Co-metabolic biodegradation of 4-chlorophenol by photosynthetic bacteria

Binchao Lu,Liang Wang,Xin Zheng,Zhongce Hu,Zhiyan Pan

Environmental contamination by 4-chlorophenol (4-CP) is a major concern. Photosynthetic bacteria have the ability to biodegrade 4-CP under dark aerobic conditions. In this study, we found that using different carbon sources (i.e. glucose, sodium acetate, sodium propionate sucrose, and malic acid) as co-metabolic substrates accelerated the biodegradation of 4-CP, and this acceleration was especially pronounced in the glucose treatment. A maximum degradation rate of 96.99% was reached under a concentration of 3.0 g·L−1 after 6 days of culture. The optimum conditions were pH 7.5, a temperature of 30°C, and a rotation speed of 135 rpm. The biodegradation of 4-CP was achieved at a range of salinities (0–3.0% NaCl, w/v). The biodegradation kinetics agreed with the Haldane model, and the kinetic constants were rmax = 0.14 d−1, Km = 33.9 mg·L−1, and Ki = 159.6 mg·L−1. Additionally, the coexistence of phenol or 2,4-dichlorophenol (2, 4-DCP) had a certain impact on the degradation of 4-CP under dark aerobic conditions. When the coexisting phenol concentration reached 100 mg·L−1, the maximum degradation rate of 4-CP reached 90.20%. The degradation rate of 4-CP decreased as the concentration of coexisting 2, 4-DCP increased.

A novel immobilized bacteria consortium enhanced remediation efficiency of PAHs in soil: Insights into key removal mechanism and main driving factor

Yuqian Li,Haomin Chen,Wei Li,Beidou Xi,Caihong Huang

The remediation of sites co-contaminated with polycyclic aromatic hydrocarbons (PAHs) and heavy metals (HMs) poses challenges for efficient and ecofriendly restoration methods. In this study, three strains ( Pseudomonas sp. PDC-1, Rhodococcus sp. RDC-1, and Enterobacter sp. EDC-1) were isolated from sites contaminated with PAHs and HMs. The constructed bacteria consortium was then immobilized using biochar, bentonite, and peat. The immobilized bacteria consortium (IBC) demonstrated efficient removal ability of phenanthrene (58.1 %-73.4 %) and benzo[ a ]pyrene (69.6 %-83.5 %) during 60 days. Additionally, the IBC decreased soil bacterial richness and diversity, but increased the relative abundance of Proteobacteria phylum and Ochrobactrum genus, which were capable of degrading PAHs. Soil microbial co-occurrence network with IBC was classified into three main modules, and 14 genera were identified as keystone taxa linked to PAHs degradation and HMs resistance. The IBC enhanced the dioxygenase metabolic pathways for PAHs degradation, including phthalic acid and salicylic acid pathways, which became the main driving factor affecting PAHs removal efficiency based on the structural equation modeling analysis. This study confirmed the potential application of the constructed IBC in the bioremediation of soil co-contaminated with PAHs-HMs, and provides insights into key removal mechanism and main driving factor of the enhanced elimination of PAHs.

Studies on the characteristics and mechanism of aerobic biodegradation of tetrabromobisphenol A by Irpex lacteus F17

Jie Chen,Juan Wu,Luosheng Fan,Rong Jia

The study investigated the characteristics of aerobic degradation of tetrabromobisphenol A (TBBPA) by Irpex lacteus F17 ( I. lacteus F17) under four different cometabolic substrates (phenol, glucose, sodium pyruvate, and sodium citrate). The biodegradation of TBBPA by I. lacteus F17 could be enhanced via cometabolism, and glucose (8 g/L) was confirmed to be the optimum carbon source. For different initial solution pH ranging from 3.0 to 8.0, the results showed that I. lacteus F17 could be applied to biodegrade TBBPA in a wide pH range of 4.0–8.0, and the degradation rate could reach the maximum 75.31%, while the debromination rate reached the maximum 12.40% under pH 5.0. In addition, it has been confirmed that Mn 2+ (50 μmol/L) could promote the secretion of manganese peroxidase and TBBPA biodegradation efficiency. Seven intermediates were identified by gas chromatography–mass spectrometry analysis, and the possible degradation pathways were proposed, which indicated the biodegradation of TBBPA might be subjected to debromination, β -scission, hydroxylation, deprotonation, and oxidation reactions.

