Sewage effluent and reclaimed water usually contain elevated concentrations of bacteria-derived endotoxins, which may have serious public health implications upon exposure. Both conventional disinfection methods, such as chlorine, ultraviolet, and ozone disinfection, and the new potassium ferrate disinfection process have limited endotoxin removal capacity. Therefore, it is necessary to find new methods of removing endotoxins. In this study, we investigated the effects of ultraviolet/ferrate (UV/Fe(VI)) combination treatment on endotoxin elimination and inhalation hazard control. Our results indicate that UV or Fe(VI) alone cannot sufficiently eliminate endotoxins but can instead exacerbate endotoxin release during bacterial inactivation. However, the UV/Fe(VI) combination rapidly eliminated approximately 85.6 % of both pure and bacteria-derived endotoxins. The optimal conditions for this disinfection process were determined to be 10 mg/L of Fe(VI) and 160 μW/cm 2 of UV exposure for 15 min. Furthermore, the pH value, bicarbonate (HCO 3 – ), humic acid, and turbidity were determined as potential factors that may affect UV/Fe(VI)-mediated endotoxin removal in reclaimed water. The potential mechanism underlying UV/Fe(VI)-induced endotoxin degradation involves several strongly oxidizing species, among which Fe(IV)/Fe(V) is regarded as the primary contributor, followed by the superoxide anion ( • O 2 – ) and hydroxyl ( • OH) radicals. These active substances induce the breakdown of the endotoxin molecular structure into smaller, non-toxic fragments. Furthermore, pure endotoxins, Escherichia coli , and Pseudomonas aeruginosa were disinfected with UV/Fe(VI), and then these aerosolized samples were inhaled exposed to mice. Our results demonstrated that combined use of UV and Fe(VI), rather than administration alone, remarkably blunted bacterial- and endotoxin-induced lung injury. In conclusion, these results demonstrate that the UV/Fe(VI) approach successfully eliminates endotoxins and related inhalation hazards from reclaimed water.

Endotoxins (LPS) are components of the cell wall of Gram-negative bacteria that are released when bacterial cell membrane is destroyed, the bacteria and the released LPS have led to serious diseases such as sepsis and septic shock. Histidine functionalized adsorbents have been used to remove LPS, however, the present hemoperfusion adsorbents have drawbacks such as inadequate LPS adsorption capability, high cost of raw material, and so on. Inspired by the structure of histidine and incorporated antibacterial molecules, we synthesize allylamine-modified polyacrylonitrile (PAD)/poly (imidazolium bromide) (PMIBr) composite (PAI) microspheres as hemoperfusion adsorbents. The PAI microspheres show excellent antibacterial ability with high killing efficiency of 98.8% against S. aureus and 95.4% against E. coli . In addition, the adsorption capacity of the microspheres for LPS in PBS is 433.5 EU/g within 3 h, and the LPS concentration could also be reduced from 7.34 EU/mL to below 0.5 EU/mL in the plasma environment. The PAI microspheres exhibit outstanding blood compatibility, antibacterial property and endotoxin removal capability, which have great potential in the treatment of sepsis patients.

A polysulfone (PSF)-based hollow fiber affinity membrane for in vitro endotoxin elimination was explored and prepared successfully. This depended on the well-known hemocompatibility of PSF membrane. The PSF hollow fiber membranes were firstly activated by a chloromethylation reaction, and treated with ethylenediamine and glutaraldehyde successively. L-Serine (Ser) was finally immobilized on the surface of the membranes as the functional ligand. The modified membrane was confirmed by X-ray photoelectron spectroscopy, attenuated total reflectance Fourier transform infrared spectroscopy, and contact angle analysis. L-Ser density up to 8.4 μmol cm−2 on PSF membranes could be achieved. The adsorption performance was preliminarily studied by comparing the PSF membrane and PSF–Ser membrane in deionized water and sodium phosphate buffer, showing that the PSF–Ser membrane had improved endotoxin adsorption capacity with decreased Bovine Serum Albumin (BSA) adsorption. The PSF–Ser membrane was further investigated in human plasma, and showed excellent removal efficiency and specificity for endotoxin. These results demonstrate the promising application potential of this novel affinity membrane in sepsis therapy.

