Enhanced sulfonamides removal via microalgae-bacteria consortium via co-substrate supplementation

Yue Wang,Jinghua Li,Yao Lei,Rong Cui,Aiping Liang,Xiaoqiang Li,Yoong Kit Leong,Jo-Shu Chang

Both co-cultivation and co-substrate addition strategies have exhibited massive potential in microalgae-based antibiotic bioremediation. In this study, glucose and sodium acetate were employed as co-substrate in the cultivation of microalgae-bacteria consortium for enhanced sulfadiazine (SDZ) and sulfamethoxazole (SMX) removal. Glucose demonstrated a two-fold increase in biomass production with a maximum specific growth rate of 0.63 ± 0.01 d −1 compared with sodium acetate. The supplementation of co-substrate enhanced the degradation of SDZ significantly up to 703 ± 18% for sodium acetate and 290 ± 22% for glucose, but had almost no effect on SMX. The activities of antioxidant enzymes, including peroxidase, superoxide dismutase and catalase decreased with co-substrate supplementation. Chlorophyll a was associated with protection against sulfonamides and chlorophyll b might contribute to SDZ degradation. The addition of co-substrates influenced bacterial community structure greatly. Glucose enhanced the relative abundance of Proteobacteria , while sodium acetate improved the relative abundance of Bacteroidetes significantly.

Improved Bio-Synthesis of 2,5-bis(hydroxymethyl)furan by Burkholderia contaminans NJPI-15 With Co-substrate

Chang Siyuan,He Xuejun,Li Bingfeng,Pan Xin

Upgrading of biomass derived 5-hydroxymethylfurfural (HMF) has attracted considerable interest recently. A new highly HMF-tolerant strain of Burkholderia contaminans NJPI-15 was isolated in this study, and the biocatalytic reduction of HMF into 2,5-bis(hydroxymethyl)furan (BHMF) using whole cells was reported. Co-substrate was applied to improve the BHMF yield and selectivity of this strain as well as HMF-tolerant level. The catalytic capacity of the cells can be substantially improved by Mn2+ ion. The strain exhibited good catalytic performance at a pH range of 6.0–9.0 and a temperature range of 25°C–35°C. In addition, 100 mM HMF could be reduced to BHMF by the B. contaminans NJPI-15 resting cells in presence of 70 mM glutamine and 30 mM sucrose, with a yield of 95%. In the fed-batch strategy, 656 mM BHMF was obtained within 48 h, giving a yield of 93.7%. The reported utilization of HMF to produce BHMF is a promising industrially sound biocatalytic process.

Co-substrate fermentation of lignocellulose with added glucose and xylose to increase production of poly (β-l-malic acid)

Zhang Yutian,Zeng Dongdong,Li Jianing,Ma Xiaojun,Li Dongna,Li Yachao

Co-substrate fermentation has become an essential approach to achieving high-efficiency and low-cost production of poly (β- l -malic acid) (PMA). Herein, sustainable rubberwood ( Hevea brasiliensis ) hydrolysate (RWH) was co-fermented by A. pullulans to synthesize PMA using glucose and xylose as co-substrate carbon sources. Moreover, vitamin B was explored as nutrient supplementation in this study. Results showed that compared with the single RWH substrate, the PMA production of co-substrate fermentation increased by 38–52%. Furthermore, the addition of VB 6 promoted the utilization of substrates and shortened the fermentation cycle of PMA. During the scale-up fermentation, the PMA yield reached 0.50–0.54 g/g. Compared with traditional glucose culture medium, co-substrate fermentation improved the conversion rate of carbon sources and reduced PMA production cost by 14.9–36.6%. This work provides a new path for high-efficiency and economical industrial fermentation technology of PMA.

Aerobic and Anaerobic Biodegradation of 1,2-Dibromoethane by a Microbial Consortium under Simulated Groundwater Conditions

Qing Wang,Miaoyan Yang,Xin Song,Shiyue Tang,Lei Yu

This study was conducted to explore the potential for 1,2-Dibromoethane (EDB) biodegradation by an acclimated microbial consortium under simulated dynamic groundwater conditions. The enriched EDB-degrading consortium consisted of anaerobic bacteriaDesulfovibrio, facultative anaerobeChromobacterium, and other potential EDB degraders. The results showed that the biodegradation efficiency of EDB was more than 61% at 15 °C, and the EDB biodegradation can be best described by the apparent pseudo-first-order kinetics. EDB biodegradation occurred at a relatively broad range of initial dissolved oxygen (DO) from 1.2 to 5.1 mg/L, indicating that the microbial consortium had a strong ability to adapt. The addition of 40 mg/L of rhamnolipid and 0.3 mM of sodium lactate increased the biodegradation. A two-phase biodegradation scheme was proposed for the EDB biodegradation in this study: an aerobic biodegradation to carbon dioxide and an anaerobic biodegradation via a two-electron transfer pathway of dihaloelimination. To our knowledge, this is the first study that reported EDB biodegradation by an acclimated consortium under both aerobic and anaerobic conditions, a dynamic DO condition often encountered during enhanced biodegradation of EDB in the field.Keywords:1,2-dibromoethane;biodegradation;microbial consortium;co-substrates;rhamnolipid;aerobic and anaerobic conditions

Evaluation of microbial p-chloroaniline degradation in bioelectrochemical reactors in the presence of easily-biodegrading cosubstrates: Degradation efficiency and bacterial community structure

Hao Xu,Xuesong Zhao,Shaobin Huang,Han Li,Na Tong,Xiangyu Wen,Congcong Sun,Saima Fazal,Yongqing Zhang

This study aimed to illustrate p -Chloroaniline ( p -CIA) biodegradation efficiencies in bioelectrochemical reactors under stimulation by a low-voltage electric field (0.2 V versus Ag/AgCl) in the presence of easily-degrading cosubstrates including glucose and acetate. The biodegradation efficiencies of closed-circuit bioreactors were compared with those of open-circuit reactors. Experimental results showed that the six different bioreactors provided different p -CIA biodegradation efficiencies. The highest biodegradation efficiency of 38.5 ± 10.3 mg/l was obtained in a closed-circuit bioreactor with acetate and the lowest biodegradation efficiency of 15.7 ± 9.4 mg/l was obtained in an open-circuit bioreactor. This difference may be attributed to the presence of electrical stimulation and acetate. The results for generated current and biodegradation efficiency indicated that acetate is a better cosubstrate than glucose. High-throughput sequencing technologies were used to characterise the bacterial community structure of the six bioreactors and revealed that different bacterial communities resulted in different treatment efficiencies.
