Chitosan-based packaging films with antibacterial-sterilization integrated continuous activity for extending the shelf life of perishable foods

Yongsheng Ni,Yumeng Li,Mengyi Wang,Haoran Li,Wendi Zhang,Lijun Tan,Jinsong Zhao,Baocai Xu

The current food packaging films can be preservative but lack the function of combining antibacterial and sterilization which lead to films can not maximize prolong shelf life of perishable foods. This study provided a new strategy to realize prolonging shelf life of perishable foods by integrating antibacterial and sterilization which focused on applying photodynamic inactivation to films with continuous activity, where curcumin (CUR) and sodium copper chlorophyll (SCC) were loaded into chitosan (CS) films. Compared to pure CS films, the barrier capacity (oxygen permeability and water vapor permeability) and mechanical properties of composite films were improved by introducing CUR and SCC. In addition, the composite film can effectively against food-borne pathogenic bacteria and significantly prolong the shelf life of cherries and pork. The provided strategy has potential application prospects in food preservation packaging.

Antimicrobial photodynamic inactivation with curcumin against Staphylococcus saprophyticus, in vitro and on fresh dough sheet

Ziyuan Wang,Yating Jia,Wanyi Li,Min Zhang

Photodynamic inactivation (PDI), as a new non-thermal decontamination and preservation technology, exhibits the advantages of low cost, free of residue and non-microbial resistance. Curcumin (CUR) as a natural photosensitizer could effectively inactivate food spoilage bacteria at an absorbance wavelength (420–470 nm) light illumination. In this study, the in vitro influence of PDI process parameters on the inactivation of Staphylococcus saprophyticus and the antimicrobial mechanism was analyzed. Approximate 5 log CFU/ml reduction was obtained when treated with 25 μM CUR and 4.32 J/cm 2 of blue LED. Obvious cell wrinkles, cell membrane disruption, intracellular contents leakage and increased membrane permeability of S. saprophyticus cell were observed after CUR-PDI treatment. Further, generation of intracellular reactive oxygen species, along with decreased intracellular enzyme activities and increased degree of cell membrane depolarization verified cell damage. The in vivo photo-antibacterial effect (1.04 log CFU/g and 0.81 log CFU/g) was validated on fresh millet dough sheet at two initial bacterial loads (5 log CFU/g and 3 log CFU/g, respectively). These findings illustrated that CUR mediated PDI could be applied to inhibit S. saprophyticus growth and possibly prevent spoilage of fresh dough products.

Highly Efficient AIE-Active palmatine photosensitizer for photodynamic inactivation of Listeria monocytogenes in apple juice preservation

Shiyu Peng,Qianying Li,Hui Wei,Ping Li,Shuyuan Liu,Lijuan Nie,Yuankui Leng,Xiaolin Huang

Photodynamic sterilization is a promising alternative to conventional antibacterial approaches with merits of high efficiency and safety. The exploration of aggregation-induced emission (AIE)-type photosensitizers from natural sources is highly valued by researchers and food industry. Herein, the use of palmatine (PA), an aggregation-induced emission-active natural product from traditional Chinese medicine, as photosensitizers for photodynamic inactivation of a tenacious foodborne pathogen of Listeria monocytogenes is investigated. Antibacterial and antibiofilm activity against L. monocytogenes of PA-mediated photodynamic process were first assessed. The results showed that PA-mediated photodynamic process could inhibit the growth of L. monocytogenes , and the minimum bactericidal concentration was determined as 80 μM. At this PA dosing level, well-established biofilms of L. monocytogenes can be effectively destroyed under light irritation. Afterward, cell- and gene-level investigations were conducted to explore the antibacterial mechanism. It is concluded that the exceptional photodynamic antibacterial activity of PA against L. monocytogenes is attributed to the disruption of the cell wall and membrane, increased cell permeability, leakage of functional proteins, and damage to DNA structure. Subsequently, PA-mediated photodynamic process was applied to reduce bacterial activity in apple juice while preserving its quality. Overall, this work highlights the significant potential of PA-mediated photodynamic strategy for controlling foodborne pathogens in food systems.

