The co-fermentation of Actinomucor elegans and Monascus purpureus for the production of meat flavor with soybean protein

Xuelian Yang,Rui Zhang,Zichun Qin,Jintao Yao,Tianxin Zhang

This study aimed to prepare meat flavor essence through Monascus purpureus and Actinomucor elegans co-fermentation. The optimal fermentation conditions were determined based on the changes in physicochemical properties during the fermentation process, combined with sensory evaluation. Subsequently, the flavor components of the meat flavor essence were analyzed through solid phase microextraction after undergoing the Maillard reaction from the fermentation broth. Gas chromatography–mass spectrometry analysis identified 62 volatile compounds, among which 25 aroma-active substances were detected by aroma extraction dilution analysis/gas chromatography–olfactometry. By calculating the odor activity values (OAVs), seven aroma-active compounds with OAVs > 1 were identified as the most significant contributors to the aroma. Through aroma reconstitution and omission experiments, the accuracy of these key flavor compounds was verified, elucidating each substance's contribution to the essence's overall aroma. This study serves as a foundation for future investigations into the correlation between the flavor of fermented flavorings and their precursor substances, optimizing the mixed-culture fermentation process and enhancing the quality of meat flavor essences.

Co-fermentation of a sugar mixture for microbial lipid production in a two-stage fermentation mode under non-sterile conditions

Wujun Liu,Weifeng Mao,Cuili Zhang,Xi Lu,Xujie Xiao,Zinan Zhao,Jintao Lin

The economical production of biodiesel from lignocellulose requires an effective co-fermentation of lignocellulose-derived sugars, such as glucose and xylose. Our present work discovered that Lipomyces starkeyi AS 2.1560 could produce microbial lipid effectively from co-fermentation of glucose and xylose in a two-stage culture mode under non-sterile conditions. High lipid production was achieved by a feed-batch fermentation process without sterility in the second fermentation stage. After incubation for 46 h in a bioreactor utilizing unsterile glucose and xylose solution, the cell biomass, lipid and lipid content reached 98.3 g L−1, 62.7 g L−1 and 63.8%, respectively. The results showed that the two-stage fermentation method should be a promising process to produce lipid from lignocellulose hydrolysates.

Optimization of fungi co-fermentation for improving anthraquinone contents and antioxidant activity using artificial neural networks

Jiao Yang,Yun Huang,Haiyu Xu,Dongyu Gu,Fa Xu,Jintian Tang,Chen Fang,Yi Yang

The fermentation products of edible fungi are rich in anthraquinones and have a variety of activities, including the antioxidant activity. Because of the large number of combinations, it is very difficult to obtain the optimal multi-strains co-fermentation to improve the yield of anthraquinone. In the present study, an intelligent model based on artificial neural networks (ANNs) using backpropagation (BP) and radial basis function (RBF) algorithms was developed and validated to predict the anthraquinone contents in 136 two fungi and 680 three fungi co-fermented products. After experimental validation of the anthraquinone contents, the mean absolute error and the mean bias error of the results from RBF ANN were lower than those from BP ANN. The results indicated that the anthraquinone contents in A. bisporus , C. comatus and H. erinaceus co-fermentation product was the highest (2.11%). Furthermore, this co-fermentation product showed strong antioxidant activity.

Assessing microbial systems and process configurations for improved ethanol production from sugary stovers by integrating soluble sugars and holocellulose

Hossain M. Zabed,Suely Akter,Yuehui Tian,Mudasir A. Dar,Junhua Yun,Mei Zhao,Arthur J. Ragauskas,Jia Li,Xianghui Qi

This study evaluated microbial systems and technological approaches to configure the whole slurry co-fermentation process for ethanol biosynthesis from a novel stover system rich in sugars. Two approaches, namely separate and simultaneous hydrolysis and co-fermentation (SHCF and SSCF, respectively), were investigated using Escherichia coli monoculture and E. coli- yeast coculture. The SSCF with E. coli monoculture produced 32.75 g/L ethanol, representing only 44.87% yield, which left 65.13 g/L of total sugars unconverted and exhibited limited xylose consumption. Subsequently, a coculture-based SHCF significantly enhanced sugar consumption, leading to increase in ethanol yield and concentration to 66.94% and 48.86 g/L, respectively. Nevertheless, xylose utilization remained minimal due to the preference for glucose and the inhibitory effects of certain compounds. Thereafter, modification of the medium composition by supplementing betaine and sodium metabisulfite improved ethanol production to 53.18 g/L by reducing the toxic effects of inhibitors. Finally, a dual-phase SSCF (DP-SSCF) was explored by allowing the consumption of sugars from the pretreated slurry in the first phase, followed by concurrent cellulose hydrolysis and utilization of the resulting glucose in the second phase. This strategy increased ethanol titer to 63.14 g/L, with 84.3% yield and 0.88 g/L/h productivity.

