Comparative pharmacokinetic study of twelve phenolic acids and flavonoids from red wine between control and coronary heart disease model rats by UFLC-MS/MS

Yu-Ye GAO,Xie-An YU,Bing WANG,Li-Fei GU,Yuan-Yuan GE,Guang-Lin ZHU,Kun-Hui SUN,Yi LU,Tie-Jie WANG,Kai-Shun BI

Red wine was famous for the potential effects on coronary heart disease (CHD), but the pharmacokinetic study of the main active compounds was unclear. In this research, a simple, rapid and sensitive ultra-fast liquid chromatography coupled with triple quadrupole mass spectrometry (UFLC-MS/MS) method was established for simultaneous determination of six phenolic acids and six flavonoids after oral administration of red wine in rat plasma. Moreover, the pharmacokinetic differences of the six active compounds in control and CHD model rats after oral administration of red wine were investigated. Rosmarinic acid and puerarin were selected as internal standards. Extraction of plasma sample was by protein precipitation using acetonitrile . An HSS T3 column (1.8 µm, 2.1 mm × 100 mm) was used to separation at a flow rate of 0.3 mL/min with mobile phase consisting of 0.1% formic acid aqueous solution and 0.1% formic acid acetonitrile. The developed method was successfully applied to comparative pharmacokinetic study between control and CHD model rats. The results showed that some pharmacokinetic parameters were significantly different among the control and CHD model rats. Accordingly, it was expected that the results of this study could provide a scientific basis for further exploring red wine for the prevention and treatment of CHD.

The negative effect of ANGPTL8 on HDL-mediated cholesterol efflux capacity

Luo Mengdie,Zhang Ziyu,Peng Yani,Wang Shuai,Peng Daoquan

Background It is well known that angiopoietin-like protein 8 (ANGPTL8) exerts its effects on lipid metabolism through the inhibition of lipoprotein lipase and subsequent elevation of plasma triglyceride. However, it is not clear whether ANGPTL8 could affect lipid metabolism via other pathways. The study was aimed to investigate the effects of ANGPTL8 on the function of high-density lipoprotein (HDL), which plays a protective role in atherosclerosis progression. Methods Two hundred and ten subjects were recruited. Plasma ANGPTL8 was measured by enzyme-linked immunosorbent assays. Cholesterol efflux capacity was chosen as the biomarker of HDL function and measured via H3-cholesterol loading THP-1 cell models. Results ANGPTL8 exhibited no significant difference between CAD group and nonCAD group, but ANGPTL8 in DM group was significantly higher than that in the nonDM group [568.3 (406.2–836.8) vs 458.2 (356.8–755.6), P = 0.023]. Compared to controls, subjects in CAD group and DM group exhibited significantly lower cholesterol efflux capacity [CAD: 14.58 ± 2.06 vs 12.51 ± 2.83%, P < 0.0001; DM: 13.62 ± 2.57 vs 12.34 ± 3.16%, P = 0.0099]. ANGPTL8 was inversely correlated with cholesterol efflux capacity (r = − 0.188, P < 0.01). Regression analysis revealed that plasma ANGPTL8 was an independent contributor to cholesterol efflux capacity (standardized β = − 0.143, P = 0.023).Conclusion ANGPTL8 presents a negative effect on HDL-mediated cholesterol efflux capacity.

Association of Coronary Artery Disease and Metabolic Syndrome: Usefulness of Serum Metabolomics Approach

Jing Ziwei,Liu Liwei,Shi Yingying,Du Qiuzheng,Zhang Dingding,Zuo Lihua,Du Shuzhang,Sun Zhi,Zhang Xiaojian

