Carboxymethyl chitosan based redox-responsive micelle for near-infrared fluorescence image-guided photo-chemotherapy of liver cancer

Tingsheng Yan,Wenxue Hui,Siyuan Zhu,Jinmei He,Zhonghua Liu,Jinju Cheng

High-efficient vectors for the co-delivery of photosensitizers and chemotherapeutics were urgently needed for the combination therapy. In this work, a redox-responsive micelle (PCL-SS-CMC-GA) was prepared for the co-delivery of doxorubicin (DOX) and pheophorbide A (PHA). Poly-ε-caprolactone was linked to carboxymethyl chitosan through a disulfide bond, which was easily broken in the reductive solution to release the payloads. The charge conversion property and glycyrrhetinic acid (GA) targeting ligand of the micelles effectively extended the average residence time (up to 18 times) in circulation and improved their intracellular uptake by HepG2 cells. The micelles exhibited an enhanced tumor accumulation and near infrared (NIR) imaging performance. More interestingly, this nanoplatform could fully exert the synergistic effect of DOX and PHA to improve the inhibition efficiency (with an inhibitory rate of 80.5 %) in vivo . With impressive photo-chemo theranostic and NIR imaging capability, [email protected] /PHA showed great potential in image-guided treatment of liver cancer.

Self-assembled metallo-supramolecular nanoflowers for NIR/acidic-triggered multidrug release, long-term tumor retention and NIR-II fluorescence imaging-guided photo-chemotherapy

Xianglian Liao,Youshi Zheng,Ziguo Lin,Yi Shen,Huiyun Lin,Xiaolong Liu,Da Zhang,Buhong Li

On-demand delivery of drug to deep tumor tissues with long-term tumor retention has directly influence on therapeutic efficiency and prognosis of patients, which still remains challenge in cancer therapy. Here we introduce metallo-supramolecular nanoflowers through multi-drugs and metal coordination effect for near-infrared (NIR) or acidic-triggered multi-drugs release, long-term accumulation at tumor tissues and NIR-II fluorescence imaging-guided photo-chemotherapy. The nanoflowers (refer to ICG⊃EDOX) are constructed by (−)-epigallocatechin-3-gallate (EGCG), Cu 2+ ions, indocyanine green (ICG) and doxorubicin hydrochloride (DOX) through coordination effect, π-π stacking and hydrophobic force. Due to strong intermolecular interaction, the ICG⊃EDOX is superior stable in physiological conditions, while releases DOX/ICG on-demand upon the acidic/NIR laser irradiation. Moreover, ICG⊃EDOX could penetrate deep tumor tissues with ultralong tumor retention (>8 d) by microvesicle (MVs)-mediated intercellular interaction. Irradiated by 808 nm laser, the higher NIR-II fluorescence signal from ICG⊃EDOX in tumors can effectively guide local phototherapy, and the synergistic therapeutic efficiency of nanoflowers with completely elimination of tumors is achieved in comparison with DOX or DOX/ICG treated 4T1-bearing mice . Overall, the excellent therapeutic effect of nanoflowers may hold great promise for the clinic translation.

Multifunctional Hypoxia-Involved Gene Silencing Nanoplatform for Sensitizing Photochemotherapy

Huimin Wang,Yuqi Chen,Jinhua Shang,Hong Wang,Min Pan,Xiaoqing Liu,Xiang Zhou,Fuan Wang

The integration of combinational photochemotherapy could achieve a promoted therapeutic performance by eliminating the frustrating side effects of each constitute, yet it is still confronted with severe therapeutic resistances that are associated with tumor hypoxia. Herein, we designed a multifunctional gene-therapy-promoted photochemotherapy strategy for specifically alleviating the hypoxia-associated therapeutic resistance. The pH-responsive zeolitic imidazolate framework-8 (ZIF-8) nanoparticles (CDHNs) facilitated the effective delivery of therapeutic agents (chlorin e6 (Ce6), doxorubicin (DOX), and HIF-1α siRNA) into the tumor for relieving specifically photochemotherapy resistance via sequential activation manners under the guidance of fluorescence imaging. The present all-in-one compact CDHNs system provides a smart and versatile multiply gained therapeutic strategy to activate the specific cellular pathways and is anticipated to boost future cancer diagnosis and post-treatment.

Cu2O Nanocubes Embedded in Polycaprolactone Nanofibers for Photo–chemotherapeutic Wound Disinfection and Regeneration

Hui Zhao,Yang Yang,Jing An,Huilong Xin,Yinan Xiao,Zhaojun Jia,Yuandong Wu,Liyuan Sheng,Min Wen