Major Active Metabolite Characteristics of Dendrobium officinale Rice Wine Fermented by Saccharomyces cerevisiae and Wickerhamomyces anomalus Cofermentation

Li Yao,Xueqin Shi,Hang Chen,Lin Zhang,Lanyan Cen,Lian Li,Yiyi Lv,Shuyi Qiu,Xiangyong Zeng,Chaoyang Wei

Rice, supplemented withDendrobium officinale, was subjected to cofermentation usingSaccharomyces cerevisiaeFBKL2.8022 (Sc) andWickerhamomyces anomalusFBKL2.8023 (Wa). The alcohol content was determined with a biosensor, total sugars with the phenol–sulfuric acid method, reducing sugars with the DNS method, total acids and total phenols with the colorimetric method, and metabolites were analyzed using LC-MS/MS combined with multivariate statistics, while metabolic pathways were constructed using metaboAnalyst 5.0. It was found that the quality of rice wine was higher with the addition ofD. officinale. A total of 127 major active substances, mainly phenols, flavonoids, terpenoids, alkaloids, and phenylpropanoids, were identified. Among them, 26 substances might have been mainly metabolized by the mixed-yeasts fermentation itself, and 10 substances might have originated either fromD. officinaleitself or from microbial metabolism on the newly supplemented substrate. In addition, significant differences in metabolite could be attributed to amino acid metabolic pathways, such as phenylalanine metabolism and alanine, aspartate, and glutamate metabolism. The characteristic microbial metabolism ofD. officinaleproduces metabolites, which are α-dihydroartemisinin, alantolactone, neohesperidin dihydrochalcone, and occidentoside. This study showed that mixed-yeasts cofermentation and fermentation withD. officinaleboth could increase the content of active substances in rice wine and significantly improve the quality of rice wine. The results of this study provide a reference for the mixed fermentation of brewer’s yeast and non-yeast yeasts in rice wine brewing. Keywords:Dendrobium officinale;cofermentation;Saccharomyces cerevisiae;Wickerhamomyces anomalus;metabolites

Industrial cellulase performance in the simultaneous saccharification and co-fermentation (SSCF) of corn stover for high-titer ethanol production

Zhang Qiang,Bao Jie

Cellulase enzymes contribute to the largest portion of operation cost on production of cellulosic ethanol. The industrial cellulases available on the industrial enzyme market from different makers and sources vary significantly in hydrolysis and ethanol, and finally lead to the changes of enzyme cost. Therefore, the selection of the proper industrial cellulase enzymes for commercial-scale production of cellulosic ethanol is crucially important in terms of high performance and cost reduction. In this study, three major cellulase enzyme products available on the Chinese industrial enzyme market were selected and evaluated as the biocatalysts for the biorefining process of lignocellulose biomass into high-titer ethanol. The cellulase enzymes included Cellic CTec 2.0 from Novozymes (Beijing), and LLC 4 from Vland (Qingdao), as well as # 7 from an industrial enzyme maker. The detailed assays on the filter paper activity, the cellobiase activity, and the total protein contents of the enzymes were conducted according to the standard protocols. When the cellulase enzymes were applied to the practical hydrolysis and ethanol-fermentation operation under the conditions of high solids loading and low range of cellulase dosage, the hydrolysis yield shows the significant difference, and the difference was narrowed in the final ethanol yield. The commercially available cellulase enzymes showed different performances in the activities, the cellulose hydrolysis yield, and the ethanol fermentation yields based on the protein dosage per gram of cellulose of corn stover. In general, the industrial cellulase products give satisfactory performance and can be applied for the practical cellulosic ethanol production on commercial scale.

