Purpose This study aims to broaden the application of nano-contrast agents (NCAs) within the realm of the musculoskeletal system. It aims to introduce novel methods, strategies, and insights for the clinical management of ischemic muscle disorders, encompassing diagnosis, monitoring, evaluation, and therapeutic intervention. Methods We developed a composite encapsulation technique employing O-carboxymethyl chitosan (OCMC) and liposome to encapsulate NCA-containing gold nanorods (GNRs) and perfluoropentane (PFP). This nanoscale contrast agent was thoroughly characterized for its basic physicochemical properties and performance. Its capabilities for in vivo and in vitro ultrasound imaging and photothermal imaging were authenticated, alongside a comprehensive biocompatibility assessment to ascertain its effects on microcirculatory perfusion in skeletal muscle using a murine model of hindlimb ischemia, and its potential to augment blood flow and facilitate recovery. Results The engineered GNR@OCMC-liposome/PFP nanostructure exhibited an average size of 203.18±1.49 nm, characterized by size uniformity, regular morphology, and a good biocompatibility profile. In vitro assessments revealed NCA’s potent photothermal response and its transformation into microbubbles (MBs) under near-infrared (NIR) irradiation, thereby enhancing ultrasonographic visibility. Animal studies demonstrated the nanostructure’s efficacy in photothermal imaging at ischemic loci in mouse hindlimbs, where NIR irradiation induced rapid temperature increases and significantly increased blood circulation. Conclusion The dual-modal ultrasound/photothermal NCA, encapsulating GNR and PFP within a composite shell-core architecture, was synthesized successfully. It demonstrated exceptional stability, biocompatibility, and phase transition efficiency. Importantly, it facilitates the encapsulation of PFP, enabling both enhanced ultrasound imaging and photothermal imaging following NIR light exposure. This advancement provides a critical step towards the integrated diagnosis and treatment of ischemic muscle diseases, signifying a pivotal development in nanomedicine for musculoskeletal therapeutics.

The limited tumor tissue penetration of many nanoparticles remains a formidable challenge to their therapeutic efficacy. Although several photonanomedicines have been applied to improve tumor penetration, the first near-infrared window mediated by the low optical tissue penetration depth severely limits their anticancer effectiveness. To achieve deep optical tissue and drug delivery penetration, a near-infrared second window (NIR-II)-excited and pH-responsive ultrasmall drug delivery nanoplatform was fabricated based on BSA-stabilized CuS nanoparticles (BSA@CuS NPs). The BSA@CuS NPs effectively encapsulated doxorubicin (DOX) via strong electrostatic interactions to form multifunctional nanoparticles (BSA@CuS@DOX NPs). The BSA@CuS@DOX NPs had an ultrasmall size, which allowed them to achieve deeper tumor penetration. They also displayed stronger NIR II absorbance-mediated deep optical tissue penetration than that of the NIR I window. Moreover, the multifunctional nanoplatform preferentially accumulated in tumor sites, induced tumor hyperthermia, and generated remarkably high ROS levels in tumor sites upon NIR-II laser (1064 nm) irradiation. More importantly, our strategy achieved excellent synergistic effects of chemotherapy and phototherapy (chemophototherapy) under the guidance of photothermal imaging. The developed nanoparticles also showed good biocompatibility and bioclearance properties. Therefore, our work demonstrated a facile strategy for fabricating a multifunctional nanoplatform that is a promising candidate for deep tumor penetration as an effective antitumor therapy.

Bioactive metal–organic frameworks (bio-MOFs) built from biofunctional metal ions and linkers show a new strategy to construct multifunctional theranostic platforms. Herein, a bio-MOF is synthetized via the self-assembling of Fe 3+ ions and doxorubicin hydrochloride (DOX) molecules. Then, through a stepwise assembly strategy, another bio-MOFs structure consisting of Gd 3+ ions and 1,3,5-benzenetricarboxylic acid (H 3 BTC) is wrapped on the surfaces of Fe-DOX nanoparticles, followed by adsorbing photosensitizer indocyanine green (ICG). Specifically, the Gd-MOF shell structure can not only act as a contrast agent for magnetic resonance imaging (MRI), but also provides protection for Fe-DOX cores, controlling the release of DOX. The photoacoustic and photothermal imaging (PAI and PTI) methods are successfully introduced to the platform by loading ICG, providing potential applications for multimodal biological imaging. The in vitro and in vivo outcomes indicate that the Fe-DOX@Gd-MOF-ICG nanoplatform exhibits outstanding synergistic antitumor performance via MR/PA/PT imaging guided chemotherapy, photothermal and photodynamic combination therapy. The work may encourage further exploration of bio-MOFs based multifunctional theranostic platforms for multimodal imaging guided compound antitumor therapy, which will open an avenue of MOFs toward biological applications.

