Perfluorocarbon groups or related compounds are usually used to modify polymer materials because of their low surface energy properties. In this paper, a novel perfluorocarbon-containing compound was synthesized and used as a capping agent for the preparation of perfluorocarbon chain terminated poly(ether sulfone). The regulation of the fluorine content was achieved by controlling the molecular weight. FTIR and 1H NMR spectra were used to confirm the existence of perfluorocarbon terminated groups in the polymer chains. In addition, the ratios of F/C were acquired by integral area calculation from the 1H NMR spectrum. XPS characterization showed that the surface migration phenomenon of fluorine became apparent along with the increase of the F/C ratio. Further testing found that the introduction of perfluorinated end groups into the PES chains had little influence on the polymers' mechanical and thermal performance, by contrast, their surface energy decreased efficiently from approximately 40 mJ m−2 to 16 mJ m−2 with the F/C ratio on the surface increased from 0 to 0.556, which could develop PES as a low surface energy material for industrial applications.

Excessive blood loss and infections are the prominent risks accounting for mortality and disability associated with acute wounds. Consequently, wound dressings should encompass adequate adhesive, hemostatic, and bactericidal attributes, yet their development remains challenging. This investigation presented the benefits of incorporating a perfluorocarbon nanoemulsion (PPP NE) into a silk-fibroin (SF)-based hydrogel. By stimulating the β-sheet conformation of the SF chains, PPP NEs drastically shortened the gelation time while augmenting the elasticity, mechanical stability, and viscosity of the hydrogel. Furthermore, the integration of PPP NEs improved hemostatic competence by boosting the affinity between cells and biomacromolecules. It also endowed the hydrogel with ultrasound-controlled bactericidal ability through the inducement of inner cavitation by perfluorocarbon and reactive oxygen species (ROS) generated by the sonosensitizer protoporphyrin. Ultimately, we employed a laparotomy bleeding model and a Staphylococcus aureus-infected trauma wound to demonstrate the first-aid efficacy. Thus, our research suggested an emulsion-incorporating strategy for managing emergency wounds.

Biomedical nanoplatforms have been widely investigated for ultrasound (US) imaging and cancer therapy. Herein, perfluorocarbon (PFC) is encapsulated into biocompatible polydopamine (PDA) to form a theranostic nanosystem, followed by the modification of polyethylene glycol (PEG) to stabilize the nanoparticle via a facile one-pot method. Under 808 nm near-infrared laser irradiation, PDA can generate hyperthermia to transform PFC droplets to bubbles with high US imaging sensitivity. The US imaging detection of the PFC-PDA-PEG nanosystem is achievable in a time span of up to 25 min in vitro at a low US frequency and mechanical index, manifesting a US imaging performance for in vivo application. Moreover, tumor cells incubated with the nanosystem are ablated effectively under laser irradiation at 808 nm. The results illustrate the potential of the PDA-based theranostic agent in US imaging-guided photothermal therapy of tumor.

The low oxygen concentration limits the therapeutic efficacy of photodynamic therapy in treating cancer cells in hypoxia, since the cytotoxic 1 O 2 can't be effectively generated in this condition. To overcome this, we load photosensitizer into perfluorocarbon nanodroplet, which has a high oxygen capacity to enrich O 2 for photodynamic consumption. Under the well-controlled hypoxic condition, we test its efficacy both in vitro and in vivo . This method can be successfully used for destroying cancer cells in hypoxic condition.

A novel organic/inorganic hybrid nanovesicle as an ultrasound imaging agent is synthesized via facile emulsion and silica deposition methods. This nanovesicle, hyaluronate (HA)–docetaxel (DTX)/perfluoro- n -pentane (PFP)@SNC, consists of an encapsulated liquid PFP core, loaded DTX, and an HA-decorated silica shell. The HA–DTX/PFP@SNC has a narrow size distribution of 274.5 ± 3.25 nm, a negative zeta potential of −11.6 ± 0.47 mV, and an entrapment efficiency of 86.70% ± 1.42%. HA–DTX/PFP@SNC possesses an ultrasound (US)-triggered drug release and a temperature-dependent size change behavior. Compared with DTX/PFP@soybean phosphatidylcholine (SPC), which has no silica shell, the HA–DTX/PFP@SNC is more stable under various conditions. The MTT assay indicates that the blank HA–PFP@SNC vehicle has no cytotoxicity to A549 cells. Furthermore, due to the HA-mediated tumor-targeting ability, the HA–DTX/PFP@SNC shows obvious cytotoxicity to A549 cells. In vitro and in vivo US imaging results indicate that HA–DTX/PFP@SNC has a stronger and more durable echo signal than DTX/PFP@SPC. Moreover, the in vivo echo signal of HA–DTX/PFP@SNC is stronger than that of DTX/PFP@SNC due to the HA-mediated tumor targeting. Therefore, this novel organic/inorganic hybrid vesicle is a US contrast agent candidate.