Enhancing efficiency in ethanol production from high solid corn stover: Insights into enzymatic hydrolysis parameters and cellulase recovery

Xiwen Jia,Dong Liu,Hao Lin,Hui Zhang,Xueyan Liang,Kaili Ding,Guanya Ji,Lujia Han,Weihua Xiao

Background The simultaneous saccharification and co-fermentation (SSCF) at high solid loading is an effective approach to improve the economic feasibility of bioethanol. Methods In this study, the high-solid SSCF of superfine grinding corn stover was treated with cheap and readily available deionized water (DW) instead of sodium citrate buffer solution (SCBS). Additionally, the evaluation of temperature, enzyme loading, solid loading, and cellulose recovery were carried out to determine their impact on ethanol production. Significant Findings The results of the analysis indicated that replacing SCBS with DW had negligible effects on the SSCF of ethanol and byproducts (acetic acid and glycerol). The ethanol yield increased by 18 % at 35 °C compared to the yield at 30 °C. As enzyme loading increased, more cellulose active sites in corn stover were bound by cellulase , which increased the ethanol yield by improving the efficiency of enzymatic hydrolysis. The SSCF process didn't incorporate any buffer, resulting in an even higher ethanol titer (55.42 g/L). Furthermore, the cellulase productivity increased by 71 % after recovery, compared to the non-recovery. The results indicated that the optimized SSCF process is a potential technique for producing bioethanol on a commercial level because it improves the production efficiency while keeping costs low.

Dry biorefining maximizes the potentials of simultaneous saccharification and co-fermentation for cellulosic ethanol production

Gang Liu,Qiang Zhang,Hongxing Li,Abdul S. Qureshi,Jian Zhang,Xiaoming Bao,Jie Bao

Despite the well-recognized merits of simultaneous saccharification and co-fermentation (SSCF) on relieving sugar product inhibition on cellulase activity, a practical concomitance difficulty of xylose with inhibitors in the pretreated lignocellulose feedstock prohibits the essential application of SSCF for cellulosic ethanol fermentation. To maximize the SSCF potentials for cellulosic ethanol production, a dry biorefining approach was proposed starting from dry acid pretreatment, disk milling, and biodetoxification of lignocellulose feedstock. The successful SSCF of the inhibitor free and xylose conserved lignocellulose feedstock after dry biorefining reached a record high ethanol titer at moderate cellulase usage and minimum wastewater generation. For wheat straw, 101.4 g/L of ethanol (equivalent to 12.8% in volumetric percentage) was produced with the overall yield of 74.8% from cellulose and xylose, in which the xylose conversion was 73.9%, at the moderate cellulase usage of 15 mg protein per gram cellulose. For corn stover, 85.1 g/L of ethanol (equivalent to 10.8% in volumetric percentage) is produced with the overall conversion of 84.7% from cellulose and xylose, in which the xylose conversion was 87.7%, at the minimum cellulase usage of 10 mg protein per gram cellulose. Most significantly, the SSCF operation achieved the high conversion efficiency by generating the minimum amount of wastewater. Both the fermentation efficiency and the wastewater generation in the current dry biorefining for cellulosic ethanol production are very close to that of corn ethanol production, indicating that the technical gap between cellulosic ethanol and corn ethanol has been gradually filled by the advancing biorefining technology.

Potassium ferrate pretreatment enhances short-chain fatty acid production and phosphorus recovery in co-fermentation system of waste activated sludge and corn stover

Xintian Yu,Wei Zeng,Jiayu Zhang,Sijia Lu,Yongzhen Peng

Developing efficient technologies to recover carbon (C) and phosphorus (P) from waste activated sludge (WAS) is crucial for promoting low-carbon development and addressing P resource shortages. This study proposes a potassium ferrate (PF) pretreatment method to enhance the co-fermentation of WAS and corn straw (CS), facilitating the efficient and stable recovery of short-chain fatty acids (SCFAs) and vivianite, while exploring the underlying mechanisms and potential applications of this approach. PF pretreatment effectively promoted WAS hydrolysis and increased CS bioavailability. SCFAs yield in the co-fermentation group reached 6410.4 mg/L, representing a 79.6 % increase. Efficient SCFAs production was achieved under a SRT of 4–5 days. In the system, SCFAs-consuming bacteria were reduced and SCFAs-producing fermentative genera, such as Paraclostridium , Terrisporobacter and Mobilitalea , were enriched. Additionally, the presence of Fe promoted the enrichment of the Clostridium genus, which is capable of both fermentation and Fe reduction. PF pretreatment enhanced the release of organic matter, thereby shortening the fermentation time. P recovery was effectively achieved through chemical acidification and precipitation techniques. Under optimal recovery conditions (Fe/P = 1.5, pH = 7, 300 rpm), the P recovery efficiency reached 81.6 %, with vivianite accounting for 68.2 %–71.8 %. This method provides a theoretical foundation and technical support for the effective utilization of WAS

Improving natto quality through Co-fermentation of functionally complementary Bacillus and Lactiplantibacillus species

