Bionic wet-adhesive tooth band-aid with bidirectional anchoring to promote dentin remineralization

Jieyu Zhu,Li Zhen,Yuan Gao,Jun Luo,Jianshu Li,Jiaojiao Yang,Jiyao Li

Tooth defects are regarded as “wounds” on the tooth. While remineralization therapies aim to rebuild tooth structures, challenges remain in protecting defects and delivering remineralization templates due to the wet and dynamic oral environment. Drawing from wound dressing, this study intends to construct a tooth dressing for the oral environment. Tannic acid (TA) was screened to crosslink fibroin protein (SF) and form internal anchoring through hydrogen bonding to fabricate a SF-TA (ST) tooth dressing matrix. Assisted by the drainage effect of the SF hydrophobic side chain on the interface, the system forms external anchoring to the tooth surface via phenolic hydroxyl groups and side chains. Notably, the bidirectional anchoring effect can be enhanced by calcium-phosphorus-enriched saliva, ensuring effective wet adhesion in the oral cavity. Loaded with an octuplet repeat of aspartate-serine-serine (8DSS) as a remineralization template, the SF-TA-8DSS (ST-8DSS) “tooth band-aid” maintains strong wet-adhesion to tooth surface under liquid contamination (bursting pressure above 780 mmHg) and delivers 8DSS into dentinal tubules (up to 100 µm over 14 days), promoting dentin remineralization. The proposed tooth band-aid offers an effective strategy for repairing tooth defects, combining mechanical reinforcement and wet adhesion through bidirectional anchoring for sustained mineralization under fluid contamination.

Applications of photothermally mediated nanohybrids for white spot lesions in orthodontics

Xinwei Lu,Yaxin Qu,Tingting Zhu,Xiaoying Qu,Zhe Zhang,Yanling Yu,Yuanping Hao

In orthodontic treatment, cariogenic bacteria in the oral cavity are the main cause of enamel white spot lesions (WSLs). Therefore, to effectively prevent and treat WSLs, it is crucial to inhibit the cariogenic bacterial activity while promoting the remineralization of demineralized tooth enamel . However, fluoride preparations commonly used for the prevention and treatment of WSLs can induce dental fluorosis if ingested in excess, and their remineralization effect is limited by the residual hydroxyapatite (HAp) content and salivary Ca 2+ and PO 4 3- levels. In this study, we propose a strategy (CMCS/ [email protected] ) for antibacterial and remineralization of WSLs by a nanohybrid of carboxymethyl chitosan (CMCS)-stabilized amorphous calcium phosphate (ACP) loaded polydopamine nanoparticles (PDA NPs) based on biomimetic remineralization techniques and biocompatible near-infrared (NIR) photoactivation therapy. The nanohybrid utilizes the excellent photothermal conversion ability of polydopamine for antimicrobial purposes, while CMCS with its own positive and negative charges (-NH 3 + and -COO - ) acts as a biomimetic mineralizing agent to stabilize ACP , supplemented with abundant Ca 2+ and PO 4 3- for remineralization of demineralized enamel. The results showed that CMCS/ [email protected] could effectively inhibit the adhesion of cariogenic Streptococcus mutants ( S. mutants ) with high bactericidal rates. In addition, the remineralization of demineralized enamel by nanohybrid was more effective after 7 days of in vitro mineralization . This study provides a theoretical and experimental basis for the use of CMCS/ [email protected] nanohybrid materials as potential materials against WSLs.

The effects and mechanisms underlying collagen-binding peptides MDPs on dentin remineralization: an in vitro study

Li Huanying,Liu Gengbo,Liu Xiaohao,Wu Buling

Section Background Currently, many biomimetic mineralization materials fail to bind effectively to collagen, which significantly limits their remineralization efficacy. To address this limitation, we designed MMP2-derived peptides (MDPs) on the basis of the strong affinity domain of collagen: MDP-a and MDP-3DSS. Section Methods Surface plasmon resonance (SPR) was used to measure the binding force of MDPs to collagen. Inductively coupled plasma-mass spectrometer (ICP-MS) was used to evaluate the effects of peptides on the adsorption of calcium ions by collagen. Transmission electron microscopy (TEM) and thermogravimetric analysis (TGA) were employed to analyze the stabilizing effect of MDPs on amorphous calcium phosphate (ACP) and the mineralization effect on collagen. Scanning electron microscopy (SEM), mechanical tests, and the CCK-8 assay were employed to evaluate the remineralization effect of MDPs on demineralized dentin and their biocompatibility. Section Results The results of the present study demonstrated that MDPs strongly bind to collagen (18 µM for MDP-a and 18.9 µM for MDP-3DSS) and can promote calcium ion adsorption by collagen (p < 0.0001). Furthermore, MDPs have been shown to stabilize ACP and induce intrafibrillar mineralization. In vitro studies revealed that both surface and cross-sectional dentin slices treated with MDPs presented dense mineral deposition. This deposition effectively occluded the dentinal tubules and resulted in enhanced mechanical properties. The tubule plugging rate, the elastic modulus and microhardness of the MDPs group were greater than those of the control group (p < 0.01). Moreover, MDP-3DSS demonstrated the most significant remineralization potential. Additionally, MDPs treatment did not affect the activity of human dental pulp stem cells (hDPSCs) (p > 0.05). Section Conclusion s The present study successfully developed and evaluated two novel biomimetic mineralized peptides, MDP-a and MDP-3DSS. It is necessary to conduct further research to explore the possibility of clinical transformation of MDPs.

