Effects of Secondary Metabolites of Rice on Brown Planthopper and Its Symbionts

Ziyuan Deng,Chengling Lai,Jun Zhang,Fan Sun,Danting Li,Peiying Hao,Xuping Shentu,Kun Pang,Xiaoping Yu

The brown planthopperNilaparvata lugens(Stål) (BPH) is a main rice pest in China and many other Asian countries. In the control of BPH, the application of insect-resistant rice has proven to be quite effective. Secondary metabolites are essential weapons in plants’ defense against phytophagous insects. Studies have found that differences in the content of secondary metabolites play a crucial role in determining whether rice exhibits resistance or susceptibility to BPH. Simultaneously, symbionts are essential to the BPH. Nevertheless, there is limited research on the impact of secondary metabolites on the symbionts within BPH. Therefore, investigating the influence of secondary metabolites on both BPH and their symbionts is significant for the control of BPH. In this experiment, newly emerged female adults of BPH were fed artificial diets containing 10 different secondary metabolites. The results indicated that methyl jasmonate had inhibitory effects on the survival rate, weight gain, and reproductive capacity of BPH. Using qPCR methods, it was discovered that the number of symbiotic fungi (Ascomycetessymbionts) within BPH significantly decreased under methyl jasmonate stress. In conclusion, this experiment has preliminarily revealed the inhibitory effects of methyl jasmonate on BPH and its symbionts, demonstrating its potential for controlling BPH.Keywords:brown planthopper;secondary metabolites;symbionts;methyl jasmonate

Enhancing treatment performance of Chlorella pyrenoidosa on levofloxacin wastewater through microalgae-bacteria consortia: Mechanistic insights using the transcriptome

Shasha Zhao,Jin Qian,Bianhe Lu,Sijing Tang,Yuxuan He,Yin Liu,Yitong Yan,Shuai Jin

Microalgae-bacteria consortia (MBC) system has been shown to enhance the efficiency of microalgae in wastewater treatment, yet its effectiveness in treating levofloxacin (LEV) wastewater remains unexplored. This study compared the treatment of LEV wastewater using pure Chlorella pyrenoidosa (PA) and its MBC constructed with activated sludge bacteria. The results showed that MBC improved the removal efficiency of LEV from 3.50–5.41 % to 33.62–57.20 % by enhancing the growth metabolism of microalgae. The MBC increased microalgae biomass and extracellular polymeric substance (EPS) secretion, yet reduced photosynthetic pigment content compared to the PA. At the phylum level, Proteobacteria and Actinobacteriota are the major bacteria in MBC. Furthermore, the transcriptome reveals that the growth-promoting effects of MBC are associated with the up-regulation of genes encoding the glycolysis, the citrate cycle (TCA cycle), and the pentose phosphate pathway. Enhanced carbon fixation, coupled with down-regulation of photosynthetic electron transfer processes, suggests an energy allocation mechanism within MBC. The up-regulation of porphyrin and arachidonic acid metabolism, along with the expression of genes encoding LEV-degrading enzymes, provides evidence of MBC’s superior tolerance to and degradation of LEV. Overall, these findings lead to a better understanding of the underlying mechanisms through which MBC outperforms PA in treating LEV wastewater.

Photosynthetic Living Fiber Fabrication from Algal-Bacterial Consortia with Controlled Spatial Distribution

Zitong Sun,Huilin Wen,Zhengao Di,Yang Zhang,Shaobin Zhang,Zhiqian Zhang,Jing Zhang,Ziyi Yu

Living materials that combine living cells and synthetic matrix materials have become promising research fields in recent years. While multicellular systems present exclusive benefits in developing living materials over single-cell systems, creating artificial multicellular systems can be challenging due to the difficulty in controlling the multicellular assemblies and the complexity of cell-to-cell interactions. Here, we propose a coculture platform capable of isolating and controlling the spatial distribution of algal-bacterial consortia, which can be utilized to construct photosynthetic living fibers. Through coaxial extrusion-based 3D printing, hydrogel fibers containing bacteria or algae can be deposited into designated structures and further processed into materials with precise geometries. In addition, the photosynthetic living fibers demonstrate a significant synergistic catalytic effect resulting from the immobilization of both bacteria and algae, which effectively optimizes sewage treatment for bioremediation purposes. The integration of microbial consortia and 3D printing yields functional living materials with promising applications in biocatalysis, biosensing, and biomedicine. Our approach provides an optimized solution for constructing efficient multicellular systems and opens a new avenue for the development of advanced materials.