Introduction: Individuals with metabolic syndrome (MetS) are at increasing risk of coronary artery disease (CAD). We investigated the common metabolic perturbations of CAD and MetS via serum metabolomics to provide insight into potential associations. Methods: Non-targeted serum metabolomics analyses were performed using ultra high-performance liquid chromatography coupled with Q Exactive hybrid quadrupole-orbitrap high-resolution accurate mass spectrometry (UHPLC-Q-Orbitrap HRMS) in samples from 492 participants (272 CAD vs. 121 healthy controls (HCs) as cohort 1, 55 MetS vs. 44 HCs as cohort 2). Cross-sectional data were obtained when the participants were recruited from the First Affiliated Hospital of Zhengzhou University. Multivariate statistics and Student’s t test were applied to obtain the significant metabolites [with variable importance in the projection (VIP) values >1.0 and p values <0.05] for CAD and MetS. Logistic regression was performed to investigate the association of identified metabolites with clinical cardiac risk factors, and the association of significant metabolic perturbations between CAD and MetS was visualized by Cytoscape software 3.6.1. Finally, the receiver operating characteristic (ROC) analysis was evaluated for the risk prediction values of common changed metabolites. Results: Thirty metabolites were identified for CAD, mainly including amino acids, lipid, fatty acids, pseudouridine, niacinamide; 26 metabolites were identified for MetS, mainly including amino acids, lipid, fatty acids, steroid hormone, and paraxanthine. The logistic regression results showed that all of the 30 metabolites for CAD, and 15 metabolites for MetS remained significant after adjustments of clinical risk factors. In the common metabolic signature association analysis between CAD and MetS, 11 serum metabolites were significant and common to CAD and MetS outcomes. Out of this, nine followed similar trends while two had differing directionalities. The nine common metabolites exhibiting same change trend improved risk prediction for CAD (86.4%) and MetS (90.9%) using the ROC analysis. Conclusion: Serum metabolomics analysis might provide a new insight into the potential mechanisms underlying the common metabolic perturbations of CAD and MetS.

Magnetic Ligand Fishing Using Immobilized Cyclooxygenase-2 for Identification and Screening of Anticoronary Heart Disease Ligands From Choerospondias axillaris

Chi Miaomiao,Wang Hongsen,Yan Zhankuan,Cao Lei,Gao Xun,Qin Kunming

Inhibition of cyclooxygenase-2 (COX-2) activity is an effective way for treatment of coronary heart disease. And as an important source of COX-2 inhibitors, bioactive compounds of Choerospondias axillaris and pharmacological mechanisms remained lacking in prospective researches. Therefore, for the purpose of accelerating the discovery of natural products targeting designed inhibitors, the COX-2 microreactor composed of functionalized microspheres and magnetic ligand fishing was developed and applied in Choerospondias axillaris, and the physicochemical properties of the COX-2 functionalized microspheres were characterized using Fourier transform infrared spectroscopy (FT-IR), vibrating sample magnetometer (VSM), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). Furthermore, the bioactive compounds singled out from ethanol decoction without prepurification were dissociated and identified by ultraperformance liquid chromatography plus Q-Exactive Orbitrap tandem mass spectrometry (UPLC-Q-Exactive Orbitrap-MS/MS). Consequently, 21 bioactive compounds consisting of 6 organic acids, 8 flavonoids, and 7 others were separated and characterized from Choerospondias axillaris, which were reported to participate in the COX-2 inhibitory pathway to varying degrees. Therefore, this method could provide a prospective solution for the extraction and identification of active pharmaceutical ingredients and the rapid screening of some enzyme inhibitors in the complex mixtures.

Nonoccupational Exposure to Pyrethroids and Risk of Coronary Heart Disease in the Chinese Population

Jiajun Han,Liqin Zhou,Mai Luo,Yiran Liang,Wenting Zhao,Peng Wang,Zhiqiang Zhou,Donghui Liu

