Papaya fruit is easily affected by low temperature, causing chilling injury (CI). In this study, the exogenous arginine was applied in the storage of postharvest papaya fruit and it was found that 0.8 mM arginine treatment significantly alleviated CI of papaya fruit. Arginine treatment promoted the accumulation of polyamines (PAs), which might be ascribed to the increased activities of arginase, arginine decarboxylase (ADC), ornithine decarboxylase (ODC), and S-adenosylmethionine decarboxylase (SAMDC). A higher accumulation of proline was found in arginine-treated papaya fruit, which may be due to increased activity of ornithine-δ-aminotransferase (OAT) and Δ 1 -pyrroline-5-carboxylate acid synthase (P5CS) and inhibited activity of proline dehydrogenase (PDH). In addition, arginine treatment increased higher nitric oxide synthase (NOS) activity and promoted the generation of nitric oxide (NO). The results of this study indicate that exogenous arginine may improve the chilling tolerance of postharvest papaya fruit by regulating arginine and proline metabolism. Practical applications In recent years, chilling injury of tropical fruits is a significant problem, including papaya. Many studies on polyamine, proline, nitric oxide, and chilling injury of fruits, but arginine, as a common precursor of polyamine, proline, and nitric oxide, are rarely studied, especially in papaya fruit. Our findings demonstrated that the exogenous arginine can regulate endogenous arginine metabolism, induce polyamine, putrescine, and nitric oxide accumulation, and coordinate to alleviate chilling injury of papaya fruit.

The occurrence of chilling injury is closely related to changes in wax. In this study, we investigated the relationship in plum fruit between wax presence (CK) and removal of epicuticular wax (REW) based on micromorphology and metabolomics. The results showed that wax reduced the incidence of chilling injury in plum fruit and slowed down the rate of loss of fruit firmness, and that the abundant wax structure not only protected the epidermal cells but also inhibited high-intensity respiration of the fruit. The correlation analysis between wax components and chilling injury illustrated that triterpene components were positively correlated with the occurrence of chilling injury. In summary, the present study revealed that wax components were involved in regulating chilling injury in plums, which provides a research idea to delay and reduce the occurrence of chilling injury in plums.

Apricot is highly susceptible to chilling injury during low temperature storage, while methyl salicylate (MeSA) can alleviate chilling injury and enhance the postharvest quality of fruit. The apricots were preharvest sprayed twice with 0.05 mmol L −1 , 0.1 mmol L −1 and 0.2 mmol L −1 of MeSA solution, and then were harvested and stored at 2 °C (humidity of 70 %). After cold storage for every 4 d, apricots were transferred to 20 °C for 2 d to determine the fruit quality. Our results indicated that 0.2 mmol L −1 MeSA treatment exhibited the best efficacy. Preharvest MeSA treatment could reduce decay rate and chilling injury (CI) index, and maintain firmness, soluble solid content (SSC), organic acids and antioxidant capacities at a relative higher level. Preharvest MeSA treatment inhibited the increase of electrolyte leakage, malondialdehyde (MDA) and H 2 O 2 content in apricot. The chilling temperature of 2 °C could lead to the degradation of sucrose to fructose and glucose in apricot. At the same time, preharvest MeSA treatment effectively alleviated CI symptoms of apricot by reducing the degradation rate. The enhanced chilling tolerance of apricot treated with MeSA could be affected by the increased sucrose content, which was regulated by the higher activities of sucrose phosphate synthase (SPS) and sucrose synthase-synthesis (SS-S), and the lower activities of acid invertase (AI), neutral invertase (NI) and sucrose synthase-cleavage (SS-C). We proposed that preharvest MeSA treatment could enhance the chilling tolerance by regulating sugar metabolism and further improve the postharvest quality of apricot during low temperature storage.