Enhanced sulfonamides removal via microalgae-bacteria consortium via co-substrate supplementation

Yue Wang,Jinghua Li,Yao Lei,Rong Cui,Aiping Liang,Xiaoqiang Li,Yoong Kit Leong,Jo-Shu Chang

Both co-cultivation and co-substrate addition strategies have exhibited massive potential in microalgae-based antibiotic bioremediation. In this study, glucose and sodium acetate were employed as co-substrate in the cultivation of microalgae-bacteria consortium for enhanced sulfadiazine (SDZ) and sulfamethoxazole (SMX) removal. Glucose demonstrated a two-fold increase in biomass production with a maximum specific growth rate of 0.63 ± 0.01 d −1 compared with sodium acetate. The supplementation of co-substrate enhanced the degradation of SDZ significantly up to 703 ± 18% for sodium acetate and 290 ± 22% for glucose, but had almost no effect on SMX. The activities of antioxidant enzymes, including peroxidase, superoxide dismutase and catalase decreased with co-substrate supplementation. Chlorophyll a was associated with protection against sulfonamides and chlorophyll b might contribute to SDZ degradation. The addition of co-substrates influenced bacterial community structure greatly. Glucose enhanced the relative abundance of Proteobacteria , while sodium acetate improved the relative abundance of Bacteroidetes significantly.

Collaborative Response of the Host and Symbiotic Lignocellulytic System to Non-Lethal Toxic Stress in Coptotermes formosanus Skiraki

Wenhui Zeng,Bingrong Liu,Wenjing Wu,Shijun Zhang,Yong Chen,Zhiqiang Li

Simple SummaryCoptotermes formosanusShiraki is a wood feeding lower termite and is widely distributed in many areas. The dynamic adjustment of theC. formosanusdigestive system to unfavorable survival environments was investigated via non-lethal toxic feeding. The toxic stress did not change the dominant role of microbial lignocellulases in cellulose degradation ofC. formosanus. The core symbiotic community was stable in abundance during the tolerance to the toxic treatment. However, a large number of low abundance taxa were significantly enriched by the low toxic feeding. These rare bacterial lineages likely contribute to toxic stress tolerance of termite. Above all, these findings add important new knowledge to our understanding of environmental adaptation of the lignocellulose hydrolysis system in termites.AbstractDisturbing the lignocellulose digestive system of termites is considered to be a promising approach for termite control. The research on the tolerance mechanism of the termite lignocellulose digestive system to harmful environment conditions is limited. In this study, we keepCoptotermes formosanusSkiraki under a non-lethal toxic condition by feeding the termites with filter paper containing the kojic acid (a low toxic insecticide). The effects of low toxic stress on the activities and gene expressions of host/symbiotic originated lignocellulases, and on the symbiotic microbial community structure ofC. formosanuswere explored. Our result showed that the low toxic stress would lead to the synchronous decrease of cellulase and hemicellulase activities, and supplementary increase of corresponding gene expressions. The symbiotic community maintained its role as the main force in the lignocellulolytic system ofC. formosanus. Meanwhile, a large number of rare taxa were significantly enriched by kojic acid treatment. These numerically inconspicuous bacterial populations might be responsible for the functions similar to phenoloxidase or insecticide detoxification and enableC. formosanusto tolerate the harmful environment. Overall, our data suggested that the digestive adaptation ofC. formosanusto physiotoxic feeding is closely related to the triple collaboration of termites–flagellates–bacteria. Keywords:lignocellulolytic system;non-lethal stress;symbiont;termite;toxic tolerance

Contaminant removal performance and lipid productivity of a cyanobacteria-bacteria consortium containing exogenous phytohormones during the treatment of antibiotic-polluted wastewater

Youshuai Fang,Guannan Lin,Ying Liu,Jian Zhang

In this study, a cyanobacteria-bacteria consortium containing native wastewater bacteria and immobilized Synechococcus sp. was constructed. The cyanobacterial cellular responses (including growth, biomass and lipid productivity) and contaminant removal ability (for TN, TP, COD and antibiotics) in the consortium were evaluated during the advanced treatment of wastewater containing 10–50 μg/L of mixed antibiotics (amoxicillin, tetracycline , erythromycin , sulfadiazine and ciprofloxacin) with the addition of a certain phytohormone (indole-3-acetic acid, gibberellin A3 or 6-benzylaminopurine) at trace level within a period of four days. Each phytohormone promoted the growth of Synechococcus sp. and increased the tolerance of Synechococcus sp. to mixed antibiotics. Indole-3-acetic acid coupled to moderate antibiotic stress could elevate lipid productivity and lipid content of Synechococcus sp. to 33.50 mg/L/day and 43.75%, respectively. Phytohormones increased the pollutant removal performance of the cyanobacteria-bacteria consortium through the stimulation of cyanobacterial growth and the regulation of cyanobacteria-bacteria interaction, which increased the abundances of microalgae-associated bacteria including Flavobacterium , Allorhizobium-Neorhizobium-Pararhizobium-Rhizobium , Bosea , Sphingomonas and Emticicia . Up to 80.83%, 98.06%, 83.26%, 99.84%, 99.50%, 89.41%, 65.61% and 60.65% of TN, TP, COD, amoxicillin , tetracycline , erythromycin , sulfadiazine and ciprofloxacin were removed by the consortium with the addition of phytohormones. In general, indole-3-acetic acid was the optimal phytohormone for enhancing lipid production and contaminant removal performance of the cyanobacteria-bacteria consortium.