Integrating nanomaterials with different properties into a single nanoplatform is an ingenious strategy to obtain multifunctional nanocomposites. Herein, we report an experimental strategy to encapsulate a two-dimensional structured vanadium sulfide (VS2) in a europium-based hyaluronic acid coordination polymer to obtain a high photothermal conversion efficiency nanocomposite (VS2@EuCP) while addressing the limitations of VS2 due to hydrophobicity for application under physiological conditions. The photothermal conversion efficiency of VS2@EuCP nanoparticles under 980 nm (0.5 W/cm2) laser irradiation was as high as 49.2%, and the material exhibited excellent resistance to photobleaching. The intrinsic paramagnetism, strong and broad absorbance in the near-infrared region, and downconversion fluorescence properties of VS2@EuCP materials enable VS2@EuCP to perform four-mode imaging functions, namely, conventional downconversion fluorescence imaging, magnetic resonance imaging, photoacoustic imaging, and photothermal imaging. This highly integrated functional VS2 nanocomposite structure has a promising future in the field of contrast-guided applications of photothermal materials.

Reactive oxygen species (ROS)-mediated therapy utilizes the slightly acidic environment of tumor microenvironment and the high amount of H 2 O 2 to disrupt the redox balance of tumor cell while minimizing damage to normal cell. However, this strategy was greatly compromised by the powerful antioxidant capacity of tumors and the limited ROS generation rate of bimetallic metal-organic frameworks (BMOFs). In pursuit of innovative strategies for precise diagnosis and effective treatment, a smart nanotheranostics Zr-TCPP(Co)/ICG/PEG/Apt-M (ZTCIPA) based on Zr/Co porphyrin MOFs was designed. Under 808 nm laser irradiation, ZTCIPA demonstrated exceptional photothermal conversion efficiency alongside robust singlet oxygen ( 1 O 2 ) generation, ensuring potent tumor ablation. Meanwhile, the temperature elevation significantly accelerated the Co 2+ mediated catalytic decomposition of H 2 O 2 to generate •OH. Large amounts of •OH and 1 O 2 formed ROS storm effectively damaged mitochondria and further resulted in tumor cell death. Furthermore, ZTCIPA achieved tumor targeted specificity while enabling real-time fluorescence imaging (FLI)/photothermal imaging (PTI) for accurate tracking. The results in vitro and in vivo demonstrated that ZTCIPA could effectively inhibit tumor growth by photothermal amplified ROS storm under 808 nm laser irradiation. Thus, this novel strategy has enormous potential by solving the bottleneck of ROS-mediated therapies.

Amongst the various cancer treatment methods such as surgery, radiotherapy and chemotherapy, chemodynamic therapy (CDT) is considered as a tumor-specific technique that exploits the use of highly reactive oxygen species (ROS, such as hydroxyl radical •OH generated in Fenton-like catalytic reaction) to trigger cancer cell death. Herein, a strategy for the photothermal-augmented imaging-guided CDT is proposed. The manganese/copper ions doped silicate nanoparticles (MCMSN) are firstly prepared. Polydopamine is covered on the outside of the silicate nanoparticles to give the polydopamine-coated manganese/copper silicate nanoparticles (MCMSN@PDA). After entering the tumor site, the MCMSN@PDA decomposes and releases manganese and copper ions. The Cu(II) ion is reduced by GSH to Cu(I), which further reacts with H 2 O 2 to generate toxic hydroxyl radicals (•OH). And manganese ion is employed as a magnetic resonance imaging reagent to realize the imaging-guided therapy. As a traditional photothermal material, polydopamine can generate heat under 808 nm laser irradiation, which causes tumor cell apoptosis. In addition, the generated heat can also enhance the CDT effect, and be used for photothermal imaging (PTI). The material achieves excellent anti-tumor therapeutic effect both in vitro and in vivo , which provides an effective paradigm for future design of multifunctional anticancer reagents.

