Ensuring excellent anticoagulant, anti-inflammatory, and endothelialization properties in vascular stents through coating construction is crucial for their satisfactory performance post-implantation. In this study, we propose a cell-membrane mimetic multifunctional hyaluronic acid (HA)-based coating by combining the aminolyzed methacryloyloxyethyl phosphorylcholine (MPC) copolymer with oxidized hyaluronic acid (Ox-HA) through Schiff base reaction. Compared with traditional anti-fouling design, the composite coating present a stage-specific ability, which can resist the adhesion of blood components, while mediating vascular cell fate with the incorporation of HA. The coating exhibited promoted endothelial cell (EC) growth and inhibited the proliferation of smooth muscle cells (SMCs) by inducing a phenotype change. Besides, the multifunctional HA-based coating can mediate macrophages to a M2 phenotype. The coated stents were implanted in rabbits, and exhibited ideal capabilities in promoting endothelialization and inhibiting inflammation and restenosis in vivo , offering a potential approach to address multifunctionality for vascular implants.

Coagulation, inflammation, and hyperplasia frequently restrict the effectiveness of cardiovascular stents . It is critical to modify stent surfaces to allow for in situ tissue healing. Interrelated coagulation and inflammation can impede endothelialization and are critical to the long-term performance of cardiovascular implants . This study proposes a relatively bio-inert “Anti-fouling plus” coating that is effective in anti-coagulation and anti-inflammation at the early stage of implantation and provides mild inflammatory responses during tissue remodeling process. The phosphorylcholine (PC)-based polymer PM(PCLA) is reacted onto a polydopamine-treated polylactic acid (PLA) surface. PC-based polymers that mimic cell membranes present remarkable antiinflammation and anticoagulation properties. The composite of epigallocatechin gallate (EGCG) and tempol is loaded on the PM(PCLA) coating to achieve “anti-fouling plus” which can mediate inflammatory responses by scavenging reactive oxygen species. In vitro hemocompatibility and cell tests demonstrate the effectiveness of coatings in inhibiting thrombus and inflammation. Interestingly, this study originally reveals a novel strategy that the anti-fouling plus coating can direct vascular tissue healing on stent via promoted endothelialization and mediated contractile smooth muscle cell phenotype by switching inflammatory cells to a pro-repair phenotype during tissue healing process, even if the coating is relatively bio-inert.

Stent implantation is one of the most effective methods for the treatment of atherosclerosis. Nitinol stent is a type of stent with good biocompatibility and relatively mature development; however, it cannot effectively achieve long-term anticoagulation and early endothelialization. In this study, nitinol surfaces with the programmed assembly of heparin, exosomes from endothelial cells, and endothelial affinity peptide (REDV) were fabricated through layer-by-layer assembly technology and click-chemistry, and then exosomes/REDV-modified nitinol interface (ACC-Exo-REDV) was prepared. ACC-Exo-REDV could promote the rapid proliferation and adhesion of endothelial cells and achieve anticoagulant function in the blood. Besides, ACC-Exo-REDV had excellent anti-inflammatory properties and played a positive role in the transformation of macrophage from the pro-inflammatory to anti-inflammatory phenotype. Ex vivo and in vivo experiments demonstrated the effectiveness of ACC-Exo-REDV in preventing thrombosis and hyperplasia formation. Hence, the programmed assembly of exosome interface could contribute to endothelialization and have potential application on the cardiovascular surface modification to prevent stent thrombosis and restenosis.

Congenital heart defects (CHDs) are one of the most common congenital malformations, accounting for ≈30% of all congenital malformations. Interventional implantation of occlusion devices is becoming the preferred treatment for CHDs. However, current occlusion devices suffer from serious problems, such as thrombosis, slow endothelialization, imprecise localization, abrasion, displacement, etc. Here, a multifunctional drug-carrying fiber platform with structural similar to the extracellular matrix is innovatively designed to develop 4D printed cardiac occlusion devices, with characteristics of efficient anticoagulation, proendothelialization, and precise localization. Biomimetic ligament structures are designed to achieve a similar mechanical response to myocardial tissue, which helps to synergize deformation and reduce tissue wear. A structural design method for biomimetic personalized multilevel occlusion devices is proposed, facilitating further improvement of sealing reliability. The radiopaque 4D printed shape memory composites are developed, realizing the complete visualization and precise localization of the device in vivo. The novel 4D printed cardiac occlusion device provides an effective way to reduce the risk of complications and contributes to versatility. It is expected to be a next-generation multifunctional repair device for personalized treatment of CHDs.

