Self-pumping, pH-responsive Janus fibrous dressing for enhanced immunomodulation and accelerated diabetic wound healing

Zhou Sha,Jing Li,Yuheng Song,Hongchuang Li,Hongmei Liu,Jiahui Fan,Xueyong Li,Xiang Fei,Meifang Zhu

Diabetic wounds, characterized by excessive exudate accumulation and dysregulated inflammation, pose a global healthcare challenge due to their persistent non-healing nature. Traditional therapies often fail to simultaneously address three critical healing requirements: precise exudate management, coordinated inflammation-immune modulation, and pro-regenerative microenvironment establishment. Here, we present an innovative self-pumping Janus fibrous dressing integrating curcumin-loaded polycaprolactone nanofibers with cotton gauze. Its gradient architecture enables spontaneous exudate drainage and dynamic microenvironment modulation, establishing an optimal milieu for accelerated wound healing. Comprehensive in vitro investigations validate the dressing’s multifunctionality, including sensitive colorimetric pH response, antibacterial properties (>99 % inhibition against S. aureus ), and antioxidant capabilities (>90 % DPPH radical scavenging). In vivo studies in a diabetic mice model demonstrate the dressing’s superior wound healing efficacy, with ∼47 % reduction in wound edge maceration, mitigated inflammatory responses, and enhanced angiogenesis compared to commercial cotton dressings. This novel approach provides a promising foundation for the development of advanced wound dressings tailored for the management and healing of chronic wounds.

The structural discrepancy in the immunomodulatory potency of the polysaccharides RFHP-1 and RFHP-2 derived from Radix Fici Hirtae

Tiankai Bai,Zhilong Shi,Shuguang Bao,Xiaorong Hou,Yuping Zhao,Wai L.W. Leung,Yuhui Li,Sirigunqiqige Pan,Ying Xin,Ye Luo,Xiaohe Xiao,Laxinamujila Bai,Huifang Li

Radix Fici Hirtae (RFH), a traditional Chinese medication, is said to be useful in controlling immunity. However, it remained unclear whether its polysaccharides components are associated with these immunomodulatory properties. Herein, two polysaccharides, designated RFHP-1 and RFHP-2, were isolated from RFH using water-soluble alcohol precipitation and column chromatography. Structural characterization by monosaccharide content analysis, methylation, and nuclear magnetic resonance spectroscopy (NMR) revealed that the main chain of RFHP-1 was formed by the interconnection of →4)-α-D-Glc p -(1→, →3,6)-β-D-Glc p -(1→, →3)-β-D-Glc p -(1→, and →4,6)-α-D-Glc p -(1→, and the main chain of RFHP-2 was formed by the interconnection of →4)-α-D-Glc p -(1→, →3,6)-β-D-Gal p -(1→, →3)-β-D-Gal p -(1→, →4,6)-α-D-Glc p -(1→ and →6)-β-D-Gal p -(1→. Both RFHP-1 and RFHP-2 raised the CD4/CD8 ratio and improved the expression of immune factors in immunocompromised mice. By activating the NF-κB pathway (promote phosphorylation of p65 and IκB), RFHP-1 and RFHP-2 both promoted release cytokines (NO, TNF-a, IL-6, and IFN-γ) at RAW 264.7 macrophages. The results of molecular docking (−8.4 kcal/mol vs. -6.0 kcal/mol) and isothermal titration calorimetry (ITC) (−7.75 kcal/mol vs. -7.46 kcal/mol) demonstrated that RFHP-1 exhibited stronger binding affinity with MD2, suggesting greater immunoactivating potential. These findings indicate that RFHP polysaccharides, particularly RFHP-1, may serve as promising candidates for development as dietary supplements for individuals with compromised immune systems.