Gut microbiota plays an important role in orchestrating the host immune response. We previously reported that gut microbiota-derived rhamnose enhances the phagocytosis of macrophages, upon which we further asked whether rhamnose has modulatory effects on inflammation. Here, we show that, in an LPS-induced endotoxic mouse model, plasma rhamnose levels are increased. This bacteria-derived sugar alone does not impact inflammatory cytokine homeostasis or cause organ damage. In contrast, it is able to alleviate endotoxin-induced systemic inflammation and organ damage. Mechanistically, in macrophages in vitro , rhamnose binds to the V39, D40, and T101 sites of carcinoembryonic antigen-associated cell adhesion molecule 1 (CEACAM1), subsequently promoting the interaction between CEACAM1 and galectin 9 (LGALS9), which increases the protein level of dual-specificity protein phosphatase 1 (DUSP1). This inhibits p38 phosphorylation and thus attenuates the LPS-triggered expressions of proinflammatory factors. Collectively, our results suggest that rhamnose signals via the CEACAM1/LGALS9-p38 axis, which suppresses endotoxemia-associated inflammation, and that rhamnose is a candidate anti-inflammatory agent for the control of infection-induced organ damage.

Endotoxin adsorption has received extensive attention in the field of blood purification. However, developing highly efficient endotoxin adsorbents with excellent hemocompatibility remains challenging. In this study, we propose a new approach for developing the functional polyethersulfone (PES) membrane to remove endotoxins. First, the PES polymer is grafted with polyethylene glycol methyl acrylate (PEG-MA) in a homogeneous phase system via γ irradiation, and PES-g-PEG can be directly used to prepare the membrane by the phase inversion method. Then, polydopamine (PDA) is coated as an adhesive layer onto a PES-g-PEG membrane in an alkaline aqueous solution, and lysozyme (Lyz) is covalently immobilized with PDA through the Schiff base reaction. Lysozyme acts as an affinity adsorption ligand of endotoxin through charge and hydrophobic action. Our study reveals that the PEG branched chain and the PDA coating on the PES membrane can maintain the secondary structure of lysozyme, and thus, the immobilized Lyz can maintain high activity. The adsorption capacity of endotoxins for the PES-g-PEG/PDA/Lyz membrane is 1.28 EU/mg, with an equilibrium adsorption time of 6 h. Therefore, the PES-g-PEG/PDA/Lyz membrane shows great potential application in the treatment of endotoxemia.

Tendon-derived type I collagen is an essential biomaterial for various biomedical devices due to its inherent bioactivity and favorable environment for cells. Correspondingly, microbial and endotoxin contamination can be easily introduced during the collagen extraction process, which is generally overlooked in fundamental scientific research, especially the endotoxin residue. Conventional approaches for mitigating endotoxin exhibit limited effectiveness when applied to biomacromolecule products because of viscosity, clogging, and diminished bioactivity. In this study, we developed a bifunctional sheet that can simultaneously reduce the microbe and endotoxin contamination in collagen solution by co-incubation and subsequent magnetic separation, avoiding the issues of blockage and bioactivity impairment. The bifunctional sheet was successfully fabricated by modifying the magnetic graphene oxide with histamine. Collagen products treated by sheets exhibited reduced microbial and endotoxin contamination while maintaining their bioactivity for encapsulated cell growth. Additionally, inflammatory stimulation of collagen was decreased in vitro and in vivo after treatment. This work may present a facile approach for diminishing microbe and endotoxin residues in collagen products in basic research, obviating the non-essential use of a sterile workshop and facilitating the development of tissue-derived collagen research.

