Development of ultraviolet-B long-lived persistent phosphors in Pr3+-doped garnets

Shao Yan,Yanjie Liang,Jingwei Liu,Dongxun Chen,Shihai Miao,Jianqiang Bi,Kangning Sun

Inorganic luminescent materials that emit in the ultraviolet-B (UVB) region (280–320 nm) have gained considerable attention in recent years because of the growing demands for applications in photochemistry and photomedicine. However, most of the available UVB phosphors are photoluminescent, for which continuous external excitation is essential. This common but very inconvenient luminescence form hinders the further development of UVB luminescence technology. Here, we report the design and development of a series of Pr3+-doped garnet-based UVB persistent phosphors by combining conduction band engineering, the vacuum referred binding energy (VRBE) diagram and persistent energy transfer control. The developed UVB persistent phosphors can not only be charged using a standard 254 nm UV lamp but also natural sunlight, leading to an intense and long-lasting UVB afterglow of more than 60 h. Besides, the persistent luminescence properties and trap charging and detrapping processes are comprehensively investigated using persistent luminescence excitation spectra and thermoluminescence experiments to uncover the possible luminescence mechanism in these phosphors. More importantly, the unique UVB persistent light emission from these garnet phosphors can be clearly imaged and recorded using a UVB camera in a bright indoor-lighting environment because of the zero-background noise from indoor ambient light, indicating that these UVB persistent phosphors have important application potential for optical tagging and optical data storage in a bright environment.

Differential Diagnosis and Precision Therapy of Two Typical Malignant Cutaneous Tumors Leveraging Their Tumor Microenvironment: A Photomedicine Strategy

Peiru Wang,Weitao Yang,Shuzhan Shen,Chao Wu,Long Wen,Qian Cheng,Bingbo Zhang,Xiuli Wang

Elevated hydrogen peroxide (H2O2) in biological tissues is generally recognized to be relevant to the carcinogenesis process that regulates the proliferative activity of cancer cells and the transformation of malignant features. Inspired by this observation, it can be hypothesized that imaging H2O2 in the tumor microenvironment (TME) could help diagnose tumor types and malignancy, and even guide precise therapy. Thus, in this study, a noninvasive photomedicine strategy is demonstrated that leverages the different levels of H2O2 in the TME, and two representative skin cancers, malignant melanoma (MM, clinically higher incidence of metastasis and recurrence) and cutaneous squamous cell carcinoma (cSCC, relatively less dangerous), are differentially diagnosed. The working probe used here is one we previously developed, namely, intelligent H2O2 responsive ABTS-loaded HRP@Gd nanoprobes (iHRANPs). In this study, iHRANPs have advantages over ratiometric imaging due to their bimodal imaging elements, in which the inherent magnetic resonance imaging (MR) mode can be used as the internal imaging reference and the H2O2 responsive photoacoustic (PA) imaging modality can be used for differential diagnosis. Results showed that after intravenous injection of iHRANPs, the tumor signals on both MM and cSCC are obviously enhanced without significant difference under the MR modality. However, under the PA modality, MM and cSCC can be easily distinguished with obvious variations in signal enhancement. Particularly, guided by PA imaging, photothermal therapy (PTT) can be precisely applied on MM, and a strong antitumor effect was achieved owing to the excessive H2O2 in the TME of MM. Furthermore, exogenous H2O2 was injected into cSCC to remedy H2O2 deficiency in the TME of cSCC, and an evident therapeutic efficacy on cSCC can also be realized. This study demonstrated that MM can be differentially diagnosed from cSCC by noninvasive imaging of H2O2 in the TME with iHRANPs; meanwhile, it further enabled imaging-guided precision PTT ablation, even for those unsatisfactory tumor types (cSCC) through exogenously delivering H2O2.

