In this study, a self-cooling laboratory system was used for pressure–shift freezing (PSF), and the effects of pressure–shift freezing (PSF) at 150 MPa on the quality of largemouth bass (Micropterus salmoides) during frozen storage at −30 °C were evaluated and compared with those of conventional air freezing (CAF) and liquid immersion freezing (LIF). The evaluated thawing loss and cooking loss of PSF were significantly lower than those of CAF and LIF during the whole frozen storage period. The thawing loss, L* value, b* value and TBARS of the frozen fish increased during the storage. After 28 days storage, the TBARS values of LIF and CAF were 0.54 and 0.65, respectively, significantly higher (p< 0.05) than the 0.25 observed for PSF. The pH of the samples showed a decreasing trend at first but then increased during the storage, and the CAF had the fastest increasing trend. Based on Raman spectra, the secondary structure of the protein in the PSF-treated samples was considered more stable. The α-helix content of the protein in the unfrozen sample was 59.3 ± 7.22, which decreased after 28 days of frozen storage for PSF, LIF and CAF to 48.5 ± 3.43, 39.1 ± 2.35 and 33.4 ± 4.21, respectively. The results showed that the quality of largemouth bass treated with PSF was better than LIT and CAF during the frozen storage. Keywords:high pressure freezing;specialty freezing techniques;largemouth bass;freezing preservation;frozen food quality

The cryopreservation of red blood cells (RBCs) holds great potential for ensuring timely blood transfusions and maintaining an adequate RBC inventory. The conventional cryoprotectants (CPAs) have a lot of limitations, and there is an obvious need for novel, efficient, and biocompatible CPAs. Here, it is shown for the first time that the addition of dimethylglycine (DMG) improved the thawed RBC recovery from 11.55 ± 1.40% to 72.15 ± 1.22%. We found that DMG could reduce the mechanical damage by inhibiting ice formation and recrystallization during cryopreservation. DMG can also scavenge reactive oxygen species (ROS) and maintain endogenous antioxidant enzyme activities to decrease oxidative damage during cryopreservation. Furthermore, the properties of thawed RBCs were found to be similar to the fresh RBCs in the control. Finally, the technique for order performance by similarity to ideal solution (TOPSIS) was used to compare the performance of glycerol (Gly), hydroxyethyl starch (HES), and DMG in cryopreservation, and DMG exhibited the best efficiency. This work confirms the use of DMG as a novel CPA for cryopreservation of RBCs and may promote clinical transfusion therapy. Keywords:cryopreservation;dimethylglycine;red blood cells;cryoprotectant

Over the past 40 years there has been a worldwide critical change in the field of assisted reproduction technology (ART), leading to the increased application of single blastocyst transfer, which is extremely important to avoid the risks of multiple pregnancy and associated complications for both mother and babies. Indeed, advancements in ART over the last few decades have been obtained thanks to several improvements, including ovarian stimulation, embryo culture conditions and, of course, progress in cryopreservation methods, especially with the application of vitrification. The ability to cryopreserve human embryos has improved significantly with vitrification compared to the initially adopted slow-freezing procedures. Since the introduction of vitrification, it has become the gold standard method to effectively cryopreserve human blastocysts. However, some new protocols are now being explored, such as the short warming procedure and even shorter exposure to the equilibration solution before vitrification, which seem to provide optimal results. Therefore, the main aim of the current narrative review, will be to illustrate the benefit of vitrification as an effective method to cryopreserve the human blastocyst and to illustrate new protocols and variations which in future may increase the performance of vitrification protocols.

Poly(ethylene glycol) (PEG) cryogels with aligned porous structures were prepared by unidirectional freezing and subsequently cryopolymerization. The aqueous reaction precursor of poly(ethylene glycol) diacrylate (PEGDA) and the initiator was first unidirectionally frozen using liquid nitrogen or other cooling agents such as frozen ethyl alcohol and frozen acetic ether, followed by radical polymerization at subzero temperature. The morphology of the obtained cryogels was observed by scanning electron microscopy (SEM) and confocal laser scanning microscope (CLSM). The pore structure of the obtained cryogels was the replica of the unidirectional ice crystals formed during the unidirectional freezing. Microtubular pores aligned along the freezing direction were observed in both dry and swollen states. The obtained cryogels showed anisotropic compressive strength according to the pore directions. The effect of the PEGDA molecular weight and the freezing temperature on morphology of the orientation-structured cryogels were also studied. The pore diameter could be adjusted from 10 to 50 μm.

