Formate, which can be produced from CO 2 with green energy, has received increasing attentions as a carbon source for bioconversion. Microalgae have the native advantages of use captured light energy to facilitate the formate assimilation. However, the low efficient natural conversion and inhibition to the electron transfer chain obstacle its applications’ development. Here, using green microalga C. reinhardtii 21gr as the model, an approach to improve formate-based biomass and protein production was raised and verified through isotope labeling experiments, and the pathway of formate which presented in photosynthetic organisms was related to amino acids. By the enhancement of chloroplast glyceraldehyde-3-phosphate dehydrogenase (cGAPDH) expression, together with the adaptive laboratory evolution (ALE) over regular lethal dose of formate, the yields of the biomass and proteins were promoted with 34 % and 55 % to the wild type, respectively. Indicated by the photosynthetic activity variations, it was proposed that the balance of cellular energy and reduction power is necessary for microalgae to achieve high productivity with formate. Further investigated by proteomics, 6-phosphogluconate dehydrogenase (6-PGD) was identified as the key enzyme to formate and nitrogen’s co-assimilation.

Multispecies biofilm exhibited high resistance to nanotoxicity by secreting extracellular polymeric substances (EPS) and undergoing alterations in the community composition. Scarce information was available to assess how these changes could further influence the transfer of nanoparticles (NPs) through the biofilm-based food chain. Photic biofilm was exposed to two distinct NPs (ZnO and TiO 2 ) and subsequently grazed by snails. Exposure to different NPs led to variations in biomass, chlorophyll content, EPS productivity, alpha diversity, and community composition of the photic biofilm. The presence of ZnO NPs facilitated the growth of phylum Cyanobacteria while TiO 2 promoted EPS productivity of photic biofilm. EPS were capable of embedding NPs (TiO 2 and ZnO) within its matrix, thereby mitigating their aggregation within the biofilm matrix. These alterations were subsequently confirmed to have an impact on the trophic transfer factors (TTF) of NPs through the constructed biofilm-snail food chain. The TTF of ZnO was lower than that of TiO 2 in feeding scenario 1 (only fed on TiO 2 or ZnO biofilm) but higher than that of TiO 2 in feeding scenario 2 (fed on TiO 2 and ZnO biofilm simultaneously), which was attributed to the shifts in the algae composition and a smaller size of ZnO NPs in EPS. This study demonstrated that the response of photic biofilm to NPs further affected the TTFs of NPs through the food chain.

The soil-water interface is replete with photic biofilm and iron minerals; however, the potential of how iron minerals promote biotic nitrate removal is still unknown. This study investigates the physiological and ecological responses of photic biofilm to hematite (Fe 2 O 3 ), in order to explore a practically feasible approach for in-situ nitrate removal. The nitrate removal by photic biofilm was significantly higher in the presence of Fe 2 O 3 (92.5%) compared to the control (82.8%). Results show that the presence of Fe 2 O 3 changed the microbial community composition of the photic biofilm, facilitates the thriving of Magnetospirillum and Pseudomonas , and promotes the growth of photic biofilm represented by the extracellular polymeric substance (EPS) and the content of chlorophyll. The presence of Fe 2 O 3 also induces oxidative stress (•O 2 − ) in the photic biofilm, which was demonstrated by electron spin resonance spectrometry . However, the photic biofilm could improve the EPS productivity to prevent the entrance of Fe 2 O 3 to cells in the biofilm matrix and mitigate oxidative stress. The Fe 2 O 3 then promoted the relative abundance of Magnetospirillum and Pseudomonas and the activity of nitrate reductase , which accelerates nitrate reduction by the photic biofilm. This study provides an insight into the interaction between iron minerals and photic biofilm and demonstrates the possibility of combining biotic and abiotic methods to improve the in-situ nitrate removal rate.

Photo-bioelectrochemical fuel cell (PBFC) represents a promising technology for enhancing removal of antibiotic pollutants while simultaneously sustainable transformation of organic wastes and solar energy into electricity. In this study, simultaneous antibiotic removal and bioelectricity generation were investigated in a PBFC with daily light/dark cycle using oxytetracycline (OTC) as a model compound of antibiotic. The specific OTC removal rate increased by 61% at an external resistance of 50 Ω compared to that in the open-circuit control, which was attributed to bioelectrochemically enhanced co-metabolic degradation in the presence of the bioanode. The OTC removal was obviously accelerated during illumination of cathode in contrast with a dark cathode due to the higher driving force for anodic bioelectrochemical reaction by using photosynthetic oxygen as cathodic electron acceptor during illumination than that using nitrate in dark. The bioelectrocatalytic activity of anodic biofilm was continuously enhanced even at an initial OTC concentration of up to 50 mg L −1 . The degradation products of OTC can function as mediators to facilitate the electron transfer from bacteria to the anode, resulting in 1.2, 1.76 and 1.8 fold increase in maximum power output when 10, 30 and 50 mg L −1 OTC was fed to the bioanode, compared to the OTC-free bioanode, respectively. The OTC feeding selective enriched OTC-tolerant bacterial community capable of degrading complex organic compounds and producing electricity. The occurrence of ARGs during bioelectrochemical degradation of OTC was affected more greatly by the succession of the anodic bacterial community than the initial OTC concentration.

The development of photosynthetic biohybrid systems (PBSs) integrating inorganic light absorbers with non-photosynthetic bacteria innovate wastewater valorization via organics bioconversion, yet interfacial electron transfer bottlenecks limit efficiency. To address this, we regulated crystal facet exposure ratios in cadmium sulfide/reduced graphene oxide (CdS/RGO) through hydrothermal synthesis time control, observing facet-dependent activity trends. The optimized biohybrid achieved a maximum hydrogen yield of 2195.3 μmol (233.6 µmol·g −1 ·h −1 over 8 h), representing a 295 % enhancement over unmodified PBSs. Density functional theory (DFT) calculations revealed that increased exposure of high-activity (1 0 3) and (1 1 2) facets correlated with reduced work function values, promoting electron emission. Synergistically combined with RGO’s electron-shuttling function, this facilitated charge transfer to bacterial outer membrane proteins. These results demonstrate facet engineering as a tunable strategy for enhancing electron donation capacity in PBSs, offering design principles for biohybrid systems.

Algal-bacterial photo-bioelectrochemical system (ABPBS) operated with daily light/dark cycle provides a novel approach for sustainable treatment of wastewater through the uses of solar energy and bioenergy . However, it can not be effectively driven when the photosynthetic oxygen supply by algae at the cathode stops at night. In this study, a solar photovoltaic capacitor discharging was integrated into the ABPBS to achieve all-weather synchronous enhanced antibiotic and nitrogen removal from wastewater. The results showed that 3.3 F, 10 F and 100 F capacitor discharge increased the degradation rate of florfenicol (FLO) by 44%, 89%, and 582% at the anode, the removal rate of ammonia nitrogen by 20.4%, 39.8%, and 55.6%, the accumulated nitrate was decreased by 70.0%, 86.5%, and 93.3%, and the accumulated nitrite was decreased by 48.6%, 45.7%, and 87.1% at the biocathode, respectively. There was a significant positive correlation between the capacity of the capacitor and its ability to promote FLO degradation and nitrogen removal. The capacitor discharge promoted the photosynthetic oxygen release and biocatalytic oxygen reduction , and stimulated the growth of functional bacteria capable of degrading complex organics through extracellular electron transfer in the anode and diverse nitrogen removal bacteria in the cathode, but there were structural differences of functional bacteria groups induced by the different capacities of capacitor discharges.
