Endogenous oxygen-evolving bio-catalytic fabrics with fortified photonic disinfection for invasive bacteria-caused refractory cutaneous regeneration

Ziyou Wang,Bingwen Du,Xiangyu Gao,Yiling Huang,Meng Li,Zhaohan Yu,Jiyao Li,Xiuyuan Shi,Yi Deng,Kunneng Liang

Invasive bacterial infection interferes the normal orchestrated course of healing cascades during cutaneous regeneration. Hypoxia in the deep-seated infection microenvironment (IME) considerably precludes photonic disinfection efficacy, especially antibacterial photodynamic therapy (aPDT). Herein we develop IME-unlocked oxygen-evolving bio-catalytic fabrics composed of electrospun poly(ε-caprolactone) scaffolds, MXene/SnS 2 bio-heterojunctions (MX/SnS bio-HJs), and lactate oxidase (LOx) for deep-seated bacteria-infected cutaneous regeneration. LOx can deplete lactic acid produced by bacteria at infected site and yield hydrogen peroxide (H 2 O 2 ). The MX/SnS bio-HJs not only catalyze H 2 O 2 to produce hydroxyl radicals (·OH) through Fenton-like reaction, but also generate oxygen (O 2 ) to fortify aPDT under Near-infrared (NIR) light irradiation, consequently, expeditious disinfection is achieved. Moreover, in vivo results reveal that the bio-catalytic fabrics exert benign cutaneous regeneration ability by slaughtering bacteria, promoting hemostasis and collagen deposition. This work provides a tactic to instill fabrics with an IME-unlocked catalytic sterilization capability through endogenous O 2 evolution for remedying infected wounds.

Copper ferrite heterojunction coatings empower polyetheretherketone implant with multi-modal bactericidal functions and boosted osteogenicity through synergistic photo/Fenton-therapy

Junchuan Zhang,Xiangyu Gao,Daichuan Ma,Shuai He,Bingwen Du,Weizhong Yang,Kenan Xie,Lu Xie,Yi Deng

Drug-free antibacterial therapy, such as photodynamic therapy and chemodynamic therapy, has become a mainstream in the treatment of implant-associated infection (IAI) due to its considerable potential on effective eradication of pathogenic bacteria via yielding overwhelming reactive oxygen species (ROS). However, superfluous ROS after therapeutical process may elicit a detrimental effect on subsequent tissue regeneration. To address this tough issue, we innovatively devise a photo-activated copper ferrite (CuFe 2 O 4 ) heterojunction coating on bioinert polyetheretherketone (PEEK) implant through hydrothermal treatment and π-π-π stacking. The heterojunction coatings not only generate localized hyperthermia and ROS upon 808 nm NIR illumination, but also trigger Fenton(-like) reaction and glutathione (GSH) depletion in bacterial infectious microenvironment (IME), all leading to robust and cyclic antibacterial property through synergistic photothermal/photodynamic/chemodynamic therapy. Besides, the decorated implants are demonstrated to show superior cytocompatibility and osteogenic-promoting capability. The mice subcutaneous implantation further confirms the excellent in vivo photo-disinfection. The advantage of the heterojunction coatings lies on its enhancement in tissue generation and osteogenesis after antibacterial dynamic therapy because the liberation of micronutrient element (Cu, Fe) is significantly augmented exposed to NIR light, showing photo-enhanced osteogenicity. As envisaged, this work provides a new strategy to empower PEEK implants with an integrative feature of bacteriostasis and osteogenicity to combat intractable IAI through synergistic photo/Fenton-therapy.

Nanocatalytic Hydrogel with Rapid Photodisinfection and Robust Adhesion for Fortified Cutaneous Regeneration

Yimeng Su,Xiumei Zhang,Yan Wei,Yu Gu,Huilun Xu,Ziming Liao,Liqin Zhao,Jingjing Du,Yinchun Hu,Xiaojie Lian,Weiyi Chen,Yi Deng,Di Huang

Chronic inflammation caused by invasive bacterial infections severely interferes with the normal healing process of skin regeneration. Hypoxia of the infection microenvironment (IME) seriously affects the antibacterial effect of photodynamic therapy in phototherapy. To address this serious issue, a nanocatalytic hydrogel with an enhanced phototherapy effect consisting of a hydrogel polyvinyl alcohol (PVA) scaffold, MXene/CuS bio-heterojunction, and polydopamine (PDA) for photothermal antibacterial effects and promoting skin regeneration is designed. The MXene/CuS bio-heterojunction has a benign photothermal effect. Singlet oxygen (1O2) and hydroxyl radicals (·OH) were generated under near-infrared light, which made the hydrogel system have good antioxidant and antibacterial properties. The addition of PDA further improves the biocompatibility and endows the nanocatalytic hydrogel with adhesion. Additionally, in vivo assays display that the nanocatalytic hydrogel has good skin regeneration ability, including ability to kill bacteria, and promotes capillary angiogenesis and collagen deposition. This work proposes an approach for nanocatalyzed hydrogels with an activated IME response to treat wound infections by enhancing the phototherapeutic effects.

Pd nanoparticle-modified TiO2-x composites as efficient visible-light active photocatalysts for tetracycline degradation and microbial disinfection

Jingtao Zhang,Mengwan Zhai,Xueying Wang,Shurui Liu,Lijuan Zhang,Xing Liu,Yao Geng,Xiaodong Sun,Bingkun Liu

In the present work, Ti 3+ -self-doped TiO 2 heterostructure photocatalysts were prepared by coupling with different additions of palladium quantum dots by means of deposition-reduction methodology. The electron paramagnetic resonance (EPR) analysis indicated the existence of Ti 3+ in synthesized samples. The transmission electron microscopy (TEM) images presented the modification of TiO 2-x nanoparticles by coupling with nano palladium. Photoluminescence (PL) results demonstrated the lowest charge carrier recombination rate in 5% Pd/TiO 2-x composites. Photocatalytic performances of photocatalysts were evaluated by degrading Tetracycline (TC) and inactivating Escherichia coli ( E. coli ) and Fusarium oxysporum f. sp. Vasinfectum ( Fov ) with the presence of visible light , in which an enhanced photocatalytic degradation and inactivation activity was found in all Pd coupled composites with the highest efficiency in 5% Pd/TiO 2-x . The improved photocatalytic activity of Pd/TiO 2-x was associated to the generation of more hydroxyl radicals (•OH) and superoxide anions radicals (•O 2- ) during photocatalytic process which were demonstrated by the electron spin-resonance spectroscopy (ESR) technique. Moreover, 5% Pd/TiO 2-x composites were found to remain high photocatalytic properties in a circulating photo-disinfection system.