Wound-associated infections represent an arduous global healthcare challenge, especially in the “post-antibiotic” era, necessitating the development of advanced antimicrobial dressing materials that simultaneously dictate the use of rapid yet effective disinfection, low drug resistance and side effects, and the potency to foster tissue healing. Nanoantimicrobial-armored nanostructured functional dressing systems have piqued extensive interest to satisfy these criteria. Herein, fusing electrospinning nanotechnology and an emerging photo–chemo synergistic sterilization paradigm, a smart “all-in-one” multifunctional wound-managing dressing platform was constructed, through incorporating photochemically active nanocrystalline cuprous oxide (Cu2O) into modified polycaprolactone (mPCL) electrospun fibrous mats. The resulting materials were systematically characterized pertaining to physiochemical analysis, photoabsorption and photocurrent measurements, and photocatalytic reactive oxygen species (ROS) generation. The capability of this nanoplatform to elicit rapid, photoactivated, catalytically augmented bacterial disinfection was corroborated, displaying ∼99–100% broad-spectrum antibacterial efficacies against Staphylococcus aureus and Escherichia coli in 25 min. Benefiting from the release of Cu2+, mPCL-Cu2O impairs bacterial contamination even under dark conditions. Furthermore, these materials also exhibited favorable in vitro/-ovo biocompatibility toward NIH3T3 fibroblasts, red blood cells, and chick chorioallantoic membrane, alongside appreciable pro-healing capacity in bridging scratch-based wounds, which could be attributed to nanofiber-afforded biomimetic topography and controllable liberation of micronutrient Cu ions. This study provides insights into the design and fabrication of simple yet smart antibacterial nanoplatforms for biomedical applications.

Porous silica nanocarriers with gold/carbon quantum dots for photo-chemotherapy and cellular imaging

Yunyun Chen,Yanbao Zhao,Xueyan Zou,Lei Sun

The integration of multiple treatment methods for improving therapy efficiency has been widely concerned. Herein, porous silica (pSiO 2 ) carriers with center-radial channels were designed and synthesized, then the photosensitizer carbon dots (CDs) and antitumor drug doxorubicin (Dox) were adsorbed into the channels and Au nanoparticles (NPs) were also loaded in the pores of the pSiO 2 carriers. Biocompatible hyaluronic acid/alginate/chitosan (HCA) multilayers were coated onto the pSiO 2 carriers to seal the loaded-drug pores. The CDs are capable of generating singlet oxygen and multicolor imaging, and Au NPs have good photothermal effect. The obtained CDs/Dox-pSiO 2 -Au/HCA not only has enhanced synergistic chemotherapy, photothermal therapy (PTT) and photodynamic therapy (PDT), but also exhibits pH/redox/enzyme-triggered drug release.

Artificial Assembled Macrophage Co-Deliver Black Phosphorus Quantum Dot and CDK4/6 Inhibitor for Colorectal Cancer Triple-Therapy

Yuxiao Fang,Zipeng Zhang,Yang Liu,Tong Gao,Shuang Liang,Qihui Chu,Li Guan,Weiwei Mu,Shunli Fu,Huizhen Yang,Na Zhang,Yongjun Liu

In recent years, therapeutic strategies based on macrophages have been inspiringly developed, but due to the high intricacy and immunosuppression of the tumor microenvironment, the widespread use of these strategies still faces significant challenges. Herein, an artificial assembled macrophage concept (AB@LM) was presented to imitate the main antitumor abilities of macrophages of tumor targeting, promoting the antitumor immunity, and direct tumor-killing effects. The artificial assembled macrophage (AB@LM) was prepared through an extrusion method, which is to fuse the macrophage membrane with abemaciclib and black phosphorus quantum dot (BPQD)-loaded liposomes. AB@LM showed good stability and tumor targeting ability with the help of macrophage membrane. Furthermore, AB@LM reversed the immunosuppressive tumor microenvironment by inhibiting regulatory T cells (Tregs) and stimulating the maturation of antigen-presenting cells to activate the antitumor immune response through triggering an immunogenic cell death effect. More importantly, in the colorectal tumor model in vivo, a strong cooperative therapeutic effect of photo/chemo/immunotherapy was observed with high tumor inhibition rate (95.3 ± 2.05%). In conclusion, AB@LM exhibits excellent antitumor efficacy by intelligently mimicking the abilities of macrophages. A promising therapeutic strategy for tumor treatment based on imitating macrophages was provided in this study.

Multistage targeted “Photoactive neutrophil” for enhancing synergistic photo-chemotherapy

Yurui Xu,Xiaomin Zhang,Getian Hu,Xiaotong Wu,Yuanyuan Nie,Heming Wu,Desheng Kong,Xinghai Ning

Cell-based drug delivery system holds a great promise in anticancer treatment, due to its potential of maximizing therapeutic efficacy while minimizing adverse effects. However, current cell system can only deliver drugs in tumor lesions, but lack an ability to target subcellular locus of therapeutic actions, thereby compromising anticancer efficacy. Herein, we bioengineered living neutrophils as a novel type of “Photoactive neutrophil” (PAN) with capabilities of self-amplified multistage targeting and inflammation response for enhancing mitochondria-specific photo-chemotherapy. PAN encapsulated multifunctional nanocomplex (RA/Ce6) of RGD-apoptotic peptide conjugate (RA) decorated liposomal photosensitizer Ce6, and could overcome tumor barriers to selectively release RA/Ce6 within tumor. Consequently, RA/Ce6 actively entered cancer cells and accumulated in mitochondria to trigger combined photodynamic therapy (PDT) and RA-induced mitochondrial membrane disruption, resulting in enhanced therapeutic effects. Importantly, PAN exhibited inflammation amplified tumor targeting after PDT, and initiated combined photo-chemotherapy to suppress tumor growth without adverse effects, leading to prolonged mice survival. Therefore, PAN represents the first multistage targeted cell therapy , and brings new insights into cancer treatment.