Biodegradable materials are a next-generation invention for the treatment of congenital heart diseases. However, the corresponding technology used to develop ideal biomaterials still presents challenges. We previously reported the first biodegradable atrial septal defect (ASD) occluder made of poly-lactic acid (PLLA). Unfortunately, the PLLA occluder had a limited endothelialization effect. In this study, the surface of the occluder membrane was coated with sericin/CD34 antibodies to promote the growth of endothelial cells and the regeneration of defective tissue and enhance the repair of the atrial septal defect. The physicochemical properties of the coat on the surface of the fiber membrane were characterized. The sericin coat successfully covered the fiber surface of the membrane, and the thickness of the membrane increased with the sericin concentration. The swelling rate reached 230%. The microscopic observation of fluorescently labeled CD34 antibodies showed that the antibodies successfully attached to the fiber membrane; the fluorescence intensity of PLLA-SH5 was particularly high. The in vitro experiment showed that the PLLA-SH-CD34 fiber membrane was biocompatible and promoted the adhesion and proliferation of endothelial cells. According to our findings, the PLLA-SH-CD34 membrane provides a theoretical and technical basis for the research and development of novel biodegradable occluders.Keywords:biodegradable;CD34 antibody;endothelialization;fiber membrane

Drug-eluting magnesium (Mg) alloy stents have a slower degradation rate and lower restenosis rate compared with uncoated stents, demonstrating good clinical efficacy. However, the release of anti-hyperplasia drugs from coatings delays endothelial tissue repair, thus leading to late stent thrombosis. To address these issues, a dual self-healed coating with various biological properties was fabricated on magnesium fluoride/polydopamine (MgF 2 /PDA)-treated Mg alloys by spraying-assisted layer-by-layer (LBL) self-assembly of chitosan (CS), gallic acid (GA), and 3-aminobenzeneboronic acid-modified hyaluronic acid (HA-ABBA). The LBL coating, approximately 1.50 μm thick, exhibited a uniform morphology with good adhesion strength (~ 1065 mN). The annual corrosion rate ( P i ) of LBL samples was ~ 1400 times slower than that of the Mg substrate, due to the physical barrier function provided by MgF 2 /PDA layers and the dual self-healed ability of LBL layers. The rapid self-healing ability (with a healing period of ~ 4 h under dynamic/static conditions) resulted from the synergistic interplay between the recombination of diverse chemical bonds within the LBL coating and the coordination of LBL-released GA with Mg 2+ , as corroborated by computer simulations. Compared with the drug-eluting coatings, the LBL sample demonstrated substantial advantages in anti-oxidation, anti-denaturation of fibrinogen, anti-platelet adhesion, anti-inflammation, anti-hyperplasia, and promoted-endothelialization. These benefits effectively address the limitations associated with drug-eluting coatings. Graphical abstract

Stent implantation remains a cornerstone of interventional cardiology, providing a minimally invasive solution to restore blood flow in occluded vessels. However, current stents face persistent challenges in simultaneously preventing neointimal hyperplasia and promoting reendothelialization, compromising their long-term efficacy. To address these limitations, we developed an in situ H2S-releasing polymer brush-coated stent that actively modulates material–blood interactions, creating a favorable microenvironment for vascular healing. H2S enhances the stent’s antithrombotic properties by inhibiting fibrinogen binding and platelet activation, while also mitigating oxidative stress and promoting macrophage polarization toward the anti-inflammatory M2 phenotype. In vivo, the H2S-releasing stents significantly improved vascular healing by accelerating endothelialization and inhibiting smooth muscle cell overproliferation, resulting in a thinner neointima with functional endothelial coverage. Transcriptomic analysis further elucidated the underlying mechanisms, revealing H2S-mediated modulation of key biological pathways that support vascular healing. These findings underscore the potential of in situ H2S release as an effective strategy for optimizing vascular implants and improving long-term outcomes.

The growing prevalence of cardiovascular diseases leads to the increasing need for vascular patches. In this work, we prepared a polyurethane/polyurethane-sodium triphosphate (PU/PU-STPP) nanoparticle composite film and subsequently a PU/PU-STPP/decellularized scaffold vascular patch
(PSVP) using a simple and low-cost method-cosedimentation. The PU/PU-STPP nanoparticle composite film exhibited the improved blood and cell compatibilities, reduced platelet adhesion, and enhanced cell attachment and proliferation. Furthermore, we explored the performance of PSVP in vivo,
using a murine model. Thirty days after the implantation with PSVP, the patched abdominal artery maintained patency, and the formation of endothelial tissue, e.g., endothelialization was observed, demonstrating its potential in vascular repair and regeneration. Moreover, our data showed that
the material had the surface similar to the luminal surface of a blood vessel, and this similarity and the synergetic effect of PU-STPP nanoparticles significantly improved the blood and cell compatibilities of the material, effectively inhibited platelet adhesion and enhanced cell attachment,
thus promoting endothelialization. To sum up, these findings implied that the PSVP may be used as a promising vascular patch material.