Effects of exogenous vitamin B12 on nutrient removal and protein expression of algal-bacterial consortium

Ji Xiyan,Luo Xin,Zhang Jibiao,Huang Deying

Chlorella vulgaris and Bacillus licheniformis consortium was added to synthetic wastewater with exogenous vitamin B 12 . In the presence of 100 ng/L vitamin B 12 , removal efficiencies of TN, NH 3 -N, PO 4 3— P, and COD were 80.1%, 76.8%, 87.9%, and 76.7%, respectively. The functional groups on the cell surface of the consortium, including -NH, -CH 3 , C=O, C=C, and P-O-C, increased with 100 ng/L vitamin B 12 . These functional groups improved the biological adsorption of the consortium; however, higher concentrations of vitamin B 12 resulted in an occlusion of the functional groups. Furthermore, there were 5 significantly enriched protein pathways, namely carbon fixation in photosynthetic organisms; amino acid metabolic pathways; the pathway of one carbon pool by folate; nitrogen metabolism; and photosynthesis. Most proteins in these pathways were upregulated, which enhanced carbon fixation and photosynthesis in the algal cells. Simultaneously, B 12 promoted significant upregulation of proteins associated with the quorum-sensing pathway, which promoted the interaction between algae and bacteria.

Effects of Fungicide Propiconazole on the Yeast-Like Symbiotes in Brown Planthopper (BPH, Nilaparvata lugens Stål) and Its Role in Controlling BPH Infestation

Shentu Xuping,Wang Xiaolong,Xiao Yin,Yu Xiaoping

Yeast-like symbiotes (YLS), harbored in the abdomen fat-body cells of the rice brown planthopper (BPH), Nilaparvata lugens Stål (Hemiptera: Delphacidae), are vital to the growth and reproduction of their host. It is feasible to manipulate BPH infestation on rice by inhibiting YLS using fungicide. In this study, the fungicide propiconazole was injected into the hemolymph of BPH thorax via microinjection to investigate its effect on YLS, especially the dominant species, Hypomyces chrysospermus, and their host BPH. Propiconazole markedly reduced the total number of YLS and H. chrysospermus in BPH hemolymph and fat body, thereby leading to an obvious higher mortality and lower fecundity of BPH than the negative control (PBS, phosphate buffer solution). After microinjecting propiconazole, the survival rate of BPH nymphs at the 5th instar was significantly lower than that obtained after PBS treatment. Eight days after propiconazole microinjection, the BPH survival rate dropped to 40%, only half of BPH survival rate treated with PBS microinjection. For female adults (1-day-old), there were significant differences in the survival rates between BPHs treated with propiconazole and those treated with PBS at days 5–8. The fecundity of BPH decreased significantly by microinjecting propiconazole and averaged only 229 eggs per female, which was 20% less than that of the negative control. Furthermore, we reared BPH on the susceptible variety TN1 sprayed with propiconazole to prove the feasibility manipulating field occurrence of BPH by inhibiting YLS using fungicides. The number of YLS and H. chrysospermus in BPH obviously declined. Subsequently, the survival rate and fecundity of BPH significantly decreased after feeding on rice treated with propiconazole. Meanwhile, the propiconazole residue was detected in the hemolymph and gut of BPH by HPLC analysis within 1 day of feeding. Inhibiting YLS using fungicides was a novel and effective way to control BPH infestation.