Endotoxin (lipopolysaccharide, LPS), widely distributed in the atmospheric environment with strong immunogenicity, is an important biological component of ambient particulate matter. However, whether LPS participates in atmospheric chemistry and how its biological health impacts change with the relevant processes are poorly understood. In this study, we employed the rat model to investigate the impact of ozone oxidation on the biological toxicity of LPS and used Fourier transform infrared spectroscopy and high-resolution electrospray mass spectrometry to study the underlying reaction mechanisms. The results show that the LPS can be oxidized by ozone and the resulting reactant greatly enhanced inflammatory anemia with a 177% capacity increase despite a minor influence on its immunogenicity. In contrast to the control, rats exposed to oxidized LPS were observed to release characteristic exhaled biomarkers, indicating that the formed reactant indeed altered the biological effects of LPS. Mechanistic investigation reveals that ozone oxidation of the hydroxyl group in the key toxic part of LPS, kdo2-lipid A, can cause dysregulation of iron homeostasis in rats, which is the mechanism of oxidized LPS-enhanced anemia. Unfortunately, these chemical structure changes and the resulting health impacts cannot be detected by the conventional LPS analysis method. This study highlights the changes in the toxicity of LPS and its health impacts when oxidized by ozone and the need to broadly consider the involvement of bioaerosol in atmospheric chemistry.

Electrostatic spinning technology can prepare nanofiber materials with high specific surface area and unique microstructure. Cellulose acetate (CA) has excellent membrane formation and processability, while chitosan (CS) is positively charged due to the abundance of amino groups and has certain biological activities. In this paper, CA/CS nanofiber membranes with different CS contents were prepared by solution blending and electrostatic spinning techniques and used in hemoperfusion (HP) for targeted removal of endotoxins. The CA/CS nanofibrous membranes were systematically characterized using infrared spectroscopy, scanning electron microscopy, etc., and the clearance ability of CA/CS nanofibrous membranes for endotoxin was tested under static and dynamic simulated HP conditions. The results showed that the addition of CS changed the structure of CA nanofibers, significantly increased the specific surface area of CA/CS nanofiber membrane, and provided more adsorption sites for the membrane material. The positive charge of CS interacted with the negatively charged endotoxin in the blood through electrostatic attraction, which enabled the CA/CS nanofiber membranes to adsorb endotoxin efficiently and exhibit higher endotoxin clearance ability. In the static adsorption experiment, when the CS content was 3.75%, the endotoxin adsorption and clearance rate could reach 580.0 ± 32.2 EU/g and 65.1 ± 3.6%, respectively. In the dynamic adsorption experiment, when the CS content was 2.50%, the CA/CS nanofiber membrane had a good effect on the adsorption of endotoxin. Meanwhile, CA/CS nanofiber membrane also has good blood compatibility. Therefore, the CA/CS nanofiber membrane is potentially valuable in blood purification.

Endotoxins or lipopolysaccharides (LPS) are potent inflammatory agents that pose serious health risks even at low concentrations, thus requiring sensitive and reliable detection methods. Current methods for endotoxin detection, primarily the Limulus Amebocyte Lysate (LAL) assay and Enzyme-linked Immunosorbent Assays (ELISA) are limited by complex procedures, prolonged assay time, and susceptibility to interference. To address these challenges, we developed a single-reporter probe-based dual-mode biosensor integrating MXene-Au nanosheets with the CRISPR/Cas12a system for endotoxin detection. The sensing platform was constructed by conjugating ssDNA-Cy5 reporters onto MXene-Au nanosheets. MXene-Au nanosheets served dual functions, including efficient quenching of fluorescence and strong enhancement of SERS signals. In the presence of target endotoxins, CRISPR/Cas12a remains inactive, leaving the ssDNA-Cy5 reporters intact on the MXene-Au surface. The close proximity of Cy5 to the MXene-Au surface results in quenched fluorescence (“FL off”) but enhanced SERS signals (“SERS on”). Conversely, in the absence of endotoxins, activated CRISPR/Cas12a cleaves the ssDNA-Cy5, releasing Cy5 from the surface, thus leading to fluorescence recovery (“FL on”) but diminished SERS signals (“SERS off”). By measuring the changes in SERS and FL signals and analysing the ratiometric I SERS /I FL signals, the biosensor achieved a lower limit of detection (LOD) of 15.9 pg/mL within 30 minutes compared to single detection modes. Validation studies in complex matrices, including spiked water and milk samples, showed performance comparable to commercial endotoxin detection kits. Overall, our CRISPR-Cas12a-mediated fluorescent-SERS dual-mode biosensor not only improves detection sensitivity but also provides built-in quality control, improving the reliability of results through cross-validation.