Xiaoxu Ren,Xiaofu Wang,Xinping Lin,Sufang Zhang,Chaofan Ji

Natto, a nutritious fermented soybean product, is known for its health benefits. Modern production methods use Bacillus subtilis as the starter culture to ensure consistent quality. However, this method overlooks the potential positive effects of other microbial species present in traditionally, naturally fermented natto. This study explored the effects of ARTP-induced mutants of B. subtilis var. natto and several strains of lactic acid bacteria (LAB) on natto quality. The results showed that these two types of bacteria have complementary roles in fermentation. B. subtilis var. natto , with enhanced extracellular protease activity, promotes the hydrolysis of soy protein and enhances the product's ACE inhibitory and fibrinolytic activities. Meanwhile, two strains of Lactiplantibacillus plantarum reduced the content of total volatile basic nitrogen (TVB-N) while increasing the content of folic acid in the final product. In addition, B. subtilis var. natto and LAB produce different profiles of volatile compounds (VOCs). Subsequently, B. subtilis var. natto and Lpb. plantarum were used to co-ferment natto in different ratios. The results showed that the two types of bacteria could co-colonize and enhance fermentation quality through their respective strengths. Specifically, Lpb. plantarum strains co-metabolized with B. subtilis var. natto to elevate the content of pyrazines, characteristic VOCs in natto. The optimal quality of natto was achieved when Lpb. plantarum HIS6, Lpb. plantarum LP3 and B. subtilis var. natto Y15-11 were combined in a 2:1:2 ratio. This study not only identified effective combinations of starter cultures with complementary benefits but also elucidated the metabolic pathways of key volatile compounds. Our findings also provide technical insights for improving natto quality, enhancing consumer acceptance, and supporting industrial production.

A potentially functional yogurt co-fermentation with Gnaphalium affine

Hao-Xiang Gao,Zhi-Long Yu,Qiang He,Shan-Hu Tang,Wei-Cai Zeng

A potentially functional yogurt co-fermentation with Gnaphalium affine was prepared and its quality and antioxidant properties were investigated. According to the classic procedure, a novel yogurt was successfully prepared with Gnaphalium affine extract (EGA). During the storage period, the pH value and titratable acidity of yogurt showed no obvious changes compared with control. However, the texture properties of yogurt, such as firmness, adhesiveness, cohesiveness and springiness, could be effectively improved. Moreover, the yogurt possessed the lower syneresis susceptibility and higher viability of Lactobacillus bulgaricus and Streptococcus thermophilus than the control. Furthermore, the yogurt showed remarkable antioxidant potential to scavenge 2, 2′-azinobis-3-ethylbenzthiazoline-6-sulphonate (ABTS), 1,1-diphenyl-2-picrylhydrazyl (DPPH), superoxide and hydroxyl radicals. Meanwhile, the yogurt could enhance the activities of antioxidant enzymes, including superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px), and it could also lead to a significant decrease of malondialdehyde (MDA) level in animal serums. The current results suggest that the yogurt co-fermentation with EGA possesses high quality properties and remarkable antioxidant potential, and thus has the great potential to be used as a novel functional food product in food industry.

Methane production performances of different compositions in lignocellulosic biomass through anaerobic digestion

Shuaishuai Ma,Hongliang Wang,Jingxue Li,Yu Fu,Wanbin Zhu

Lignocellulosic biomass holds great potential in the production of renewable energy such as biomethane via anaerobic digestion . However, the digestion performance of the three main compositions of lignocellulose during biogas production is not well understood so far, and this has greatly hindered the efficient use of biomass by anaerobic fermentation. In this study, the anaerobic digestion performances of glucose, cellulose , hemicellulose , lignin, and their combinations in biomethane production were investigated. The biomethane production potential of cellulose was higher than that of hemicellulose and lignin, and high crystallinity cellulose shows adverse impact on methane production. Hemicellulose had a fast hydrolysis rate and was easier to be digested than cellulose, but it also easily led to excessive acids accumulation. Lignin showed a very limited potential in biomethane production. The co-fermentation of cellulose and hemicellulose could promote cellulose conversion and eliminate the over-acidification of hemicellulose, resulting in efficient and stable biogas production . The co-fermentation of lignin with cellulose and hemicellulose showed an inhibition in biomethane production, and lignin with different structure and sulfur content had different degree of inhibition. This study provides insights into the mechanisms of anaerobic fermentation of lignocellulosic biomass, and paves the way for the better use of biomass.