Polysaccharide network protection strategy enhances photodynamic stability: Unlocking new applications of edible pigments in fruit preservation

Bingzhi Li,Yiqi Wang,Linpin Luo,Liang Zhang,Yiya Ping,Bangfeng Fu,Wetao Zhang,Lei Liu,Jianlong Wang

Edible antibacterial packaging films, due to their excellent biosafety and environmental friendliness, exhibit significant application potential in food preservation. However, current edible antibacterial films primarily rely on natural bacteriostatic compounds, which are limited by high dosage requirements, inconsistent performance, and narrow antibacterial spectra, making it challenging to effectively control microbial contamination. To address these challenges, we introduced erythrosine B (ERY), an FDA-approved pigment with strong photodynamic inactivation performance, combined with gellan gum (GG) to develop a novel edible photodynamic antibacterial film. Under light irradiation, ERY can rapidly generate reactive oxygen species (ROS) at a concentration as low as 5 μg/mL, thereby killing 100 % of bacteria and significantly inhibiting fungal growth. Notably, the gel network of GG serves a dual purpose by restricting ERY molecular movement and creating a protective barrier, effectively mitigating photodegradation of ERY caused by excessive ROS, thus enhancing the photostability of ERY. Experimental results demonstrate that, under identical light intensity, GG-ERY (GGE) exhibits negligible photobleaching after 30 min, showing an 8.26-fold improvement in inhibition efficacy compared to ERY, facilitating the sustained production of ROS. Owing to its efficient and long-lasting antibacterial performance, the GGE film extends the postharvest storage period of grapes to 14 days, representing a 1.75-fold increase compared to the control group. This study not only significantly enhances the antibacterial efficacy of edible films but also provides novel insights into the application of edible pigments in photodynamic antibacterial and food preservation.

Turmeric carbon quantum dots enhanced chitosan nanocomposite films based on photodynamic inactivation technology for antibacterial food packaging

Fangzhou Wen,Peiyuan Li,Hongjun Yan,Wei Su

The increased demand for food quality and safety has led the food industry to pay urgent attention to new packaging materials with antimicrobial activity . In this study, we combined photodynamic inactivation of bactericidal technology in food packaging materials by incorporating fluorescent carbon quantum dots (CDs) prepared from the natural plant turmeric into a chitosan matrix to prepare a series of active composite food packaging films (CDs-CS). The chitosan film containing CDs had better mechanical properties, UV protection and hydrophobicity . Under irradiation with a 405 nm light source, the composite film was able to produce abundant reactive oxygen species , and the CDs-CS 2 film exhibited reductions of approximately 3.19 and 2.05 Log 10 CFU/mL for Staphylococcus aureus and Escherichia coli respectively within 40 min. In cold pork storage applications, CDs-CS 2 films showed inhibition of the growth of colonization in pork and retarded the spoilage of pork within 10 days. This work will provide new insights to explore safe and efficient antimicrobial food packaging.

Sustained photodynamic H2O2 generation via disodium 5′-inosinate enhances cooked meat preservation by membrane-targeted antibacterial action

Lijun Tan,Shasha Zheng,Yong Xie,Wendi Zhang,Baocai Xu

Photodynamic inactivation (PDI) is a promising non-thermal antimicrobial technology known for broad-spectrum efficacy and low resistance potential. However, conventional PDI face limitations in bactericidal efficacy due to the short-lived and poorly diffusing reactive oxygen species (ROS). This study proposes a novel food-grade PDI strategy that incorporates disodium 5′-inosinate (IMP) into a vitamin B2 (VB2)-mediated system, significantly enhancing H 2 O 2 production. Acting as an electron donor, IMP promotes type I photochemical reactions, enabling sustained H 2 O 2 generation and achieving >6 log CFU/mL inactivation of Bacillus safensis , a spoilage bacterium often overlooked in cooked meat products. Mechanistic analyses showed that H 2 O 2 oxidized key phospholipids in the cell membrane, increased permeability, caused membrane potential hyperpolarization and ion leakage, and ultimately disrupted multiple membrane domains. Moreover, this strategy also exhibited persistent antimicrobial activity, effectively delaying spoilage in cooked meat products stored at 4 °C and 25 °C. Overall, this study introduces a safe and effective non-thermal sterilization method with promising industrial potential and offers a novel pathway to enhance PDI efficacy via intensified type I ROS mechanisms.

