Background Silver and photothermal therapy (PTT) have been widely used for eradicating the drug-resistant bacteria. However, the risks of excess of silver for humans and the low efficiency of PTT still limit their in vivo therapeutic application. Integration of two distinctive bactericides into one entity is a promising platform to improve the efficiency of antimicrobial agents. Results In this study, a chemo-photothermal therapeutic platform based on polydopamine (PDA)-coated gold nanorods (GNRs) was developed. The PDA coating acquired high Ag+ ions loading efficiency and Cy5-SE fluorescent agent labeled glycol chitosan (GCS) conjugation (Ag+-GCS-PDA@GNRs). This platform became positively charged in the low pH environment of the abscess, allowing their accumulation in local infection site as revealed by thermal/florescence imaging. The loaded Ag+ ions was released in a pH-sensitive manner, resulting in selective Ag+ ions delivery to the abscess environment (pH ~ 6.3). More importantly, the ultralow dose of Ag+ ions could effectively damage the bacterial membrane, causing the permeability increase and the heat resistance reduction of the cell membrane, leading to the large improvement on bactericidal efficiency of PTT. On the other hand, the hyperthermia could trigger more Ag+ ions release, resulting in further improvement on bactericidal efficiency of chemotherapy. Combinational chemo-hyperthermia therapy of Ag+-GCS-PDA@GNRs could thoroughly ablate abscess and accelerate wound healing via a synergistic antibacterial effect. Conclusion sOur studies demonstrate that Ag+-GCS-PDA@GNRs is a robust and practical platform for use in chemo-thermal focal infection therapy with outstanding synergistic bacteria ablating.

Psoriasis is a chronic inflammatory dermatosis with excessive activation of toll-like receptors (TLRs). Small interfering RNA (siRNA) has been considered a promising therapeutic approach by significantly suppressing mRNA expression. However, the low skin barrier penetration and the poor stability of siRNA pose a key challenge. Herein, we developed a novel and near-infrared (NIR) activated nanosystem (termed as GD-siMYD88@SPN/DA) that utilizes the gold nanorod (GNR) with highly photothermal conversion efficiency to realize localized thermal effects and photothermal-controlled release of siMYD88. Meanwhile, the shell grafted desmosome core-glycoprotein3 (Dsg3) antibody endowed it with the ability to target keratinocytes. The endocytosis of GD-siMYD88@SPN/DA by HaCaTs was significantly higher than that of the non-targeted nanosystem (GD-siMYD88@SPN), and the GD-siMYD88@SPN/DA exhibited enhanced stability in the skin. The double strand formed by the GNR-linked DNA fragment and siMYD88 serves as controlled element in response to photothermal stimulation, achieving controlled release of siMYD88. Knockdown of the MYD88 gene could effectively inhibit the TLR/MYD88/NF-κB signaling pathway, thereby reducing the production of psoriasis-related inflammatory factors. Notably, the synergistic therapeutic effects of mild-photothermal therapy combined with GD-siMYD88@SPN/DA were observed both in vitro and in vivo . Overall, this spatiotemporally controllable approach provides a versatile strategy for controllable gene therapy and mild-photothermal therapy for psoriasis.

Two-dimensional MoS 2 (2D-MoS 2 ) and its nanocomposites have been extensively studied in cancer therapy research since their prominent surface area and NIR absorption ability. In this work, Eu 3+ -incorporated MoS 2 (MoS 2 :Eu 3+ ) nanoflowers with enhanced photothermal conversion efficiency (PCE) were successfully developed by a one-step bottom-up hydrothermal method. Under 808 nm near-infrared (NIR) laser irradiation, the photothermal agents MoS 2 :5%Eu 3+ nanoflowers have a PCE at ~ 49.05%, which is 1.75 times higher than the pristine MoS 2 . MoS 2 :5%Eu 3+ as efficient photosensitizers also promote generation of cytotoxic reactive oxygen species (ROS) under 808 nm laser irradiation, thus achieving photodynamic therapy (PDT) for cancers at the same time. Enhanced in vitro breast cancer therapy through the combination of photothermal therapy (PTT) and PDT is clearly proved by using the MoS 2 :Eu 3+ nanosheet as a multifunctional nanoprobe. The unique MoS 2 :Eu 3+ works as a promising nanoprobe for PDT/PTT cancer therapies, and also provides a novel strategy for PCE improvement of the 2D-MoS 2 .