In situ monitoring of the photothermal (PT) effect at the cellular level is of great importance in the photothermal (PT) treatment of cancer. Herein, we report a class of DNA-templated gold nanoparticle (GNP)–quantum dot (QD) complexes (GQC) for PT sensing in solution and in cancer cells in vitro. Specifically, the QD photoluminescence (PL) could be activated at elevated temperature with a wide thermo-responsive range between 45 °C and 70 °C, which fits the temperature threshold for effective cancer cell ablation. The general applicability of GQC for intracellular PT sensing is explored using three types of PT agents (gold nanorods (GNRs), gold nanostars (GNSs), and Prussian blue nanoparticles (PBNPs)) with various PT performances. We show that the intracellular QD PL is gradually activated with increasing near-infrared (NIR) irradiation time, providing a good correlation with the surrounding medium temperature for PT sensing. Moreover, we demonstrate that the GQC sensor could be used for specific photothermal labeling and imaging of cancer cells. The QD PL signal is retained in the cells post-treatment, thereby potentially enabling persistent photothermal labeling of cancer cells for post-treatment cell tracking and imaging-guided therapy evaluation.

Purpose Malignant melanoma is one of the most devastating types of cancer with rapid relapse and low survival rate. Novel strategies for melanoma treatment are currently needed to enhance therapeutic efficiency for this disease. In this study, we fabricated a multifunctional drug delivery system that incorporates dacarbazine (DTIC) and indocyanine green (ICG) into manganese-doped mesoporous silica nanoparticles (MSN(Mn)) coupled with magnetic resonance imaging (MRI) and photothermal imaging (PI), for achieving the superior antitumor effect of combined chemo-photothermal therapy. Materials and Methods MSN(Mn) were characterized in terms of size and structural properties, and drug loading and release efficiency MSN(Mn)-ICG/DTIC were analyzed by UV spectra. Photothermal imaging effect and MR imaging effect of MSN(Mn)-ICG/DTIC were detected by thermal imaging system and 3.0 T MRI scanner, respectively. Then, the combined chemo-phototherapy was verified in vitro and in vivo by morphological evaluation, ultrasonic and pathological evaluation. Results The as-synthesized MSN(Mn) were characterized as mesoporous spherical nanoparticles with 125.57±5.96 nm. MSN(Mn)-ICG/DTIC have the function of drug loading-release which loading ratio of ICG and DTIC could reach to 34.25±2.20% and 50.00±3.24%, and 32.68±2.10% of DTIC was released, respectively. Manganese doping content could reach up to 65.09±2.55 wt%, providing excellent imaging capability in vivo which the corresponding relaxation efficiency was 14.33 mM −1 s −1 . And outstanding photothermal heating ability and stability highlighted the potential biomedical applicability of MSN(Mn)-ICG/DTIC to kill cancer cells. Experiments by A375 melanoma cells and tumor-bearing mice demonstrated that the compound MSN(Mn)-ICG/DTIC have excellent biocompatibility and our combined therapy platform delivered a superior antitumor effect compared to standalone treatment in vivo and in vitro. Conclusion Our findings demonstrate that composite MSN(Mn)-ICG/DTIC could serve as a multifunctional platform to achieve a highly effective chemo-photothermal combined therapy for melanoma treatment.

Multimodal imaging-guided synergistic anticancer strategies have attracted increasing attention for efficient diagnosis and therapy of cancer. Herein, a multifunctional nanotheranostic agent FePtMn-Ce6/FA (FPMCF NPs) is constructed by covalently anchoring photosensitizer chlorin e6 (Ce6) and targeting molecule folic acid (FA) on ultrasmall homogeneous ternary FePtMn nanocrystals. Response to tumor microenvironment (TME), FPMCF NPs can release Fe 2+ to catalyze H 2 O 2 into •OH by Fenton reaction and simultaneously catalyze hydrogen peroxide (H 2 O 2 ) into O 2 to overcome the tumor hypoxia barrier. Released O 2 is further catalyzed into 1 O 2 under 660 nm laser irradiation with Ce6. Thus, the FPMCF NPs exhibit superior dual-ROS oxidization capability including ferroptosis chemodynamic oxidization and 1 O 2 -based photodynamic oxidization. Interestingly, FPMCF NPs reveal strong photothermal conversion efficiency exposed to an 808 nm laser, which can assist dual-ROS oxidization to suppress solid tumor remarkably. Additionally, Mn 2+ can be released from FPMCF NPs to enhance longitudinal relaxivity (T 1 -weighted magnetic resonance (MR) imaging) and Fe-synergistic transverse relaxivity (T 2 -weighted MR imaging), which is convenient for diagnosis of solid tumors. Meanwhile, the fluorescent/photothermal (FL/PT) imaging function of FPMCF NPs can also accurately monitor tumor location. Therefore, FPMCF NPs with multimodal MR/FL/PT imaging-guided synergistic chemodynamic/photodynamic/photothermal cancer therapy capability have potential bioapplication in bionanomedicine field.