Preparation, characterization, and application of gallic acid-mediated photodynamic chitosan-nanocellulose-based films

Kun Lin,Yu-zhang Zhu,Hua-wei Ma,Jincheng Wu,Chao-nan Kong,Yang Xiao,Hong-cun Liu,Li-lan Zhao,Xiao-lin Qin,Li-fang Yang

In this study, an active film composed of gallic acid (GA), chitosan (CS), and cellulose nanocrystals (CNC) was prepared using a solution casting method and synergistic photodynamic inactivation (PDI) technology. Characterization of the film showed that the CS-CNC-GA composite film had high transparency and UV-blocking ability. The addition of GA (0.2 %–1.0 %) significantly enhanced the mechanical properties, water resistance, and thermal stability of the film. The tensile strength increased up to 46.30 MPa, and the lowest water vapor permeability was 1.16 × e −12 g/(cm·s·Pa). The PDI-treated CS-CNC-GA1.0 composite film exhibited significantly enhanced antibacterial activity, with inhibition zone diameters of 31.83 mm against Staphylococcus aureus and 21.82 mm against Escherichia coli. The CS-CNC-GA composite film also showed good antioxidant activity. Additionally, the CS-CNC-GA 1.0 composite film generated a large amount of singlet oxygen under UV-C light irradiation. It was found that using the CS-CNC-GA 1.0 composite film for packaging and storage of oysters at 4 °C effectively delayed the increase in pH, total colony count, and lipid oxidation in oysters. In conclusion, the CS-CNC-GA composite film based on PDI technology has great potential for applications in the preservation of aquatic products.

Fabrication of gelatin-based photodynamic food packaging with dual-antimicrobial activity for enhanced pork preservation

Lijuan Xu,Rongqiang Chen,Tian Ren

Microbial spoilage is a major reason of global food waste. Photodynamic inactivation (PDI) showed promise as an antimicrobial strategy owing to the reactive oxygen species (ROS) produced by photosensitizer, but the short lifespan of ROS limited its penetration into cells and accordingly hindered its effectiveness against Gram-negative bacteria. In this study, a photodynamic composite film with aggregation-induced emission behavior and dual antimicrobial activity was developed by incorporating natural photosensitizer nanoparticles with ε-Polylysine (PL) into a matrix composed of gelatin and dialdehyde nanocellulose. The advantage of this design lay in the PL's role as a " forward team”, facilitating ROS penetration through perforating the external membrane or cell wall of bacteria, thereby enhancing the PDI efficiency and achieving a 6 log CFU/sample bacterial reduction within 15 min of light exposure. Moreover, the composite film exhibited minimal ultraviolet transmittance, high transparency (exceeding 86.57 %), hydrophobicity, and enhanced mechanical strength. Pilot studies further confirmed the composite film's effectiveness in inhibiting microbial growth, protein oxidation, lipid oxidation, and pH of pork samples, successfully extending the shelf life of fresh pork to 10 days at 4 °C. Therefore, this study proposes a green and efficient synergistic antimicrobial strategy with PDI, demonstrating a potential application for enhanced pork preservation.

A Biosynthesized Near-Infrared-Responsive Nanocomposite Biomaterial for Antimicrobial and Antibiofilm Treatment

Guiyuan Chen,Yihan Wu,Kai Jin,Hongfei Lu,Mingyue Tao,Tiantian Wang,Jing Zhang,Xiaohui Zhu,Jinliang Liu,Yong Zhang

Photodynamic inactivation (PDI) has become an appealing alternative strategy to treat infections without developing resistance to microbes. In PDI treatment, near-infrared (NIR) light is preferred because it causes less damage to normal tissues and leads to better penetration in deep tissues. Here, we develop an NIR-responsive nanomedicine for efficient broad-spectrum antimicrobial photodynamic treatment. By harnessing the biosynthetic capability of a bacterial cellulose-producing microorganism, we construct a nanocomposite biomaterial to deliver and recycle the nanomedicine. Our simple one-step biosynthetic approach does not impede the antimicrobial potency of the nanomedicine under NIR activation and requires no chemical modification. The resulting nanocomposite has been tested in antimicrobial treatment of different microorganisms, exhibiting a great potential to eliminate pathogens in biofilms and to treat in vivo infections.