Enhanced degradation of phenolic compounds in coal gasification wastewater by activated carbon-Fe3O4 nanoparticles coupled with anaerobic co-metabolism

Yajie Li,Weikang Kong,Hongbo Liu,Yaoliang Hong,Tianyin Huang

The aim of our research was to investigate the enhanced anaerobic degradation process of coal gasification wastewater (CGW) by conductive material activated carbon and Fe 3 O 4 nanoparticles (Fe 3 O 4 NPs) with the assistance of co-substrate. The optimal ratio of activated carbon to Fe 3 O 4 NPs (3:1) and the optimal addition of co-substrate (glucose concentration =1500 mg/L) were explored through batch tests. During the long-run reactor experiments, 1500 mg/L glucose was mixed into the influent, and activated carbon-Fe 3 O 4 NPs in a ratio of 3:1 were mixed into the anaerobic sludge. The concentration of total phenols in the effluent was kept around 50 mg/L, with a removal rate of 96% and an increase in methane production to 226 mL/day in the experimental reactor. Compared with the control reactor, the methane yield was increased by 64.6%. Toxicity analysis indicated that the toxicity of CGW can be greatly alleviated by conductive material activated carbon and Fe 3 O 4 NPs coupled with anaerobic co-digestion conditions. Microbial community analysis demonstrated that direct interspecies electron transfer (DIET) may have occurred between exoelectrogens ( Clostridium , Anaerolinea, Mesotoga, Synergistia) and methanogenic endoelectrogens ( Methanobacterium , Methanothrix ) for syntrophic metabolism through activated carbon-Fe 3 O 4 nanoparticles coupled with anaerobic co-metabolism.

Reduction characteristic of chlorobenzene by a newly isolated Paenarthrobacter ureafaciens LY from a pharmaceutical wastewater treatment plant

Nan Liu,Yan-yan Yao,Jin Zhang,Ji-guo Zhang,Chao Wu,Du-juan Ouyang,Chang-yong Zou,Zhen-qiang Yang,Ji-xiang Li

A highly efficient chlorobenzene-degrading strain was isolated from the sludge of a sewage treatment plant associated with a pharmaceutical company. The strain exhibited a similarity of over 99.9% with multiple strains of Paenarthrobacter ureafaciens . Therefore, the strain was suggested to be P. ureafaciens LY. This novel strain exhibited a broad spectrum of pollutant degradation capabilities, effectively degrading chlorobenzene and other organic pollutants, such as 1, 2, 4-trichlorobenzene, phenol, and xylene. Moreover, P. ureafaciens LY co-metabolized mixtures of chlorobenzene with 1, 2, 4-trichlorobenzene or phenol. Evaluation of its degradation efficiency showed that it achieved an impressive degradation rate of 94.78% for chlorobenzene within 8 h. The Haldane–Andrews model was used to describe the growth of P. ureafaciens LY under specific pollutants and its concentrations, revealing a maximum specific growth rate ( μ max ) of 0.33 h −1 . The isolation and characterization of P. ureafaciens LY, along with its ability to degrade chlorobenzene, provides valuable insights for the development of efficient and eco-friendly approaches to mitigate chlorobenzene contamination. Additionally, investigation of the degradation performance of the strain in the presence of other pollutants offers important information for understanding the complexities of co-metabolism in mixed-pollutant environments. Significance statement The degradation efficiency of chlorobenzene in waste gas is difficult to deal with its high volatility and molecular stability. However, the research on treatment technology of typical chlorine-containing volatile organic compounds is still concerned. A highly efficient chlorobenzene-degrading strain, identified as Paenarthrobacter ureafaciens LY, was successfully isolated from the sludge of a sewage treatment plant associated with a pharmaceutical factory. Its degradation efficiency was evaluated and found that it achieved an impressive degradation rate of chlorobenzene within 8 h. We believe that our study makes a contribution to the insight into solving the pollution of chlorobenzene.