Atherosclerosis is often accompanied by inflammation and oxidative stress. Excessive reactive oxygen species (ROS) can damage the vascular endothelium, leading to endothelial dysfunction and reduced nitric oxide (NO) bioavailability. Further accumulation of ROS contributes to vascular cell damage, lipid peroxidation, and extracellular matrix deposition. Thus, clearing excess ROS and reshaping the oxidative microenvironment is essential for treating atherosclerosis (AS). In this study, Ebselen, which mimics glutathione peroxidase and possesses redox capabilities, was successfully synthesized. Subsequently, a multifunctional coating was designed using a combination of Ebselen and poly (trimethylene carbonate) (PTMC), capable of protecting cells from ROS-induced damage, promoting vascular endothelialization, and exhibiting anti-proliferative properties. The Ebselen-loaded coating effectively scavenges free radicals (with an elimination rate of 89 %), catalytically releases NO (0.96 × 10⁻¹⁰ to 1.26 × 10⁻¹⁰ mol/cm²/min), and sustainably delivers Ebselen to the lesion site through a redox cycle. Notably, this coating shows excellent hemocompatibility and cytocompatibility. Subcutaneous implantation results indicated that the fibrous capsule thickness of PTMC10 was the lowest, at just 47.7 % of that of PTMC. Therefore, the Ebselen-loaded coating presents promising applications in cardiovascular stents.

In recent years, valvular heart disease has become a serious disease threatening human life and is a major cause of death worldwide. However, the glutaraldehyde (GLU)-treated biological heart valves (BHVs) fail to meet all requirements of clinical application due to disadvantages such as valve thrombus, cytotoxicity, endothelialization difficulty, immune response, and calcification. Encouragingly, there are a large number of carboxyls as well as a few amino groups on the surface of GLU-treated BHVs that can be modified to enhance biocompatibility. Inspired by natural biological systems, we report a novel approach in which the heart valve was cross-linked with erythrocyte membrane biomimetic drug-loaded nanoparticles. Such modified heart valves not only preserved the structural integrity, stability, and mechanical properties of the GLU-treated BHVs but also greatly improved anti-coagulation, anti-inflammation, anti-calcification, and endothelialization. The in vitro results demonstrated that the modified heart valves had long-term anti-coagulation properties and enhanced endothelialization processes. The modified heart valves also showed good biocompatibility, including blood and cell biocompatibility. Most importantly, the modified heart valves reduced the TNF-α levels and increased IL-10 compared to GLU-treated BHVs. In vivo animal experiments also confirmed that the modified heart valves had an ultrastrong resistance to calcification after implantation in rats for 120 days. The mechanism of anti-calcification in vivo was mainly due to the controlled release of anti-inflammatory drugs that reduced the inflammatory response after valve implantation. In summary, this therapeutic approach based on BHVs cross-linking with erythrocyte membrane biomimetic nanoparticles sparks a novel design for valvular heart disease therapy.

Combining the advantages of the fibrous nanostructure of carbon nanotubes (CNTs) and the bioactivities of heparin/heparin-mimicking polyanions, functional nanofibrous heparin or heparin-mimicking multilayers were constructed on PVDF membrane with highly promoted endothelialization and antithrombogenic activities. Oxidized CNT (oCNT) was first functionalized with water-soluble chitosan (polycation), then enwrapped with heparin or a typical sulfonated heparin-mimicking polymers (poly(sodium 4-styrenesulfonate-co-sodium methacrylate)) to construct the multilayers. Then, the surface-deposited multilayers were constructed via electrostatic layer-by-layer assembly of the functionalized oCNTs. The scanning electron microscope and atom force microscope images confirmed that the coated multilayers exhibited nanofibrous and porous structure. The live/dead cell staining and cell viability assay results indicated that the coated nanofibrous multilayers had excellent compatibility with endothelial cells. The cell morphology observation further confirmed the promotion ability of surface endothelialization due to the coated heparin/heparin-mimicking multilayers. Further systematical evaluation on blood compatibility revealed that the surface heparin/heparin-mimicking multilayer-coated membranes also had significantly improved blood compatibility including restrained platelet adhesion and activation, prolonged blood clotting times, and inhibited activation of coagulation and complement factors. In summary, the proposed nanofibrous multilayers integrated endothelialization and antithrombogenic properties; meanwhile, the heparin-mimicking coating validated comparable performances as heparin coating. Herein, it is expected that the surface coating of nanofibrous multilayers, especially the facilely constructed heparin-mimicking coating, may have great application potential in biomedical fields.

In this study, we designed and synthetized artificial vascular scaffolds based on nanofibers of collagen functionalized with hyaluronic acid (HA) in order to direct the phenotypic shape, proliferation, and complete endothelization of mouse primary aortic endothelial cells (PAECs). Layered tubular HA/collagen nanofibers were prepared using electrospinning and crosslinking process. The obtained scaffold is composed of a thin inner layer and a thick outer layer that structurally mimic the layer the intima and media layers of the native blood vessels, respectively. Compared with the pure tubular collagen nanofibers, the surface of HA functionalized collagen nanofibers has higher anisotropic wettability and mechanical flexibility. HA/collagen nanofibers can significantly promote the elongation, proliferation and phenotypic shape expression of PAECs. In vitro co-culture of mouse PAECs and their corresponding smooth muscle cells (SMCs) showed that the luminal endothelialization governs the biophysical integrity of the newly formed extracellular matrix (e.g., collagen and elastin fibers) and structural remodeling of SMCs. Furthermore, in vitro hemocompatibility assays indicated that HA/collagen nanofibers have no detectable degree of hemolysis and coagulation, suggesting their promise as engineered vascular implants. Keywords:collagen;hyaluronic acid;vascular endothelial cells;nanofibers;endothelization