An activatable type I photosensitizer for NIR fluorescence imaging and photodynamic therapy of drug-resistant bacterial infections

Zhongkun Wang,Rui Zhu,Chenrui Li,Weiwei Xu,Liangyou Zhao,Guangjin Gao,Weihao An,Xiang Dong,Qingqing Miao,Qing Li

Bacterial infections pose a significant global health challenge, further exacerbated by the emergence of antibiotic-resistant strains, thereby necessitating the development of alternative therapeutic strategies. Type I photosensitizers, as an emerging phototheranostic modality, hold great promise for antibacterial treatment but are limited by poor specificity. Herein, we report a hydrogen sulfide (H 2 S)-activatable theranostic probe (CyIN) that integrates near-infrared (NIR) fluorescence imaging and type I photodynamic therapy (PDT) functionalities into a single platform for the precise treatment of drug-resistant bacterial infections. CyIN is rationally constructed by masking the hydroxyl group of iodine-substituted hemicyanine with a H 2 S-cleavable 2,4-dinitrophenyl group, which efficiently quenches NIR fluorescence and suppresses reactive oxygen species generation. Such molecular configuration allows CyIN to turn on its NIR fluorescence signal and type I photodynamic action upon interaction with bacterial-derived H 2 S, achieving distinguishable detection of drug-resistant bacterial infection, potent bacterial clearance (>99 %), and accelerated healing of bacteria-infected wounds. Furthermore, histological analyses confirm that the CyIN treatment can reduce inflammation, enhance collagen deposition, and promote tissue regeneration without systemic toxicity. Thus, this work presents a promising H 2 S-responsive phototheranostic platform for precise bacterial detection and efficient photodynamic eradication of drug-resistant bacterial infections.

Battery-Free Optoelectronic Patch for Photodynamic and Light Therapies in Treating Bacteria-Infected Wounds

Zhao Xue,Wenxin Chou,Yixuan Xu,Ziyi Cheng,Xuechun Ren,Tianzhen Sun,Wenbin Tong,Yang Xie,Junyu Chen,Nuohan Zhang,Xing Sheng,Yongtian Wang,Hongyou Zhao,Jian Yang,He Ding

Light therapy is an effective approach for the treatment of a variety of challenging dermatological conditions. In contrast to existing methods involving high doses and large areas of illumination, alternative strategies based on wearable designs that utilize a low light dose over an extended period provide a precise and convenient treatment. In this study, we present a battery-free, skin-integrated optoelectronic patch that incorporates a coil-powered circuit, an array of microscale violet and red light emitting diodes (LEDs), and polymer microneedles (MNs) loaded with 5-aminolevulinic acid (5-ALA). These polymer MNs, based on the biodegradable composite materials of polyvinyl alcohol (PVA) and hyaluronic acid (HA), serve as light waveguides for optical access and a medium for drug release into deeper skin layers. Unlike conventional clinical photomedical appliances with a rigid and fixed light source, this flexible design allows for a conformable light source that can be applied directly to the skin. In animal models with bacterial-infected wounds, the experimental group with the combination treatment of metronomic photodynamic and light therapies reduced 2.48 log 10 CFU mL –1 in bactericidal level compared to the control group, indicating an effective anti-infective response. Furthermore, post-treatment analysis revealed the activation of proregenerative genes in monocyte and macrophage cell populations, suggesting enhanced tissue regeneration, neovascularization, and dermal recovery. Overall, this optoelectronic patch design broadens the scope for targeting deep skin lesions, and provides an alternative with the functionality of standard clinical light therapy methods.

Metal–Organic Frameworks-Derived Carbon Nanoparticles for Photoacoustic Imaging-Guided Photothermal/Photodynamic Combined Therapy

Peng Yang,Ye Tian,Yongzhi Men,Ranran Guo,Haibao Peng,Qin Jiang,Wuli Yang

Combination of photothermal therapy (PTT) and photodynamic therapy (PDT) has become a promising cancer treatment in recent years. However, their applications are limited by complex synthetic protocols and low efficacy. Hence, optimizing experimental approach and improving the efficiency of phototherapy is the current research focus. In this work, various pyrolysis temperatures and sizes of zeolitic imidazolate framework-8 (ZIF-8) derived carbon nanoparticles (ZCNs) are obtained by a simple direct pyrolysis of the ZIF-8 nanoparticles. Meanwhile, the ZCNs can be used as photothermal agents and photosensitizers to produce heat and reactive oxygen species simultaneously upon near-infrared laser irradiation. Moreover, it is observed that the phototherapy effects and photoacoustic (PA) signal of ZCNs could be enhanced with the increase in the nanoparticle size. Subsequently, guided by PA imaging, the therapeutic effect of ZCNs is investigated on a small animal model, where tumors are entirely eliminated with minimal side effect, demonstrating the high efficacy of the larger size of ZCNs through combination of PTT and PDT. Therefore, it is expected that the ZCN is a simple and highly effective phototherapeutic platform for oncotherapy, and the concept of size-dependent enhanced behavior of phototherapy and PA imaging will be very useful in the development of nanomaterials for cancer therapy.