Breast cancer is a malignant tumor that poses a significant threat to women's health and single therapy fails to play a good oncological therapeutic effect. Synergistic treatment with multiple strategies may make up for the deficiencies and has gained widespread attention. In this study, sulfhydryl-modified hyaluronic acid (HA-SH) was covalently crosslinked with polydopamine (PDA) via a Michael addition reaction to develop an injectable hydrogel, in which PDA can be used not only as a matrix but also as a photothermal agent. After HSA and paclitaxel were spontaneously organized into nanoparticles via hydrophobic interaction, hyaluronic acid with low molecular weight was covalently linked to HSA, thus conferring effectively delivery. This photothermal injectable hydrogel incorporates PTX@HSA-HA nanoparticles, thereby initiating a thermochemotherapeutic response to target malignancy. Our results demonstrated that this injectable hydrogel possesses consistent drug delivery capability in a murine breast cancer model, collaborating with photothermal therapy to effectively suppress tumor growth, represented by low expression of Ki-67 and increasing apoptosis. Photothermal therapy (PTT) can effectively stimulate immune response by increasing IL-6 and TNF-α. Notably, the treatment did not elicit any indications of toxicity. This injectable hydrogel holds significant promise as a multifaceted therapeutic agent that integrates photothermal and chemotherapeutic modalities.

In cancer treatment, although prodrugs have been developed to overcome obstacles such as undesirable pharmacokinetic performance and serious off-target toxicity of most chemotherapeutics, the combination of prodrugs with nanotechnology could be used to overcome the shortcomings of conventional prodrug strategies and facilitate more efficient drug delivery and accumulation of anticancer prodrugs at specific sites/tissues. Phototherapy is a method that uses near-infrared (NIR) light to achieve local and non-invasive tumor therapy. The combination of chemotherapy and phototherapy has shown many therapeutic advantages, including synergistic therapeutic effects and the reduction of toxic and side effects of drugs through dose reduction. In this work, a hydrazone linkage was reported to conjugate Ti 3 C 2 T x MXene nanosheets with doxorubicin hydrochloride (DOX) and methoxy poly(ethylene glycol) benzaldehyde (mPEG-CHO) through a chemical surface modification to form a two-dimensional (2D) nano-prodrug Ti 3 C 2 T x -DOX/PEG (TDP) for synergistic chemo-photothermal therapy. First, hydrazide functionalized Ti 3 C 2 T x was obtained. Then, the carbonyl group of DOX and the aldehyde group of mPEG-CHO can form a pH-responsive hydrazone bond with hydrazide-functionalized Ti 3 C 2 T x . The high surface area of Ti 3 C 2 T x could overcome the reduced drug loading caused by conjugation. Aqueous stability and biocompatibility were enhanced by the introduction of PEG, making the material capable of prolonged circulation in the body's bloodstream. By achieving excellent antitumor results with this nano-prodrug, we have added another brick to the edifice of Ti 3 C 2 T x MXene as an anticancer drug carrier, and given new vitality to the study of anticancer prodrugs combined with nanotechnology.

Microneedles (MNs) are an attractive strategy for the efficient, painless transdermal delivery of drugs with low systemic toxicity. In this work, a novel near-infrared (NIR) light-triggered separable microneedle system was fabricated to supply controlled chemo-thermal therapy to superficial tumors by co-loading a self-developed NIR-II fluorescence fluorophore Flav7 and the antitumor drug doxorubicin (DOX) in phase-change polycaprolactone (PCL) arrowheads on a soluble base. For the MNs we proposed, Flav7 offers MNs with NIR light-triggered heating generation and drug release properties as well as a NIR-II imaging ability for guiding combined chemo-thermal therapy, and the soluble supporting base ensures that the needle arrows are embedded in the skin after insertion and endows the MNs with a separation ability to reduce the contact time of the MNs with the skin. In vitro studies showed that Flav7 + DOX-loaded MNs exhibited adequate insertion abilities, stable thermal properties, and desirable light-activated melting effects and drug release capabilities. In vivo studies showed that the Flav7 + DOX-loaded MNs presented a remarkable NIR-II imaging ability, a good biosafety to the major organs, and a strong tumor-killing ability. It is worth noting that all of the treated breast cancer mice survived for 50 days without tumor recurrences or major organ damage. This study presented a promising MN system loaded with the multipurpose small molecule Flav7 that can be used for high-efficiency light-activated drug release for chemotherapy and high-performance light-controlled thermotherapy of tumors and also for remarkable NIR-II fluorescence imaging to guide chemo-thermal therapy.