Antiviral effects of rhein on largemouth bass ranavirus (LMBRaV)

Mengmeng Chen,Mingyang Xue,Yong Zhou,Chen Xu,Nan Jiang,Yuding Fan,Yan Meng

Largemouth bass ranavirus (LMBRaV), also known as largemouth bass virus (LMBV), is a highly damaging viral pathogen that causes widespread mortality in cultured largemouth bass and poses a significant threat to the farming industry in China. However, there is a paucity of effective preventive methods for this disease by LMBRaV. Research has established that herbal extracts possess antiviral activity, potentially exerting a controlling effect on viruses in aquaculture. Therefore, the identification of simple and structurally safe herbal extract immunomodulators is critical for largemouth bass health and development. Rhein, an herbal compound, exhibits a broad spectrum of pharmacological effects in herbal medicine. This study aimed to evaluate the properties and effects of rhein against LMBRaV in epithelioma papilloma cyprinid (EPC) and largemouth bass. In vitro CCK-8 assays were performed to determine the highest safe concentrations. Significantly, 50 mg/L rhein effectively inhibited cytopathic effects (CPE) in EPC induced by LMBRaV, resulting in a 97 % reduction in viral replication rates. Furthermore, a six-point dose-response experiment revealed that rhein doses as low as 6.4 mg/L inhibited LMBRaV and suppressed LMBRaV infection with a dose-dependent manner. Subsequent experiments demonstrated that sustained rhein activity effectively alleviated CPE and nuclear damage caused by LMBRaV infection. Additionally, rhein exerted protective effects against declines in LMBRaV-induced mitochondrial membrane potential. In vivo experiments validated that largemouth bass diets supplemented with 0.075 % rhein exerted a protective effect against LMBRaV infection. Moreover, LMBRaV MCP gene expression levels were lower in several organs of fish in the rhein supplementation group. Immunogene analysis determined that rhein upregulated relevant gene expression levels in multiple organs of largemouth bass, with higher activities observed for the Mx, INF-γ, and IRF3 genes. Histopathological results displayed that a supplemental dosage of 0.075 % rhein significantly mitigated LMBRaV infection-induced pathological damage in the liver, spleen, and kidney. Taken together, these in vivo and in vitro results support the effective use of rhein as a promising candidate for the development of antiviral treatment against LMBRaV infection, highlighting its role in aquatic animal husbandry as a wide-spectrum antiviral and immune response booster.

Preparation, Structure Characteristics, Antioxidant, Antitumor, and Immunomodulatory Activity of Polysaccharides From Rheum officinale Baill. and Their Derivatives

Xiaoliang Zhao,Yanmei Liu,Haili Wang,Kaili Qi,Haidong Wen,Yuxin Wang,Yuanyuan Ma,Cunjin Wang,Jing Zhang,Yonggang Wang,Weijie Zhang

In this study, three polysaccharides (RHC, RHH, and RHA) were sequentially extracted from Rheum officinale Baill., under different conditions, and their sulfated, phosphorylated, and selenated derivatives were successfully synthesized. Results indicated that the sequential extraction method effectively separated polysaccharides with distinct structures. According to their charge properties, these three polysaccharides were identified as acidic polysaccharides with different structural characteristics. Activity assays showed that the phosphorylation modification of polysaccharides significantly enhanced the scavenging of hydroxyl, superoxide, and DPPH radicals, as well as the inhibitory effects on HepG2 and HeLa cells. Phosphorylated derivatives also promoted proliferation, phagocytosis, and nitric oxide (NO) release in RAW264.7 macrophages. In the 50–800 µg/mL range, these polysaccharides and their derivatives exhibited no cytotoxicity to RAW264.7 cells and significantly promoted cell proliferation, phagocytosis, and NO release. Notably, RHC and its derivatives have the most prominent antitumor and immunomodulatory effects. Overall, R. officinale polysaccharides and their derivatives, especially phosphorylated ones, have potential applications in health food and pharmaceuticals. Graphical

Antidepressant and immunosuppressive activities of two polysaccharides from Poria cocos (Schw.) Wolf