Mesoporous semiconductors combined with up-conversion nanoparticles for enhanced photodynamic therapy under near infrared light

Fan Yang,Jun Liu,Xue Jiang,Weiwei Wu,Zhenni Wang,Qi Zeng,Ruichan Lv

Photodynamic therapy (PDT) is a promising and effective method for tumor therapy that relies on the reactive oxygen species (ROS) produced by photosensitizers at specific wavelengths to inhibit tumor cells. Inorganic semiconductive materials are potential photosensitizers that can excellently absorb ultraviolet light to produce ROS to kill cancer cells. However, this strategy is still limited in terms of practical applications due to the weak penetration of ultraviolet light through biological tissue, as well as the hypoxic tumor microenvironment, largely decreasing ROS generation. In this research, novel PDT agents made with mesoporous lanthanide-semiconductor composites are developed to obtain a remarkable amount of generated ROS under near-infrared (NIR) laser irradiation. Due to the larger size (about 120 nm) of the up-conversion material (UCM) used as the substrate, coated with different amounts of semiconductors with mesoporous morphologies, this platform could emit higher blue emission under a 980 nm laser. Meanwhile, both of the semiconductors (SnO2 and TiO2) used have wide absorbance bands in the ultraviolet region, and the ultraviolet fluorescence emitted from the UCM core under NIR laser excitation can be used as the energy donor. Electron transfer processes in SnO2 and TiO2 are generated via the above platforms and produce ROS through photochemical action. Furthermore, the coated semiconductors are mesoporous with larger surface areas (about 302 m2 g−1) and various channels; this is beneficial to obtain enough oxygen to generate more ROS under a hypoxic environment. The PDT efficiency of a typical NaYF4@SnO2 sample is studied using a DPBF detector, in vitro MTT assays, and in vivo tumor inhibition experiments, revealing that this lanthanide-semiconductor platform could be potentially used as a PDT agent under NIR excitation.

Diagnostic and Therapeutic Integration Prodrug: Dynamic Probe Engineering via Cys/GSH Depletion to Amplify Type I Photodynamic Therapy for Breast Cancer

Yongfei Huang,Junping Liang,Wenbo Lu,Chenzhong Li,Zhefeng Fan

Cancer is a serious threat to human health, and it is essential to find effective methods for detecting and treating cancer. Diagnostic integration is a new strategy for cancer diagnosis and treatment that enables localized detection and treatment of tumors in a single administration and effectively reduces the toxic side effects on normal tissues. It remains a challenge for small-molecule probes to achieve multifunctionality in sensing, targeting, imaging, and therapy. Photodynamic therapy (PDT), which uses photosensitizers to generate reactive oxygen species under specific wavelengths of light to ablate tumor tissue, has demonstrated unique advantages in tumor treatment. However, the high expression of Cys/GSH in the tumor region will consume the reactive oxygen species generated by photosensitizers during PDT and reduce the therapeutic effect of the tumor. In this study, the highly expressed Cys/GSH in the tumor microenvironment was used as the activation source, and benzopyrylium salt and coumarin dyes were selected as fluorophores, with the ether bond and aldehyde group as recognition sites, to construct a near-infrared-activated Cys/GSH fluorescent probe (Bpl-O-Cou). Based on the intramolecular rearrangement mechanism and intramolecular cyclization mechanism, Bpl-O-Cou could discriminate the detection of Cys/GSH accompanied by different fluorescence emissions. Under near-infrared laser irradiation, Bpl-O-Cou + Cys/GSH could produce •OH and O2–•, leading to 4T1 cell death. In vivo experiments further illustrated the interventional role of Bpl-O-Cou in the PDT process. This study provides a new idea for the design of multifunctional probes for cancer diagnostics and therapeutics.