The specific-targeting approach could promote the specificity of diagnosis and the accuracy of cancer treatment. The choice of a specific-targeting receptor is the key step in this approach. Glypican-3 (GPC3) is an oncofetal proteoglycan anchored on the cell membrane. It is overexpressed even in the early stage of hepatocellular carcinoma (HCC), whereas it shows almost no expression in the healthy adult liver. Therefore, GPC3 may be applied as a specific-targeting receptor for HCC theranostics. In this study, a GPC3 specific-targeting theranostics nanodevice, GPC3 targeting peptide (named G12)-modified liposomes co-loaded with sorafenib (SF) and IR780 iodide (IR780), was developed (GSI-Lip), which aims to realize early diagnosis and precise chemo-photothermal therapy of HCC. SF was the first-line chemotherapy drug for the treatment of HCC. IR780 was used for photothermal therapy and near-infrared fluorescence imaging. The evaluation of early diagnosis verified that early-stage tumors (3.45 ± 0.98 mm3, 2 days after 5 × 105 H22 cells’ inoculation in mice) could be clearly detected using GSI-Lip, which was significantly more sensitive than folic acid-modified liposomes (p < 0.01, 32.90 ± 10.01 mm3, 4 days after 1 × 106 H22 cells’ inoculation in mice). The study of the endocytic pathway indicated that specific G12/GPC3 recognition may induce caveolae-mediated endocytosis of GSI-Lip. Notably, the accumulation of GSI-Lip in tumors was significantly increased compared with that observed with folic acid-modified liposomes (p < 0.01). Specific-targeting endowed the precise antitumor effect of GSI-Lip. GSI-Lip showed a higher antitumor efficacy in comparison with folic acid-modified liposomes (inhibition rate: 90.52% vs 84.22%, respectively; p < 0.01). During a period of 21 days, the synergistic chemo-photothermal therapy (GSI-Lip + laser) exhibited a better antitumor effect versus GSI-Lip without laser (inhibition rate: 94.93% vs 90.52%, respectively; p < 0.01). Overall, GPC3-targeted GSI-Lip promoted the sensitivity and specificity of HCC early diagnosis and achieved synergistic efficacy of chemo-photothermal theranostics, which has potential clinical applications. Furthermore, the present study revealed that a more specific-targeting ligand could further improve the efficacy of theranostics against HCC.

Janus particles, consisting of two or more chemically distinct composites within a single structural system, have attracted significant attention for their solid surfactant functionality, as well as their potential applications in micro/nanomotors and functional materials. Here, we present a simple and robust method to prepare plasmonic Janus particles consisting of a polystyrene-tethered gold nanorod (AuNRs@PS) head and a poly(4-vinylpyridine) (P4VP) head through emulsion confined assembly. The balance of the Janus particles can be finely tuned by adjusting the volume ratio of the AuNRs@PS solution and P4VP solution. The result shows that the diameter ratio (r) of AuNRs@PS5k/P4VP is proportional to the volume ratio (R) of the AuNRs@PS and P4VP solutions. Furthermore, the obtained Janus particles with AuNR head have a peak absorbance of around 800 nm, which can be applied in photothermal therapy. Additionally, multifunctionality can be achieved by reducing nanoparticle (NP) precursors on a prefabricated scaffold of P4VP or co-assembling P4VP-tethered NPs with AuNRs@PS building blocks. These multifunctional Janus particles hold great potential for applications in micro/nanomotors, catalysts, and biological materials.

Bacterial infections present a significant threat to human health, a challenge that is intensified by the slow pace of novel antibiotic development and the swift emergence of bacterial resistance. The development of novel antibacterial agents is crucial. Indocyanine green (ICG), a widely used imaging dye, efficiently generates reactive oxygen species (ROS) and heat for treating bacterial infections but suffers from aggregation and instability, limiting its efficacy. In this study, tungsten disulfide (WS₂) nanosheet with a high surface area was used to load ICG, creating a multifunctional nanocomposite, WS 2 /ICG, aimed at treating bacteria-infected wounds. The two-dimensional surface structure of WS₂ provides dispersible binding sites for ICG, and the synthesized nanocomposite exhibits excellent stability. Under near-infrared (NIR) laser excitation, the generated heat further synergistically enhances the yield of singlet oxygen. Additionally, the WS₂/ICG nanoplatform synergistically combines photothermal effect with photodynamic effect, achieving a “1 + 1 > 2” enhancement. Upon NIR laser excitation, the nanocomposite disrupts bacterial cell membranes through localized heating and ROS accumulation, leading to energy metabolism system disruption and subsequent bacterial lysis and death. The findings demonstrate WS₂/ICG's outstanding antibacterial properties and biocompatibility, effectively treating skin infections and promoting tissue regeneration, providing a simple and promising solution for bacteria-infected wounds.

