Target-triggered aggregation of gold nanoparticles for photothermal quantitative detection of adenosine using a thermometer as readout

Yingzhou Tao,Fang Luo,Longhua Guo,Bin Qiu,Zhenyu Lin

Colorimetric platform using the aggregation of gold nanoparticles (AuNPs) is a pretty simple method for biosensing, but advanced instruments such as specterophotometer is still needed to achieve accurately quantitative readout. Aggregated AuNPs exhibit excellent photothermal properties under near-infrared laser irradiation, which is significantly different from non-aggregated AuNPs. Herein, given the different photothermal effect, we translated the AuNPs-based colorimetric assay into a photothermal assay for the quantitative detection of adenosine using a thermometer as readout. Short single-stranded DNA (ssDNA, adenosine aptamer) was adsorbed on the surface of AuNPs and hence prevented the aggregation of AuNPs under high ionic concentration. The presence of adenosine caused the structural change of ssDNA and the AuNPs became aggregated. The enhanced temperature under NIR-laser irradiation has a linear response to the concentration of adenosine in the range of 2.0–50.0 μM. The detection limit was 1.7 μM. This proposed method is portable, easy and applicable to the quantitative assay of other targets by simply replacing of the sequence of ssDNA.

Development of a fast and ultrasensitive black phosphorus-based colorimetric/photothermal dual-readout immunochromatography for determination of norfloxacin in tap water and river water

Shuyue Ren,Qiaofeng Li,Jingyi Wang,Bingyan Fan,Jialei Bai,Yuan Peng,Shuang Li,Dianpeng Han,Jin Wu,Jiang Wang,Kang Qin,Zunquan Zhao,Baoan Ning,Tie Han,Huanying Zhou,Zhixian Gao

A rapid and ultrasensitive method for colourimetric/photothermal dual-readout detection was developed using an 808 nm NIR laser and a thermal imaging app on mobile phone. Norfloxacin was used as a model contaminant to demonstrate this universal rapid detection method. It is innovatively, to use the advanced two-dimensional material black phosphorus as a colourimetric/photothermal reagent for the first time. The samples were added to the strip, and the analytes were selectively captured on the conjugate pad by monoclonal antibody-modified magnetic/upconversion nanocomposites . The samples flowed through the strips by capillary action until reaching the control line, where immune complex formation occurred due to the presence of secondary antibody. The added black phosphorus could be captured by the the antigens which were directly exposed to the test line and a brown band could be observed by naked eye. Upon illumination by NIR light for 1 min, the real-time temperature is obtained for quantitative analysis through the thermal imaging performed by mobile phone camera. This method can achieve the detection of norfloxacin in water samples within 20 min, and the detection limits of colorimetric and photothermal readout can reach 45 pg mL −1 . Compared with conventional strips, this method provided an increased sensitivity by about two orders of magnitude, with a integrated portable laser and a mobile phone. It is a valuable method for rapid detection and can be applied to other environmental contaminants as well.

Colorimetric and photothermal dual-modal analysis of a cancer-related gene based on G-quadruplex-hemin cascade signal amplification

Hongling Tao,Yanyue Chen,Jiawen Liu,Jiali Liu,Yunhui Yang,Rong Hu

p53 gene is one of the most important anticancer genes and has been recognized as a typical biomarker for the early diagnosis of cancers. In this paper, a portable point-of-care testing (POCT) platform was developed for the colorimetric and photothermal dual-modal analysis of the p53 gene. Long concatemer G4 structures with high catalytic activity were formed by a target-driven hybridization chain reaction, and their catalytic properties can further be enhanced with the help of iodide ions. This dual-modal method realized the sensitive multicolor visual point-of-care detection of the p53 gene, where color variations from colourless to blue were easily distinguished by the naked eye. The temperature output was achieved by effective photothermal conversion and regulation guided by the accumulated concatemer G4 complexes in the TMB–H2O2 system, avoiding the need for complex instruments. The proposed dual-modal assay for p53 gene detection is cost-effective, simple, highly sensitive (3.34 pM), stable, and remarkably selective. It was also applied to determine the concentration of the p53 gene in spiked serum samples.

