While it is known that temperature sensors trigger calcium (Ca 2+ ) signaling to confer cold tolerance in cells, less is known about sensors that couple with other secondary messengers. Here, we identify a cold sensor complex of CHILLING-TOLERANCE DIVERGENCE 6 (COLD6) and osmotin-like 1 (OSM1), which triggers 2′,3′-cyclic adenosine monophosphate (2′,3′-cAMP) production to enhance cold tolerance in rice. COLD6, which is encoded by a major quantitative trait locus (QTL) gene, interacts with the rice G protein α subunit (RGA1) at the plasma membrane under normal conditions. Upon exposure to chilling, cold-induced OSM1 binds to COLD6, kicking out RGA1 from interaction. This triggers an elevation of 2′,3′-cAMP levels for enhancing chilling tolerance. Genetic data show that COLD6 negatively regulates cold tolerance and functionally depends on OSM1 in chilling stress. COLD6 alleles were selected during rice domestication. Knockout and natural variation of COLD6 in hybrid rice enhanced chilling tolerance, hinting design potential for breeding. This highlighted a module triggering 2′,3′-cAMP to improve chilling tolerance in crops.

Self-healability and cold-resistance will greatly improve the reliability of an energy-storage device in case of physical damage and subzero environment, which bring higher and higher requirement to electrolyte. Herein, a multifunctional supramolecular gel polymer electrolyte is prepared, which includes the first cross-linking network between PVA and TA through hydrogen bond cross-linking, and the second PVA network via the freezing-thawing cycle. The synergistic effects of two networks endow the gel polymer electrolyte with outstanding mechanical properties such as high tensile strengths (1.19 MPa), elongations (18.35 mm mm −1 ), self-recovery and excellent self-healability. Moreover, a high conductivity of gel polymer electrolyte is also obtained. A supercapacitor based on the gel polymer electrolyte can significantly restore its electrochemical performances after 10 cutting-healing cycles without external treatment at room temperature. In addition, the supercapacitor presents great controllability and can be tailored into different patterns without property deterioration. Besides, even at −20 °C, the SC is able to maintain 68.3% of its room temperature capacitance. The investigation offers a facile and versatile strategy to construct an intrinsically self-healable and cold-resistant energy-storage device which has potential applications for portable and wearable electronics, smart apparels or flexible robots, and so on.

As an important gas signaling molecule, hydrogen sulfide (H2S) plays a crucial role in regulating cold tolerance. H2S cooperates with phytohormones such as abscisic acid, ethylene, and salicylic acid to regulate the plant stress response. However, the synergistic regulation of H2S and auxin in the plant response to cold stress has not been reported. This study showed that sodium hydrosulfide (NaHS, an H2S donor) treatment enhanced the cold stress tolerance of cucumber seedlings and increased the level of auxin.CsARF5, a cucumber auxin response factor (ARF) gene, was isolated, and its role in regulating H2S-mediated cold stress tolerance was described. Transgenic cucumber leaves overexpressingCsARF5were obtained. Physiological analysis indicated that overexpression ofCsARF5enhanced the cold stress tolerance of cucumber and the regulation of the cold stress response byCsARF5depends on H2S. In addition, molecular assays showed thatCsARF5modulated cold stress response by directly activating the expression of the dehydration-responsive element-binding (DREB)/C-repeat binding factor (CBF) geneCsDREB3, which was identified as a positive regulator of cold stress. Taken together, the above results suggest that CsARF5 plays an important role in H2S-mediated cold stress in cucumber. These results shed light on the molecular mechanism by which H2S regulates cold stress response by mediating auxin signaling; this will provide insights for further studies on the molecular mechanism by which H2S regulates cold stress. The aim of this study was to explore the molecular mechanism of H2S regulating cold tolerance of cucumber seedlings and provide a theoretical basis for the further study of cucumber cultivation and environmental adaptability technology in winter. Keywords:ARF;auxin;cold stress;cucumber;DREB;hydrogen sulfide;module;resistance

Chilling injury (CI) is a bottleneck factor constraining the refrigeration and cold chain logistics of mango fruit. Methyl jasmonate (MeJA) is an important phytohormone that can regulate multiple abiotic stresses. This study evaluated the potential effects of MeJA on CI in ‘Guifei’ mango fruit stored at 4 °C and its underlying mechanisms involved in the regulation of antioxidation and energy status. The application of 50 µM MeJA mitigated CI symptoms and improved ripening-related quality during refrigeration, as indicated by a lowered CI index, retarded declines in chlorophyll fluorescence ( Fv/Fm ) and chromaticity L * and advanced color transformation (from green to yellow), softening and accumulation of soluble solids. MeJA treatment repressed the increases in relative conductivity (RC), malondialdehyde (MDA) content and reactive oxygen species (ROS) (O 2 -. and H 2 O 2 ). Reduced ROS generation in MeJA-treated fruit was a consequence of the enhancement of antioxidant enzyme activities , including superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) and glutathione reductase (GR). Ultrastructural observation revealed that MeJA treatment protected mitochondrial structure. MeJA treatment promoted intracellular energy synthesis and utilization, as indicated by increments in intracellular adenosine triphosphate (iATP) level, activities of isocitrate dehydrogenase (IDH), α-ketoglutarate dehydrogenase (α-KGDH), succinate dehydrogenase (SDH), cytochrome C oxidase (CCO), H + -ATPase and Ca 2+ -ATPase as well as contents of reduced coenzymes (NADH and FADH 2 ). In addition, the expression levels of genes ( MiAPYs , MiENTs , MiPUPs and MiAPRTs ) related to extracellular ATP (eATP) hydrolysis, eATP transport and iATP regeneration were upregulated in response to MeJA, contributing to maintenance of eATP/iATP homeostasis . These findings suggest that MeJA could reinforce cold tolerance in mango fruit by inhibiting oxidative stress and promoting energy metabolism.