The rapid freezing technique for porous foods using tissue pre-degassing followed by ultrasound-assisted freezing (UF) was developed, and its effects on quality attributes of radishes including tissue air volume, hardness, total calcium contents, bonded calcium contents, retention rates of bonded calcium and microstructures were investigated. To evaluate the freezing efficiency, parameters including total freezing time, phase transition time, and the increases of freezing rate and phase transition rate were determined. Besides, multivariate statistical analyses including principal component analysis (PCA) and hierarchical cluster analysis (HCA) were performed to visualize and further analyze the quality differences of radishes under different treatments. Results suggested that decreasing tissue air volumes can significantly shorten the phase transition time of UF. Samples treated by pre-degassing for 5 min at −0.09 MPa followed by UF ( D - 0.09 M P a 5 m i n -UF) showed the freezing rate and phase transition rate increased by 28.8% and 29.8%, respectively, as compared with the same pre-degassed samples frozen by immersion freezing ( D - 0.09 M P a 5 m i n -IF). Retention rates of bonded calcium positively correlated with the sample hardness, announcing the importance of bonded calcium maintenance during radish freezing. Both PCA and HCA indicated that D - 0.09 M P a 5 m i n -UF endowed radishes with quality attributes more similar to the fresh ones, which was further verified by microstructure analysis, showing remarkably alleviated plasma membrane puncture, cell separation and deformation in D - 0.09 M P a 5 m i n -UF samples. The current study proved that the technique of tissue pre-degassing followed by UF could effectively improve the freezing efficiency and quality attributes of frozen radishes, and thus have great potentials in rapid freezing of porous foods.

This study aimed to analyze the cryoprotective effect of a ovalbumin (OVA) and β-cyclodextrin (βCD) mixture (3:1, OVA/βCD) on the structure, rheology and gelling properties of myofibrillar proteins (MPs) during 90 days of frozen storage. A mixture of OVA/βCD at different concentrations (0, 2, 4, and 6%) was added to MPs and stored at −18 °C for 90 days. The addition of OVA/βCD significantly decreased the sulfhydryl contents while it increased the surface hydrophobicity, which was closely connected with tertiary structural changes. Circular dichroism analysis showed that the addition of OVA/βCD enhanced the stability of the secondary structure by inhibiting the decline in the α-helix. Rheological properties analysis indicated that 6% OVA/βCD treatment showed better storage modulus (G’) and loss modulus (G”). In addition, treatment of OVA/βCD showed better gel forming properties than the control group (0%), helping to form a homogeneous and denser gel network. The results proved that 6% OVA/βCD could be act as a promising cryoprotectant, which can improve the structure and gel behavior ofCulter alburnusMPs during frozen storage. Moreover, OVA/βCD could be a potential alternative to conventional cryoprotectants at the industrial level to increase the economic and commercial values of seafood products. Keywords:myofibrillar proteins;ovalbumin;β-cyclodextrin;cryoprotectant;frozen storage

Poly(ethylene glycol)-poly(D,L-lactide) (PEG-PLA), a type of copolymer widely used for drug delivery, was synthesized in the present work based on previous studies, which is used for bovine serum albumin (BSA)-loaded micelle formulation. The micelles exhibited less than 100 nm in size with narrow polydispersity indices (PDI) of smaller than 0.3, and showed an encapsulation efficiency of 69%. BSA was encapsulated in the PEG-PLA micelle successfully exhibiting intact structure indicated by native polyacrylamide gel electrophoresis (Native-PAGE), fluorescence spectroscopy and fourier transform infrared spectroscopy (FTIR) analysis methods. It investigated the effects of freeze-drying, varying amount of cryoprotectants (sucrose, mannitol and lactose), and ultrasonication treatment on the size, and size distribution. The micelles were agglomerated in the freeze-drying process demonstrated by transmission electron microscope (TEM) and dynamic light scattering (DLS) analysis. Five percent of mannitol and ultrasonication for 90 s were found producing a significant reduction in agglomeration as well as a sharp decrease in size. The results demonstrated suitable cryoprotectant, and ultrasonication are beneficial to control the size and distribution of protein-loaded PEG-PLA micelle.

Successful cryopreservation and revival rely on effective cooling and rewarming strategies. Rewarming processes are more vital, because insufficient rewarming normally leads to devitrification or recrystallization, resulting in irreversible damage to biomaterials. Conventional rewarming methods often fail to achieve the required super-high rates. Laser warming has emerged as a promising approach for achieving ultra rapid and internalized heating. Most studies on laser rewarming rely heavily on simulations and lack experimental validation. Moreover, conventional procedures require the manual transfer of vitrified samples from liquid nitrogen to laser exposure spot, which introduces operational risks. To address these limitations, an integrated platform combining vitrification-based cryopreservation with automated laser rewarming was developed. A fast-response temperature measurement system was incorporated to monitor the real-time thermal changes in samples during laser irradiation. The laser rewarming rate of a 1 μL droplet was measured to be 3.5 × 10 6 K/min, which is far beyond those achieved by conventional water bath methods and significantly higher than the required critical warming rate. Three distinct temperature evolution modes were observed: insufficient, appropriate, and excessive heating. We directly confirmed a temperature overshoot followed by rapid thermal decay immediately after laser exposure, which has never been reported before. By varying the laser energy and beam size, the extent of temperature overshoot and subsequent thermal relaxation were controlled, which enabled the optimization of laser rewarming strategy. The findings can be used for the refinement of numerical models and will contribute to the development of safer and more effective rewarming protocols in the cryopreservation field.

