Co-culture of Cronobacter sakazakii and Staphylococcus aureus: Explore the influence of mixed biofilm formation and regulation of Cronobacter sakazakii biofilm formation genes

Danliangmin Song,Ai Jia,Xuehe Qi,Kai Dong,Shiyu Liu,Chaoxin Man,Xinyan Yang,Yujun Jiang

Bacterial biofilm is a protective matrix composed of metabolites secreted by bacteria that envelop bacteria. By forming a biofilm, bacteria can considerably improve their environmental tolerance. In food-related processing environment, different types of microorganisms are often present in biofilms. The main contaminating strain in the powdered infant formula (PIF) processing environment, Cronobacter sakazakii and Staphylococcus aureus continues to pollute the PIF processing environment after biofilm production. This study selected Cronobacter sakazakii with a weak biofilm-forming ability as one of the test organisms. The coexistence of Cronobacter sakazakii and Staphylococcus aureus on the surface of production equipment was simulated to analyze the interaction. Biofilm formation in the co-culture group was significantly higher than the others. In-depth study of the effect of Staphylococcus aureus on the biofilm formation genes of Cronobacter sakazakii . Results show two bacteria can coexist on the surface of a metal device, forming a more compact hybrid biofilm structure. Under co-culture conditions, S. aureus increased bcsA and fliD expression in Cronobacter sakazakii , whereas decreased bcsC expression. Signaling molecules produced by Staphylococcus aureus (Autoinducer 2) significantly promoted the biofilm formation of Cronobacter sakazakii at the concentration of 0–500 ng/mL (0.099–0.177) and up-regulated the expression of bcsA , filD and flhD genes.

Weissella confusa XG-3 EPS: production intensification by co-culturing with Candida shehatae and partial characterization

Shuhan Yang,Shouqi Zhao,Yingying Ning,Dan Zhao

Previous studies have shown that co-culture methods can enhance exopolysaccharide production, no studies have reported this enhancement in co-cultures between Weissella confusa and Candida shehatae. This study developed an efficient co-culture system combining W. confusa XG-3 and C. shehatae (1:1 ratio), and employed response surface methodology to optimize both exopolysaccharide yield and properties. The initial co-culture produced 56.82 ± 0.12 g/L exopolysaccharide, a 1.63-fold increase over single culture. Optimization (sucrose: 105.97 g/L; yeast extract: 5.81 g/L; peptone: 6.24 g/L) further increased the yield to 115.66 g/L, a 2.04-fold improvement. The co-cultured exopolysaccharide had high water solubility (60.45 ± 0.70%) and water holding capacity (614.74 ± 9.26%). The co-cultured EPS showed significant antioxidant activity. At 5 mg/mL, the scavenging rates of ABTS, DPPH, hydroxyl, superoxide, H2O2 and total reducing power reached the highest 52.22 ± 0.47%, 50.79 ± 2.86%, 55.80 ± 2.56%, 36.05 ± 2.07%, 66.85 ± 0.30%, and 0.39 ± 0.01. It also had a superior effect on the proliferation of several probiotics compared with commercial prebiotics and exhibited high thermostability with a degradation threshold at 292.5 °C. These results demonstrate a novel, scalable strategy for producing multifunctional EPS with broad applications in the food industry.

Enhanced co-extraction of glycyrrhizic acid and glabridin from Glycyrrhiza glabra L. through enzyme-focused techniques

Mei-yuan Li,Zhi-peng Jia,De-min Li,Man-ping Luo,Ying-ying Meng,Kang- xu Wang,Bei Wang,Bing Liu,Xin-guo Zhang,Qin Li