Wuxia Zhang,Lu Chen,Peng Li,Jinzhong Zhao,Jinyou Duan

Pursuit of novel effective antidepressants is an urgent task. Poria cocos ( Schw .) Wolf (PCW) is a traditional herb with antidepressant effects . Polysaccharides designated PCWPW and PCWPS were purified from PCW by DEAE-52 cellulose chromatography. Their structures were characterized using physicochemical and spectral methods. Chemical analysis indicated that PCWPW (37,154 Da) and PCWPS (186,209 Da) were mainly composed of mannose , glucose, galactose and fucose . Their antidepressant activities were evaluated by tail suspension test (TST), forced swimming test (FST) with chronic unpredictable mild stress (CUMS) model mice. To investigate the antidepressant mechanism, the neuroprotective and immunomodulation effects were assessed by MTT method. Results showed that PCWPW and PCWPS possess obvious antidepressant effects and can suppress ConA-stimulated T cell proliferation in a dose-dependent manner. In addition, PCWPS could protect the PC12 cells from H 2 O 2 -induced damage and suppress LPS-induced B cell proliferation . These findings implied that PCWPW and PCWPS might be a candidate for developing antidepressant or immunosuppressive agents in food and pharmaceutical industries.

Immunomodulatory effect of a formula developed from American ginseng and Chinese jujube extracts in mice

Zhuo-ping Yu,Dong-dong Xu,Lai-feng Lu,Xiao-dong Zheng,Wei Chen

Background: American ginseng ( Panax quinquefolius L.) and Chinese jujube ( Zizyphus jujuba Mill.) are commonly used in traditional Chinese medicine to enhance immune function. Objective: The present study aimed to develop one Chinese prescription, Shenzao Cha (SZC), consisting of American ginseng and Chinese jujube, and systematically investigate its immunomodulation in healthy ICR mice. Methods: Normal ICR mice received intragastric administration of SZC (1.3, 2.6, and 5.2 g raw material/kg body weight) once daily for four weeks, while a control group received the same amount of sterile water. Results: SZC significantly increased the spleen and thymus indices and T-lymphocyte proliferation, while the T-lymphocyte proliferation in the 5.2 g/kg group was 1.4-fold higher than that in the control. Further, 1.3 g/kg SZC could markedly improve hemolytic activity by 25.2%, and 2.6 g/kg SZC increased the NK cell activity by 78.6% relative to the control. In addition, the activities of antioxidant enzymes (superoxide dismutase, catalase, and glutathione peroxidase), that participated in modulating oxidative stress, were significantly increased in the liver, spleen, thymus, and serum, while the contents of malondialdehyde were dramatically decreased. Conclusions: SZC exhibited potent immunomodulatory effects on innate and adaptive immunity in healthy ICR mice, as well as potential antioxidant activity for prevention of oxidative stress, which was suggested to partly contribute to the immune enhancement.

Immunoregulation and antioxidant activities of a novel acidic polysaccharide from Radix Paeoniae Alba

Zhang Wuxia,Hu Yihua,Zhao Jinzhong,Zhang Yongpo,Guo Dongdong,Gao Chunyan,Duan Jinyou,Li Peng

Radix Paeoniae Alba is widely used in Chinese traditional medicine to treat various diseases such as gastrointestinal disorders, immunomodulatory, cancer, and other diseases. In this paper, a novel acidic polysaccharide RPAPS purified from Radix Paeoniae Alba was evaluated for its structural features and potential of immunomodulatory and antioxidant activities. RPAPS (molecular weight: 1.0× 10 5  Da) was mainly composed of α-(1 → 4)-Glc p , α-Ara p , α-Gal p , α-Rha p , β-D-Glc p , α-(1 → 6)-linked Glc p and GalA. Immunological tests indicated that RPAPS could improve RAW264.7 phagocytic activity and LPS-induced splenocyte proliferation. For antioxidant activities, RPAPS showed reducing power and DPPH scavenging activity in dose dependent. Moreover, RPAPS could significantly protect the PC12 cells from H 2 O 2 damage. These data implied polysaccharides RPAPS had the potential to be novel natural antioxidative and immunopotentiating agents for using in functional foods or medicine.