The potent role of endogenous melatonin (MEL) in affecting chilling injury (CI) and related chilling tolerance regulatory mechanisms in harvested litchi fruit during refrigeration at 4 °C were investigated in this study. In the first experiment, exogenous MEL exposure exerted an inhibitory impact on the development of CI severity in refrigerated litchi fruit, as indicated by the lower CI index and delayed red color loss of pericarp, whereas treatment with p -chlorophenylalanine ( p -CPA), an inhibitor of MEL biosynthesis, aggravated the degree of CI. The suppression of CI afforded by exogenous MEL was abated under the combined application of exogenous MEL and p -CPA. In the second experiment, the effects of p -CPA alone treatment on changes in biochemical parameters during refrigeration were analyzed. The results showed that p -CPA down-regulated the expressions of MEL biosynthesis genes, including LcTDC , LcT5H and LcSNAT , contributing to reduced generation of endogenous serotonin and MEL. p -CPA treatment accelerated oxidative stress-triggered membrane deterioration, as reflected by increases in relative leakage rate (RLR), reactive oxygen species (ROS) production, lipoxygenase (LOX) activity and malondialdehyde (MDA) accumulation. The membrane damage caused by p -CPA further resulted in loss of subcellular compartmentalization and enzymatic oxidation, as demonstrated by increased activities of polyphenol oxidase (PPO) and peroxidase (POD) in parallel with reduced contents of total phenolics and anthocyanin. Compared with control fruit, lower activities of antioxidant enzymes, including superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR) and glutathione reductase (GR), were observed in fruit receiving p -CPA. Additionally, p -CPA suppressed the expression levels of genes encoding oxidized protein repair-related enzymes, including LcMsrA1 , LcMsrA2 , LcMsrB1 and LcMsB2 . These results suggest that endogenous MEL might confer a protective role against chilling stress by modulating cellular redox status in harvested litchi fruit during refrigeration.

Snap bean ( Phaseolus vulgaris L.) is very sensitive to low temperature during postharvest storage. Methyl jasmonate (MeJA) is a volatile hormone ubiquitous in many kinds of plants and plays important role in regulating plant physiological functions. The alleviating effect of MeJA treatment on chilling injury (CI) in postharvest snap bean was studied. We found MeJA treatment markedly decreased the index of chilling injury, electrolyte leakage and content of malondialdehyde. MeJA-treated snap bean exhibited higher levels of total soluble solids and titratable acidity. Similarly, MeJA could prevent the degradation of chlorophyll and ascorbic acid. Not only total antioxidant capacity but also metal chelating ability of MeJA-treated snap bean was stronger than untreated control. MeJA-treated snap bean showed higher scavenging activities on superoxide radical (O 2 .- ), hydroxyl radical (·OH) and 1,1-diphenyl-2-trinitrophenylhydrazine radical either. MeJA application stimulated the activities of ascorbate peroxidase (APX), peroxidase (POD), superoxide dismutase (SOD) and catalase (CAT) in snap bean. In conclusion, this study reveals MeJA can be used to relieve chilling injury and maintain the quality of snap bean during low-temperature storage. 8 µM MeJA is the optimal concentration to alleviate chilling injury of snap bean.

The influence of melatonin treatment on the quality and chilling injury of guavas during storage at 4 ± 1 °C were evaluated. Compared with control group, fruit of guava cv. Xiguahong exposed to various concentrations (50, 100, 150, and 200 μmol/L) of melatonin showed a significantly lower fruit respiration rate, weight loss, cell membrane permeability, and chilling injury index, but a higher commercially acceptable fruit rate, higher peel L*, h ° value, and chlorophyll content. Melatonin treatment also delayed the decreases of fruit firmness, sucrose, total soluble sugar, vitamin C, titratable acidity, and total soluble solids. These data indicate that melatonin treatment could increase chilling tolerance and retain quality of cold-stored guavas. Among various concentrations of melatonin treatment, 100 μmol/L melatonin-treated guavas showed the preferable quality properties and lowest chilling injury index. Thus, melatonin may be a novel method of postharvest handling to enhance cold resistance and extend storage-life of cold-stored guava fruit.