Hierarchical structure and mechanical properties of remineralized dentin

Yi Chen,Jianming Wang,Jian Sun,Caiyun Mao,Wei Wang,Haihua Pan,Ruikang Tang,Xinhua Gu

It is widely accepted that the mechanical properties of dentin are significantly determined by its hierarchical structure. The current correlation between the mechanical properties and the hierarchical structure was mainly established by studying altered forms of dentin, which limits the potential outcome of the research. In this study, dentins with three different hierarchical structures were obtained via two different remineralization procedures and at different remineralization stages: (1) a dentin structure with amorphous minerals incorporated into the collagen fibrils, (2) a dentin with crystallized nanominerals incorporated into the collagen fibrils, and (3) a dentin with an out-of-order mineral layer filling the collagen fibrils matrix. Nanoindentation tests were performed to investigate the mechanical behavior of the remineralized dentin slides. The results showed that the incorporation of the crystallized nanominerals into the acid-etched demineralized organic fibrils resulted in a remarkable improvement of the mechanical properties of the dentin. In contrast, for the other two structures, i.e. the amorphous minerals inside the collagen fibrils and the out-of-order mineral layer within the collagen fibrils matrix, the excellent mechanical properties of dentin could not be restored.

Polyaspartic Acid-Calcium-Lanthanum Complexes Induce Antibacterial Remineralization of Dentin and In-Depth Occlusion of Dentinal Tubules

Ling Zhu,Wei Liu,Yizhou Zhang,Zhifang Wu,Yiru Wang,Yuedan Xu,Haiyan Zheng,Hongli Zhang,Mengfei Yu,Xiaoting Jin,Zhe Wang,Zihuai Zhou,Baiping Fu

Dental caries is a global oral dilemma leading to demineralization of tooth hard tissues and exposure of dentinal tubules (DTs). Multifunctional restorative strategies, including remineralization, antibacterial properties, and DT occlusion, are urgently needed for dental caries management but remain a significant challenge. Herein, the polyaspartic acid-calcium-lanthanum suspension (5 g L −1 -3.06 м-0.34 м) followed by phosphate solution (2.04 м) is adopted to conduct polyelectrolyte-cation complexes pre-precursor process. This strategy, for the first time, successfully induces La-doped intrafibrillar mineralization of collagen fibrils with lanthanum-doped hydroxyapatite (La-HAp). After sequential application of two mineralization media, the demineralized dentin is fully remineralized and the DTs are deeply occluded to a depth of 100 µm with La-HAp after 7 d of incubation in artificial saliva or oral cavity of rabbit. The remineralized dentin demonstrates antibacterial properties against cariogenic bacteria ( Streptococcus mutans ) both in vitro and in vivo, and its mechanical properties are almost restored to those of intact dentin. The mineralization media cause no apparent irritation to oral mucosa or dental pulp. Hence, this multifunctional strategy confers the remineralized dentin surface with antibacterial properties and blocks the invasion of cariogenic bacteria through DTs, providing insight into the clinical management of dentin caries accompanied by dentin hypersensitivity.

Lanthanum-Loaded ZIF Nanomaterials for Caries Prevention

Han Xiao,Yuping Qian,Lijun Zhao,Yiman Tang,Yuan Pan,Yunzi Long,Shihan Li,Yukun Chen,Jiale Sun,Chao Yuan,Ludan Zhang,Wenshu Ge