Photodynamic inactivation of Listeria monocytogenes using a natural aggregation-induced emission photosensitizer and its application in salmon preservation

Qianying Li,Baichuan Wang,Shiyu Peng,Hui Wei,Ping Li,Yuankui Leng,Xiaolin Huang

Photodynamic sterilization has been regarded as a promising alternative to conventional antibacterial approaches. However, the photodynamic efficiency of conventional photosensitizer is often compromised by aggregation-induced quenching effects. Photosensitizers, especially natural products, with aggregation-induced emission (AIE) properties open a new avenue for photodynamic sterilization. Herein, we investigated the photodynamic antibacterial performance and mechanism of the AIE-type natural product of berberine (BBR) against Listeria monocytogenes ( L. monocytogenes ). The bactericidal effect was assessed through the determination of minimum bactericidal concentration and implementation of anti -biofilm experiments. The results showed that BBR can significantly suppress the proliferation of L. monocytogenes and can efficiently remove the L. monocytogenes -derived biofilm in a concentration-dependent trend. The mechanism of killing L. monocytogenes has been speculated based on bacterial morphology characterization, viability staining results, enzyme activity assays, DNA electrophoresis analysis, and molecular docking. The excellent anti-bacterial activity of BBR mediated photodynamic process may account for its abilities to the irreversibly disruption of bacterial membrane, inhibition of intracellular enzyme activity and decomposition of intracellular DNA molecules. We also demonstrated that BBR embedded sodium alginate/chitosan packaging film is feasible to maintain the freshness of salmon under light illumination by inhibiting the bacterial proliferation.

Photodynamic inactivation of curcumin combined with ascorbic acid against Penicillium italicum in vitro and on fresh-cut orange

Yang Wang,Yuling Zhao,Rongrong Wu,Jingwen Gao,Menghan Chen,Yue Cui,Jianxiong Hao,Junhua Han,Karl Matthews

This study investigated the antifungal effect of curcumin-mediated photodynamic inactivation (PDI) with ascorbic acid (Vc) as a synergist on Penicillium italicum and its biofilms, and evaluated the efficacy in inactivating P. italicum on fresh-cut orange. Results showed that the number of P. italicum spores decreased by 4.61 Log CFU/mL when the concentration of curcumin, Vc and illumination time was 75 μmol/L, 250 mg/L and 20 min, respectively. After PDI treatment, the cell structure of P. italicum was destroyed and leakage of cell contents was observed. Mature P. italicum biofilms were formed after 72 h of incubation, and PDI could reduce the spores in biofilms by 3.23 Log CFU/mL. In addition, under the optimal photodynamic conditions, the number of P. italicum spores in fresh-cut orange decreased by 2.48 Log CFU/g, and the sensory quality was well maintained. In conclusion, curcumin-mediated photodynamic technology could better maintain the freshness of fresh-cut orange. Therefore, this study deepened the understanding of the antimicrobial efficacy of curcumin-mediated PDI on food borne pathogens and its impact on food quality.

Curcumin-loaded polycaprolactone/carboxymethyl chitosan nanofibers with photodynamic inactivation of Staphylococcus aureus developed by solution blow spinning

Chaoyi Shen,Jiahao Zhong,Fengjun Guo,Changfeng Zhang,Changqing Zhu,Di Wu,Kunsong Chen

The purpose of the present study was to utilize solution blow spinning to fabricate curcumin (CUR)-loaded polycaprolactone (PCL)/carboxymethyl chitosan (CMCH) nanofibers with photodynamic inactivation activity for inactivation of S. aureus . The results of ATR-FTIR and XRD spectra demonstrated the successful encapsulation of CUR without disrupting the structure of the nanofibers and increasing the crystallinity of the nanofibers. After the encapsulation of CUR, the thermal stability, tensile strength, and hydrophobicity of the PCL/CMCH nanofibers were improved. Besides, the CUR-loaded PCL/CMCH nanofibers possessed good biocompatibility and low toxicity, and could release CUR sustainably over 120 h. Moreover, the obtained nanofibers showed better inhibition of S. aureus under blue light irradiation at a wavelength of 450 nm compared to the no-light condition. The above results suggested that the developed nanofibers with photodynamic inactivation can be applied as food packaging materials to reduce the impact of S. aureus on human wellness.