Research on the Combined Antibacterial and Wound-Healing Effects of Oxygen-Carrying Hydrogel Photodynamic and Photothermal Therapy-Targeting Biofilms

Weiwei Zhang,Yongqi Yang,Xuanjun Zhang,Ping Song,Xiaokuo Shen,Lin Gui,Longbao Zhu,Dongdong Sun,Fei Ge,Wanzhen Li

Biofilms present a significant obstacle in the treatment of bacterial infections, because of their ability to evade the host immune response and to resist conventional antibacterial drugs. This study introduces an innovative approach to overcoming these challenges by targeting the biofilm microenvironment using a multifunctional therapeutic platform. The platform leverages perfluorohexane (PFH) as an oxygen carrier to alleviate the multifunctional therapeutic platform. It combines the photosensitizer indium Chlorin e6 (Ce6) and the photothermal agent molybdenum disulfide (MoS2) co-encapsulated within a liposome nanoparticle structure, further stabilized by Zeolitic imidazolate framework-8 (ZIF-8). This configuration, termed MoS2–Ce6-PFH@ZIF-8 (MCP@ZIF-8) improves the stability, functionality, and therapeutic potential of the nanoparticles. Additionally, the nanoparticles are embedded into a hydrogel matrix (MCP@ZCP formed by cross-linking carboxymethyl chitosan (CMCS) with pluronic F-127 (PF-127) for localized delivery. MCP@ZIF-8 exhibited a high photothermal therapy conversion efficiency of approximately 52.7%, thereby enabling effective heat generation under near-infrared (NIR) light. MCP@ZIF-8 produced higher reactive oxygen species levels than Ce6 alone under NIR irradiation at 660 nm. This dual photothermal and photodynamic mechanism achieves an antibacterial efficacy of up to 99%, effectively disrupting biofilms. Animal studies demonstrated that MCP@ZCP promotes superior wound healing, compared with conventional treatments. Transcriptome analysis demonstrated that MCP@ZCP disrupts bacterial metabolic pathways including amino acid synthesis, carbon metabolism, nitrogen metabolism, and other pathways, thereby inhibiting bacterial growth and proliferation. Comprehensive biosafety assessments have confirmed that MCP@ZCP is biocompatible and nontoxic, indicating its suitability for clinical applications. Thus, oxygen-carrying hydrogels can be used as an approach for treating bacterial infections.

An AIE cyanine photosensitizer achieving hypoxic tumor ablation through synergistic treatment of type i photodynamic and photothermal therapy

Peng-Wei Chen,Lin-Song Li,Yu-Meng Zhang,Bang-Bang Liu,Xianjiao Tang,Liping Chen,Xiaojing Yang,Mei-Xia Zhao

In recent years, most photosensitizers suffer from poor photothermal conversion efficiency (PCE), vulnerability to aggregated-caused quenching under physiological conditions, and are highly dependent on oxygen, which seriously limit their application in cancer therapy. In this study, DTIR780 was an aggregation induced emission (AIE) photosensitizer, which was synthesized through the originality of molecular engineering. Because DTIR780 had a special organelle targeting lysosomes and mitochondria, it had excellent PCE (PCE = 28.9 %), and could simultaneously generate Type I reactive oxygen species (ROS): O 2 ·- and ·OH and Type II ROS: 1 O 2 , showing significantly enhanced tumor phototoxicity in both normoxia and hypoxic tumor microenvironments. In vivo testing demonstrated that DTIR780 NPs was efficiently accumulated at the tumor site while significantly inhibiting tumor growth in a subcutaneous H22 tumor mouse model. In summary, this lysosome and mitochondria targeted DTIR780 NPs has great potential in clinically image-guided type I photodynamic therapy (PDT) and photothermal therapy (PTT), offering a novel avenue for developing high-performance type I PDT/PTT photosensitizers for the efficient treatment of hypoxia tumors.

