus, a novel colourimetric strategy has been developed for the selective and rapid visual detection of catalase by using the decelerating etching of gold nanorods (NRs). In the presence of hexadecyltrimethylammonium bromide and SCN−, which can reduce the redox potential of Au(I)/Au (0), Au NRs would be etched by H2O2 along the longitudinal direction. As a consequence, the colour of solutions changed from bluish green to pink accompanied by the localized surface plasmon resonance (LSPR) extinction peak being shifted to short wavelength. In contrast, the etching process would be prohibited in the presence of catalase, and the colour and LSPR extinction peak of Au NRs would remain unchanged. Furthermore, this Au NR based sensor is applied to test paper that could be used in the detection of catalase in real samples. The discussion of the mechanism indicates that such a sensing method has an ideal performance in terms of sensitivity, selectivity, and linearity.

Eye-Drop Nano-Formulation of Catalase Self-Assembled with Thiolated Chitosan for Effective Treatment of Dry Eye Disease

Ming Shao,Yu Chai,Yutong Jiang,Xinyi Wu,Wenjie Xie,Jiayi Lu,Xuehui Fu,Yao He,Xiaofeng Zhang,Han Zhang,Zhuang Liu

Dry eye disease (DED), the most prevalent ophthalmological condition worldwide, can cause severe ocular discomfort and even visual impairment. Effective yet safe therapeutics for severe DED are still highly demanded. Herein, considering the important role of excessive reactive oxygen species (ROS) in triggering DED, an eye-drop nano-formulation of catalase (CAT) self-assembled with cysteine-modified chitosan (CS-Cys) is designed for DED treatment. Upon eye-drop administration of CS-Cys/CAT nanoparticles, CS-Cys can form disulfide bonds with abundant thiols in the mucin layer of the tear film, anchoring catalase to the corneal surface. Thus the excess ROS accumulated on the ocular surface can be effectively eliminated, resulting in a regulated tear microenvironment. In mouse and rabbit models, it is verified that CS-Cys/CAT eye drops can offer excellent therapeutic effects, especially in promoting the recovery of damaged epithelium and increasing tear secretion. Remarkably, CS-Cys/CAT eye drops showed notably better therapeutic performance than clinically used cyclosporin and dexamethasone, as well as several new DED drugs in clinical trials. Thus, the work presents a unique nanoparticulate eye-drop-based ocular delivery system to allow prolonged ocular retention of protein therapeutics, and such nanoformulation formulated by fully biocompatible/biodegradable components possesses significant translational potential for effective and safe DED treatment.

Plasmonic Enzyme-Linked Immunosorbent Assay Using Nanospherical Brushes as a Catalase Container for Colorimetric Detection of Ultralow Concentrations of Listeria monocytogenes

Rui Chen,Xiaolin Huang,Hengyi Xu,Yonghua Xiong,Yanbin Li

Plasmonic enzyme-linked immunosorbent assay (pELISA) based on catalase (CAT)-mediated gold nanoparticle growth exhibits ultrahigh sensitivity for detecting disease-related biomarkers using sandwich formats. However, the limit of detection (LOD) of this strategy for Listeria monocytogenes is only around 103 CFU/mL, which considerably exceeds the amount of L. monocytogenes commonly present in food products (<100 CFU/g). Herein, we report an improved pELISA method for detection of L. monocytogenes at ultralow concentrations with the sandwich formats using silica nanoparticles carrying poly(acrylic acid) brushes as a “CAT container” to increase enzyme loading for enhancing the detection signal. Under optimal conditions, the proposed pELISA exhibits good specificity and excellent sensitivity for L. monocytogenes with a LOD of 8 × 101 CFU/mL in 0.01 M phosphate-buffered saline, via a reaction that can be discriminated by the naked eye. The LOD obtained by this method was 2 and 5 orders of magnitude lower than that of conventional CAT-based pELISA and horseradish peroxidase (HRP)-based conventional ELISA, respectively. Coupled with large-volume immunomagnetic separation, the LOD for L. monocytogenes-spiked lettuce samples reached 8 × 101 CFU/g. The improved pELISA also exhibited a great potential in detecting a single cell of L. monocytogenes in 100 μL of solution.

Dual-aptamer-based enzyme linked plasmonic assay for pathogenic bacteria detection

Lei Zhan,Chun Mei Li,Zhi Feng Fu,Hong Yan Zou,Cheng Zhi Huang

Development of rapid, sensitive, and selective method for pathogenic bacteria detection is of great importance for food safety, medical diagnostic, and environmental monitoring. Currently, most techniques for low numbers of bacteria detection require advanced instrumentation or skilled operators. Herein, we present a facile colorimetric detection platform for bacterial detection using Ag nanoplates as chromogenic substrate, which takes advantages of the high specificity and affinity of aptamer and the ability of catalase to hydrolyze H 2 O 2 that can etch Ag nanoplates. By introducing catalase to the sandwich structure composed by dual-aptamer recognition strategy, bacteria detection signal is converted to the peak shift of LSPR and colorimetric change. This proposed method allows a fast naked-eye detection of S. aureus at the concentration of 60 CFU/mL based on the combination of streptavidin-biotin system and inherent sensitivity of plasmonic Ag nanoplates. Owing to the high selectivity and sensitivity, as well as the low-cost and good adaptability, this plasmonic assay is expected to be suitable for pathogenic bacteria detection in resource-limited settings.