Control of Rhizobia Endosymbiosis by Coupling ER Expansion with Enhanced UPR

Jing Ren,Qi Wang,Xiaxia Zhang,Yongheng Cao,JingXia Wu,Juan Tian,Yanjun Yu,Qingqiu Gong,Zhaosheng Kong

Legumes establish symbiosis with rhizobia by forming a symbiotic interface that enables cross-kingdom exchanges of signaling molecules and nutrients. However, how host organelles interact with symbiosomes at the symbiotic interface remains elusive during rhizobia endosymbiosis. Here, symbiotic cells are reconstructed using 3D scanning electron microscopy (SEM) and uncover that the host endoplasmic reticulum (ER) undergoes dynamic expansion to gradually enwrap symbiosomes, facilitating their compartmentalization and endosymbiosis. Consistently, altering ER lamellar expansion by overexpressing MtRTNLBs , the reticulons responsible for ER tubulation, impairs rhizobia accommodation and symbiosome development. Intriguingly, unfolded protein response (UPR)-marker genes, bZIP60 and IRE1A/B , show continuously activated expression during nodule development, and the two UPR-deficient mutants, ire1b , and bzip60 , exhibit compromised ER biogenesis and defective symbiosome development. Collectively, the findings underpin ER expansion and UPR activation as two key events in rhizobia accommodation and reveal an intrinsic coupling of ER morphology with proper UPR during root nodule symbiosis.

Adaptation responses of Dunaliella sp. and its natural symbiotic bacteria to sulfamethoxazole

Xiao Tan,Gang Pan,Wenxue Che,Ying Man,Liyi Wang,Jinting Li,Hui Zhao

Microalgal-bacterial consortia provide a sustainable, low-cost solution for antibiotic-contaminated wastewater remediation. Microalgae cultivation typically involves natural symbiotic bacteria, which are often overlooked. To fill this gap, this study evaluated the adaptive response of Dunaliella sp. and its natural symbiotic bacteria to sulfamethoxazole (SMX) through cultivation experiments and 16S rDNA sequencing. The results demonstrated strong SMX tolerance but limited removal efficiency by the consortium. The adaptive responses were characterized by increased bacterial diversity and connectivity, with quorum sensing bacteria-driven community changes and biofilm formation playing a critical role in this process. Dunaliella sp. exhibited strong antioxidant capacity (with catalase playing a key role) and increased extracellular polymeric substances secretion, which not only contributed to its tolerance but also promoted microalgae-bacteria interactions, enhancing consortium adaptability. These findings strengthen our understanding of the responses of natural symbiotic bacteria to antibiotics and advance the development of microalgae-bacteria symbiosis technology for wastewater treatment.

Enhanced biodegradation of chlortetracycline via a microalgae-bacteria consortium

Yue Wang,Yuanyuan He,Xiaoqiang Li,Dillirani Nagarajan,Jo-Shu Chang

Microbial removal of Chlortetracycline (CTC) at low CTC concentrations (in the order of 10–20 mg/L) has been reported. In this study, a novel microalgae-bacteria consortium was developed for effective CTC biodegradation at higher concentrations (up to 80 mg/L). The microalgae-bacteria consortium is resistant to up to 80 mg/L CTC, while the pure microalgal culture could only tolerate 60 mg/L CTC. CTC removal in the initial 12 h was primarily via biosorption by the microalgae-bacteria consortium and the adsorption capacity increased from 61.71 to 102.53 mg/g biomass in 12 h. Further, CTC biodegradation by the microalgae-bacteria consortium was catalyzed by extracellular enzymes secreted under antibiotic stress. The symbiotic bacterial diversity was analyzed by high throughput sequencing. The aerobic bacteria Porphyrobacter and Devosia were the dominant genera in the consortium. In the presence of CTC, a microbial community shift occurred with Chloroptast , Spingopyxis, and Brevundimonas being the dominant genera.

Effects of terminal electron acceptors on the biodegradation of waste motor oil using Chlorella vulgaris-Rhodococcus erythropolis consortia: Kinetic and thermodynamic windows of opportunity analysis

Yongrui Pi,Wenpeng Jia,Shengkai Chi,Hongke Meng,Yongzheng Tang

The Chlorella vulgaris - Rhodococcus erythropolis consortia was constructed for the biodegradation of waste motor oil (WMO), combined with thermodynamic calculations and stoichiometric analyses. The microalgae-bacteria consortium was constructed as C. vulgaris : R. erythropolis = 1:1 (biomass, cell/mL), pH = 7, 3 g/L WMO. Under the same condition, the terminal electron acceptors (TEAs) play a crucial role in the WMO biodegradation, which follows Fe 3+ >SO 4 2- > none. The biodegradation of WMO fitted well with the first-order kinetic model under experimental temperatures with different TEAs ( R 2 >0.98). The WMO biodegradation efficiency reached 99.2 % and 97.1 % with Fe 3+ and SO 4 2− as TEAs at 37 °C, respectively. Thermodynamic methanogenesis opportunity windows with Fe 3+ as TEA are 2.72 times fold as large as those with SO 4 2- . Microorganism metabolism equations demonstrated the viability of anabolism and catabolism on WMO. This work lays the groundwork for the implementation of WMO wastewater bioremediation and supports research into the biochemical process of WMO biotransformation .