A novel polyvinyl alcohol-amino multi-walled carbon nanotube (PVA-AMWCNT) nanocomposite microsphere was prepared successfully for the first time and used for endotoxin removal. The resulting AMWCNT modified PVA microsphere was characterized by SEM, Raman spectrum and fluorescence image, which indicated AMWCNT was dispersed into the macropores of PVA microsphere uniformly. The PVA-AMWCNT microspheres showed better adsorption capability and faster adsorption equilibrium for endotoxin in aqueous solution when compared to the PVA microsphere with polymyxin B (PMB) as ligand. More noteworthy, the PVA based microspheres had little nonspecific adsorption in simulated serum. Therefore, PVA-AMWCNT nanocomposite microsphere with an excellent haemocompatibility has a great potential application in clinical blood purification.

A novel magnetic chitosan microsphere (MCM) was prepared from chitosan and Fe3O4 through inverse suspension crosslinking, epichlorohydrin activation, and grafted with ligands. The magnetometer measurements data indicated that the magnetic microspheres had superparamagnetic property as well as fast magnetic response without the magnetic hysteresis effect and thus can be well used in magnetic separation. The adsorption results showed that the magnetic chitosan microsphere grafted with ligand had good adsorption capacity on endotoxin up to 554.1 EU/g under the condition of 25.0 mg/mL adsorbent, 17.2 EU/mL endotoxin. It is expected that this new microsphere will have a feasible and useful application in adsorbing endotoxin from biological products.

NF-κB and MAPK are classic inflammation signaling pathways which regulate inflammation signal transmission and induce the expression of many inflammatory factors. Based on the potent anti-inflammatory activity of benzofuran and its derivatives, several new heterocyclic/benzofuran hybrids were first designed and synthesized by molecular hybridization. Their structure was confirmed by1H NMR,13C NMR, HRMS or X-single crystal diffraction. The anti-inflammatory activity of these new compounds was screened by compounds; compound5dexhibited an excellent inhibitory effect on the generation of NO (IC50= 52.23 ± 0.97 μM), and low cytotoxicity (IC50> 80 μM) against the RAW-264.7 cell lines. To further elucidate the possible anti-inflammatory mechanisms of compound5d, the hallmark protein expressions of the NF-κB and MAPK pathways were studied in LPS-stimulated RAW264.7 cells. The results indicate that compound5dnot only significantly inhibits the phosphorylation levels of IKKα/IKKβ, IKβα, P65, ERK, JNK and P38 in the classic MAPK/NF-κB signaling pathway in a dose-dependent manner, but also down-regulates the secretion of pro-inflammatory factors such as NO, COX-2, TNF-α and IL-6. Further, the in vivo anti-inflammatory activity of compound5dindicated that it could regulate the involvement of neutrophils, leukocytes and lymphocytes in inflammation processes, and reduce the expression of IL-1β, TNF-α and IL-6 in serum and tissues. These results strongly suggest that the piperazine/benzofuran hybrid5dhas a good potential for developing an anti-inflammatory lead compound, and the anti-inflammatory mechanism might be related to the NF-κB and MAPK signaling pathways. Keywords:benzofuran derivatives;piperazine;anti-inflammatory;NF-κB/MAPK signaling pathway;endotoxemic mice
