The stratum corneum of the outermost skin is an important barrier impeding transdermal
permeation, and permeation enhancers can reduce the barrier resistance
of the stratum corneum and enhance the permeation of drugs in tissues.
The optical imaging depth, signal intensity, and scattering
coefficient variation rules of skin tissues in time dimension are
obtained by using optical coherence tomography (OCT). The effect of
optical clearing agents (OCAs) on OCT imaging is obtained by
quantitatively analyzing the changes in the optical properties of
tissues. D-fructose, one of the monosaccharides, and sucrose, one of
the disaccharides, were selected for the ex vivo
optical clearing experiments on pig skin tissues utilizing the
dimethyl sulfoxide (DMSO) carrier effect. We find that DMSO synergized
with sugars applied to skin tissue has a more significant increase in
the optical imaging depth and signal intensity, and a reduction in the
scattering coefficient with an increasing concentration of DMSO. DMSO
with a high concentration and D-fructose with saturated concentration
(10:1; v/v) effectively reduce light attenuation in OCT imaging and
improve the image quality. This operation will also shorten the
application time to minimize skin damage from hyperosmotic agents.

Rationale: Intravital optical imaging is a significant method for investigating cerebrovascular structure and function. However, its imaging contrast and depth are limited by the turbid skull. Tissue optical clearing has a great potential for solving this problem. Our goal was to develop a transparent skull window, without performing a craniotomy, for use in assessing cerebrovascular structure and function. Methods: Skull optical clearing agents were topically applied to the skulls of mice to create a transparent window within 15 min. The clearing efficacy, repeatability, and safety of the skull window were then investigated. Results: Imaging through the optical clearing skull window enhanced both the contrast and the depth of intravital imaging. The skull window could be used on 2-8-month-old mice and could be expanded from regional to bi-hemispheric. In addition, the window could be repeatedly established without inducing observable inflammation and metabolic toxicity. Conclusion: We successfully developed an easy-to-handle, large, switchable, and safe optical clearing skull window. Combined with various optical imaging techniques, cerebrovascular structure and function can be observed through this optical clearing skull window. Thus, it has the potential for use in basic research on the physiopathologic processes of cortical vessels.

Various tissue optical clearing techniques have sprung up for large volume imaging. However, there are few methods showed clearing and imaging data on different organs while most of them were focused on mouse brain, and as a result, it is difficult to select the suitable method for organs in practical applications due to lack of quantitative evaluation and comprehensive comparison. Therefore, it is necessary to evaluate and compare the performances of clearing methods for different organs. In this paper, several typical optical clearing methods were applied, including 3DISCO, uDISCO, SeeDB, FRUIT, CUBIC, Sca l eS and PACT to clear intact brain, heart, kidney, liver, spleen, stomach, lung, small intestine, skin and muscle. The clearing efficiency, sample deformation, fluorescence preservation and imaging depth of these methods were quantitatively evaluated. Finally, based on the systemic evaluation of various parameters described above, the appropriate clearing method for specific organ including kidney or intestine was screened out. This paper will provide important references for selection of appropriate clearing methods in related researches.

Three-dimensional (3D) cell cultures provide an important model for various biological studies by bridging the gap between two-dimensional (2D) cell cultures and animal tissues. Microfluidics has recently provided controllable platforms for handling and analyzing 3D cell cultures. However, on-chip imaging of 3D cell cultures within microfluidic devices is hindered by the inherent high scattering of 3D tissues. Tissue optical clearing techniques have been used to address this concern but remain limited to fixed samples. As such, there is still a need for an on-chip clearing method for imaging live 3D cell cultures. Here, to achieve on-chip clearing for live imaging of 3D cell cultures, we conceived a simple microfluidic device by integrating a U-shaped concave for culture, parallel channels with micropillars, and differentiated surface treatment to enable on-chip 3D cell culture, clearing, and live imaging with minimal disturbance. The on-chip tissue clearing increased the imaging performance of live 3D spheroids with no influence on cell viability or spheroid proliferation and demonstrated robust compatibility with several commonly used cell probes. It allowed dynamic tracking of lysosomes in live tumor spheroids and enabled quantitative analysis of their motility in the deeper layer. Our proposed method of on-chip clearing for live imaging of 3D cell cultures provides an alternative for dynamic monitoring of deep tissue on a microfluidic device and has the potential to be used in 3D culture-based assays for high-throughput applications.