Dental caries represents one of the most widespread oral health challenges worldwide, impacting people of every age demographic. Traditional anticaries strategies primarily rely on fluoride, yet its chronic overuse may lead to health issues such as dental fluorosis. Lanthanum (La), a rare earth element, has emerged as a promising fluoride alternative due to its low toxicity and strong anticaries activity. However, studies have shown that the direct use of free lanthanum under physiological conditions may cause cellular structural damage, dose-dependent hemolytic reactions, and even liver and kidney fibrosis, highlighting the urgent need to optimize its delivery method. In this study, zeolitic imidazolate framework nanoparticles (La@ZIF) were used to encapsulate lanthanum, enabling its controlled and sustained release, thereby proposing a novel fluoride-free anticaries strategy. Three La@ZIF nanoparticles with varying La incorporation levels (20%, 40%, and 60%) were synthesized via a one-pot method and systematically characterized. Results demonstrated their excellent degradation performance and acid-neutralizing capacity under acidic conditions. In vitro experiments confirmed the favorable biocompatibility of La@ZIF and its ability to effectively promote enamel remineralization and restore surface hardness, achieving comparable efficacy to traditional fluoride treatments. In vivo studies further validated the remineralization potential of La@ZIF in a rat model, with no adverse effects observed on major organs. La@ZIF nanoparticles exhibit remarkable anticaries performance and biosafety, offering a new direction for developing fluoride-free anticaries materials.

The glycerol stabilized calcium phosphate cluster for rapid remineralization of tooth enamel by a water-triggered transformation

Luo Nan,Lu Bing-Qiang,Deng Yu-Wei,Zeng Hua,Zhang Yu,Zhan Jing-Yu,Xu Xiao-Chen,Cao Gui-Zhi,Wen Jin,Zhang Zhiyuan,Feng Xi-Ping,Jiang Xinquan,Chen Feng,Chen Xi

Remineralization is a common strategy for the repair of early demineralized tooth enamels, but the harsh dynamic oral environment often hampers its efficacy. Rapid remineralization is expected to address this challenge, however, the stabilizers of remineralization materials often resist their transformation required for repair. Here, by dissolving the ions of calcium and phosphate in glycerol-dominant solvents, we obtain the calcium phosphate clusters (1–2 nm), which are stabilized by glycerol (with high viscosity and affinity to clusters), but can perform a fast enamel repair via the water-triggered transformation in both static and dynamic environments. Upon the in vitro and in vivo (female Sprague-Dawley rats) studies, the clusters swiftly enter the nano-/micro-sized enamel defect sites, then form a compact hydroxyapatite repair layer within a short time (30 min, much faster than the conventional materials), and significantly recovers mechanical properties. This material is promising for large-scale preparation and applications in dental remineralization.

Polydopamine Promotes Dentin Remineralization via Interfacial Control

Yinan Qu,Tianyi Gu,Qiaolin Du,Changyu Shao,Jing Wang,Biao Jin,Weijing Kong,Jian Sun,Chaoqun Chen,Haihua Pan,Ruikang Tang,Xinhua Gu

Biomineralization has intrigued researchers for decades. Although mineralization of type I collagen has been universally investigated, this process remains a great challenge due to the lack of mechanistic understanding of the roles of biomolecules. In our study, dentine was successfully repaired using the biomolecule polydopamine (PDA), and the remineralized dentine exhibited mechanical properties comparable to those of natural dentine. Detailed analyses of the collagen mineralization process facilitated by PDA showed that PDA can promote intrafibrillar mineralization with a decreased heterogeneous nucleation barrier for hydroxyapatite (HAP) by reducing the interfacial energy between collagen fibrils and amorphous calcium phosphate (ACP), resulting in the conversion of an increasing amount of nanoprecursors into collagen fibrils. The present work highlights the importance of interfacial control in dentine remineralization and provides profound insight into the regulatory effect of biomolecules in collagen mineralization as well as the clinical application of dentine restoration.

Modified casein-stabilized amorphous calcium phosphate nanoparticles prevent dental caries by inhibiting the growth of Streptococcus mutans and promoting the remineralization of tooth enamel

Pengchao Fang,Xiaoxing Ye,Leijie Tian,Yibiao Chen,Xiaolan Li,Haiyan Hu

Dental caries, a prevalent oral disease, is caused by acid- producing plaque microorganisms-notably Streptococcus mutans ( S. mutans )-which drive enamel demineralization. Current prevention strategies, such as fluoride and chlorhexidine, primarily target planktonic bacteria but exhibit limited efficacy against biofilms and neglect enamel remineralization, thereby reducing their therapeutic efficacy. To address these limitations, we developed dual-functional nanoparticles (AXCP NPs) that can eradicate S. mutans biofilm and promote enamel remineralization. The amphiphilic polymer Arg-XOS-CPP (AXC) was synthesized by covalently conjugating oligosaccharide carbonyl groups to the amino residues of L-arginine and casein. Self-assembly of AXC with calcium and phosphate ions yielded hybrid nanoparticles (AXCP NPs). Experimental results indicated that AXCP NPs had no effect on the viability of HaCaT cells. Under acidic conditions, protonation of arginine triggered a surface charge reversal from negative to positive, enabling targeted binding to negatively charged biofilms. As expected, AXCP NPs exhibited potent anti- S. mutans activity (MIC: 640   μg/mL) and remarkable biofilm eradication efficacy (>80% clearance). In vitro release and remineralization assays demonstrated that AXCP NPs continuously released calcium and phosphorus ions, regulating the mineralization process and effectively restoring enamel functionality. Furthermore, in mice caries model, localized application of AXCP NPs significantly suppressed biofilm accumulation on tooth surface and restored mineral density in demineralized enamel, inhibiting caries progression.

