Photo-controlled Hierarchical Assembly and Fusion of Coumarin-containing Polydiacetylene Vesicles

Jingguo Li,Hao Jiang,Wenlong Hu,Hongyan Xia,Gang Zou,Qijin Zhang

Herein, we synthesize a coumarin-substituted diacetylene monomer (CODA) and report the novel photo-controlled reversible assembly and disassembly behavior of the polymerized CODA (PCODA) vesicles. The photo-triggered dimerization and cleavage reactions of the coumarin groups within the surface of the adjacent PCODA vesicles can be utilized as the driving force to induce assembly and disassembly of PCODA vesicles. Moreover, the boundary of PCODA vesicles in the aggregates becomes more obscure when the irradiation time exceeds 30 min. Fusion occurs upon close docking of target membranes, driven by sufficient dimerization of the coumarin groups within the surface of PCODA vesicles.

A DNA tetrahedron-based light-controlled intramolecular CHA for spatiotemporal imaging of miRNA in living cells with high sensitivity and imaging contrast

Jing Peng,Simin Liu,Ziyue Qiu,Luyin Zhang,Ziyan Dong,Qianying Zhou,Ting Huang,Yanfei Zhang,Jingyu Shi,Jin-Xiang Chen,Zong Dai,Jun Chen

miRNA is an effective disease diagnostic biomarker. Developing efficient miRNA in living cells analysis methods, holds significant importance for disease diagnosis and understanding disease progression. Strategies combining DNA nanostructures and enzyme-free amplification have been developed for highly sensitive and rapid imaging of miRNA. However, due to nonspecific amplification of enzyme-free amplification inherent in the probe assembly process and during transfection, these strategies suffer from significant background signals and reduced sensitivity, limiting their further application. To overcome these limitations, we have developed a DNA tetrahedral-based and photocontrolled intramolecular CHA strategy (TCHA-P) to achieve spatiotemporal, highly sensitive, and accurate imaging of miRNA in living cells. In this strategy, the DNA tetrahedron not only serves as a transfection vector but also enhances the local concentration of reactant probes, boosting the reaction kinetics and sensitivity of CHA. Additionally, the light-controlled group (PC-linker) can trigger CHA at selected times and locations, enhancing the spatiotemporal precision of miRNA imaging and ensuring high sensitivity and accuracy by eliminating nonspecific amplification during the assembly and transfection processes. Using miR-10b as a proof-of-concept model, the proposed strategy demonstrated higher signal-to-noise ratio compared to traditional CHA, with a detection limit of 28 pM. Living cell imaging experiments showed that the strategy could effectively differentiate the expression of miR-10b between normal breast cells and breast cancer cells, providing high imaging contrast. The strategy holds promise for broadening the path of nucleic acid amplification-based imaging and is expected to become an effective tool in nucleic acid-based disease diagnosis, treatment, and drug development.

A photo-controlled hyaluronan-based drug delivery nanosystem for cancer therapy

Feifei Sun,Peng Zhang,Yinyin Liu,Chunbo Lu,Yuanhao Qiu,Haibo Mu,Jinyou Duan

In this paper, a novel photo-controlled drug-loaded nanomicelles were self-assembled by the amphiphile of hyaluronan- o -nitrobenzyl-stearyl chain (HA-NB-SC) with doxorubicin (DOX) encapsulated within the hydrophobic core. DOX-loaded HA-NB-SC nanomicelles are ∼139 nm in diameter. CD44-overexpressed HeLa cells can easily take up HA-NB-SC micelles through recognition of HA moiety. DOX-loaded HA-NB-SC nanomicelles could be disassembled upon UV light (365 nm) and consequently, release DOX at desired pathological sites. Furtherly, nitrosobenzaldehyde derivative, photo-induced products of HA-NB-SC and DOX could inhibit the proliferation of HeLa cells together. This strategy may shed some light on delivery of hydrophobic anti-cancer drugs in a controlled manner.

