γ -Glutamyltransferase (GGT, EC number: 2.3.2.2) is an enzyme critical for glutathione metabolism and serves as a key biomarker for liver function and disease. Diabetes mellitus, a prevalent metabolic disorder, often complicates liver function and increases the challenge of accurately monitoring hepatic GGT levels due to fluctuating metabolic conditions. In this study, we introduce a sulfonate-modified hemicyanine dye-based probe, HDP-GGT, designed for the targeted and real-time assessment of GGT activity in the liver. HDP-GGT combines the fluorescence and photoacoustic advantages of hemicyanine dyes with a specific peptide sequence that enhances sensitivity and specificity for GGT detection. Utilizing both in vitro and in vivo methodologies, the sulfonate-modified HDP-GGT probe enables dual-modal imaging of hepatic and serum GGT activity by photoacoustic and fluorescence models, respectively. To evaluate the clinical translational potential of HDP-GGT, we applied it to detect GGT in mouse serum, finding that the fluorescence signal intensity was highest in the diabetes mellitus (DM) group and significantly reduced in the diabetes mellitus + metformin (DM + Met) group, suggesting that HDP-GGT has promising clinical application prospects. Our results indicate that HDP-GGT is a potent tool for non-invasive monitoring of liver health and could play a crucial role in diagnosing and managing liver-related disorders, particularly in the context of diabetes.

Copper ions, implicated in processes such as oxidative stress and inflammation, are believed to play a crucial role in cardiovascular disease, a prevalent and deadly disease. Despite this, current diagnostic methods fail to detect early stage cardiovascular disease or track copper ion accumulation, limiting our understanding of the disease’s progression. Therefore, the development of noninvasive techniques to image copper ions in cardiovascular disease is urgently needed to enhance diagnostic precision and therapeutic strategies. In this study, we report the successful synthesis and application of a copper ion-activated photoacoustic probe, CS-Cu, which exhibits high sensitivity and selectivity toward copper ions both in vitro and in vivo. CS-Cu was able to noninvasively monitor the changes in copper ion levels and differentiate between different mice based on copper ions in urine. Furthermore, the probe demonstrated good photoacoustic stability and exhibited no significant toxicity in the mice. These findings suggest that CS-Cu could be a promising tool for early detection and monitoring of Cu2+ levels in vivo and urine, providing a new perspective on the role of copper ions in cardiovascular disease.

Recent investigations have revealed the tight connections between carboxylesterase (CE) and the lipid metabolism-associated disorders. In this work, based on the previous reports, a photoacoustic (PA) probe Hcy-CE was developed for the in vivo imaging of CE in two lipid metabolism disorders. The reaction with CE led to the departure of the carbamate quencher together with the linkage, thus to release of the reporter Hcy to exhibit the turn-on signals of PA reporting accompanied with the absorbance and fluorescence modes. The responses in all three modes showed high consistency. In the PA mode, the linear correlation range was CE level 0–10 U/L, while the limit of detection value was 0.52 U/mL. The optical performance of the probe Hcy-CE in the solution system was practical with high steadiness and selectivity. Moreover, Hcy-CE exhibited potential capability of monitoring the CE level in the induced foam cells. In particular, Hcy-CE realized the in vivo imaging of the CE level from multiple views and continuous cross-sections in the model mice of different groups. The combination of the PA imaging with the blood biochemical tests and immunohistochemically staining methods might help distinguish the three statuses of different mice from each other. This work provided meaningful information for the accurate diagnosis and regulation of the lipid metabolism-related disorders.

Nanocatalytic medicine, which aims to accurately target and effectively treat tumors through intratumoral in situ catalytic reactions triggered by tumor-specific environments or markers, is an emerging technology. However, the relative lack of catalytic activity of nanoenzymes in the tumor microenvironment (TME) has hampered their use in biomedical applications. Therefore, it is crucial to develop a highly sensitive probe that specifically responds to the TME or disease markers in the TME for precision diagnosis and treatment of diseases. In this work, a chiral photoacoustic (PA) nanoprobe (D/L-Ce@MoO 3 ) based on the H 2 O 2 -catalyzed TME activation reaction was constructed in a one-step method using d -cysteine (D-Cys) or l -cysteine (L-Cys), polymolybdate, and cerium nitrate as raw materials. The designed and synthesized D/L-Ce@MoO 3 chiral nanoprobe can perform in situ, non-invasive, and precise imaging of pharmacological acute liver injury. In vivo and in vitro experiments have shown that the D/L-Ce@MoO 3 probe had chiral properties, the CD signal decreased upon reaction with H 2 O 2 , and the absorption and PA signals increased with increasing H 2 O 2 concentration. This is because of the catalytic reaction between Ce ions doped in the nanoenzyme and the high expression of H 2 O 2 caused by drug-induced liver injury to produce ·OH, which has a strong oxidizing property to kill tumor cells and destroy the Mo-S bond in the probe, thus converting the chiral probe into an achiral polyoxometalate (POM) with PA signal.

Increasing evidence has emphasized the pivotal role of antibiotic tolerance in the evolution of antibiotic resistance , refractory infection and infection recurrence. Moreover, satisfactory treatment of infection relies highly on early, precise detection and subsequent timely therapeutic interventions. In this study, the phenotype of increased production of H 2 S in antibiotic-tolerant Staphylococcus aureus ( S. aureus ) was identified and proven to be essential to the maintenance of antibiotic tolerance. Hence, a photoacoustic probe for detecting infection as well as an antibiotic enhancer targeting bacterial endogenous H 2 S to eradicate antibiotic-tolerant bacteria was successfully developed based on a bismuth metal-organic framework (Bi-MOF). Bi-MOF achieved early in situ and precise photoacoustic imaging of infection by generating Bi 2 S 3 inside bacteria. Moreover, Bi-MOF enhanced the eradication efficacy of methicillin against tolerant S. aureus by downregulating the intracellular H 2 S levels. Overall, Bi-MOF offers a promising diagnostic tool for early, precise infection diagnosis and a therapeutic strategy for tolerant bacteria-related refractory infection.

Cardiovascular disease (CVD) is a chronic disease characterized by the accumulation of lipids and fibrous tissue within the arterial walls, potentially leading to vascular obstruction and an increased risk of heart disease and stroke. Hydroxyl radicals play a significant role in the formation and progression of CVD as they can instigate lipid peroxidation, resulting in cellular damage and inflammatory responses. However, precisely detecting hydroxyl radicals in CVD lesions presents significant challenges due to their high reactivity and short lifespan. Herein, we present the development and application of a novel activatable optical probe, Cy-OH-LP, designed to detect hydroxyl radicals in lipid-rich environments specifically. Built on the Cy7 molecular skeleton, Cy-OH-LP exhibits near-infrared absorption and fluorescence characteristics, and its specific response to hydroxyl radicals enables a turn-on signal in both photoacoustic and fluorescence spectra. The probe demonstrated excellent selectivity and stability in various tests. Furthermore, Cy-OH-LP was successfully applied in an in vivo model to detect hydroxyl radicals in mouse models, providing a potential tool for diagnosing and monitoring AS. The biosafety of Cy-OH-LP was also verified, showing low cytotoxicity and no significant organ damage in mice. The findings suggest that Cy-OH-LP is a promising tool for the specific detection of hydroxyl radicals in lipid-rich environments, providing new possibilities for research and clinical applications in the field of oxidative stress-related diseases.