Antioxidant activity is an important feature for evaluating fruit quality and tolerance to biotic or abiotic stresses. Moreover, antioxidant activity is involved in chilling injury (CI) response and postharvest quality in fruit. Chemical elicitors can induce systemic acquired resistance in fruit against pathogens, which could partially replace synthetic fungicides. Recently, researchers have found that preharvest sprays with chemical elicitors can improve antioxidant activity, reduce CI, and maintain quality in harvested fruit. In this review, we summarize that preharvest elicitors spray improve antioxidant activity in harvested fruit by promoting antioxidant components biosynthesis as well as antioxidant ability in vitro. Moreover, preharvest elicitors spray alleviates CI in fruit by regulation of membrane lipid metabolism and reactive oxygen species metabolism. In addition, preharvest elicitors spray maintains fruit quality by modulation of respiration and ethylene release. Finally, this review points out the issues existing and proposes an outlook on preharvest elicitors spray to maintain postharvest fruit quality. Keywords:fruit;preharvest elicitor spray;antioxidant activity;chilling injury;postharvest quality

This study aims to investigate the effect of exogenous melatonin (EMT) on cold injury (CI) and the physiological indicators of cucumbers under refrigerated conditions (2 °C). After being treated with different concentrations (0, 50, 100, and 500 μmol L −1 ) of EMT, the postharvest cucumbers were stored at 2 °C. During storage, cucumbers were collected at designed intervals (0, 3, 6, 9, and 12 d) and rewarmed for 2 days at room temperature (20–22 °C). The impacts of EMT were evaluated by periodically monitoring the changes in chilling injury index and physiological properties including weight loss rate, respiratory intensity, tissue electrolyte leakage, malondialdehyde (MDA) content, vitamin C, soluble protein content, free proline, and reactive oxygen species. Compared with the control, the treatment of EMT at the concentration of 100 μmol L −1 could significantly improve the contents of soluble protein, vitamin C, and free proline content and inhibit the rise of chilling injure index, weight loss rate, respiration intensity, tissue electrolyte leakage, MDA content, and reactive oxygen species. In addition, the results indicated that EMT treatment at a specific level (100 μmol L −1 ) had an effective impact on alleviating the CI of cucumbers during storage at low temperatures.

The alteration of surface microstructures and chemical composition in cuticle of papaya fruit in response to chilling stress were comparatively studied between cultivars of ‘Risheng’ and ‘Suihuang’ after harvest. Fruit surface was covered by fissured wax layers in both cultivars. The presence of granule crystalloids was cultivar dependent, with higher abundance in 'Risheng' and lower in 'Suihuang'. Various typical very-long-chain aliphatics i.e., fatty acids, aldehydes, n -alkanes, primary alcohols, and n -alkenes dominated waxes; and cutin monomers were prominently 9/10,16-dihydroxyhexadecanoic acid in papaya fruit cuticle. Chilling pitting symptom was accompanied by modification of granule crystalloids into flat appearance and decreased primary alcohols, fatty acids, and aldehydes in ‘Risheng’, but no evident changes in ‘Suihuang’. The response of cuticle to chilling injury in papaya fruit might be not directly related to the overall amount of waxes and cutin monomers, but more likely to the alteration of appearance morphologies and chemical composition in cuticle.

Glycine betaine (GB) treatment is useful to reduce chilling injury (CI) of several kinds of fruits including peach. However, the regulatory mechanism remains unknown. In this study, peach fruit was treated with 10 mmol L −1 GB solution for 10 min. The effects of GB treatment on CI, phenolic and soluble sugar metabolism were investigated in this study. Moreover, phenylpropanoid and soluble sugar content, and enzyme activities associated with phenolic and sugar metabolisms were also measured. The results showed that GB reduced CI and maintained high levels of total phenolic and flavonoid content. The activities of phenylpropanoid metabolism-related enzymes were significantly enhanced by GB. Higher content of sucrose and lower contents of fructose and glucose were observed in GB-treated fruits. Therefore, our results showed that GB could enhance chilling tolerance of peach through regulating phenolic and sugar metabolisms, and maintaining high levels of individual phenolic and sucrose content.