Synergy of light-controlled Pd nanozymes with NO therapy for biofilm elimination and diabetic wound treatment acceleration

T. Huang,Z. Yu,B. Yuan,L. Jiang,Y. Liu,X. Sun,P. Liu,W. Jiang,J. Tang

The presence of bacteria, existing as highly organized biofilm communities, in chronic non-healing wounds has been identified as a significant impediment for wound healing. Nanozymes , with unique antimicrobial mechanisms, as a new alternative for antibiotics, have the potential to synergize with nitric oxide (NO) with enhanced antibacterial and antibiofilm ability. However, the always-on state of nanozymes and the reactivity of NO limit their clinical applications. In this context, an intelligent and multifunctional Pd-MOF@PAzo@SNP nanoplatform was fabricated using UiO-66 as a palladium (Pd) nanozyme-loading vehicle, then a surface modification with photosensitive polyazobenzene (PAzo), and the adsorption of the NO donor sodium nitroprusside (SNP) via a host-guest interaction between β -cyclodextrin-modified hyaluronic acid ( β -CD-HA) and azobenzene. The activity of Pd-nanozyme was easily controlled via ultraviolet (UV) light, and its photosensitivity was regulated by changing the side-chain unit length of PAzo. Furthermore, NO was released in response to the UV irradiation and played a synergistic role with the peroxidase activity of Pd nanozyme, exhibiting excellent antibacterial and antibiofilm activity in the presence of 0.01 mM hydrogen peroxide (H 2 O 2 ). In vivo , Pd-MOF@PAzo@SNP accelerated the healing of a biofilm-infected diabetic wound by dispersing the biofilm, reducing bacterial burden, and promoting angiogenesis and collagen deposition. Overall, the nanoplatform provides a reliable and highly efficient strategy to develop an intelligent nanozyme synergy with NO therapy in chronic wound management.

Refractive index adjustable intraocular lens design to achieve diopter control for improving the treatment of ametropia after cataract surgery

Yueze Hong,Dong Liu,Haoyu Zou,Qingqing Jia,Sihan Tang,Quankui Lin

Intraocular lens (IOL) implantation is currently the most effective clinical treatment for cataracts. Nevertheless, due to the growth of the eye axis in patients with congenital cataracts during the process of growth and development, the progressive incapacity of an IOL with a fixed focus does not meet the demands of practical usage, leading to the occurrence of ametropia. This work describes an innovative class of an IOL bulk material that offers good biosafety and light-controlled refractive index adjustment. Acrylate materials were synthesized for the preparation of IOLs by free radical polymerization of ethylene glycol phenyl ether methacrylate (EGPEMA), hydrophilic monomer 2-(2-ethoxyethoxy) ethyl acrylate (EA), and functional monomer hydroxymethyl coumarin methacrylate (CMA). Under 365/254 nm ultraviolet (UV) irradiation, the coumarin group could adjust the polymer material's refractive index through reversible photoinduced dimerization/depolymerization. Meanwhile, the potential for the IOL use is enabled by its satisfactory biosafety. Such a light-induced diopter adjustable IOL will be more appropriate for implantation during cataract surgery since it will not require the correction needed for ametropia and will offer more accurate and humane treatment. Statement of significance • A coumarin group containing-copolymer was synthesized and fabricated into intraocular lenses (IOL). • A functional IOL is diopter adjustable by light irradiation through triggering the [2 + 2] cycloaddition of the coumarin groups. • Such light-triggered diopter-adjustable copolymer IOLs have a great potential in clinical application.

