Chronic hypoxia affects stem cell function during tissue repair. Thus far, the hypoxia-associated impact on periosteal stem cells (PSCs), the main contributor to bone repair, remains unknown, and a tailored oxygen modulation strategy for optimizing PSC function is lacking. Here, PSCs exhibit time-dependent proliferation and survival upon hypoxic exposure and a critical 48-h time-point is identified at which hypoxia transitions from beneficial to detrimental. Then, a photothermal-sensitive coaxial fiber-reinforced membrane containing oxygen and pravastatin is constructed to function as an intelligent oxygen supply system. Leveraging near-infrared light as an ON/OFF switch, the system noninvasively scales up oxygen release beginning 48 h post-implantation, counteracting prolonged hypoxia and mitigating its adverse effects on PSCs. The sustained release of pravastatin from the membrane accelerates early neovascularization both directly through its pro-angiogenic effect and indirectly by stimulating vascular endothelial growth factor secretion from PSCs, ensuring a continuous oxygen supply after exogenous oxygen exhaustion. Notably, pravastatin steers PSCs toward robust osteogenic differentiation and provides multifunctional bioactive cues for advanced bone regeneration in vivo. This time-scheduled approach to modulate oxygen supply noninvasively could be applicable beyond bone regeneration for hypoxia-related diseases and multi-tissue repair. Graphical

Elaborately designed biocompatible nanoplatforms simultaneously having diverse therapeutic and imaging functions are highly desired for biomedical applications. Herein, a Bi2Se3 nanoagent with a special morphology as a nanoscale spherical sponge (NSS) has been fabricated and investigated in vitro and in vivo. The highly porous NSS exhibits strong, steady, and broad-band absorbance in the near-infrared range as well as high efficiency and stability of photothermal conversion, resulting in high antitumor efficacy for photothermal therapy (PTT). Together with a high X-ray attenuation coefficient (218% that of the clinically used iopromide), the NSS shows excellent performance on triple-modal high-contrast imaging, including X-ray-computed tomography, multispectral optoacoustic tomography, and infrared thermal imaging. Furthermore, the high surface area and porous structure impart the NSS a competent drug loading capability as high as 600% of that on Bi2Se3 nanoplates, showing a bimodal pH/photothermal sensitive drug release and pronounced synergetic effects of thermo-chemotherapy with a tumor inhibition ratio even higher than that of PTT alone (∼94.4% vs ∼66.0%). Meanwhile, the NSS is highly biocompatible with rather low in vitro/in vivo toxicity and high stability, at variance with easily oxidized Bi2Se3 nanoagents reported previously. Such biocompatible single-component theranostic nanoagents produced by a facile synthesis and highly integrated multimodal imaging and multiple therapeutic functions may have substantial potentials for clinical antitumor applications. This highly porous nanostructure with a large fraction of void space may allow versatile use of the NSS, for example, in catalysis, gas sensing, and energy storage, in addition to accommodating drugs and other biomolecules.

A novel multifunctional graphene oxide nano drug carrier (GO@LM-SP-FA) was constructed by connecting a linear maltodextrin polymer (LM-SP) and folic acid (FA) to the surface of graphene oxide (GO) nanoparticles. Doxorubicin hydrochloride (DOX) as a model drug was loaded onto the GO@LM-SP-FA through π–π stacking, hydrogen bonding and hydrophobic interaction. The GO@LM-SP-FA nanomaterial possessed excellent photothermal conversion efficiency, double-targeted drug delivery and near-infrared light (NIR)/pH triggered drug release. More importantly, GO@LM-SP-FA combined the cancer cell targeting of FA, showing excellent cancer-killing efficiency in HeLa (78%) and MCF-7 (61%). Therefore, the novel nano-drug carrier synthesized in this paper has potential practical application value in the treatment of cancer, especially in cervical cancer which has high FA expression.

The tumor microenvironment, characterized by hypoxia, supports the efficacy of anaerobic bacteria like attenuated S. typhimurium in cancer therapies. These bacteria target and penetrate deep tumor regions, significantly reducing tumor size but often lead to tumor regrowth due to limited long-term efficacy. To enhance the therapeutic impact, a novel biohybrid system, S@UIL, has been developed by coating S. typhimurium with a zirconium-based nanoscale metal–organic framework (UiO-66-NH 2 ) loaded with indocyanine green (ICG) and luteolin (LUT). This system maintains the bacteria’s tumor-targeting ability while increasing the penetration and therapeutic effectiveness through excessive autophagy and mild hyperthermia. In a subcutaneous colon cancer model, the integration of LUT and ICG promotes autophagic cell death and photothermal sensitization, leading to the release of damage-associated molecular patterns (DAMPs). These DAMPs activate immune responses through dendritic cells and T -cells, enhancing immunogenic cell death (ICD) and potentially reducing immune evasion by tumors. This single-administration approach also integrates multimodal imaging capabilities, providing a promising strategy for improved tumor ICD induction and cancer progression inhibition.
