Rapid on-site diagnosis of PEDV and PoRV co-infection by gold magnetic nanoparticles-based SERS immunochromatography

Zhaoyuan He,Xiaoge Duan,Zhi Zhao,Yingkai Chen,Chengxiang Fu,Fuxin Zhang,Jinzi Wang,Jianyuan Feng,Nanxin Lin,Hailan Chen

Porcine epidemic diarrhea virus (PEDV) and porcine rotavirus (PoRV) are the two main pathogens causing porcine diarrhea, which are characterized by high morbidity and mortality. Most of the diagnostic methods available are limited to the laboratory or fail to highlight their advantages in terms of target species, detection time, sensitivity, and stability. To meet the demand for rapid on-site diagnosis of PEDV and PoRV co-infection, a surface-enhanced Raman scattering (SERS) immunochromatographic sensor based on gold magnetic nanoparticles (MNPs) was developed. The sensor is dual-mode, detecting on the basis of color signals by the naked eye and Raman signals. After a series of optimizations, the constructed sensor could perform simultaneous qualitative and quantitative detection of PEDV and PoRV in just 18 min, with visualized (color signals observed by the naked eye) limits of detection (LOD) of 3.13 × 10 2 TCID 50 /mL and 4.69 × 10 2 copies/μL, respectively. The LOD based on Raman signal analysis was as low as 4.63 × 10 1 TCID 50 /mL and 3.30 × 10 2 copies/μL for PEDV and PoRV, respectively. In addition, the sensor exhibited excellent specificity without cross-reactivity with common pathogens. The overall compliance rate with RT-PCR was 92.1 % (35/38) for 38 clinical samples. Therefore, the sensor is characterized by high sensitivity, specificity, reproducibility, and accuracy, making it suitable for the simultaneous rapid on-site detection of PEDV and PoRV.

Microfluidic detection based on size-encoded microbeads and rolling circle amplification for multiplex respiratory virus nucleic acid detection

Xingmeng Chen,Yi Wang,Zhun Lin,Huiling Zhang,Longqiang Yang,Fuyan Hong,Jieye Huang,Yuntao Liu,Yuanqing Zhang,Peifeng Ke

Acute respiratory tract diseases (ARDs) are predominantly caused by viral infections, with one of their hallmark characteristics being multiple viral co-infections. Such multiple viral infections not only complicate therapeutic interventions but also lead to an increase in mortality rates. Most traditional bioassays, however, are limited to identifying a single type of virus, leading to missed diagnoses in samples with multiple respiratory pathogens. Consequently, there is an urgent need for the development of a detection method that exhibits high sensitivity, allows for constant-temperature operation, is user-friendly, and is capable of simultaneously detecting multiple viral nucleic acid targets. To address this challenge, we developed a novel method for the simultaneous detection of multiple respiratory viral nucleic acids using size-encoded microbeads and rolling circle amplification. Additionally, we designed a microfluidic chip featuring distinct spacer arrays that separate the microbeads into different regions based on their size, thereby generating characteristic signals indicative of varying biomarker levels. Rolling circle amplification significantly enhances the detection sensitivity on the microbeads, achieving a detection limit as low as fmol L−1. In representative clinical samples, both the sensitivity and specificity for distinguishing target viral RNA from other respiratory diseases reached 100%. Importantly, this strategy enables simultaneous and in situ sensitive analysis of four viral nucleic acids. The throughput of pathogen nucleic acid detection can be further enhanced by incorporating additional microbeads of varying sizes and implementing a corresponding microchip design. We anticipate that this strategy holds substantial promise for clinical diagnosis of multiple respiratory viral infections.