A NIR-responsive upconversion implant for wireless photodynamic therapy of tumors

Cijun Shuai,Jie Zeng,Zhaoxiong Zhang,YouWen Yang,MingLi Yang,Hengyun Ye,Pan He,Jun Zan,Jun Zhang

Photosensitizer-mediated photodynamic therapy (PDT) enables wireless tumor inactivation by releasing cytotoxic reactive oxygen species (ROS). Nevertheless, the activation of conventional photosensitizers typically depends on visible light with limited tissue penetration, thereby restricting their therapeutic efficacy in deep-seated tumors. To overcome this limitation, an upconversion implant was engineered to function as a flexible internal light-emitting device by converting deeply penetrating near-infrared-I (NIR-I) light into visible light. Specifically, a composite PDT nanosystem was constructed by linking NaYF 4 :Yb,Er upconversion nanoparticles with the rose bengal photosensitizer—whose emission and absorption spectra are well-matched—using polyvinylpyrrolidone (PVP) as a molecular bridge. This PDT nanosystem was subsequently incorporated into poly-L-lactic acid (PLLA) scaffolds fabricated via selective laser sintering. Under 980 nm laser irradiation, the upconversion implant was capable of wirelessly emitting upconverted visible light through more than 2 cm of an in vitro tissue model and generating abundant ROS via the photodynamic effect. In vitro cell experiments demonstrated that the system effectively eliminated tumor cells by damaging the cell membrane and cellular genetic material. These results suggest that the scaffolds possess significant potential as anti-tumor tissue implants and offer a promising avenue for the development of innovative tumor treatment strategies.

Exploring the singlet oxygen generation of iridium(III) complex containing phenothiazine for photodynamic therapy

Qinglin Chen,Hong Zeng,Jie Wu,Yuanyan Li,Yibao Li,Yongquan Wu

As a photochemical-based treatment approach, photodynamic therapy (PDT) relies on a photosensitiser (PS) that produces reactive oxygen species (ROS) after irradiation, which lead to irreparable cell damage and hence cell death. Cyclometalated iridium(III) complexes are often used in the development of PSs attributed to their rich photophysical properties with high quantum yields, large Stokes shifts, long-lived phosphorescence and good photostability. In addition, phenothiazine has excellent electron-donating ability and good rigid structure, its derivatives have been shown to possess activity against bacteria, malignant cells, and protozoa. In this work, we designed and synthesized two iridium(III) complexes DF-Ir-CHO and DF-Ir-PTZ. The DF-Ir-PTZ contains a novel ligand functionalized with phenothiazine. DF-Ir-PTZ has a better singlet oxygen ( 1 O 2 ) production quantum yield of 0.357 upon light irradiation and negligible dark cytotoxicity, suggesting that its biological safety is favorable. In addition, DF-Ir-PTZ was able to generate 1 O 2 and ROS in cancer cells under light conditions, leading to cell apoptosis of MCF-7.

Multistage targeted “Photoactive neutrophil” for enhancing synergistic photo-chemotherapy

Yurui Xu,Xiaomin Zhang,Getian Hu,Xiaotong Wu,Yuanyuan Nie,Heming Wu,Desheng Kong,Xinghai Ning

Cell-based drug delivery system holds a great promise in anticancer treatment, due to its potential of maximizing therapeutic efficacy while minimizing adverse effects. However, current cell system can only deliver drugs in tumor lesions, but lack an ability to target subcellular locus of therapeutic actions, thereby compromising anticancer efficacy. Herein, we bioengineered living neutrophils as a novel type of “Photoactive neutrophil” (PAN) with capabilities of self-amplified multistage targeting and inflammation response for enhancing mitochondria-specific photo-chemotherapy. PAN encapsulated multifunctional nanocomplex (RA/Ce6) of RGD-apoptotic peptide conjugate (RA) decorated liposomal photosensitizer Ce6, and could overcome tumor barriers to selectively release RA/Ce6 within tumor. Consequently, RA/Ce6 actively entered cancer cells and accumulated in mitochondria to trigger combined photodynamic therapy (PDT) and RA-induced mitochondrial membrane disruption, resulting in enhanced therapeutic effects. Importantly, PAN exhibited inflammation amplified tumor targeting after PDT, and initiated combined photo-chemotherapy to suppress tumor growth without adverse effects, leading to prolonged mice survival. Therefore, PAN represents the first multistage targeted cell therapy , and brings new insights into cancer treatment.