Glycyrrhizic acid (GA) and glabridin (GD) are very important sweeteners and whitening agents present in Glycyrrhiza glabra L. This study investigates the cellulase and pectinase produced by endophytes in G. glabra to optimize and enhance the extraction of GA and GD from licorice using enzyme-assisted extraction (EAE) and microbial co-culture-assisted extraction (MCAE) approaches. Twelve strains with cellulase and pectinase activities were isolated from G. glabra. Notably, the strain G1–5, preliminarily identified as Penicillium sp., demonstrated superior performance in enzyme production and extraction efficiency of GA. Compared to the traditional extraction methods, the EAE process showed an obvious improvement of 32.21% and 56.37% in GA and GD extraction rates, respectively. Similarly, the MCAE process with the enzyme-producing strain G1–5 exhibited an increase of 25.52% and 38.45% in GA and GD extraction rates, respectively. These findings provide valuable guidance and innovative strategies for the extraction of active ingredients.

Generation of a co-culture cell micropattern model to simulate lung cancer bone metastasis for anti-cancer drug evaluation

Huixiang Zhong,Liuyang Xuan,Dandan Wang,Jianhua Zhou,Yan Li,Qing Jiang

Lung cancer bone metastases usually involve multiple interactions between cancer cells and host organs. To mimic such a structure, a cell co-culture micropattern of A549 lung cancer cells and osteoblast cells (A549/OB) was developed using a μ-eraser strategy for anti-cancer drug evaluation and also understanding cell–cell communication. When applying the μ-eraser strategy, a PDMS stamp was pressed to induce cell lysis. Hence, the minimum pressure required to induce cell lysis was first quantified. Then the pressure and pressing time for the micropatterning process were optimized to obtain cell micropatterns of high fidelity and repeatability. For different types of cells (A549 and human mesenchymal stem cells) and different substrates (TCP and PLGA nanofiber sheets), the optimized pressure was also different (i.e., for A549 on TCP, a pressure of 24.5 kPa was pressed for 10 s). According to a live/dead assay and Alamar Blue assay, cell viability and proliferation potential were not affected by the micropatterning process. There were several advantages for the μ-eraser strategy: substrates were not pre-treated; cell micropatterns mainly relied on the pattern of the PDMS stamp along with the micropatterning process; cells in the micropattern were not restricted in specified regions. Thus the A549/OB co-culture micropattern on TCP was used to evaluate the efficacy of a therapeutic agent (doxorubicin). In the co-culture micropattern, the efficacy of doxorubicin decreased when the expression of ALP in OB was elevated. The co-culture micropattern model showed the potential to be used for new anti-cancer drug development.

Solvents production from cassava by co-culture of Clostridium acetobutylicum and Saccharomyces cerevisiae

Gaoxiang Qi,Lian Xiong,Mutan Luo,Qianlin Huang,Chao Huang,Hailong Li,Xuefang Chen,Xinde Chen

Co-culture of Clostridium acetobutylicum and Saccharomyces cerevisiae was used to enhance solvents (acetone-butanol-ethanol, ABE) production from cassava without amylolytic enzymes addition. Under the simultaneous inoculation condition of C. acetobutylicum and S. cerevisiae with the inoculation level of 5% (v/v) for both strains, 21.74   g/L solvents was produced from 75   g/L cassava with the yield of 0.36   g solvents/g glucose. The solvents titer and yield obtained in the co-culture process increased by 14.3% and 9.1%, respectively, compared to those achieved in ABE fermentation. For further improvements in total solvents titer and yield from cassava, ABE fermentation and co-culture process from higher cassava concentration media were conducted. 42.56   g/L total solvents was achieved from 150   g/L cassava by co-culture with the yield of 0.46   g solvents/g glucose. The solvents titer and yield obtained in the co-culture process increased by 141.0% and 43.8%, respectively, compared to those obtained from ABE fermentation. The results showed great potential for the use of co-culture of C. acetobutylicum and S. cerevisiae for solvents production from starchy substrate.