Introducing additional oxidizers in aluminized explosives is an effective way to improve energy performance by enhancing the secondary combustion reaction. In this work, perfluorocarbon as oxidizer has been introduced into TATB/Al based energetic materials to improve combustion reaction and energy output performance. The uniform TATB/Al/perfluorocarbon composites are prepared through acoustic resonance mixed technology, and the energy and pressure output performance are fully studied in air blast and underwater explosion. A significantly exothermic peak derived from the reaction between fluorine and Al is observed between 540 and 580℃ for TATB/Al/perfluorocarbon. The heat of explosion of TATB/Al/perfluorocarbon is increased first and then decreased with F/Al ratio from 0 to 0.608. The highest heat of explosion is 6021 J/g for TATB/Al/perfluorocarbon with F/Al ratio of 0.274. Furthermore, the enhanced working capacity, the significantly enlarged fireball and relatively high overpressure are obtained for TATB/Al/perfluorocarbon. More importantly, the shock impulse and first bubble oscillation time of TATB/Al/perfluorocarbon have been increased by 23.2% and 21.18% during underwater explosion, respectively. The results further illustrate that adding perfluorocarbon is a feasible approach to enhance energy output due to combustion reaction between fluorine and Al, which can be applied in explosives and propellants.

Ferrocene, known for its 'sandwich' structure and Fenton catalytic activity for tumor treatment, its two-dimensional (2D) self-assembly has not yet been exploited for therapeutic applications. Perfluorocarbons (PFCs) can carry and transport oxygen to alleviate tumor hypoxia. Nevertheless, designing ferrocene derivatives with PFC chains to alleviate tumor hypoxia and improve cancer treatment remains a challenge. Here, we first synthesized ferrocene derivatives with different PFC chains (nF-Fc, n = 5, 9, 13) by conjugating perfluorocarbonic acids with 2-(ferrocenylethynyl)aniline (Fc). These derivatives were able to self-assemble into 2D nanosheets, with hydrated particle size decreasing as the chain length increased. The chain lengths also influenced the oxygen-carrying capacity, iron release capacity, and glutathione consumption capacity, all of which impacted their ability to alleviate tumor hypoxia, produce intracellular reactive oxygen species, and enhance chemodynamic therapeutic efficacy. Notably, PFC incorporation altered the mechanism of induced cell death. By elucidating the effects of ferrocene derivatives with different PFC chains on the hypoxic tumor microenvironment (TME) and their therapeutic efficacy, this research advances the development of ferrocene-based 2D materials for cancer treatment and offers a novel approach to optimize compound structures better regulation of the hypoxic TME, ultimately improving therapeutic outcomes.

Stimuli-responsive microcarriers have received considerable attention in a variety of fields, including disease diagnosis, drug delivery, sensing, and imaging. Here, we report the generation of multiple-responsive perfluorocarbon-loaded magnetic hydrogel microcapsules (PMHMs) with uniform size for magnetic controlled ultrasound (US) and laser activation. The PMHMs are fabricated by a novel coaxial interface shearing (CIS) method based on the mechanism of liquid bridge formation and fracture. Perfluorocarbon and iron oxide magnetic nanoparticles are used as US-responsive and photothermal absorption medium, respectively, and magnetic nanoparticles are also used for magnetic-controlled targeting. Moreover, the size, structure, and function of the prepared biocompatible PMHMs can be precisely controlled by adjusting the process parameters of CIS. It is indicated that the PMHMs have different US- and light-responsive characteristics, mainly because of the difference of their activation mechanisms. It is demonstrated that laser has better activation resolution and can achieve site-specific activation and drug release of PMHMs. The multiple-responsive features imply that the PMHMs fabricated by CIS may provide an effective drug release platform for biomedical and pharmaceutical applications.

Hypoxia-based agents (HBAs), such as anaerobic bacteria and bioreductive prodrugs, require both a permeable and hypoxic intratumoural environment to be fully effective. To solve this problem, herein, we report that perfluorocarbon nanoparticles (PNPs) can be used to create a long-lasting, penetrable and hypoxic tumour microenvironment for ensuring both the delivery and activation of subsequently administered HBAs. In addition to the increased permeability and enhanced hypoxia caused by the PNPs, the PNPs can be retained to further achieve the long-term inhibition of intratumoural O 2 reperfusion while enhancing HBA accumulation for over 24 h. Therefore, perfluorocarbon materials may have great potential for reigniting clinical research on hypoxia-based drugs.