Solar energy is a promising renewable energy with the potential for the sustainable development of the world. Efficient photo-thermal conversion is essential for harvesting and conversion of solar energy, therefore, the main challenge is the development of efficient and low-cost photothermal conversion materials. Carbon framework can be considered as a candidate but somehow its application potential can be still constrained due to the limited absorption of near-infrared (NIR) light. Herein, we propose a general strategy for preparing two-dimensional (2D) transition metal dichalcogenides nanosheets and three-dimensional (3D) carbon framework composites (2D/3D ReS 2 @C) as a photothermal material, which has an excellent broadband light absorption performance (in the wavelength range from 200 to 2500 nm). A small thermoelectric (TE) module with an area of 4 × 4 cm 2 is integrated with annealed ReS 2 @C as a light absorber for the investigation of photo-thermoelectric conversion. The open-circuit voltage of the assembled device increases clearly under solar illumination and reaches the maximum value of 136.3 mV, which is ∼ five times larger than that without the absorber. In addition, 20 TE modules coated with ReS 2 @C absorber layers are connected in series, which can produce a maximum open-circuit voltage of 2.12 V (∼66.25 V/m 2 ) to light up a red light-emitting diode (LED) under natural sunlight. Moreover, the annealed ReS 2 @C powder demonstrates a rapid and strong photothermal response under NIR light (wavelength >800 nm), which indicates a great application potential in photothermal imaging and photothermal cancer therapy.

Targeted chemo-phototherapy has received widespread attention in cancer treatment for its advantages in reducing the side effects of chemotherapeutics and improving therapeutic effects. However, safe and efficient targeted-delivery of therapeutic agents remains a major obstacle. Herein, we successfully constructed an AS1411-functionalized triangle DNA origami (TOA) to codeliver chemotherapeutic drug (doxorubicin, DOX) and a photosensitizer (indocyanine green, ICG), denoted as TOADI (DOX/ICG-loaded TOA), for targeted synergistic chemo-phototherapy. In vitro studies show that AS1411 as an aptamer of nucleolin efficiently enhances the nanocarrier’s endocytosis more than 3 times by tumor cells highly expressing nucleolin. Subsequently, TOADI controllably releases the DOX into the nucleus through the photothermal effect of ICG triggered by near-infrared (NIR) laser irradiation, and the acidic environment of lysosomes/endosomes facilitates the release. The downregulated Bcl-2 and upregulated Bax, Cyt c, and cleaved caspase-3 indicate that the synergistic chemo-phototherapeutic effect of TOADI induces the apoptosis of 4T1 cells, causing ~ 80% cell death. In 4T1 tumor-bearing mice, TOADI exhibits 2.5-fold targeted accumulation in tumor region than TODI without AS1411, and 4-fold higher than free ICG, demonstrating its excellent tumor targeting ability in vivo. With the synergetic treatment of DOX and ICG, TOADI shows a significant therapeutic effect of ~ 90% inhibition of tumor growth with negligible systemic toxicity. In addition, TOADI presents outstanding superiority in fluorescence and photothermal imaging. Taken together, this multifunctional DNA origami-based nanosystem with the advantages of specific tumor targeting and controllable drug release provides a new strategy for enhanced cancer therapy.

This study reports a family of photothermal materials, metal ion/tannic acid assemblies (MITAs). MITAs from FeIII, VIII, and RuIII afford excellent photothermal efficiency (η ≈ 40%). Sharply differing from the currently existing photothermal agents, MITAs are highlighted by merits including green synthesis, facile incorporation of diagnostic metal ions, and particularly topology-independent adhesion. Owing to the adhesion nature of MITAs, various kinds of MITA-based nanoengineerings are readily available via the self-adhesion of MITAs onto diverse templates, enabling MITAs well suited as a photothermal platform for versatile combination with other therapy approaches and imaging techniques. As a proof of concept, polymeric/inorganic nanoparticle/nanovesicle-supported FeIII-tannic acid (FeIIITA) is fabricated. The photothermal effect is shown to be unaffected by the template origin and type and FeIIITA thickness on the templates. We validate the potency of nanovesicle-supported FeIIITA (PNV@FeIIITA) for tumor-specific photoactivated utilizations, including NIR photothermal therapy with complete tumor elimination, photothermal imaging (PTI), and photoacoustic imaging (PAI) in addition to T1-MRI imaging. PNV@FeIIITA can be simultaneously equipped with functionalities, including T2-MRI imaging by additionally doping MnII and NIR fluorescence imaging by encapsulating a hydrophilic NIR fluoroprobe. MITA demonstrates unparalleled superiority as a photothermal platform in engineering multimodal theranostics for advanced applications.