Enhancement of polymyxin B1 production by an artificial microbial consortium of Paenibacillus polymyxa and recombinant Corynebacterium glutamicum producing precursor amino acids

Hui-Zhong Sun,Si-Yu Wei,Qiu-Man Xu,Wei Shang,Qing Li,Jing-Sheng Cheng,Ying-Jin Yuan

Polymyxin B, produced by Paenibacillus polymyxa, is used as the last line of defense clinically. In this study, exogenous mixture of precursor amino acids increased the level and proportion of polymyxin B1 in the total of polymyxin B analogs of P. polymyxa CJX518-AC (PPAC) from 0.15 g/L and 61.8 % to 0.33 g/L and 79.9 %, respectively. The co-culture of strain PPAC and recombinant Corynebacterium glutamicum -leu01, which produces high levels of threonine, leucine, and isoleucine, increased polymyxin B1 production to 0.64 g/L. When strains PPAC and C. glu -leu01 simultaneously inoculated into an optimized medium with 20 g/L peptone, polymyxin B1 production was increased to 0.97 g/L. Furthermore, the polymyxin B1 production in the co-culture of strains PPAC and C. glu -leu01 increased to 2.21 g/L after optimized inoculation ratios and fermentation medium with 60 g/L peptone. This study provides a new strategy to improve polymyxin B1 production.

Assembled/Disassembled Modular Scaffolds for Multicellular Tissue Engineering

Xiaopeng Yu,Hongshi Ma,Yufeng Wang,Jianxin Hao,Lei Chen,Michael Gelinsky,Chengtie Wu

The behavior of tissue resident cells can be influenced by the spatial arrangement of cellular interactions. Therefore, it is of significance to precisely control the spatial organization of various cells within multicellular constructs. It remains challenging to construct a versatile multicellular scaffold with ordered spatial organization of multiple cell types. Herein, a modular multicellular tissue engineering scaffold with ordered spatial distribution of different cell types is constructed by assembling varying cell-laden modules. Interestingly, the modular scaffolds can be disassembled into individual modules to evaluate the specific contribution of each cell type in the system. Through assembling cell-laden modules, the macrophage-mesenchymal stem cell (MSC), endothelial cell-MSC, and chondrocyte-MSC co-culture models are successfully established. The in vitro results indicate that the intercellular cross-talk can promote the proliferation and differentiation of each cell type in the system. Moreover, MSCs in the modular scaffolds may regulate the behavior of chondrocytes through the nuclear factor of activated T-Cells (NFAT) signaling pathway. Furthermore, the modular scaffolds loaded with co-cultured chondrocyte-MSC exhibit enhanced regeneration ability of osteochondral tissue, compared with other groups. Overall, this work offers a promising strategy to construct a multicellular tissue engineering scaffold for the systematic investigation of intercellular cross-talk and complex tissue engineering.

Co-cultivation of microalga and xylanolytic bacterium by a continuous two-step strategy to enhance algal lipid production

Zhangzhang Xie,Weitie Lin,Jianfei Luo

To enhance microalgal lipid production, canonical two-step cultivation strategy that by transferring the microalgal cells grown in nutrient-replete medium to nutrient-depleted medium is widely used. However, the harvesting step during the transfer raises the production cost. To avoid the harvesting step, this study developed a continuous two-step (CTS) cultivation strategy. In the strategy, Chlorella sacchrarophila was grown in bioreactor while a xylanolytic bacterium Cellvibrio pealriver grown in an inner bag that embedded in the bioreactor; after the first-step co-cultivation, the inner bag is removed which then start the second-step cultivation of C. sacchrarophila . Based on the strategy, the lipid production was determined as 825.34–929.79 mg·L −1 , which were 1.7–1.9 times higher than that of cultivation in canonical two-step strategy using glucose as feedstock. During the CTS strategy, the co-cultivation using xylan as feedstock promotes the microalgal growth and the removal of inner bag produces nutrient-depleted condition for enhancing microalgal lipid production.