Stable and Photothermally Efficient Antibody-Covered Cu3(PO4)2@Polydopamine Nanocomposites for Sensitive and Cost-Effective Immunoassays

Xiaofeng Tan,Xiaoying Wang,Lianhua Zhang,Luyao Liu,Gengxiu Zheng,He Li,Feimeng Zhou

Polydopamine (PDA)-coated or encapsulating Cu3(PO4)2 (Cu3(PO4)2@PDA) nanosheets were synthesized, allowing the C-reaction protein (CRP) antibody to be attached electrostatically for immunosensing of CRP with simple photothermal detection. The antibody-covered Cu3(PO4)2@PDA nanosheets replace the antibody-conjugated enzyme in the enzyme-linked immunosorbant assays. Owing to the high surface area of the 2-D-structured Cu3(PO4)2@PDA nanosheets and the coabsorption of light in the near-IR spectrum by Cu3(PO4)2 and PDA, a small amount of Cu3(PO4)2@PDA confined in the wells of a titer plate generates an easily detectable temperature change after irradiation at 808 nm. The temperature changes, measured by an inexpensive pen-type thermometer, increased linearly with the analyte concentration from 0.42 to 16 pM. We found that the linear relationship can be fitted by the isotherm derived from responses collected from heterogeneous sensors covered with different ligand or antibody densities. The low detection limit (0.11 pM) is largely due to the attachment of a great number of antibodies onto the flat nanosheets. The antibody-covered Cu3(PO4)2@PDA nanosheets are stable and can be used under conditions that are generally unfavorable to enzymatic activities. The excellent agreement between our results and immunoturbidimetric assays of CRP in serum samples from patients and healthy donors demonstrates its utility for disease diagnosis in clinical settings. This cost-effective, biocompatible, and convenient photothermal immunosensor affords a range of possibilities for detecting diverse protein biomarkers.

Cyclodextrin reduced Fe(III) nanozyme-based colorimetric and photothermal dual-mode assay for early monitoring meat freshness by sensitive detection of hypoxanthine

Sujuan Sun,Junqing Yang,Gang-Gang Yang,Xiaoxiao Wang,Xinya Han

A simple and sensitive method for monitoring the freshness of meat products is crucial for safeguarding food hygiene and human health. Here, we synthesized an environmentally friendly β -CD@Fe nanozyme through a one-pot solvothermal method, enabling colorimetric and photothermal dual-mode detection of hypoxanthine (Hx). In the xanthine oxidase (XOD)/nanozyme enzymatic cascade system, Hx ultimately led to the oxidation of colorless TMB to blue oxTMB, accompanied by enhanced absorbance at 652 nm and an increased temperature under 808 nm laser irradiation. The proposed dual-mode assay for Hx exhibited a good linear relationship in the range of 1–40 μM, achieving low detection limits of 0.29 μM (colorimetric) and 1.0 μM (photothermal), respectively. The β -CD@Fe nanozyme, characterized by its environmental friendliness, mild synthesis conditions, and high peroxidase-like activity, was successfully applied for the sensitive analysis of Hx in actual meat samples. This advancement offers promising potential for applications in food safety supervision.

Colorimetric and photothermal dual-mode immunoassay of aflatoxin B1 based on peroxidase-like activity of Pt supported on nitrogen-doped carbon

Shuoying Huang,Wenqiang Lai,Bingqian Liu,Mingdi Xu,Junyang Zhuang,Dianping Tang,Youxiu Lin

In this work, a split-type dual-mode (colorimetric/photothermal) immunoassay method was designed for point-of-care testing (POCT) detection of mycotoxins (aflatoxin B 1 , AFB 1 as the model analyte) in foodstuffs based on Pt supported on nitrogen-doped carbon amorphous (Pt-CN). The as-synthesized Pt-CN exhibits excellent peroxidase-mimicking activity, which can catalyze the oxidization of 3,3′,5,5′-tetramethylbenzidine (TMB) into TMB ox with sensitive colorimetric readout in the presence of hydrogen peroxide (H 2 O 2 ). Moreover, the TMB ox also serves as a near-infrared (NIR) photothermal agent to convert the colorimetric readout into heat under the irradiation of an 808 nm laser. A competitive-type immunoreaction is carried out between AFB 1 and glucose oxidase (GOx)-labeled AFB 1 -bovine serum albumin (AFB 1 -BSA-GOx) conjugates. With the formation of immune complexes, the entrained GOx catalyzes the hydrolysis of glucose to generate H 2 O 2, which further involves the Pt-CN catalyzed production of TMB ox to increase colorimetric/photothermal readouts. Depending on the degree of TMB oxidation, the dual-mode immunoassay provides a linear range of 1.0 pg/mL to 10 ng/mL, with a limit of detection (LOD) of 0.22 pg/mL for the colorimetric assay and 0.76 pg/mL for the photothermal assay. Moreover, the developed method is successfully used to detect AFB 1 in peanuts with acceptable accuracy compared with commercially enzyme-linked immunosorbent assay (ELISA) kits. Significantly, the photothermal readout in this method is recorded on a mobile phone device without any expensive instruments, providing an affordable and convenient tool for food safety testing.