Bioactive nanocapsules based on lychee-like bilayer structure for synovitis treatment

Wenwen Chai,Pengfei Tian,Mengke Fan,Xiaochen Chen,Liyan Zhang,Muyan Qin,Yingbo Wang,Yansong Wang,Wei Chen,Qiang Yang,Honglong Li,Xu Cui,Deping Wang,Haobo Pan

Osteoarthritis (OA), as a degenerative disease, often necessitates intra-articular administration to alleviate pain. Synovial inflammation represents an important pathological factor in the development of OA. ZIF-8 (a type of Zeolitic imidazolate frameworks), a newly developed drug carrier, has garnered significant attention due to its high drug loading capacity and favorable biocompatibility. However, pH-sensitive burst drug release in inflammatory environments hampered its clinical application. To overcome this limitation, we have engineered a novel lychee-like double-layered bioactive nanocapsule, with ZIF-8 serving as the core and a bioactive amorphous sol as the outer shell, facilitated by Zn ion and Si–O coordination. The amorphous sol not only inhibits the rapid degradation of ZIF-8 in inflammatory conditions, preventing quick drug release, but also enhances the immunomodulatory capabilities of the functional carrier. Remarkably, our studies reveal that, in addition to sustained drug release, the synergistic effect of the functional carrier and anti-inflammatory drugs can regulate inflammation-induced lymphatic abnormalities. This is manifested by the suppression of macrophage secretion of excessive VEGF-C and iNOS, thereby modulating lymphatic density and restoring normal lymphatic rhythmic function. These findings indicate that such pathology-adaptive functional carriers possess superior drug release control and immunomodulatory potential compared to traditional carriers, presenting a promising innovative material for synovitis treatment.

Characterization, antioxidant and immunomodulatory activities of polysaccharides from Prunella vulgaris Linn

Chao Li,Qiang Huang,Xiong Fu,Xiu-Jie Yue,Rui Hai Liu,Li-Jun You

Water-soluble polysaccharides from Prunella vulgaris Linn ( P. vulgaris ) were fractionated using DEAE-Sepharose fast-flow column to obtain several eluents of water (PV-P1), 0.1   M NaCl (PV-P2) and 0.2   M NaCl (PV-P3). Structural analyses showed that PV-P1 had a higher molecular weight and degree of branching as compared to PV-P2 and PV-P3. Tertiary structure analyses indicated that PV-P1, PV-P2 and PV-P3 did not have triple-helical conformation. PV-P2 and PV-P3 showed stronger antioxidant activities than PV-P1, as measured radical scavenging capacities. PV-P1 showed stronger immunomodulatory activities than PV-P2 and PV-P3 in term of stimulation of the production of pro-inflammatory cytokines, including nitric oxide (NO), tumor necrosis factor-α (TNF-α), and interleukin-6 (IL-6) in murine macrophage RAW 264.7 cells. PV-P1, PV-P2 and PV-P3 did not exhibit cytotoxicities against RAW 264.7 at the concentrations tested. These results suggest that P. vulgaris polysaccharides could be explored as potential antioxidant and immunomodulatory agents for the complementary medicine or functional foods.

Mechanism of Ligilactobacillus salivarius GX118 in Regulating the Growth of Rainbow Trout (Oncorhynchus mykiss) and Resistance to Aeromonas salmonicida Infection