Infected wounds pose a significant global health challenge due to the persistence of bacterial biofilms and limited tissue self-repair. Nitric oxide (NO) functions as a potent antimicrobial agent, demonstrating a dual capacity for both antimicrobial action and tissue rejuvenation across varying concentrations. However, achieving controlled NO release at distinct stages of infected wound progression, simultaneously targeting biofilm removal and wound recovery, remains a formidable challenge. In this work, we introduce a smart electrospun fibrous membrane, featuring an interior laden with NO-loaded HKUST-1 particles and a porous external surface. Notably, the results reveal the photothermal property of HKUST-1 when exposed to near-infrared (NIR) light, enabling precise management of NO release contingent upon light conditions. During the initial phase of infection treatment, a significant NO release is triggered by near-infrared photothermal stimulation, synergistically complementing photothermal therapy to effectively eliminate bacterial biofilms. Subsequently, in the wound-healing phase, NO is released from the degrading fibrous membrane in a controlled and gradual manner, synergizing with trace amounts of copper ions released during MOF degradation. This collaborative mechanism accelerates the formation of blood vessels within the wound, thereby facilitating the healing process. This study suggests a promising and innovative approach for the effective treatment of infected wounds. Graphical Abstract A smart electrospinning fibrous membrane that can intelligently release NO at different stages of infected wound treatment was designed, which could eliminate biofilm and promote the healing of infected wounds.

Photothermal therapy, because of its inherent high specificity and low invasiveness, can easily and quickly inhibit infected wounds and promote repair. Herein, flexible wood (LM) was synthesized and combined with clarithromycin-carrying black phosphorus in a polyvinyl alcohol matrix to create a highly effective photothermal and flexible wood-based hydrogel (LM@PBKb), which exhibited photothermal and photodynamic bactericidal effects as well as long-lasting wound sterility for the treatment of infected skin wounds. The LM@PBKb hydrogel exhibited a photothermal conversion efficiency of 28.8%. When 808 nm laser irradiation and the long-lasting sustained release of the LM@PBKb hydrogel were combined, Escherichia coli was almost completely inhibited. After 808 nm laser irradiation in the near-infrared region, LM@PBKb used in mice with infected wounds rapidly inhibited infection, maintained a sterile environment for a long time, and accelerated wound healing. Scanning electron microscopy images show that the wounds treated with LM@PBKb were relatively flat and resembled uninfected wounds. This highly efficient photothermal flexible wood-based hydrogel is a high-potential environmentally friendly biomaterial.

Bacterial infections considerably limit the success of scaffold transplantation. CuO 2 may generate hydroxyl radicals through a self-cascade catalytic process, which is effective for sterilization. However, reducing CuO 2 -derived Cu + ions to Cu° can lead to a decline in catalytic activity. To overcome these limitations, we anchored CuO 2 onto MoS 2 to establish stable Cu-S-Mo interfacial sites. The CuO 2 /MoS 2 nanozyme was fabricated using a low-temperature co-precipitation technique and introduced into poly- l -lactic acid scaffolds using selective laser sintering. The Cu-S-Mo interfacial sites showed a triad of functions that synergistically enhanced catalytic performance. These sites stabilized the Cu atoms, augmented catalytic activity by facilitating efficient electron transfer through the Mo 4+ /Mo 6+ redox couple, and improved the conduction of heat derived from the photothermal response of MoS 2 to the Cu catalyst. The synergistic action of chemodynamic and photothermal therapy, mediated by the Cu-S-Mo sites, resulted in remarkable antibacterial efficacy, with 92.3 % and 93.5 % inhibition rates against Staphylococcus aureus and Escherichia coli , respectively. Experimental evidence and simulations demonstrated that Cu-S-Mo structure stabilized the Cu atoms and improved electron transfer via interatomic charge transfer. These results highlight the potential of the CuO 2 @MoS 2 /PLLA scaffold in eliminating bacterial infections.
