To mitigate the significant side effects and limited efficacy of traditional treatment methods, we propose a novel strategy that involves the use of nanodiamond (ND) drug loading combined with up-converted blue light irradiation to achieve highly efficient chemotherapy-photodynamic treatment of liver tumor cells, specifically those of the HepG2 cell line. ND-loaded drugs delivered a high concentration of doxorubicin to HepG2 cells, enhancing the chemotherapy effect. Upon exposure to near-infrared irradiation, blue light is excited by up-conversion nanoparticles (UCNPs), thereby activating the production of reactive oxygen qdemonstrated that the dual-mode therapy of chemotherapy and photodynamic therapy showed significantly improved anticancer effects. The inhibition rates were 77.4 % in vitro and 88.1 % in vivo for HepG2 cells, which are significantly higher than those treated with single photodynamic therapy (18.9 %, 28.6 %) or drug-loaded therapy (62.6 %, 71.4 %). It is noted that under the treatment conditions, no obvious tissue injury or inflammation was observed in the heart, liver, spleen, lung, and kidney samples collected from mice, indicating that the UCNPs and NDs exhibit good biocompatibility and are less toxic to normal cells and tissues, which would be favorable for clinical applications.

Nanoparticles (NPs) modified with targeting ligands have often shown great potential in targeted drug delivery for tumor therapy. However, the clearance of NPs by the monocyte-phagocyte system (MPS) and the relatively low cellular uptake by tumor cells have significantly limited the antitumor efficacy of a variety of nanomedicines . Tumor microenvironment-mediated multidrug resistance also reduces the antitumor efficacy of internalized nanomedicines. Herein, we developed an innovative nanomedicine for combined chemo-photodynamic therapy of melanoma through targeted drug delivery and significantly improved the cellular uptake of the nanomedicine through the charge-reversal phenomenon. An amphiphilic platinum (IV)-polyethylenimine-chlorin e6 (Pt(IV)-PEI-Ce6) polymer was designed, prepared, and self-assembled into NPs (PPC) in an aqueous solution, and these NPs were subsequently coated with hyaluronic acid (HA) to afford [email protected] The surface-coated HA provided PPC with a negatively charged surface potential to reduce the clearance by the MPS during systemic circulation and enhanced the targeted delivery of PPC to CD44-overexpressing melanoma cells. Upon accumulation in the tumor site, hyaluronidase overexpressed in the tumor induced HA degradation to release the positively charged PPC, resulting in an increased internalization of PPC into tumor cells. Bioactive Pt(II) was released in response to high glutathione level in the tumor cells for effective tumor chemotherapy. Under 650 nm laser irradiation, Ce6 produced reactive oxygen species (ROS), thus driving photodynamic therapy. Finally, [email protected] exhibited combined photodynamic-chemotherapeutic antitumor efficacy against the melanoma cells in mice. Statement of significance Tumors are one of the greatest threats to human health, and chemotherapy has been one of the most common therapeutic modalities for treating tumors; however, many challenges related to chemotherapy remain, such as low delivery efficiency, side effects, and unsatisfactory therapeutic efficacy. Nanomedicines modified with targeting ligands have often shown great potential in improving targeted drug delivery for tumor therapy; however, the clearance of nanomaterials by the monocyte-phagocyte system and the relatively low cellular uptake by tumor cells have significantly limited the antitumor efficacy of a variety of nanomedicines. Herein, we developed a novel charge-reversal-based, hyaluronic acid-coated, Pt(IV) prodrug and chlorin e6-based nanomedicine to improve systemic circulation and targeted accumulation of the nanomedicine in the tumor tissue and to enhance its intracellular uptake. This nanomedicine may provide a potential new platform to improve the drug content inside tumor cells and to effectively inhibit tumor growth through combined chemotherapy and photodynamic therapy.