Discerning promotion mechanisms of fungi Clonostachys rosea on growth of freshwater microalga Chlorella sp. by non-contact culture

Juan Liu,Shiqing Sun,Kangyu Chen,Shumei Gao,Dixing Chen,Yuejin Zhang,Changwei Hu

Fungi-microalgae consortia have great potential in microalgae harvesting and wastewater treatment. However, the symbiotic mechanisms underlying diffusible signaling metabolite production by fungi and microalgae remain unclear. Here, the positive impacts of Clonostachys rosea on Chlorella sp. were investigated using metabolomics and transcriptomics during a 5-day non-contact co-culture experiment. Co-cultured Chlorella sp. had a significantly higher growth rate and better photosynthetic performance than its mono-culture equivalent. The metabolites played important roles in the up-regulation of energy and amino acid metabolism processes in algal cells, whilst transcriptomics showed the up-regulation of genes involved in photosynthesis and signaling molecules. The biosynthesis of tryptophan and indole-3-acetic acid (IAA) was up-regulated in both algal cells and fungi, indicating that IAA is an important diffusible signaling molecule for algal growth regulation. This work provides novel insights into the signaling molecule interactions between fungi and microalgae in fungi-microalgae consortia.

Strategies for enhancing short- and medium-chain fatty acids synthesis from corn stover-based sugar: A new perspective on co-culture

Xiwen Jia,Jiajie Xu,Luoyang Liu,Xueyan Liang,Yinghui He,Lujia Han,Weihua Xiao

In this study, corn stover-based sugar served as the initial carbon source to investigate the impact of different co-culture strategies on the synthesis of short- and medium-chain fatty acids (SMCFAs) through carbon chain elongation (CCE). The results indicated that the sequential inoculation outperformed simultaneous inoculation in the M. elsdenii and Lactobacillus groups. The inclusion of Lactobacillus significantly enhanced substrate utilization, generating lactate that facilitated the integration of odd carbon carboxylates (such as propionate and valerate) into the CCE products. The caproate concentration in the M. elsdenii + L. pentosus group reached 3.4 g/L, which was 1.3 times that in the control group. Furthermore, the introduction of C. kluyveri effectively boosted the synthesis of butyrate and caproate through CCE using ethanol as an electron donor within the co-culture setup of the M. elsdenii+C. kluyveri group. Moreover, the carbon efficiency of the co-culture group increased by 10–21 % compared to the control group, and the cell protein titer was 1.5–2 times higher than that of the control group. This study has established a method for co-producing SMCFAs and cell proteins utilizing bio-sugars through a co-culture approach, offering implications for lignocellulose utilization and potential high-value applications of SMCFAs in the future.

Improved the lipopeptide production of Bacillus amyloliquefaciens HM618 under co-culture with the recombinant Corynebacterium glutamicum producing high-level proline

Xin-Yue Chen,Hui-Zhong Sun,Bin Qiao,Chang-Hao Miao,Zheng-Jie Hou,Shu-Jing Xu,Qiu-Man Xu,Jing-Sheng Cheng

The application of antibacterial lipopeptides is limited by high cost and low yield. Herein, the exogenous L-proline significantly improved lipopeptide production by Bacillus amyloliquefaciens HM618. A recombinant Corynebacterium glutamicum producing high levels of proline using genetically modifying proB and putA was used to establish consortium, to improve lipopeptide production of strain HM618. Compared to a pure culture , the levels of iturin A, fengycin, and surfactin in consortium reached 67.75, 39.32, and 37.25 mg L -1 , respectively, an increase of 3.19-, 2.05-, and 1.63-fold over that produced by co-cultures of B. amyloliquefaciens and recombinant C. glutamicum with normal medium. Commercial amylase and recombinant Pichia pastoris with a heterologous amylase gene were used to hydrolyze kitchen waste. A three-strain consortium with recombinant P. pastoris and C. glutamicum increased the lipopeptide production of strain HM618 in medium containing KW. This work provides new strategies to improve lipopeptide production by B. amyloliquefaciens.