A novel photocleavable heparin derivative with light controllable anticoagulant activity

Feifei Sun,Yinyin Liu,Dongdong Wang,Zhaojie Wang,Haibo Mu,Feng Wang,Kan Ding,Jinyou Duan

Heparin (HP) has enormous potential for clinical medication applications owing to its anticoagulant activity. However, the strong anticoagulant capacity of HP also leads to some side effects. Herein, a photocaged derivative (HP-DMNB) of HP was synthesized for light control of its anticoagulant activity. The synthesized HP-DMNB was characterized by NMR and FTIR analysis, which confirmed the successful modification of HP with the photocleavable 4,5-dimethoxyl-2-nitrobenzyl (DMNB) groups. After the modification, the molecular weight of HP-DMNB (DS = 0.34%) changed from 61 to 71 kDa. Anticoagulant activity analysis showed that HP-DMNB had a reduced anticoagulant capacity compared with commercial HP, while its anticoagulant activity will regain after releasing the free carboxyl groups of HP under UV light. In addition, HP-DMNB and its UV irradiated products were observed to possess good biocompatibility through the MTT assays and live-dead assays with HaCaT cells, which may have impacts on the clinical medication applications of HP.

Light-controlled versatile manipulation of liquid metal droplets: a gateway to future liquid robots

Hongtai Ren,Hu Jin,Jian Shu,Jie Xie,Erlong Wang,Du-An Ge,Shi-Yang Tang,Xiangpeng Li,Weihua Li,Shiwu Zhang

The controlled actuation of liquid metal (LM) droplets has recently shown great potential in developing smart actuating systems for applications in robotics. However, there is a lack of a simple approach for the precise manipulation of multiple LM droplets in a 2D plane, which hinders the development of complex control over droplets for realizing useful robotic applications. To overcome this challenge, here, a versatile and powerful light-induced manipulation of LM droplets is presented. The key principle is to selectively activate phototransistors in an electrolyte using infrared laser beams to electrically control LM droplets via Marangoni forces. This approach shows the ability of inducing concurrent motion, splitting, and merging of multiple LM droplets simply using light without complex and bulky systems. Parameters affecting the manipulation of LM droplets are thoroughly investigated. Moreover, a vehicle carrier driven by wheels composed of multiple LM droplets for making a light-controlled relay is demonstrated. We believe such a light-induced control method for manipulating LM droplets has the potential for advancing the development of future field-programmable robotics and droplet-based soft collaborative robots.

Light-controlled microbots gathering as a sterilization platform for highly efficient capturing, concentrating and killing targeted bacteria

Laibo Song,Shujie Zhang,Qimeng Wang,Wei Chen,Bo Liu,Yuan-Di Zhao

Owing to the dispersion of photothermal particles and targeted bacteria, the heat generated by the photothermal effect cannot be completely transferred to the target, resulting in a reduction in the efficiency of traditional photothermal sterilization. Herein, to solve the problem, a new photothermal sterilization platform was successfully developed for capturing and gathering bacteria based on the photothermal effect of microbots. The sterilization microbots were fabricated from carbonized sunflower pollen grains (SPG) with polydopamine (PDA) coating. Bacterial capture molecule, vancomycin, is grafted onto the outer surface of the PDA. Under the focused laser, the PDA/SPG microbot can show positive phototactic motion and bubble-induced gathering powered by photothermal-induced Marangoni effect for efficient capture, concentration, and aggregation the targeted bacteria. Moreover, the gathered microbots can generate higher temperature through photothermal effect to efficiently kill bacteria compared with the dispersed ones. In word, such sterilization microbots could provide a new proof-of-concept perspective on photothermal therapy, and be promising for various sterilization applications.

O-nitrobenzyl Caged Molecule Enables Photo-controlled Release of Thiabendazole

Wen Fu,Zhongli Shao,Zhiping Xu,Zhong Li,Xusheng Shao

Graphical NP-TBZ is developed by covalently linking o -nitrobenzyl ( NP ) with thiabendazole ( TBZ ). Compound NP-TBZ can be controlled to release TBZ in dependent to light, which resulted in controllable in-vivo activity against wheat scab. It is promising for spatiotemporal controlled-release of TBZ in factual applications, posing a theoretical guidance in development of controlled-release pesticides. Pesticides are essential in agricultural development. Controlled-release pesticides have attracted great attentions. Base on a principle of spatiotemporal selectivity, we extended the photoremovable protective group (PRPG) into agrochemical agents to achieve controllable release of active ingredients. Herein, we obtained NP-TBZ by covalently linking o -nitrobenzyl ( NP ) with thiabendazole ( TBZ ). Compound NP-TBZ can be controlled to release TBZ in dependent to light. The irradiated and unirradiated NP-TBZ showed significant differences on fungicidal activities both in vitro and in vivo. In addition, the irradiated NP-TBZ displayed similar antifungal activities to the directly-used TBZ , indicating a factual applicability in controllable release of TBZ . Furthermore, we explored the action mode and microcosmic variations by SEM analysis, and demonstrated that the irradiated NP-TBZ retained a same action mode with TBZ against mycelia growth.