Developing strategies for the treatment of bacterial biofilms is challenging due to their complex and resilient structure, low permeability to therapeutics, and ability to protect resident pathogens. Herein, we demonstrate that a polylysine-stabilized perfluorocarbon nanoemulsion is favored for penetrating biofilms and sensitizing the cavitation effect of low-intensity ultrasound, resulting in the dispersal of extracellular polymeric substances and killing of the protected cells. Through experiments, we observed a complete penetration of the nanoemulsion in a 40 μm Pseudomonas aeruginosa biofilm and demonstrated that it was induced by the fluidic perfluorocarbon, possibly attributing to its low surface tension. Furthermore, we presented an almost complete antibiofilm effect with a low-intensity ultrasound (1 MHz, 0.75 W/cm2, 5 min) in diverse cases, including cultured biofilms, colonized urinary catheters, and chronic wounds. During the treatment process, the perfluorocarbon phase enhanced the number and imploding energy of ultrasound cavities, thoroughly divided the biofilm structure, prevented biofilm self-healing, and sterilized the resident pathogens. Thus, the penetration and sensitization of the nanoemulsion might serve as a facile and potent strategy for eradicating biofilms in various applications.

Photodynamic therapy (PDT) has shown promising development due to its low toxicity, high selectivity and low side effects. However, the effectiveness of PDT is dependent on the concentration of oxygen and tumor cells have a hypoxia microenvironment that reduces the therapeutic effect of PDT. Therefore, near-infrared absorbing conjugated copolymers with perfluorocarbon side chains, 2,5-bis(2- octyldodecyl)-3,6-di(thiophen-2-yl)-2,5-dihydropyrrolo[3,4-c]pyrrole-1,4-dione- co -2,5-thiophene- co -9,9-bis( N -methylperfluorohexylsulfonamidoethyl propionate)- fluorene copolymers, with the ability to generate singlet oxygen under hypoxia conditions were designed and synthesized. The structures of the copolymers were characterized by Fourier transform infrared spectrometer (FTIR) and proton nuclear magnetic resonance spectrum ( 1 HNMR). The singlet oxygen generation ability of the copolymers and corresponding nanoparticles under organic solvents and aqueous conditions was investigated. The results show that the absorption wavelengths of the fluorine-containing conjugated copolymers are greater than 650 nm and they show strong absorption in near-infrared region. The perfluorocarbon (PFC) side chain endows the copolymers with high singlet oxygen generation capability. As the content of fluorine-containing structural units increases, the generation capability of singlet oxygen increases. When the proportion of fluorine-containing structural units is 35%mol (P 3 -F35%), the copolymer has the singlet oxygen quantum yield of 36.29%. And the singlet oxygen generation capability of the fluorine-containing copolymers is significantly enhanced even under hypoxia conditions. The nanoparticle of fluorine-containing conjugated polymer shows good singlet oxygen generation ability in aqueous suspension. The results of this study indicate that introducing fluorine-containing structure unit into near-infrared absorbing conjugated copolymers is an effective way to enhance the singlet oxygen generation capability of copolymers, which may have broad application prospects in the field of photodynamic therapy.

In the treatment of heart disease, strategies for the targeted delivery of protein therapeutics to the heart by inhalation are still immature. Perfluorocarbons (PFCs) are inert chemicals with good biocompatibility , and unique physico-chemical properties that have recently led to their applications in numerous fields. In this study, we combined the advantages of protein-phospholipid complexes and PFC emulsions and then synthesized protein-loaded PFC nanoemulsions (PNEs) to test whether, after inhalation, these nanoemulsions could deliver therapeutic proteins to the heart. After preparing protein-phospholipid complexes by lyophilization, we obtained PNEs by extrusion. The particle size and surface charge of PNEs were about 140 nm and -50 mV, respectively. In vitro results showed that the PNEs had a fine particle fraction of 35% and exhibited sustained protein release. Translocation studies were done using three types of pulmonary epithelial cells, and ~7% translocation was observed in the Calu-3 cell line. Further, they were easily absorbed by cells and had therapeutic effects in culture. In vivo results showed that the PNEs successfully delivered proteins to the myocardial tissue of rats and reduced ischemic myocardial injury caused by acute myocardial infarction (AMI). This study suggests that inhalation of PNEs is a new potential strategy to deliver proteins to cardiac tissues for treating heart diseases.