Influence of Polyphosphate on the Mineralization Balance of Tooth Enamel

Jing Ru,Xiaochen Xu,Yuxuan Cheng,Nan Luo,Shuo Tan,Xi Chen,Feng Chen,Bing-Qiang Lu

Dental minerals are in an equilibrium state of demineralization and remineralization, which can be disrupted by pathogenic bacteria to cause dental caries. While the inorganic polymer polyphosphate (polyP) is ubiquitous in living organisms and is also widely involved in mineralization regulations, its specific influence on the mineralization balance of teeth remains unclear. As a concept-and-proof study, the effects of polyP on the demineralization and remineralization of teeth are investigated on dental enamel (the highly mineralized outer covering tissue of teeth) from the perspective of mineralization balance. We found that a high concentration (containing 1.0–20 mM P element, comparable to and higher than the free phosphate ions in body fluids) of polyP has the capability to demineralize enamel in the aqueous solution, yet this effect is absent in the simulated biological environments including simulated body fluid and MEM (α-minimum essential medium) solutions. More importantly, polyP with a very low concentration (containing ≥5.0 μM P) is able to inhibit enamel mineralization significantly. This suggests that polyP could impact the mineralization balance of enamel by preferentially inhibiting the remineralization process, thereby disrupting the equilibrium necessary for maintaining enamel health.

Synthesis and characterization of an anti-caries and remineralizing fluorine-containing cationic polymer PHMB-F

Y. D. Chen,C. Shu,Z. H. Duan,J. J. Xu,X. J. Li,F. Chen,Q. J. Luo,X. D. Li

Dental caries have become a major global public health problem. Plaque control and remineralization of initial enamel lesions are paramount for the prevention and control of caries. Polyhexamethylene biguanide (PHMB) is a type of cationic amphipathic antibacterial agent with broad-spectrum antibacterial properties and good biological safety. Fluoride delays demineralization and promotes the remineralization of hard dental tissues. However, a high concentration is needed for it to function as an antibacterial agent. In order to create a PHMB with the benefits associated with fluoride, we synthesized a fluorine-containing cationic polymer, PHMB-F. Fourier transform-infrared spectroscopy and solid state nuclear magnetic resonance characterization confirmed the successful synthesis of PHMB-F. Antibacterial tests showed that PHMB-F had better antiseptic efficacy for Streptococcus mutans compared with just PHMB. Moreover, positively-charged PHMB-F allows fluoride ions to exist closer to the enamel surface with negative potential, which markedly lowers the ion concentrations in the microenvironment adjacent to hard dental tissues needed to maintain equilibrium. Thus, only low concentrations of PHMB-F are required for enamel remineralization.

Polyzwitterion Manipulates Remineralization and Antibiofilm Functions against Dental Demineralization

Jiankang He,Jianhai Yang,Min Li,Yachong Li,Yanyun Pang,Jiayin Deng,Xu Zhang,Wenguang Liu

Biomineralization technology has become a trend for the arrest and prevention of dental caries. In particular, the bioactivity and ability to release large amounts of Ca2+ and PO43– ions make amorphous calcium phosphate (ACP) for hard tissue remineralization are highly desired. However, the instability of ACP limits its clinical application. Under continuous bacterial challenge in the oral cavity, the currently developed ACP-based remineralization system lacks the ability to inhibit bacterial adhesion and biofilm formation. Here, a dual-functional nanocomposite with antibiofilm and remineralization properties was designed by combining zwitterionic poly(carboxybetaine acrylamide) (PCBAA) and ACP. The resulting nanocomposite was stable in solution for at least 3 days without any aggregation. The PCBAA/ACP nanocomposite exerted a significant inhibitory effect on the adhesion and biofilm formation of Streptococcus mutans and exhibited bactericidal activities under acidic conditions resulting from bacteria. Moreover, compared with fluoride, this nanocomposite demonstrated superior effects in promoting the remineralization of demineralized enamel and the occlusion of exposed dentinal tubules in vivo and in vitro. The present work provides a theoretical and experimental basis for the use of the PCBAA/ACP nanocomposite as a potential dual-functional agent for arresting and preventing caries.