Pyrethroids and the metabolites have been frequently observed in the environment. Animal data suggests that pyrethroids can induce adverse effect on the cardiovascular system but there are no human studies examining pyrethoids exposure as a risk for coronary heart disease (CHD). We analyzed three nonspecific pyrethroids metabolites in urine and studied the association with CHD risk. A total of 72 CHD patients and 136 healthy subjects were recruited in Shanxi province in China from 2013 to 2014 by matching age and gender. The median concentrations of urinary cis-CDDA (cis-3-(2,2-dichlorovinyl)-2,2-dimethyl cyclopropane carboxylic acid), trans-CDDA (trans-3-(2,2-dichlorovinyl)-2,2-dimethyl cyclopropane carboxylic acid) and 3-PBA (3-phenoxybenzoic acid) among healthy subjects were 1.03, 0.42, 0.74 μg/L respectively, while the median concentrations of the three metabolites among CHD patients were 1.93, 1.07, 1.09 μg/L respectively, significantly higher than healthy subjects. Upper tertile of urinary pyrethroid metabolites were associated with an increased risk of CHD compared with the lowest tertile (cis-CDDA: ORT3vsT1 = 6.86, 95% CI: 2.76–17.06, p-trend = 0.000; trans-CDDA: ORT3vsT1 = 6.94; 95% CI: 2.80–17.19; p-trend =0.000; 3-PBA: ORT3vsT1 = 3.62; 95% CI: 1.48–8.88; p-trend = 0.009; total pyrethroid metabolites: ORT3vsT1 = 4.55; 95% CI: 1.80–11.54; p-trend = 0.002). This study provides information on pyrethroids exposure in China and reveals a possible positive association between pyrethroids exposure and the risk of coronary heart disease.

Sensitive Electrochemical Immunosensor Based on MXene-Nafion/Polyaniline (PANI) for the Sensitive Determination of the Novel Biomarker sST2 for Atherosclerotic Coronary Heart Disease by Differential Pulse Voltammetry (DPV)

Jian Huang,Guolin Yuan,Dongliang Cheng,Fei Xie,Xianru Xia,Xiandong Li,He An,Chunyan Peng

Atherosclerotic coronary heart disease (CHD) is ischemic and a worldwide health issue. The soluble suppression of tumorigenicity 2 (sST2) is a novel biomarker for atherosclerotic coronary heart disease. This work develops an immunosensor with a nanocomposite consisting of polyaniline (PANI) and MXene-Nafion for the specific and sensitive determination of sST2. PANI was first electropolymerized on a glassy carbon electrode (GCE) to enhance electron transfer. Subsequently, MXene-Nafion dispersion was drop cast on the PANI/GCE substrate to further increase the electrical conductivity and immobilize anti-sST2 which captures sST2 for detection. The current response as a function of sST2 concentration was measured by differential pulse voltammetry (DPV). Under the optimal conditions, this immunosensor demonstrates a linear range from 1 pg mL−1 to 1 μg mL−1 and a detection limit of 0.149 pg mL−1. The device offers high sensitivity, excellent specificity, satisfactory reproducibility, good stability, and has significant potential for detecting clinical biomarkers. In addition, the immunosensor is able differentiate between serum of healthy people and CHD patients.

Novel Electrochemiluminescent Immunosensor Using Dual Amplified Signals from a CoFe Prussian Blue Analogue and Au Nanoparticle for the Detection of Lp-PLA2

Lixin Wang,Yuhong Liu,Jilin Yan,Huiling Li,Yifeng Tu

Coronary heart disease (CHD) poses an important threat to human health, and its pathogenesis is the formation of atheromatous plaques in coronary ventricles. Compared to other biomarkers, lipoprotein-associated phospholipase A2 (Lp-PLA2), which is involved in multiple processes of atherosclerosis, is a noticeable inflammatory biomarker related to CHD. Herein, using a multifunctional nanocomposite containing a CoFe Prussian blue analogue (PBA) and Au nanoparticles (AuNPs) (AuNPs@CoFe PBA) as a sensing substrate, an electrochemiluminescent (ECL) immunosensor was developed for the highly sensitive detection of Lp-PLA2. Benefiting from the synergistic effect of the PBA and AuNPs, the nanocomposite exhibits excellent peroxidase-like activity and can catalyze the luminol–ECL reaction, amplifying the ECL signal by ∼29-fold. Meanwhile, the enlarged specific surface area of the nanocomposite and the presence of abundant AuNPs allow the immobilization of more antibody proteins, thereby improving the sensing response of the immunosensor. When the target Lp-PLA2 is captured by the antibody on the sensor surface, the sensor emits a reduced ECL signal because of the increased mass and electron transfer resistance due to the formation of the immune complex. Under optimized conditions, the constructed ECL immunosensor exhibits a broad linear range from 1 to 2200 ng/mL and a low detection limit of 0.21 ng/mL. Additionally, the ECL immunosensor exhibits high specificity, stability, and reproducibility. This work provides a new approach to diagnose CHD and broadened the application of the PBA in the field of ECL sensors.