Sono-photodynamic therapy (SPDT) integrates the advantages of sonodynamic therapy (SDT) and photodynamic therapy (PDT), showing great potential for the minimally invasive treatment of malignant tumors. Nevertheless, the effectiveness of sensitizers in PDT is often limited by insufficient light penetration, restricting the synergistic therapeutic effects of SPDT. Herein, we present a controllable strategy using in situ ultrasound-triggered bioluminescence for enhanced SPDT. This method utilizes a mechanosensitive bioluminescent protein composed of the mechanosensitive channel of large conductance (MscL) and the aequorin (AEQ) photoprotein. Upon ultrasound stimulation, MscL responds to mechanical force by inducing Ca2+ influx, which subsequently activates AEQ to oxidize a luminogenic substrate and generate bioluminescence. By leveraging ultrasound and its associated bioluminescence, the natural sono-photosensitizer, Chlorin e6 (Ce6), exhibits efficient sono-/photocytotoxicity, induces immunogenic cell death, and synergistically enhances the antitumor immune response. This ultrasound-triggered SPDT not only suppresses primary tumor growth and metastasis but also induces a long-term immune memory to resist tumor rechallenge. The integration of SPDT with immune activation forms a multimodal “sono–photo–immuno” platform for in situ breast tumor treatment and simultaneously prevents tumor recurrence after tumor resection. These findings showcase our platform as a versatile tool for the development of more effective cancer therapies.

BackgroundThe clinical efficiency of photodynamic therapy (PDT) in combination with chemotherapy has proven to be a promising strategy for tumor treatment, yet is restricted by the high glutathione (GSH) concentration at the tumor site and nonspecific drug targeting.ObjectiveThe goal of the current research was to create a biocompatible GSH-depleting and tumor-targeting nanoparticle (denoted as DOX/CA@PCN-224@HA) for the combined photodynamic and chemo photo-chemo) therapy.MethodsThe nanoparticles were characterized by transmission electron microscopy (TEM). A UV-vis spectrophotometer was used to measure the drug loading efficiency (DE) and encapsulation efficiency (EE). The GSH-depleting ability was measured using Ellman's test. Confocal laser scan microscopy (CLSM) was used to assess the cellular uptake. MTT was adopted to evaluate the cytotoxicity of DOX/CA@PCN-224@HA against 4T1 cells.ResultsThe altered PCN-224 showed excellent monodispersing with a dimension of approximately 193 nm ± 2 nm in length and 79 nm ± 3 nm in width. The larger and spindle grid-like structure of PCN-224 obtains better dual-drug loading ability (DOX: 20.58% ± 2.60%, CA: 21.81% ± 1.98%) compared with other spherical PCN-224 nanoparticles. The ultimate cumulative drug release rates with hyaluronidase (HAase) were 74% ± 1% (DOX) and 45% ± 2% (CA) after 72 h. DOX/CA@PCN-224@HA showed GSH-consuming capability, which could improve the PDT effect. The drug-loaded nanoparticles could accurately target 4T1 cells through biological evaluations. Moreover, the released DOX and CA display cooperative effects on 4T1 cells in vitro. DOX/CA@PCN-224@HA nanoparticles showed inhibition against 4T1 cells with an IC50 value of 2.71 μg mL-1.ConclusionThis nanosystem displays great potential for tumor-targeted enhanced (photo-chemo) therapy.

Photodynamic therapy (PDT) and photothermal therapy (PTT) have been explored widely for application in cancer treatment. In this work, we describe the synthesis of CaTiO3:Yb,Er (CTO) nanofibers co-conjugated with Rose Bengal (RB) and gold nanorods (AuNRs), which offer the potential for combined upconversion photoluminescence (UCPL) and enhanced, synergistic PDT and PTT. Based on this delivery platform, RB and AuNRs served as the PDT and PTT agents, respectively. RB and AuNRs have strong and well-matched absorption with the green and red emissions of UCPL CTO nanofibers respectively, hence a single 980 nm continuous wave laser with deep tissue penetration can be employed to allow PDT and PTT to occur simultaneously. The nanocomposite can effectively convert the near-infrared (NIR) radiation from the laser into a combination of targeted hyperthermia and generation of reactive oxygen species (ROS). In comparison with PDT or PTT alone, the combined PDT/PTT treatment showed significantly enhanced suppression of the viability of Hep G2 cells in vitro, demonstrating its potential for use in oncology.