Co-metabolic degradation of tetrabromobisphenol A by Pseudomonas aeruginosa and its auto-poisoning effect caused during degradation process

Wantang Huang,Hua Yin,Yuanyuan Yu,Guining Lu,Zhi Dang,Zhanghong Chen

In this study, Pseudomonas aeruginosa was applied to degrade tetrabromobisphenol A (TBBPA) with glucose as a co-metabolic substrate. Influencing factors of co-metabolic degradation such as pH, TBBPA and glucose concentration were examined and the degradation efficiency under optimal condition reached about 50% on the 7th day. The study also proved that the extracellular action, rather than intracellular one, played a leading role in TBBPA degradation. Five metabolites including debromination and beta-scission products were identified in this study. The extracellular active substance pyocyanin was considered as the origin of H 2 O 2 and OH·. The variation of concentrations of H 2 O 2 and OH· shared the same trend, they increased in the early days and then declined gradually. On the 1st day, the OD600 of P.aeruginosa in the co-metabolic group was 6.0 times higher than the initial value while total organic carbon (TOC) decreased about 78%, which might lead to the occurrence of pyocyanin auto-poisoning. Flow cytometry was applied to detect the cellular state of P.aeruginosa during degradation . The increasing intracellular ROS showed that cells were suffering from oxidative stress and the change of membrane potential revealed that cellular dysfunction had occurred since the 1st day. This research indicated that the toxic effect on P.aeruginosa was probably not directly correlated with TBBPA, but was caused by pyocyanin auto-poisoning.

Anaerobic co-metabolic degradation of ceftriaxone sodium: Performance and mechanism

Haoran Chen,Zongping Wang,Yuxin Huang,Junchi Wei,Gang Guo,Lei Miao

Ceftriaxone sodium (CTX) is a typical β-lactam antibiotic in pharmaceutical wastewater. Although traditional biological treatments are low-cost and eco-friendly, antibiotics usually inhibit microorganisms , resulting in limited treatment performance. The biological co-metabolism process provides carbon sources and energy for microorganisms through co-substrate addition, thereby producing enzymes or cofactors to enhance the degradation of refractory pollutants. Therefore, the present study aimed to investigate the degradation performance of CTX through anaerobic biological co-metabolism. A simple co-metabolic substrate (i.e., glucose, 80 mg L −1 ) was added into an anaerobic-activated sludge system, and its mechanism was explored in terms of the variations in extracellular polymeric substances , microbial structure, and resistance genes. Additionally, the possible degradation pathways of CTX were described. The results showed that the degradation efficiency of CTX (200 mg L −1 ) was only about 35% within 8 h in the control group, while it increased to 67.6% in the experimental group with glucose. Chloroflexi , Actinobacteria , Bacteroidetes ADurb.Bin123, and Bacteroidales were the predominant bacterial species in the system. Additionally, the types of enzymes and the relative abundance of β-lactam resistance genes in the co-metabolic system significantly increased by 7 and 0.77%, respectively. Furthermore, nine primary anaerobic CTX biodegradation products were detected, indicating the existence of three possible degradation pathways. Overall, the study results provide a novel approach to treating CTX-containing wastewater.

Effects of excessive Platycodon grandiflorus root on gut microbiota and host co-metabolism in mice

Shasha Han,Zichen Luo,Shihang Bao,Zihan Xiao,Weichen Xu,Tong Xie,Chen Shi,Jin Wang,Jinjun Shan