Tumor Microenvironment Responsive Drug-Dye-Peptide Nanoassembly for Enhanced Tumor-Targeting, Penetration, and Photo-Chemo-Immunotherapy

Jinrong Peng,Qian Yang,Yao Xiao,Kun Shi,Qingya Liu,Ying Hao,Fan Yang,Ruxia Han,Zhiyong Qian

Nanomedicine constructed by therapeutics has unique and irreplaceable advantages in biomedical applications, especially in drug delivery for cancer therapy. The strategy, however, used to construct the therapeutics-based nanomedicines with tumor microenvironmental factor responsiveness is still sophisticated. In this study, an easy-operating procedure is used to construct a therapeutics-based nanosystem with active tumor-targeting, enhanced penetration, and stimuli-responsive drug release behavior as well as programmed cell death-1/programmed cell death-ligand 1 (PD-1/PD-L1) blockading mediated immunomodulation to enhance tumor immunotherapy. The matrix metalloproteinase-2 responsive peptide with the existence of Lyp-1 sequence contributes to the success of active tumor-targeting and the enhancement of the penetration of the nanoparticles in tumor tissue. The obtained nanosystem strikingly inhibits the primary tumor growth in the first 24 h (more than 97.5% of tumor cells are inhibited), and total inhibition can be achieved with the combination of photothermal therapy. IR820, which is served as the carrier for the therapeutics, is used as a photosensitizer for photothermal therapy. The progress and aggression of distal tumor has further been alleviated by a d -peptide which is an antagonist for PD-1/PD-L1 blockage. Therefore, a therapeutics-constructed multifunctional nanosystem is provided to realize a combinational therapeutic strategy to enhance the therapeutic outcome.

Charge reversal hairpin peptide modified synergy therapeutic nanoplatforms for tumor specific drug shuttling

Zhechen Fan,Zhongsheng Ji,Feng Zhang,Peng Luo,Huaqing Zhang,Jianping Zhou,Hao Cheng,Yang Ding

Given the distinct pathological features of neoplasm tissues, multifunctional responsive nanocarriers have been recently considered as promising candidates to optimize the chemotherapy regime. As a result, we propose a graphene oxide-based pH-responsive drug delivery system via covalent assembly of “hairpin-like” cell penetrating peptides with acid sensitive and charge reversal properties to realize superior tumor specificity and lower toxicity. Graphene oxide here can serve as high doxorubicin-loading nanosheets and facilitate swift drug release in response to laser irradiation, which provides an efficient platform for the synergy of photo-chemotherapy. Structurally, polyglycol conjugation on the graphene oxide surface fulfils the function of nanocomposite stabilization. After administration, the elaborately acid sensitive cell penetrating peptides maintain the hairpin structure under physiological conditions, while after entering the tumor acidic microenvironment, they undergo charge reversal and structural conversion to promote the cellular uptake of nanoparticles. The evaluation of nanocomposites in vitro revealed their negligible systematic toxicity and remarkable antitumor effects. In vivo experiments also confirmed the impressive stability and tumor-specific targeting for alleviating breast cancer. In conclusion, hairpin peptide modified graphene oxide nanoparticles show multiple merits including high drug carrying capacity, selective tumor penetration, responsive drug release and effective combination oncotherapy.

An efficient photo-chemo combination therapeutic platform based on targeted reduction-responsive self-crosslinked polymer nanocapsules

Shengbei Qi,Chaoyong Liu,Meng Qin,Cheng Tao,Jie Liu,Yuan Le,Jiexin Wang,Jianjun Zhang,Yunfeng Lu

Photo-chemo combination therapy holds great promises for cancer treatment; thus developing efficient approaches for delivering photo-chemo combination reagents has long been pursued. Herein, an amphiphilic branched copolymer, four-arm poly(ethylene glycol)-poly(propylene sulfide)-cyclic(Arg-Gly-Asp-D-Phe-Lys) (PEG-PPS-cRGD), was synthesized to prepare targeted self-crosslinked polymer nanocapsules consisting of a disulfide bond and the αvβ3 integrin-targeting ligand cRGD. Furthermore, by coloading both indocyanine green (ICG) and doxorubicin (DOX) in PEG-PPS-cRGD nanocapsules, a high-performance photo-chemo combination therapeutic platform designated as DINCs was successfully developed. The dissolution rate investigations demonstrated that DINCs exhibited sensitive reduction-responsive drug release profiles during the incubation with glutathione (GSH), while the drug release rate could be further accelerated by NIR laser irradiation. The in vitro studies showed that DINCs had outstanding cellular uptake, photothermal effects and ROS generation under laser irradiation, which significantly enhanced cytotoxicity in the treatment of squamous cells (SCC-15). Moreover, the in vivo results showed that synergistic photo-chemo combination treatment by DINCs plus laser irradiation could completely eliminate the tumor of squamous cell carcinoma with favorable biological safety.