An anti-complement homogeneous polysaccharide from Houttuynia cordata ameliorates acute pneumonia with H1N1 and MRSA coinfection through rectifying Treg/Th17 imbalance in the gut–lung axis and NLRP3 inflammasome activation

Xinxing Li,Wenxin Ding,Yan Lu,Haiyan Zhu,Weilian Bao,Yang Liu,Jiaren Lyu,Lishuang Zhou,Hong Li,Jiyang Li,Daofeng Chen

The coinfection of respiratory viruses and bacteria is a major cause of morbidity and mortality worldwide, despite the development of vaccines and powerful antibiotics. As a macromolecule that is difficult to absorb in the gastrointestinal tract, a homogeneous polysaccharide from Houttuynia cordata (HCPM) has been reported to exhibit anti-complement properties and alleviate influenza A virus (H1N1)-induced lung injury; however, the effects of HCPM without in vitro antiviral and antibacterial activities on more complicated pulmonary diseases resulting from viral-bacterial coinfection remains unclear. This study established a representative coinfection murine pneumonia model infected with H1N1 (0.2 LD 50 ) and methicillin-resistant Staphylococcus aureus (MRSA, 10 7 CFU). HCPM significantly improved survival rate and weight loss, and ameliorated gut–lung damage and inflammatory cytokine production. Interestingly, the therapeutic effect of HCPM on intestinal damage preceded that in the lungs. Mechanistically, HCPM inhibited the overactivation of the intestinal complement (C3a and C5a) and suppressed the activation of the NLR family pyrin domain-containing 3 (NLRP3) pathway, which contributes to the regulation of the Treg/Th17 cell balance in the gut–lung axis. The results indicate the beneficial effects of an anti-complement polysaccharide against viral–bacterial coinfection pneumonia by modulating crosstalk between multiple immune regulatory networks.

Rapid and Quantitative Screening of Bacteria-Virus Co-Infection in Saliva Based on Ready-to-Use Portable Resonant Sensors

Rujing Sun,Xin Li,Min Jiang,Yelongzi Cao,Yaqian Yang,Hao Wen,Zhenghan Shi,Chaobo Dai,Hao Liang,Qingjun Liu

Seasonal outbreaks of respiratory infections place a significant burden on hospital-centered diagnostic and treatment systems. Co-infections involving both bacterial and viral pathogens further complicate infection assessment, highlighting the need for rapid and accurate diagnostic tools. In this work, we present a ready-to-use, portable resonant sensing system capable of rapidly and quantitatively detecting key coinfection biomarkers in saliva, specifically C-reactive protein (CRP) and respiratory syncytial virus (RSV) fusion protein. The sensor operates via competitive immune binding within a hydrogel-based resonant structure, where biomarker binding induces shifts in resonant frequency that are wirelessly detected. To preserve antibody activity and hydrogel mechanical integrity, we developed an integrated, ready-to-use sensor system optimized for ultrasensitive detection. This platform enables analysis of unprocessed saliva samples within 5 min, achieving low limits of detection of 24 fg/mL for CRP and 31 fg/mL for RSV fusion protein. Quantitative analysis of patient samples demonstrated strong concordance with both enzyme-linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR) results. This portable sensing platform offers a promising solution for point-of-care coinfection diagnostics, supporting real-time infection monitoring and potentially easing the burden on healthcare systems.

Antimicrobial and Antiviral Nanofibers Halt Co-Infection Spread via Nuclease-Mimicry and Photocatalysis

Jieran Yao,Zhenhong Luo,Jiaying Lin,Na Meng,Jiangna Guo,Hui Xu,Rongwei Shi,Linhui Zhao,Jiateng Zhou,Feng Yan,Bin Wang,Hailei Mao

The escalating spread of drug-resistant bacteria and viruses is a grave concern for global health. Nucleic acids dominate the drug-resistance and transmission of pathogenic microbes. Here, imidazolium-type poly(ionic liquid)/porphyrin (PIL-P) based electrospun nanofibrous membrane and its cerium (IV) ion complex (PIL-P-Ce) are developed. The obtained PIL-P-Ce membrane exhibits high and stable efficiency in eradicating various microorganisms (bacteria, fungi, and viruses) and decomposing microbial antibiotic resistance genes and viral nucleic acids under light. The nuclease-mimetic and photocatalytic mechanisms of the PIL-P-Ce are elucidated. Co-infection wound models in mice with methicillin-resistant S. aureus and hepatitis B virus demonstrate that PIL-P-Ce integrate the triple effects of cationic polymer, photocatalysis, and nuclease-mimetic activities. As revealed by proteomic analysis, PIL-P-Ce shows minimal phototoxicity to normal tissues. Hence, PIL-P-Ce has potential as a “green” wound dressing to curb the spread of drug-resistant bacteria and viruses in clinical settings.