Exogenous Catalase Stimulates the Polymyxin E-Induced Rapid Killing of Paenibacillus polymyxa

Yu Zhiliang,Zhang Ling,Qin Wangrong,Yin Jianhua,Qiu Juanping

Polymyxin E (colistin) has been considered as an important peptide antibiotic for clinical treatment of Gram-negative pathogens by selectively targeting lipopolysaccharide on the outer membrane. Recent reports showed that colistin can also kill Gram-positive bacteria, but the mechanism of colistin against Gram-positive bacteria remains unclear. In this study, we found that colistin can kill its producer Paenibacillus polymyxa , a Gram-positive bacterium, by both disrupting its cell integrity and inducing oxidative stress. Enhancement of the cell permeability is the major action of colistin against P. polymyxa . In relative, colistin-induced oxidative stress plays an associated role in killing of P. polymyxa . Notably, we, for the first time, found that exogenous catalase is able to intensify the colistin-induced killing by inhibiting the intracellular catalase activity and thus increasing oxidative stress. Our findings would shed light on a not yet clearly described bactericidal mechanism of colistin against Gram-positive bacteria. More importantly, our findings pave the road for the future to lower clinical use of colistin for avoiding bacterial resistance.

Preliminary exploration of the loss of free Methylselenocysteine during selenium-rich tea processing

Jue Liu,Youjun Hu,Jie Zheng,Pan Yue,Shi-Yi Ou,Jiaqi Lu,Xiting Huang,Wanyi Cao,Hua Zhou

Methylselenocysteine (SeMC), the primary organic selenium in tea, enhances human nutrition through its bioavailability and bioactivity (e.g. antioxidant activity). This study investigated SeMC stability during tea processing: high-temperature fixation caused 48.11 % SeMC loss (vs. 25.54 % when rolling at 25 °C), following first-order kinetics ( Ea  = 65.58   kJ/mol, air). Nitrogen protection reduced degradation to <10 % at 150 °C. We first report that 29.19 % of oxidised SeMC (SeMCO) regenerates to SeMC at 25 °C, challenging the assumed irreversibility of selenium oxidation. Catalase (≥200 U/mg) synergized with nitrogen to inhibit oxidation, enabling 1) shortened thermal exposure, 2) nitrogen blanketing, and 3) enzymatic control. These results provide practical solutions to preserve selenium functionality during tea processing, thus advancing selenium-rich food production.

Oxidative stress induced by emerging halohydroxybenzonitrile disinfection byproducts and their binding interaction with catalase

Shaoyang Hu,Fangyuan Dong,Xiangxiang Li,Ning Sun,Xiaoyang Wang,Shuqi Guo,Yuntao Qi,Rutao Liu

Halohydroxybenzonitriles (HHBNs) were reported to exist in drinking water as an emerging group of nitrogen-containing aromatic disinfection byproducts (DBPs). The involvement of binding energy with catalase (CAT) as one of molecular descriptors in the quantitative structure-activity relationship model for HHBN cytotoxicity supported that oxidative stress is highly possible to be an essential cytotoxicity mechanism for HHBNs, and the disruption of cellular redox homeostasis may be partly attributed to their interactions towards CAT. However, the relevant evidence is lacking as yet. Thus, in this study, typical biomarkers for oxidative stress and damage in HHBN-treated cells were examined, and the binding interactions between HHBN and CAT were explored. Results indicated that upon exposure to 3,5-dibromo-2-hydroxybenzonitrile, the antioxidant system (e.g., CAT, superoxide dismutase, and glutathione) was disrupted, and excessive reactive oxygen species gave rise to oxidative DNA damage, which further resulted in cell apoptosis. Moreover, the binding interaction between CAT and 3,5-dibromo-2-hydroxybenzonitrile could change the structure and activity of CAT, with the formation of complexes primarily dependent on van der Waals forces and hydrogen bonding. A positive correlation was observed between HHBN cytotoxicity and inhibitory potency on the biological function of CAT, and some unidentified iodinated HHBNs warrant special attention in future owing to their potential high cytotoxicity. The above results contribute to a better understanding of the toxic mechanisms of emerging HHBN DBPs.