Xiyu Cao,Bowen Wang,Wenhao Ren,Jiang Wang,Qi Liu,Yichao Ren,Xianliang Meng

Lactic acid bacteriaLigilactobacillus salivariushas been shown to be a substitute for antibiotics in the treatment of bacterial disease in high animals. However, its beneficial mechanism in fish farming is still unclear. This study evaluated the antagonistic effects of theLigilactobacillus salivariusGX118 strain onAeromonas salmonicidaand its regulation of rainbow trout growth in vivo and in vitro. The results found that GX118 produces an antibacterial substance that can directly destroy the cell wall ofA. salmonicida. Whole-genome sequencing of GX118 revealed that Enterolysin A is a type III bacteriocin with antibacterial properties. An in vivo experiment showed that the supplementation of GX118 in diet competitively inhibited the colonization ofA. salmonicidain the intestine. In addition, it was able to improve the growth performance of rainbow trout within a 21-day feeding experiment. The supplementation of GX118 increased the diversity of gut microbiota, in which the abundance ofBacteroidota,Blautia, andRhodobacteraceaeincreased. In addition, the use of GX118 activated the expression ofIFN-γandNF-κBgenes and reduced the expression level ofIL-6andIL-8, thus exhibiting a certain effect on activating the immunity of rainbow trout. This study provides a scientific basis for the development of antibacterial probiotics in the healthy farming of rainbow trout.

Structural characterization and biological activities of two α-glucans from Radix Paeoniae Alba

Zhang Wuxia,Li Peng,Song Dan,Niu Hong,Shi Songshan,Wang Shunchun,Duan Jinyou

Radix Paeoniae Alba is widely used in Chinese traditional medicine to treat various diseases such as gastrointestinal disorders, cancer, and other diseases. In this study, two polysaccharides RPAPW1 and RPAPW2 were isolated from Radix Paeoniae Alba by DEAE-52 cellulose chromatography and G-25 sephadex. According to physicochemical methods, NMR and methylation analysis, RPAPW1 and RPAPW2 were established to be α-glucans consisting of predominant 4-linked α- Glc residues branched at O-6 and contained trace amount of protein and uronic acid. Immunological tests indicated that RPAPW1, RPAPW2 and could promote splenocyte proliferation and RAW264.7 phagocytic activity. In vitro , RPAPW1 and RPAPW2 elicited a week reducing power, DPPH scavenging activity and could not protect the PC12 cells from H 2 O 2 damage. These data implied polysaccharides RPAPW1 and RPAPW2 had the potential to be a natural immunopotentiating and antioxidant supplement for preparing functional foods and nutraceuticals.

Dual-Biomimetic Bone Adhesive with Osteoimmunomodulatory Capabilities for Anatomical Reconstruction of Comminuted Fractures

Junyao Cheng,Hufei Wang,Ming Li,Jianpeng Gao,Xiao Liu,Chuyue Zhang,Pengfei Chi,Bo Li,Yuan Xve,Daoyang Fan,Zheng Wang,Jianheng Liu,Xing Wang,Licheng Zhang

The development of bone adhesives capable of restoring natural bone structure and function represents a pioneering advancement in surgical technology, offering significant potential to improve the treatment outcomes of comminuted fractures. However, the development of a clinically demand-driven adhesive, ensuring both reliable adhesion and osteogenic activity, poses a significant challenge. Herein, a structurally and functionally dual-biomimetic bone adhesive is developed through the design of an organic–inorganic network-enhanced hydrogel, which integrates caffeic acid-grafted collagen (CAC), aminated laponite (ALAP), and N-hydroxysuccinimide ester-terminated polyethylene glycol (tetra-PEG-SC). Benefiting from nanosheet-induced strengthening, the mechanically reinforced adhesive can maintain integrity under extreme compression (98%) and tensile (600%) deformations, facilitating anatomical repositioning of rabbit radius and porcine femur fractures. Of greater importance, the adhesive accelerated fracture healing, potentially via ROS scavenging and immunomodulation, as evidenced by reduced oxidative stress and M2 macrophage polarization in vitro. Assessed through an innovative rabbit radius comminuted fracture model, the injectable bone adhesive demonstrates rapid and flexible adhesion of bone fragments in a blood-rich environment, as observed over the 2 fold improvement in biomechanical and radiological performance compared with commercially available cyanoacrylate adhesives. This intricately designed bone adhesive holds promise as a novel solution for addressing complex fracture cases in surgical treatment.