Tunable Spreading and Shrinking on Photocontrolled Liquid Substrate

Wenjie Ji,Weibin Li,Yuren Wang,Ding Lan

Droplets of n-hexadecane were observed to shrink under ultraviolet (365 nm) and spread under blue light (475 nm) irradiation on an aqueous solution of photosensitive surfactant AzoTAB. We demonstrate that the change of wettability of n-hexadecane droplet on the solution depends on the change of oil–water interface tension. According to the addition of ethanol into the substrate, the change of relative diameter ΔD/D exceeds 20%, much larger than the system without ethanol. With light-emitting diode (LED) light as a sole power source, without any other triggers, we provide a contactless and isothermal method to realize photocontrolled alternative spreading and shrinking of a droplet on a liquid surface, which provides a basis for a chromocapillary-based optical zoom liquid lens.

Preferential chiral solvation induced supramolecular chirality in optically inactive star Azo polymers: photocontrollability, chiral amplification and topological effects

Lu Yin,Yin Zhao,Shunqin Jiang,Laibing Wang,Zhengbiao Zhang,Jian Zhu,Wei Zhang,Xiulin Zhu

A series of star-shaped azobenzene (Azo)-containing side chain polymers with 3, 4 and 6 arms were synthesized by the atom transfer radical polymerization technique initiated by multifunctional initiators. The intense bisignated circular dichroism (CD) signals in the UV–vis region were observed when these star polymers aggregated in a dichloroethane/limonene mixed solvent, indicating the successful chirality transfer from limonene molecules to these star Azo polymers. The chirality originates from the preferential supramolecular chirality of well-organized optically inactive side-chain Azo units. The supramolecular chirality is closely related to the volume fraction of cosolvents, the enantiopurity (ee) of limonene, molecular weight and arm numbers of star Azo polymers. Interestingly, the maximum values of CD and gCD of the polymer aggregates increase with molecular weight in the case of 3-armed Azo polymers, and increase simultaneously with arm numbers of the polymers having similar repeating units under each optimized experimental condition. Surprisingly, the first chiral amplification behaviours were observed in the 4- and 6-armed Azo polymers, but absent in linear and 3-armed counterparts. The supramolecular chirality can be destroyed by trans–cis photoisomerization and recovered by thermal cis–trans isomerization of Azo units. The current research will provide further insight into the mechanism study and production of effective chiroptical materials in chiral solvation induced supramolecular chirality of the achiral polymers.

Light controllable chitosan micelles with ROS generation and essential oil release for the treatment of bacterial biofilm

Zhaojie Wang,Hu Bai,Chunbo Lu,Chunyan Hou,Yuanhao Qiu,Peng Zhang,Jinyou Duan,Haibo Mu

Bacterial biofilms are widely associated with persistent infections and food contamination. High resistance to conventional antimicrobial agents resulted in an urgent need for novel formulation to eliminate these bacterial communities. Herein we fabricated light controllable chitosan micelles loading with thymol (T-TCP) for elimination of biofilm. Due to the exterior chitosan, T-TCP micelles easily bind to negative biofilm through electrostatic interaction and efficiently deliver the essential oil payloads. Under irradiation, T-TCP micelles generated ROS , which triggered simultaneous thymol release and also resulted in additional ROS-inducing bactericidal effects, both effectively eradicating biofilms of Listeria monocytogenes and Staphylococcus aureus . This formulation provided a platform for other water-insoluble antimicrobials and might be used as a potent and controllable solution to biofilm fighting.