Integrating oxygen-boosted sonodynamic therapy and ferroptosis via engineered exosomes for effective cancer treatment

Mingbo Wu,Zhanlin Zhang,Dong Li,Xiaomiao Ruan,Jingwen Yang,Siyi Chen,Xin Li,Wenwu Ling

Rationale: Ferroptosis and sonodynamic therapy (SDT) are both promising therapeutic modalities, but their clinical application remains challenging due to the hypoxic tumor microenvironment and limited supply of polyunsaturated fatty acids. Developing an agent with oxygen-enhanced SDT and increased ferroptosis sensitivity is crucial for advancing tumor therapy. Methods: In this study, catalase (Cat) and Acyl-CoA synthetase long-chain family member 4 (ACSL4) highly expressed 4T1 cells were constructed via lentivirus transfection. Cat and ACSL4 enriched exosomes (EXO@CA) were then extracted and loaded with the sonosensitizer tetrakis (4-carboxyphenyl) porphyrin (TCPP) through electroporation to create engineered exosomes (EXO@CAT). We evaluated the ability of EXO@CAT to generate oxygen in a hydrogen peroxide environment and investigated its effect on motion profiles and permeability of EXO@CAT. The in vitro antitumor activity was assessed via cytotoxicity, ROS levels, live/dead staining, and apoptosis, with ferroptosis biomarkers confirming ferroptosis activation. We also evaluated the in vivo anticancer efficacy of EXO@CAT by tumor growth analysis and histological and immunohistochemical staining in mouse models bearing breast tumor. Results: EXO@CAT harnesses ultrasound stimulation to facilitate oxygen-enriched SDT, demonstrating significant capacity for singlet oxygen ( 1 O 2 ) generating, which promotes the accumulation of lipid peroxidation (LPO), ultimately leading to the induction of ferroptosis. Concurrently, ACSL4 released from EXO@CAT also increases LPO accumulation by modifying cellular lipid composition, thereby enhancing cellular sensitivity to ferroptosis. Moreover, both in vitro and in vivo experiments demonstrate that the homologous targeting ability of EXO@CAT enables its efficient accumulation in tumor tissues, and the oxygen generation catalyzed by Cat not only alleviates tumor hypoxia but also facilitates the penetration of EXO@CAT into deeper layers of tumor tissue. Conclusions: EXO@CAT combines endogenous proteins, which are prone to inactivation, with an exogenous sonosensitizer, allowing synergistic anticancer treatment of both ferroptosis and SDT with improved efficacy.

Three-dimensional ordered magnetic macroporous metal-organic frameworks for enzyme immobilization

Yuxiao Feng,Hongtong Hu,Zichen Wang,Yingjie Du,Le Zhong,Chenxi Zhang,Yanjun Jiang,Shiru Jia,Jiandong Cui

Metal-organic frameworks (MOFs) have been emerged as a promising support for immobilizing enzymes owing to the tunable porosity, high surface area, and structural diversity. However, most of these possess nanometer size and small pores, which are difficult to recover them from the reaction medium and present low immobilization efficiency and protein loading capacity, and high substrate diffusion limitations. Herein, a novel magnetic amino-functionalized zeolitic imidazolate framework-8 (ZIF-8) with 3D highly ordered macroporous structure was synthesized using the assembled polystyrene (PS) nanosphere monoliths as a template. Subsequently, catalase (CAT) molecules were immobilized on the surface of macroporous magnetic ZIF-8 and inside the macropores by precipitation, covalent binding and cross-linking. The resultant immobilized CAT showed high immobilization efficiency (58%) and protein loading capacity (29%), leading to 500% higher activity than the immobilized CAT on ZIF-8 (CAT/ZIF-8). Meanwhile, the immobilized CAT could be easily recovered with a magnet without obvious activity loss. The traditional CAT/ZIF-8 lost its activity after 6 cycles, whereas, the immobilized CAT retained 90% activity of its initial activity after reusing for 8 cycles, indicating excellent reusability. In conclusion, this study provides a facile and efficient approach to immobilize enzymes on/in MOFs with enhanced activity and excellent recyclability.

Synthesis of catalase-inorganic hybrid nanoflowers via sonication for colorimetric detection of hydrogen peroxide

Miaorong Zhang,Na Yang,Yuxiang Liu,Jianguo Tang

A catalase-inorganic hybrid nanoflowers (Cat-HNFs)-based colorimetric platform was prepared for visual detection of hydrogen peroxide (H 2 O 2 ). Catalase via coordination was self-assembled and embedded in the copper phosphate crystals by sonication. Under the designed colorimetric system, Cat-HNFs showed linear absorbance responses toward H 2 O 2 in the concentration range of 1 μM to 1 mM, and the detection limit was 1 μM. Compared to free catalase, Cat-HNFs exhibited enhanced activity and stability for H 2 O 2 detection. Moreover, after 5 cycles via centrifugal recovery, Cat-HNFs still remained 68% of their initial activity. The detection result in human serum suggested Cat-HNFs can be a promisng candidate for the detection of H 2 O 2 in biomedical field.