Luteolin Inhibits Ferroptosis of HUVEC by Regulating the Sirt1/Nrf2 Pathway

Xiang Ming,Lin Xiangdong,Chen Haiying

Coronary heart disease (CHD) is a disease caused by organic and functional coronary artery stenosis, resulting in a reduced oxygen supply to the heart. This study aimed to investigate the mechanism via which Luteolin regulates the Sirt1 / Nrf2 pathway to inhibit ferroptosis in human umbilical vein endothelial cells (HUVEC) associated with CHD. An ox-LDL-induced HUVEC cell model with Sirt1 silencing and Luteolin treatment was established. The silencing efficiency of Sirt1 was validated using quantitative reverse transcription polymerase chain reaction (qRT-PCR) and Western blot analysis. The results of molecular docking and DARTS experiments showed that Luteolin could effectively bind Sirt1 . Subsequently, we measured the expression of Nrf2 and Sirt1 , cell viability, MDA, GSH, Fe 2+ levels, lipid ROS content, expression of ferroptosis-related proteins GPX4, FTH, FTL, and cell migration parameters. The results showed that Luteolin could activate the Sirt1 / Nrf2 axis and effectively inhibit ox-LDL-induced ferroptosis in HUVEC. Experimental results revealed that Luteolin could enhance HUVEC cell viability, decrease MDA, Fe 2+ , and ROS levels, increase GSH levels, promote the expression of HO-1, GPX4, FTH, FTL, inhibit the expression of ACSL4 and TFRC, and enhance the migration capability of HUVEC cells. Moreover, silencing Sirt1 reversed the effects of Luteolin on the activation of Sirt1 and Nrf2 , confirming the dependence of these effects on the Sirt1 / Nrf2 signaling pathway. In conclusion, this study indicates that Luteolin could inhibit ferroptosis in HUVEC in CHD by modulating the Sirt1 / Nrf2 axis, providing a basis for further research on strategies for preventing and treating CHD and related diseases.

Screening of lipid metabolism biomarkers in patients with coronary heart disease via ultra-performance liquid chromatography–high resolution mass spectrometry

Fang Cai,Fandong Ren,Yunmei Zhang,Xiaoxue Ding,Guanghui Fu,Dabing Ren,Lijuan Yang,Ning Chen,Ying Shang,Yongdan Hu,Lunzhao Yi,Hong Zhang

Coronary heart disease (CHD) has a high mortality worldwide. This study aimed to screen lipid metabolism biomarkers in patients with coronary heart disease via ultra-performance liquid chromatography–high resolution mass spectrometry. Extraction and reconstitution solvents, liquid chromatographic and mass spectrometry conditions were optimized to detect more plasma lipid metabolites. In this study, the chromatographic and mass spectra characteristics of lipid metabolites were summarized. A total of 316 lipid metabolites were annotated via diagnostic fragment ion filtration, nitrogen rule filtration, and neutral loss filtration. Glycerophospholipid metabolism and sphingolipid metabolism were revealed as the main lipid disorders of CHD. This study provides a novel insight for high-throughput detection of lipid metabolites in plasma and provides a further understanding of the occurrence of CHD, which can provide valuable suggestions for the prevention of CHD.