The combination of phototherapy and chemotherapy with superior advantages is a promising strategy for cancer therapy. However, combination therapy is generally regulated by two different wavelengths of light or other stimuli, which results in complex operations and inevitable systemic side effects, even affecting therapeutic efficacy. Herein, we design a signal NIR light-regulated nanoplatform via the self-assembly process of reactive oxygen species (ROS)-sensitive prodrug (DTD), human serum albumin (HSA), and IR780 for combined photothermal/photodynamic therapy and chemotherapy. Upon 808 nm laser irradiation, IR780 in nanoparticles generates abundant ROS and a significant photothermal effect to achieve photothermal/photodynamic therapy. Meanwhile, the generating ROS further cleans up the thioketal link to release DOX for chemotherapy. Hence, signal NIR light can effectively control the process of combination therapy. In vivo and in vitro experiment results demonstrate that the multifunctional nanoparticles exhibit excellent antitumor efficacy via the combination of phototherapy and chemotherapy controlled by a signal NIR laser. Overall, the signal NIR light-regulated nanoparticles with combination therapy performance provide a versatile platform for enhancing antitumor efficacy.

The low cure rate and high mortality associated with cancer pose significant threats to human health. Photodynamic and photothermal therapies have emerged as promising treatment strategies for various types of cancers. In this study, we successfully synthesized a novel type of carbon dot (CD) using 1,2,4-aminobenzene and ethylenediamine as precursors. Surprisingly, these CDs exhibited outstanding nucleolus-targeting capabilities coupled with a remarkable photothermal effect. Through the integration of these nucleolus-targeting CDs with indocyanine green (ICG) and folic acid (FA), we created CDs-ICG-FA nanocomplexes suitable for combined photodynamic and photothermal therapy. In vitro experiments demonstrated that CDs-ICG-FA maintained a robust photothermal ability, achieving a conversion efficiency of up to 34.3%. Furthermore, CDs-ICG-FA generated abundant reactive oxygen species, effectively inducing cancer cell death and demonstrating its potential for photodynamic therapy. In MCF-7 cancer cells, CDs-ICG-FA exhibited a pronounced synergistic photothermal/photodynamic anticancer effect. Subsequent in vivo experiments in mice revealed that CDs-ICG-FA could selectively accumulate at tumor sites, significantly inhibiting tumor growth upon exposure to an 808 nm laser. These findings suggest that the developed nucleolus-targeting CDs-ICG-FA hold promising potential for cancer targeting and the application of combined photothermal/photodynamic therapy.

Although combined chemotherapy (Chemo), photothermal (PTT) and photodynamic (PDT) in cancer therapy has drawn significant attention due to its superior anticancer ability, the required high intensity of irradiation results in serious photo-toxicity to healthy neighboring cells, and thus limits its biomedical applications. Herein, we developed an ultralow-intensity near infrared (NIR) light synchronously activated collaborative Chemo/PTT/PDT nanoplatform. The nanoplatform is composed of a highly emissive upconversion (UC) core, chlorin e6 (Ce6) photosensitizer and the anticancer drug doxorubicin hydrochloride (DOX) co-loaded in a mesoporous silica (MS) shell, and polyethylene glycol-modified graphene (PGO) acts as both the photothermal reagent and smart switch for promoted drug release. Upon 808 nm NIR light exposure with ultralow intensity (0.25 W cm−2), which is below the maximum permissible exposure (MPE, 0.33 W cm−2) for skin, the mild hyperpyrexia of PGO induced both cancer cell irreversible death for PTT and greatly promoted drug release for enhanced Chemo. On the other hand, the upconverted 660 nm light from UC activated Ce6 to generate reactive oxygen species for PDT, while the upconverted 540 nm light from UC could be employed for visualizing the treatment process. The in vitro and in vivo anticancer experiments demonstrate that the ultralow-intensity NIR light synchronously activated Chemo/PTT/PDT nanoplatform exhibits remarkable therapeutic efficacy with minimal photodamage.