The plant cuticle covers almost all the outermost surface of aerial plant organs, which play a primary function in limiting water loss and responding to the environmental interactions. Banana fruit is susceptible to thermal changes with chilling injury below 13°C and green ripening over 25°C. Herein, the changes of surface morphology, chemical compositions of cuticle, and the relative expression of cuticle biosynthesis genes in banana fruit under low-temperature storage were investigated. Banana fruit exhibited chilling injury rapidly with browned peel appearance stored at 4°C for 6 days. The surface altered apparently from the clear plateau with micro-crystals to smooth appearance. As compared to normal ones, the overall coverage of the main cuticle pattern of waxes and cutin monomers increased about 22% and 35%, respectively, in browned banana stored under low temperature at 6 days. Fatty acids (C 16 –C 18 ) and ω-OH, mid-chain-epoxy fatty acids (C 18 ) dominated cutin monomers. The monomers of fatty acids, the low abundant ω, mid-chain-diOH fatty acids, and 2-hydroxy fatty acids increased remarkably under low temperature. The cuticular waxes were dominated by fatty acids (> C 19 ), n -alkanes, and triterpenoids; and the fatty acids and aldehydes were shifted to increase accompanied by the chilling injury. Furthermore, RNA-seq highlighted 111 cuticle-related genes involved in fatty acid elongation, biosynthesis of very-long-chain (VLC) aliphatics, triterpenoids, and cutin monomers, and lipid-transfer proteins were significantly differentially regulated by low temperature in banana. Results obtained indicate that the cuticle covering on the fruit surface was also involved to respond to the chilling injury of banana fruit after harvest. These findings provide useful insights to link the cuticle on the basis of morphology, chemical composition changes, and their biosynthesis regulations in response to the thermal stress of fruit during storage.

Cold storage is widely used for the postharvest preservation of fruits and vegetables; however, okra, as a tropical vegetable, is susceptible to chilling injury under low-temperature storage conditions, leading to quality deterioration, reduced nutritional value, and significant economic losses. Nitric oxide (NO), as an important signaling molecule, plays a crucial role in the postharvest preservation of fruits and vegetables. To investigate the effects of different concentrations of nitric oxide on the postharvest quality of okra under cold storage, fresh okra pods were treated with sodium nitroprusside (SNP), a commonly used NO donor, at concentrations of 0 (control), 0.5 (T1), 1.0 (T2), 1.5 (T3), and 2.0 mmol·L−1(T4). The results showed that low-concentration NO treatment (T1) significantly reduced weight loss, improved texture attributes including hardness, springiness, chewiness, resilience, and cohesiveness, and suppressed the increase in adhesiveness. T1 treatment also effectively inhibited excessive accumulation of cellulose and lignin, thereby maintaining tissue palatability and structural integrity. Additionally, T1 significantly delayed chlorophyll degradation, preserved higher levels of soluble sugars and proteins, and enhanced the activities of key antioxidant enzymes, including superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), contributing to improved oxidative stress resistance and membrane stability. In contrast, high-concentration NO treatments (T3 and T4) led to pronounced quality deterioration, characterized by accelerated membrane lipid peroxidation as evidenced by increased malondialdehyde (MDA) content and relative conductivity, and impaired antioxidant defense, resulting in rapid texture degradation, chlorophyll loss, nutrient depletion, and oxidative damage. These findings provide theoretical insights and practical guidance for the precise application of NO in extending shelf life and maintaining the postharvest quality of okra fruits.