Rapid identification, quantitation, and antioxidant activity evaluation of the components in Guanxin Shutong capsule with liquid chromatography and mass spectrometry

Ming Meng,Haofang Wan,Yida Bao,Yu He,Chang Li,Haitong Wan

Guanxin Shutong capsule (GSC) is a traditional Chinese medicinal prescription used in the treatment of coronary heart disease (CHD) and angina pectoris in clinic. However, the chemical profile of GSC is still uncovered, which hindered the progress of pharmacological study and clinical application. Herein, high performance liquid chromatography (HPLC) together with high resolution mass spectrometry (HR-MS) techniques were employed to analyze the quality consistency and to identify chemical components in GSC. As a result, a total of 111 compounds were tentatively annotated. Quantitative analysis based on HPLC-ultraviolet detection (UV) was performed for 6 main components and fingerprints of 10 different batches of GSC were established. The developed method was validated for linearity, precision, repeatability, stability, and recovery. The quality evaluation and similarity analysis of the 10 batches were also performed. Furthermore, in vitro antioxidant activity assays demonstrated that GSC exhibited potential DPPH and hydroxyl radical scavenging capacities. Especially, salvianolic acids showed the strongest free radical scavenging capacities, which might be the main component for quality control of GSC.

Study of Metoprolol-Loaded Poly(Lactic-co-Glycolic Acid)–Poly (Trimethylene Carbonate)–Poly(Glycolic Acid) Drug-Eluting Stents for Coronary Heart Disease

Rongrong Lu,Aimaitijiang Maimaiti,Tuohetiajimu Abudureheman

Background and Objective:Drug-eluting stents (DES) have become a crucial strategy for improving the outcomes of patients with coronary artery disease (CAD). This work aimed to evaluates the therapeutic efficacy of metoprolol-loaded poly (lactic-co-glycolic acid)–poly(trimethylene carbonate)–poly (glycolic acid) (PLGA–PTMC–PGA) DES (Meto-PLGA/PTMC DES) in a rabbit model of CAD.Methods:A total of 30 New Zealand white rabbits (male, weighing 4–6 kg) were randomly divided into three groups: Sham group (chest suturing without intervention), Model group (CAD animal model without DES), and stent group (treatment with the Meto-PLGA/PTMC DES). The blood samples were collected at regular intervals to assess the serum inflammatory markers, cardiac function parameters, hemodynamic parameters and coronary remodeling. Statistical analysis was performed using One-way analysis of variance (ANOVA), followed by a post hoc Tukey’s test to compare the differences between groups (p < 0.05 indicated statistically significance). Results:The Meto-PLGA/PTMC DES, through its unique co-polymer structure, achieved stable Meto loading in vivo. This stent enabled the gradual release of Meto, thus maintaining sustained drug concentrations and optimizing CAD treatment. The animal experiment results indicated that the sent group (Meto-loaded DES) exhibited markedly lower levels of interleukin-8 (IL-8), tumor necrosis factor-alpha (TNF-α), vascular cell adhesion molecule-1 (VCAM-1), and intercellular adhesion molecule-1 (ICAM-1) relative to the model group (p < 0.05). In terms of cardiac function, compared with the model group, the stent group exhibited significantly elevated left ventricular systolic pressure (LVSP), ±dp/dtmax, arterial systolic blood pressure (BPs), and diastolic blood pressure (BPd), along with a marked reduction in left ventricular end-diastolic pressure (LVEDP) compared with the model group (p < 0.05). The coronary tissue morphology in the stent group revealed notably reduced intimal thickness, intimal area and degree of stenosis versus the Model group, with a prominent increase in lumen area (p < 0.05).Conclusion:The Meto-PLGA/PTMC DES not only effectively provides sustained drug release and exhibits superior mechanical properties and excellent blood compatibility. Animal experiments further validated the crucial role of the Meto-PLGA/PTMC DES in reducing inflammatory responses, improving cardiac function and alleviating coronary artery stenosis.