By allowing more photons to reach deeper tissue, the optical clearing agent (OCA) has gained increasing attention in various optical imaging modalities. However, commonly used OCAs have high fluidity, limiting their applications in in vivo studies with oblique, uneven, or moving surfaces. In this work, we reported an OCA with high viscosity. We measured the properties of this viscous OCA, and tested its successful performances in the imaging of a living animal’s skin with two optical imaging modalities: photoacoustic microscopy and optical coherence tomography. Our results demonstrated that the viscous OCA has a great potential in the study of different turbid tissues using various optical imaging modalities.

A handheld Raman detector with operational convenience, high portability, and rapid acquisition rate has been applied in clinics for diagnostic purposes. However, the inherent weakness of Raman scattering and strong scattering of the turbid tissue restricts its utilization to superficial locations. To extend the applications of a handheld Raman detector to deep tissues, a gold nanostar-based surface-enhanced Raman scattering (SERS) nanoprobe with robust colloidal stability, a fingerprint-like spectrum, and extremely high sensitivity (5.0 fM) was developed. With the assistance of FPT, a multicomponent optical clearing agent (OCA) efficiently suppressing light scattering from the turbid dermal tissues, the handheld Raman detector noninvasively visualized the subcutaneous tumor xenograft with a high target-to-background ratio after intravenous injection of the gold nanostar-based SERS nanoprobe. To the best of our knowledge, this work is the first example to introduce the optical clearing technique in assisting SERS imaging in vivo. The combination of optical clearing technology and SERS is a promising strategy for the extension of the clinical applications of the handheld Raman detector from superficial tissues to subcutaneous or even deeper lesions that are usually “concealed” by the turbid dermal tissue.

Significance: Reflection Mueller matrix imaging is suitable for characterizing the microstructure of bulk specimens and probing dynamic processes in living animals, there are always demands for speed and accuracy for such applications to avoid possible artifacts and reveal a sample’s intrinsic properties.Aim: To demonstrate a design of collinear reflection Mueller matrix fast imaging microscope based on dual division of focal plane (DoFP) polarimeters (DoFPs-CRMMM) which has high measurement speed and accuracy.Approach: In DoFPs-CRMMM, to improve the measurement speed, we applied the dual DoFP polarimeters design on the collinear reflection system for the first time to achieve fast imaging in about 2 s. To improve the measurement accuracy, we improved the double-pass eigenvalue calibration method (dp-ECM) by background light correction, and explored the optimization of the set of reference samples. Results: DoFPs-CRMMM was applied to measure the standard polarization samples and monitor the tissue optical clearing process of an artificial layered bulk tissue. Results show that the system has satisfactory performance which can capture the variation of polarization properties during the dynamic process. Conclusions: We present the establishment and demo application of DoFPs-CRMMM. The measurement speed can be further accelerated for potential applications in monitoring dynamic processes or living biomedical systems.

Skull optical clearing window permits us to perform in vivo cortical imaging without craniotomy, but mainly limits to visible (vis)-near infrared (NIR)-I light imaging. If the skull optical clearing window is available for NIR-II, the imaging depth will be further enhanced. Herein, we developed a vis-NIR-II skull optical clearing agents with deuterium oxide instead of water, which could make the skull transparent in the range of visible to NIR-II. Using a NIR-II excited third harmonic generation microscope, the cortical vasculature of mice could be clearly distinguished even at the depth of 650 μm through the vis-NIR-II skull clearing window. The imaging depth after clearing is close to that without skull, and increases by three times through turbid skull. Furthermore, the new skull optical clearing window promises to realize NIR-II laser-induced targeted injury of cortical single vessel. This work enhances the ability of NIR-II excited nonlinear imaging techniques for accessing to cortical neurovasculature in deep tissue.