Summary The development of biocompatible ice-controlling materials for non-vitreous cryopreservation of cells is of great importance to the field of biomedicine. Here, we present a protocol to use fulvic acid (FA) for efficient non-vitreous cryopreservation of red blood cells (RBCs) that both promotes the melting of ice crystals and retards their growth/recrystallization. We describe steps for FA fractionation and performing tests for ice recrystallization and ice freezing/thawing. We then detail the freezing/thawing of RBCs, recovering RBCs, and testing their viability. For complete details on the use and execution of this protocol, please refer to Bai et al. (2022). 1

In transfusion medicine, the cryopreservation of red blood cells (RBCs) is of major importance. The organic solvent glycerol (Gly) is considered the current gold-standard cryoprotectant (CPA) for RBC cryopreservation, but the deglycerolization procedure is complex and time-consuming, resulting in severe hemolysis. Therefore, it remains a research hotspot to find biocompatible and effective novel CPAs. Herein, the natural and biocompatible inulin, a polysaccharide, was first employed as a CPA for RBC cryopreservation. The presence of inulin could improve the thawed RBC recovery from 11.83 ± 1.40 to 81.86 ± 0.37%. It was found that inulin could promote vitrification because of its relatively high viscosity and glass transition temperature (Tg′), thus reducing the damage during cryopreservation. Inulin possessed membrane stability, which also had beneficial effects on RBC recovery. Moreover, inulin could inhibit the mechanical damage induced by ice recrystallization during thawing. After cryopreservation, the RBC properties were maintained normally. Mathematical modeling analysis was adopted to compare the performance of inulin, Gly, and hydroxyethyl starch (HES) in cryopreservation, and inulin presented the best efficiency. This work provides a promising CPA for RBC cryopreservation and may be beneficial for transfusion therapy in the clinic.

To investigate the impact of frozen storage conditions on the physicochemical properties of soybean protein and explore the underlying mechanisms, this study focused on soybean isolate (SPI), ß-soybean companion globulin (7S), and soybean globulin (11S). The protein solutions were prepared at a concentration of 2% and subjected to freezing for 1 and 5 days. Subsequently, the protein content, physicochemical properties, secondary structure, sulfhydryl content, and chemical interaction forces were assessed and analyzed using UV spectrophotometry, Zeta potential measurements, SDS-PAGE, Fourier infrared spectroscopy, and endogenous fluorescence photoemission spectroscopy. The obtained results revealed that the solubility and total sulfhydryl content of SPI, 7S, and 11S exhibited a decreasing trend with prolonged freezing time. Among them, 11S demonstrated the largest decrease in solubility and total sulfhydryl content, followed by SPI, and 7S the least. During freezing, the aromatic amino acids of SPI, 7S, and 11S molecules were exposed, leading to increased hydrophobicity, protein aggregation, and particle size enlargement, and the structure of the protein changed from disordered structure to ordered structure. After freezing, the polarity of the microenvironment of SPI, 7S, and 11S increased, and their maximum fluorescence emission wavelengths were red-shifted. Notably, the largest red shift of SPI was from 332 nm to 335 nm. As freezing time increased, the contribution of hydrogen bonding increased, while the contribution of hydrophobic interactions decreased. This indicates that freezing affects the hydrophobic interactions, hydrogen bonding, and other chemical forces of the protein. The growth of ice crystals leads to the unfolding of protein molecular chains, exposure of internal hydrophobic groups, enhancement of hydrophobicity, and alters the secondary structure of the protein. Keywords:different freezing time;soybean isolate protein;7S;11S

Gelatin cryogels are good candidate scaffolds for tissue engineering because of their interconnected macroporous structure. For bone regeneration, inorganic components are chosen to reinforce gelatin cryogels: (i) to mimic the compositions of natural bone tissue and (ii) to meet the mechanical requirements of bone repairing. Cryogels were prepared from methacrylated gelatin (GelMA) in this study, and hydroxyapatite nanorods (HANRs) with surface-grafted acrylate groups (D-HANRs) were synthesized to reinforce the cryogels, in which, the crosslinking between GelMA and D-HANRs was expected. In parallel, HANRs and hydroxyapatite nanowires (HANWs) were also composited with the GelMA cryogels to investigate the effects of filler morphology and interfacial bonding on the overall properties of the resulting composite cryogels comparatively. All these composite cryogels demonstrated potential as bone repairing materials by displaying excellent performances such as high porosity, appropriate water retention, shape recovery, and fast resilience features, as well as good biocompatibility and cell affinity. In comparison with the HANR composited GelMA cryogel, the HANWs were able to ameliorate the compression and the rheology performances of the resulting composite cryogels more efficiently due to the fact that the one-dimensional HANWs played a bridging role in the gelatin matrix. Among all the preparations, however, it was the D-HANRs that achieved the strongest reinforcement efficiency in mechanical properties because the double bonds on their surface could be photo-crosslinked with GelMA to form interfacial bonding. With these findings, we concluded that it was preferable for inorganic fillers designed for cryogel-type bone repairing materials to be in a one-dimensional morphology with surface functional groups to strengthen their interfacial bonding with the polymeric matrix.