Detection of Bitter Limonin in Citrus Samples Using Plasmonic Gold Nanobipyramid-Based Dual-Mode Lateral Flow Immunoassay

Tian Zhang,Chenxi Guo,Lu Yang,Lei Liu,Di Sun,Xinqi Long,Qi Wu,Qiyang Zhao,Yaohai Zhang,Jing Li,Yongliang Cui,Yue He

The detection of limonin, a bitter compound in citrus samples, is crucial for maintaining product quality, safety, and marketability, and ultimately benefits both producers and consumers. In this assay to effectively modify the monoclonal antibody on gold nanobipyramids (AuNBP), we replaced the original hexadecyltrimethylammonium bromide coating (CTAB) with SH-PEG2000-NHS. Then AuNBP were modified with a limonin-specific monoclonal antibody (mAb) to create the AuNBP@mAb immunoprobe. We present a novel plasmonic AuNBP-based lateral flow immunoassay (LFIA) for colorimetric and photothermal assay of limonin. The limit of detection for the colorimetric mode of AuNBP is 4.8 ng/mL. The limit of detection for the photothermal mode of AuNBP is 3.8 ng/mL .Validation experiments using citrus samples demonstrate the robustness and reliability of the LFIA in real-world applications. Comparative analysis with ultra-high performance liquid chromatography tandem mass spectrometry confirms the accuracy of our LFIA.

Bimetallic organic framework nanozyme-driven aptasensor for colorimetric and photothermal dual-modal detection of kanamycin

Gengli Huang,Shuo Qi,Xiaodong Lu,Zehua Zhang,Kaiqing Wu,Zhouping Wang

Kanamycin (KAN) residues in environment and food sources pose a considerable threat to human health, creating a strong demand for developing facile yet highly sensitive analytical methods. Herein, a highly sensitive aptasensor based on a bimetallic organic framework nanozyme was constructed for dual-modal KAN detection, employing both colorimetric and photothermal signals regulated by aptamer-controlled nanozyme activity. The synthesized Zr-Fe-MOF, exhibiting peroxidase-like activity, catalyzed the oxidization of colorless 3, 3′, 5, 5′-tetramethylbenzidine (TMB), resulting in the formation of a blue-colored oxidized TMB, which could generate detectable colorimetric and photothermal signals. Once bound to aptamer, its catalytic activity was significantly enhanced due to the formation of nanozyme-aptamer conjugates with increased negative charge. However, this activity was restored through the highly sensitive recognition of the target by the aptamer, leading to a decrease in colorimetric and photothermal signals. Thus, this aptamer-modulated Zr-Fe-MOF enable effective dual-modal detection of KAN, achieving detection limits of 0.17 μM and 0.47 μM for KAN in colorimetric and photothermal modes, respectively. Recoveries of 98.34%–108.8% and 91.67%–106.80% were respectively obtained by colorimetric and photothermal methods. The proposed dual-modal aptasensor demonstrates exceptional performance, paving the way for further exploiting antibiotic residues sensing platform.

Dual-signal based immunoassay for colorimetric and photothermal detection of furazolidone

Lihong Su,Yaqian Chen,Lulu Wang,Han Zhang,Jing Sun,Jianlong Wang,Daohong Zhang

As a point-of-care testing method, lateral flow immunoassays are attracting extensive attentions for antibiotic residue detections; however, immunoassay techniques for qualitative and quantitative detections are limited by the poor sensitivity and requirement for cumbersome quantitative devices. To overcome these limitations, MnO 2 -Au, with an excellent carrying capacity of Au nanoparticles and high photothermal signals, was introduced and a dual-signal immunoassay using colorimetric/photothermal MnO 2 -Au signal probe was proposed for furazolidone (FZD) detection. The dual-signal immunoassay not only enables visual detection relying on color change, but also enables quantitative detection via converting conventional assay signals into heat signals using a thermal infrared imager recording. Based on the excellent light-to-heat conversion activity of the bifunctional MnO 2 -Au, the FZD metabolite of 3-amino-2-oxazolidinone (AOZ) can be qualitatively detected in virtue of colorimetric signals with a visual limit of 1 ng mL −1 and quantitatively detected by photothermal signals with a detection limit of 0.43 ng mL −1 . The dual-signal immunoassay biosensor was well applied in food samples with acceptable recoveries of 80.6–106 %. The immunoassay that integrates colorimetric and photothermal analysis will hold a broader application area in real-time monitoring.