Ethnopharmacological relevance Platycodon grandiflorus root, is a widely used herb in East Asia for treating respiratory diseases, but research on its oral safety is limited. Aim of the study This study examines the potential adverse gastrointestinal reactions resulting from excessive consumption of Platycodon grandiflorus root (PR) and its effects on gut microbiota and host co-metabolism. Materials and methods This study evaluated the effects of different doses (1.5, 4.5, and 7.5 g/kg/day) of PR on ICR mice through gavage. Select the 7.5 g/kg/day dosage group and the control group to assess intestinal morphology and conduct histopathological studies. Examine inflammation-related factors and tight junction proteins using WB, qPCR, and ELISA. Additionally, perform 16S rDNA sequencing and metabolomic analyses to evaluate changes in gut microbiota and endogenous metabolites. Finally, the clearance of gut microbiota with antibiotics, the effects of excessive PR on mice were investigated. Results Excessive intake of PR can lead to mortality in mice, as well as symptoms such as intestinal flatulence and slowed intestinal transit, suggesting the occurrence of chronic intestinal pseudo-obstruction accompanied by endotoxemia. It altered both α-diversity and β-diversity in the gut microbiota of mice, with increased relative abundances of Pseudomonadota, Verrucomicrobiota, Escherichia-Shigella , Akkermansia , Bacteroides , and Klebsiella , closely linked to intestinal obstruction and bacterial overgrowth. Excessive intake of PR also resulted in metabolic disturbances in mice, particularly in the levels of metabolites such as bate-hydroxybutyrate, 5,6-dihydrouracil, uridine, isoleucine, mannitol, bate-alanine, L-cysteine, L-tyrosine, and orotic acid, which may provide insights into the side effects associated with excessive consumption of PR. Clearing the gut microbiota significantly mitigated adverse effects on the intestines and restored metabolite levels. Conclusions This study demonstrates that excessive PR induces gut microbiota and metabolic disruption in normal mice, with the overgrowth of Gram-negative bacteria releasing LPS that impair smooth muscle contraction, leading to adverse effects such as chronic intestinal pseudo-obstruction.

Naphthalene Enhances Polycyclic Aromatic Hydrocarbon Biodegradation by Pseudomonas aeruginosa in Soil and Water: Effect and Mechanism

Bo Li,Hulong Liu,Xiaona Liu,Li Han,Jing Yang,Lingke Kang,Liuyuan Tang,Tianwei Qian

Bioremediation is a promising technique owing to its effectiveness, low cost, and environmental friendliness. Previous studies have focused on the degradation efficiency of polycyclic aromatic hydrocarbons (PAHs) in soil and water. However, the expression of PAH-catabolic genes in organisms involved in the degradation process has been rarely and unsystematically reported. In this study, a PAH-degrading strain—Pseudomonas aeruginosa(PQ249631)—was successfully isolated from coking-contaminated soil and used for PAH degradation in soil and water. Furthermore, the degradation of PAHs (naphthalene, fluorene, phenanthrene, anthracene, and pyrene) was investigated in single, binary, and mixture systems to explore the interaction of substrates. The results showed that when naphthalene was used as a cometabolite carbon source, the removal rates of fluorene, phenanthrene, anthracene, and pyrene increased from 14.33%, 17.25%, 6.61%, and 4.47% to 72.08%, 100.00%, 15.63%, and 6.63%, respectively. In a PAH mixture, the degradation rate of each PAH was higher when naphthalene, rather than glucose, was used as the cometabolite carbon source. Transcriptome analysis revealed significant differential expression of PAH-catabolic genes and ATP-binding cassette transporter-related genes under naphthalene stress. The enhanced degradation of PAHs could be attributed to the augmentation of the PAH metabolic pathway and membrane transportation, facilitating the transfer of PAHs to bacteria. These findings underscore the effectiveness ofP. aeruginosaas a PAH degrader and provide molecular insights into enhancing PAH degradation.

Cytochrome P450-mediated co-metabolism of fluoroquinolones by Haematococcus lacustris for simultaneously promoting astaxanthin and lipid accumulation

Xiang Wang,Zhong-Hong Zhang,Kuan-Kuan Yuan,Hui-Ying Xu,Guo-Hui He,Libin Yang,Joseph Buhagiar,Wei-Dong Yang,Yalei Zhang,Carol Sze Ki Lin,Hong-Ye Li