Precise oncotherapy requires effective cancer treatments that are guided by clinical imaging techniques. One of the most representative cases is multi-imaging-guided phototherapy. This study presents a novel multifunctional theranostic agent of SnxWO3 tungsten bronze, which is an excellent light absorber in the near infrared (NIR) range. Theoretical calculations based on density functional theory confirm that the insertion of donor Sn atoms into orthorhombic WO3 gives rise to the broadband visible-NIR absorption. Accordingly, both the photothermal effect and reactive oxygen species (ROS) production could be realized under NIR light irradiation by SnxWO3 tungsten bronze nanocrystals, thereby triggering the potent in vivo photothermal and photodynamic synergistic therapy. Meanwhile, modified SnxWO3 tungsten bronze has the functions of photoacoustic imaging (PAI), X-ray computed tomography (CT) imaging and near-infrared fluorescence (NIRF) imaging for tumor detection as well. Finally, for investigating the antitumor mechanism of in vivo solid tumors, clinical imaging modalities of B-mode ultrasonography (US) and magnetic resonance imaging (MRI) are employed to monitor the tumor evolution process after the photo-treatment, verifying a typically liquefactive necrosis process. These results indicate that the SnxWO3 tungsten bronze nanostructure is a promising theranostic agent for imaging-guided cancer therapy.

Graphical Ultrathin Two-Dimensional BiOCl-Bi 2 S 3 -Cu 2 S Ternary Heterostructures with Enhanced LSPR effect for NIR Photonic Bacterial Disinfection (Chen, Yang, et al.). : Photonic disinfection, particularly near-infrared (NIR) light triggered antibacterial, has emerged as a highly promising solution for combating pathogenic microbes due to its spatiotemporal operability, safety, and low cost of apparatus. However, it remains challenging to construct NIR-responsive antibacterial agents with high light-converting efficacy and elucidate synergistic mechanisms. In this work, ultrathin two-dimensional (2D) BiOCl-Bi 2 S 3 -Cu 2 S ternary heterostructures that can efficiently kill drug-resistant bacteria were synthesized by doping 0D Bi 2 S 3 and Cu 2 S nanoparticles in the 2D BiOCl nanosheets via a facile one-pot hydrothermal method. Notably, the incorporation of Cu 2 S nanoparticles bestows strong NIR light-harvesting capability to the composite nanosheets due to their localized surface plasmon resonance (LSPR). Upon NIR light illumination, the BiOCl-Bi 2 S 3 -Cu 2 S nanosheets can achieve enhanced photonic hyperthermia and reaction oxygen species (ROS) generation, serving as single light-activated bi-functional photothermal/photodynamic therapeutics. High-speed hot electrons and large local electronic fields caused by LSPR might play an important role in thermal vibrations and effective carrier separations, respectively. Benefiting from the unique ternary heterostructures, both the photothermal conversion and ROS generation efficacy of BiOCl-Bi 2 S 3 -Cu 2 S nanosheets are significantly improved compared to the binary BiOCl-Cu 2 S or BiOCl-Bi 2 S 3 nanosheets. Accordingly, the ternary composite nanosheets can effectively kill bacteria via the NIR-driven photonic disinfection mechanism. This work presents a new type of 2D composite nanosheets with ternary heterostructures for NIR photonic disinfection.

Biofilm-associated infections exert more severe and harmful attacks on human health since they can accelerate the generation and development of the antibiotic resistance of the embedded bacteria. Anti-biofilm materials and techniques that can eliminate biofilms effectively are in urgent demand. Therefore, we designed a type I photosensitizer (TTTDM) with an aggregation-induced emission (AIE) property and used F-127 to encapsulate the TTTDM into nanoparticles (F-127 AIE NPs). The NPs exhibit highly efficient ROS generation by enhancing intramolecular D–A interaction and confining molecular non-radiative transitions. Furthermore, the NPs can sufficiently penetrate the biofilm matrix and then detect and eliminate mature bacterial biofilms upon white light irradiation. This strategy holds great promise for the rapid detection and eradication of bacterial biofilms.

The unique optical properties of squaraine dyes make them promising for cancer phototheranostics, but the reported squaraines for in vivo treatments mainly rely on their photothermal effect, where monotherapy cannot achieve the desired therapeutic effect. Here we generated a type of squaraine capable of killing tumors through both photothermal and photodynamic effects. We optimized squaraine structure with selenium modulation and formulated it into nanoparticles that showed strong absorption of infrared light, negligible fluorescence, good photothermal conversion (66.6 %), and strong photodynamic effects even after several irradiation cycles. In addition, the nanoparticles could be tracked through their strong optoacoustic signal. In mice, the nanoparticles effectively accumulated in tumors and eliminated them upon irradiation, without causing adverse effects. Our work demonstrates the potential of selenium modulation of squaraine for precise cancer diagnosis and treatment through synergistic photothermal and photodynamic effects.
