Hydrogel capsules are widely applied in materials science, pharmaceutical science, and tissue engineering due to their tunable nature, defined core–shell architecture, and proper biocompatibility. Specially, multi-core capsules are highly functional carriers with notable advantages for co-encapsulation of diverse incompatible components without cross-contamination. However, current approaches for generating such capsules are limited by using gas/oil-water-oil systems, leading to poor biocompatibility and functions due to the gas/oil cores. Herein, a simple and new approach is proposed for flexible fabrication of multi-aqueous core hydrogel capsules (MACHC) using microfluidic aqueous two-phase system (µATPS). In this system, a flow-focusing microfluidic device is designed to create a stable laminar flow with two immiscible liquids composed of polyethylene glycol and dextran, which is sheared by oil phase to further effectively produce MACHC. The volume and components of each core can be well tailored by precisely adjusting the distinct flows, while the number of cores is determined by the orifice design of microfluidic chip. The MACHC are successfully used to co-encapsulate heterogeneous hepatic and endothelial cells, indicating good biocompatibility and multi-functionality of the multi-core carriers. The proposed µATPS-based approach is stable and convenient, holding promise for generation of multi-responsive materials, tissue engineering, and co-delivery of diverse drugs.

Combination therapy is a promising treatment for certain advanced drug-resistant cancers. Although effective inhibition of various tumor cells was reported in vitro , combination treatment requires improvement in vivo due to uncontrolled ratiometric delivery. In this study, a tumor-targeting lipodisk nanoparticle formulation was developed for ratiometric loading and the transportation of two hydrophobic model drugs, doxorubicin (DOX) and paclitaxel (PTX), in one single platform. Furthermore, a slightly acidic pH-sensitive peptide (SAPSP) incorporated into lipodisks effectively enhanced the tumor-targeting and cell internalization. The obtained co-loaded lipodisks were approximately 30 nm with a pH-sensitive property. The ratiometric co-delivery of two drugs via lipodisks was confirmed in both the drug-resistant MCF-7/ADR cell line and its parental MCF-7 cell line in vitro , as well as in a tumor-bearing mouse model in vivo compared with a cocktail solution of free drugs. Co-loaded lipodisks exerted improved cytotoxicity to tumor cells in culture, particularly to drug-resistant tumor cells at synergistic drug ratios. In an in vivo xenograft mouse model, the anti-tumor ability of co-loaded lipodisks was evidenced by the remarkable inhibitory effect on tumor growth of either MCF-7 or MCF-7/ADR tumors, which may be attributed to the increased and ratiometric accumulation of both drugs in the tumor tissues. Therefore, tumor-specific lipodisks were crucial for the combination treatment of DOX and PTX to completely exert a synergistic anti-cancer effect. It is concluded that for co-loaded lipodisks, cytotoxicity data in vitro could be used to predict their inhibitory activity in vivo , potentially enhancing the clinical outcome of synergistic therapy.

The co-delivery of curcumin (CUR) with other polyphenol bioactives often has synergistic benefits. However, conventional co-encapsulation carriers formed by precipitation frequently exhibit limited loading properties, particularly for bioactives with different solubilities. In this study, we designed a highly efficient vehicle for the co-delivery of hydrophobic CUR and hydrophilic procyanidins (PCs), adopting a multiple interactions–based strategy via a two-step flash nanoprecipitation (FNP) process. The optimal nanoparticles (NP), composed of a Fe II crosslinked PCs/CUR nanocore (NC) and a soy protein isolates (SPI) shell, showed high individual encapsulation efficiency (∼95%) and a total loading content of ∼38%. Compared with the conventional production process, FNP endowed our NP with a small size (133 nm) and good uniformity (polydispersity index = 0.267). Ultraviolet–visible and Fourier transform infrared spectra revealed multiple interactions (coordination and hydrogen bonding) among the carrier materials and bioactives, which played a central role in achieving excellent loading properties for both PCs and CUR. The core–shell structure of the NP greatly enhanced the thermal–light stability of the encapsulated PCs and CUR. Synergistic antioxidation effect was found between PCs and CUR, and the nano-formulation further increased the antioxidation efficiency. In vitro release experiments showed the favorable pH responsiveness of our NP in the vast gastrointestinal pH gradient effectively preventing the leakage of wrapped bioactives in the stomach pH environment, which benefits the intestinal delivery of the bioactives. Our nanoparticulate co-delivery system fabricated using the FNP process provides new insights into the formulation strategy of multiple bioactives and shows great potential for application in the fields of functional foods and dietary supplements.