Sensitive detection of butyrylcholinesterase activity based on a separation-free photothermal assay

Zerui Ma,Lili Cao,Yifei Xing,Anrui Feng,Peng Zhang,Wei Li,Hailiang Nie,Yu-e Shi,Zhenguang Wang

Butyrylcholinesterase (BChE), a regulator for the cholinesterase activity in serum, is an important index for clinical diagnosis of organophosphorus poisoning and liver related disease. However, the assays for BChE activity are highly dependent on expensive and bulk equipment and professional operators, which limits its straightforward and portable detection. In this work, we proposed a straightforward and sensitive photothermal assay for the detection of BChE activity, avoiding the use of expensive instrument, professional operators, and separation process. Polydopamine (PDA) nanoparticles can be produced through the oxidation of dopamine by MnO 2 nanosheets, which show a rather strong photothermal features. Hydrolyzation of substrate into thiocholine and butyrate happens after introducing BChE. The generated thiocholine and butyrate inhibit the formation of PDA nanoparticle through reducing MnO 2 into Mn 2+ and creating an acidic environment, respectively. Those effects lead to the decreasing of photothermal efficiency of the reaction system, resulting in the decreasing on temperature change. Quantitative detection of BChE activity is achieved by recording the temperature change of system, using a common device, with a limit of detection of 40.65 U/L calculated. The presented results provide a clue for developing straightforward and portable photothermal assay for BChE, which shows great potential for their clinical applications.

Rapid photothermal detection of foodborne pathogens based on the aggregation of MPBA-AuNPs induced by MPBA using a thermometer as a readout

Laibao Zheng,Wenjia Dong,Chaochuan Zheng,Yunqiu Shen,Ruolan Zhou,Zhenxing Wei,Zhixuan Chen,Yongliang Lou

Rapid and portable detection of foodborne pathogens is of great significance for food safety and public health. The colorimetric methods based on naked-eye have been demonstrated to be a suitable qualitative method for point-of-care testing (POCT). However, analytical instruments like a microplate reader must be needed for the quantitative assay. To overcome its limitation, we herein report a novel photothermal method for foodborne pathogens based on the photothermal effect of aggregated mercaptophenylboronic acid-functionalized AuNPs (MPBA-AuNPs) induced by MPBA to translate the colorimetric detection into a simple temperature measurement using thermometers as the readout. The aggregated AuNPs show higher photothermal conversion efficiency than well-separated AuNPs under 660 nm laser irradiation. In the presence of bacteria, MPBA-AuNPs will attach to the surface of bacteria and keep separated from aggregation induced by excess MPBA, resulting in a lower temperature increase under 660 nm laser irradiation. Using E. coli O157:H7 as a model target, a good linear relationship is observed between temperature increase and bacteria concentration from 1.00 × 10 5 -1.00 × 10 9 cfu mL −1 (R 2 = 0.9877) with a detection limit of 1.97 × 10 4 cfu mL −1 , which is three orders of magnitude lower than of the MPBA-AuNPs-based colorimetric assays . The proposed photothermal method provided a universal platform for rapid and portable detection of broad-spectrum bacteria strains in real samples.

In situ generation of polydopamine as photothermal reporter for on-site testing of azodicarbonamide in flour using an integrated 3D-printed device

Guo Lan,Zhao Dong-Mei,Hu Bao-Hua,Wu Shuo,Chen Shuai,Yu Yong-Liang

The detection of the illegal additive azodicarbonamide (ADA) in flour and related products is crucially important due to the harmful effects of excessive consumption on human health. However, the requirement for expensive instruments and trained operators poses a major challenge for on-site testing of ADA, particularly in field enforcement and consumer self-monitoring. Herein, an economical and portable 3D-printed photothermal (PT) device constructed by a 3D-printed sample bracket, a separable 96-well plate, an 808 nm laser, and a thermometer was rationally designed for quantitative on-site testing of ADA in flour. Since the target ADA could convert reducing glutathione (GSH) into non-reducing glutathione disulfide (GSSG), manganese dioxide nanosheets (MnO 2 NSs) in this sensing system remain intact to catalyze the conversion of dopamine (DA) into polydopamine nanoparticles (PDA NPs). During the detection process, the PT signal changes induced in the sample driven by the 808 nm laser could be easily recorded with a digital thermometer. The proposed PT method demonstrates an excellent linear response for ADA in the range of 30–1000 μM, with a detection limit as low as 10 μM, which was significantly lower than the maximum allowable ADA level in flour extracts (388 μM). This portable and cost-effective PT sensing system shows great potential for quantitative on-site testing, enabling immediate food safety identification in resource-limited regions. Graphical