Microalgae-based antibiotic removal treatment has attracted attention because of its low carbon and sustainable advantages. The microalgal co-metabolism system with a suitable carbon source leads to enhanced performance of pollutant removal. However, currently, limited knowledge is available for the removal of fluoroquinolone using a microalgae-mediated co-metabolism system. In this study, we first investigated that the biotic processes by alga Haematococcus lacustris in the co-metabolism system by adding glycerol would be the main contributors responsible for the removal of 10 mg/L ofloxacin (OFL) with the efficiency of 79.73% and the removal of 10 mg/L enrofloxacin (ENR) with the efficiency of 54.10%, respectively. Furthermore, we found that pyruvate from glycerol was converted into substrates and precursors, thereby resulting in the significant accumulations of microalgal astaxanthin and lipid. The astaxanthin content of H. lacustris was achieved at 4.81% and 4.69% treated with OFL and ENR in the presence of glycerol, with 16.04% and 14.55% of lipid content, respectively. The proposed metabolites and pathways were identified to plausibly explain the biodegradation of fluoroquinolone by H. lacustris . The molecular analyses demonstrated that cytochrome P450 (CYP450) enzymes are responsible for the biodegradation of fluoroquinolone, and it was further verified that fluoroquinolones might insert into CYP450 to finally form an efficient and tight binding conformation by molecular dynamic simulation. These findings provide a microalgae-based route for feasible and sustainable biodegradation of antibiotics using a co-metabolism strategy comprising glycerol as a carbon source, with the synergistic accumulation of valuable products.

Advantages of residual phenol in coal chemical wastewater as a co-metabolic substrate for naphthalene degradation by microbial electrolysis cell

Peng Ding,Ping Wu,Qihao Cao,Hongbo Liu,Chongjun Chen,Min-Hua Cui,He Liu

The use of co -metabolic substrates is effective for polycyclic aromatic hydrocarbons (PAHs) removal, but the potential of the high phenol concentrations in coal chemical wastewater (CCW) as a co-metabolic substrate in microbial electrolysis cell (MEC) has been neglected. In this study, the efficacy of varying phenol concentrations in comparison to simple substrates for degrading naphthalene in MEC under comparable COD has been explored. Results showed that phenol as a co-metabolic substrate outperformed sodium acetate and glucose in facilitating naphthalene degradation efficiency at 50 mg-COD/L. The naphthalene removal efficiency from R P , R A , and R G was found to be 84.11 ± 0.44 %, 73.80 ± 0.27 % and 72.43 ± 0.34 %, respectively. Similarly, phenol not only enhanced microbial biomass more effectively, but also exhibited optimal COD metabolism capacity. The addition of phenol resulted in a stepwise reduction in the molecular weight of naphthalene, whereas sodium acetate and glucose led to more diverse degradation pathways. Some bacteria with the potential ability to degrade PAHs were detected in phenol-added MEC, including Alicycliphilus , Azospira , Stenotrophomonas , Pseudomonas , and Sedimentibacter . Besides, phenol enhanced the expression of ncrA and nmsA genes, leading to more efficient degradation of naphthalene, with ncrA responsible for mediating the reduction of the benzene ring in naphthalene and nmsA closely associated with the decarboxylation of naphthalene. This study provides guidance for the effective co-degradation of PAHs in CCW with MEC, demonstrating the effectiveness of using phenol as a co-substrate relative to simple substrates in the removal of naphthalene.

Efficient anaerobic bioremediation of high-concentration benzo[a]pyrene in marine environments

Qingxue Leng,Jun Mu,Guangfeng Yang

Benzo[a]pyrene (BaP), a persistent organic pollutant that may accumulate in sea sediments after oil spill or BaP chemical leakage accidents, considerably harms marine ecosystems and human health. Previous studies have been predominantly focused on its degradation at low concentrations, while the remediation of BaP pollution with high concentrations was neglected. Additionally, the metabolic pathways associated with its anaerobic degradation remain unclear. As a first attempt, super-efficient systems for BaP anaerobic degradation were established, and the corresponding metabolic pathways were elucidated in this study. The results showed that the BaP removal rate in BaP-only system with initial concentrations of 200 mg/L reached 3.09 mg/(L·d) within 45 days. Co-solvent, acetone promoted anaerobic BaP degradation (4.252 mg/(L·d)), while dichloromethane showed a newly-discovered co-metabolic effect. In the system with 500 mg/L of BaP and dichloromethane addition, the removal rate increased drastically (14.64 mg/(L·d)) at 400 mg/L turn point of BaP. Additionally, the corresponding microbial community-level metabolic network was firstly proposed.
